Abstract
The invention discloses a children oral care composition and application thereof, wherein the children oral care composition is composed of xylitol, a calcium-containing compound and probiotics, the weight ratio of the xylitol to the calcium-containing compound is 1: 2-50: 1, and the weight ratio of the xylitol to the probiotics is 1: 8-50: 1. The composition of the invention fully exerts the synergistic effect of the xylitol, the calcium-containing compound and the probiotics, prevents decayed teeth in different links, has the decayed tooth preventing capability obviously superior to that of a single active substance, can provide oral bacteria inhibiting capability superior to that of the single active substance, can maintain normal oral microbial flora at the same time, guarantees oral health, and finally achieves the effects of dispelling peculiar smell of the oral cavity, preventing decayed teeth, promoting calcification of the teeth, firming the teeth, nourishing gingiva and healthy periodontium. The children oral care composition can be used for preparing children oral care products such as children toothpaste, children mouthwash or children mouth cleaning agent.
Description
Technical Field
The invention relates to the technical field of oral care products, in particular to oral care for children, and specifically relates to a composition suitable for oral care for children and application thereof.
Background
The prevalence rate of oral diseases of children in China is high, wherein caries and periodontal diseases are the most common oral diseases of children. The third national oral health epidemiological survey in 2005 shows that the prevalence rates of caries in 5-year-old deciduous teeth and 12-year-old children's permanent teeth in China are respectively as high as 66.0% and 28.9%. The teeth of the infants or children are thin in hard tissues, low in mineralization degree, high in solubility and strong in permeability, and improper feeding modes and poor oral hygiene of the children are added, so that the teeth are easy to be diseased in the growth and development process, the teeth become good-onset people with dental caries, oral diseases such as swelling and pain of gum and halitosis are accompanied, and the children directly influence the learning, life, physical and psychological health.
Fluoride is also a commonly used anticaries substance, but because of poor self-control ability of children and incomplete swallowing function, it is easy to ingest excessive fluorine during long-term use of toothpaste, which is not good for health.
Xylitol, as a commonly used anticaries substance, can inhibit the growth and acid production of Streptococcus mutans, a major cariogenic bacterium, so that xylitol can reduce the number of Streptococcus mutans, and has the effects of promoting remineralization and inhibiting demineralization. However, xylitol, while having anticaries efficacy, must be added in a certain amount to have significant anticaries efficacy, increasing formulation cost and stability risks.
The Chinese patent application with publication number CN105434322A discloses a toothpaste containing oral cavity probiotics, which can reduce the permanent planting of harmful bacteria, inhibit the growth and reproduction of pathogenic bacteria, and reduce the number of harmful bacteria such as cariogenic bacteria, periodontal pathogenic bacteria and oral candida after entering the oral cavity along with the toothpaste. The probiotics can also generate aggregation reaction with harmful bacteria such as tooth decay bacteria, periodontal bacteria, oral odor bacteria and the like, and the encapsulated probiotics are taken out of the oral cavity. However, the use of a single probiotic in toothpaste is somewhat limited: 1. probiotics generally need to be stored in a low-temperature environment to ensure effective activity, and low-temperature storage of toothpaste in the processes of production, logistics, sale and use is almost impossible. 2. When the probiotics is added into the toothpaste, part of the toothpaste can generate yellowing phenomenon and faint yellow aggregates with the diameter of about 1-3mm which can be seen by naked eyes appear, the attractiveness and the use of the toothpaste are seriously influenced, in addition, through single live probiotics for preventing caries, the live bacteria preparation usually has the obvious curative effect only by having a certain quantity of live bacteria, a large quantity of probiotics can influence the appearance, the use and the stability of the toothpaste, and the stability of the live probiotics in the toothpaste is uncontrollable.
In summary, although the prior art has a research on the dental caries prevention effect of the oral care product, no report has been found on a research on a safe, non-toxic and stable oral care product which is specially developed to inhibit oral bacteria, prevent dental caries and promote calcification of teeth according to the characteristics of oral care of children.
Disclosure of Invention
The invention aims to overcome the defects of the prior art and provides an oral care composition which is used for children, can effectively inhibit oral bacteria, remove oral odor, prevent decayed teeth, promote calcification of teeth, firm teeth, and has good safety performance and stability.
It is another object of the present invention to provide the use of the above oral care composition in the manufacture of a children's oral care product.
The above-mentioned invention is realized by the following scheme:
a children oral care composition, the oral care composition is composed of xylitol, calcium-containing compound and probiotics; the weight ratio of the xylitol to the calcium-containing compound in the oral care composition is 1: 2-50: 1, preferably 1: 2-15: 1, and the weight ratio of the xylitol to the probiotics in the oral care composition is 1: 8-50: 1, preferably 1: 4-6: 1.
In the children oral care composition, xylitol is a commercial product.
In the children oral care composition, the calcium-containing compound is any one or a combination of two or more of commercially available soluble calcium or slightly soluble calcium in any proportion; the calcium-containing compound is preferably any one of calcium lactate, calcium glycerophosphate or calcium gluconate; the calcium-containing compound is further preferably calcium lactate.
In the children oral care composition, the probiotics are probiotic inactivated freeze-dried powder sold in the market; the probiotics are preferably one or a combination of two or more of bifidobacterium adolescentis freeze-dried powder, bifidobacterium lactis freeze-dried powder, bifidobacterium bifidum freeze-dried powder, lactobacillus crispus freeze-dried powder, lactobacillus bulgaricus freeze-dried powder, lactobacillus gasseri freeze-dried powder, lactobacillus helveticus freeze-dried powder, lactobacillus johnsonii freeze-dried powder, lactobacillus plantarum freeze-dried powder, lactobacillus reuteri freeze-dried powder, lactobacillus rhamnosus freeze-dried powder, lactobacillus salivarius freeze-dried powder or streptococcus thermophilus freeze-dried powder in any proportion; the probiotic is further preferably lactobacillus salivarius freeze-dried powder.
Experiments prove that the children oral care composition has good stability and good synergy of the three components, and can realize the effects of inhibiting bacteria in the oral cavity of children, removing peculiar smell in the oral cavity, preventing decayed teeth of children, promoting calcification of teeth of children and firming teeth according to the characteristics of the oral cavity of children.
The children's oral care compositions of the present invention can be formulated into children's oral care products with orally acceptable carrier components well known in the art; the carrier component is surfactant, colorant, humectant, thickener, perfume, flavoring agent, chelating agent, antiseptic, foaming agent, friction agent, sweetener or nutrient substance, etc.; the oral care product is common dosage forms such as toothpaste, mouthwash, tooth powder or oral cleaning agent; when the oral care product is prepared by matching the oral care composition for children with other carrier components, the dosage of the oral care composition is 0.1-5%, preferably 0.2-3% of the total weight of the oral care product.
In order to optimally achieve the efficacy of the children's oral care composition of the present invention, the inventors have conducted studies on carrier components with which the children's oral care composition can be combined, so as to ensure that the selected carrier components can be more synergistic when used in combination with the children's oral care composition of the present invention to prepare children's oral care products, as shown in the following:
the carrier component is a preservative which is any one of sodium benzoate, potassium sorbate, benzoic acid or nipagin ester; the preservative is preferably sodium benzoate; the preservative is 0.05-5% of the total weight of the oral care product for children, and the preferable amount is 0.1-2% of the total weight of the oral care product for children.
The carrier component is a sweetening agent which is any one or the combination of two or more than two of saccharin sodium, sucralose, mogroside or stevioside in any proportion; the amount of the sweetening agent is 0.05-5% of the total weight of the oral care product for children, and the preferable amount is 0.1-2% of the total weight of the oral care product for children.
The carrier component is a colorant, and the colorant adopts food-grade pigment; the amount of the colorant is 0.0001-0.004% of the total weight of the children care oral product.
The carrier component is a humectant which is any one or a combination of two or more than two of glycerol, polyethylene glycol, sorbitol or propylene glycol in any proportion; the dosage of the humectant accounts for 10-80% of the total weight of the oral care product for children, and the preferred dosage of the humectant accounts for 25-70% of the total weight of the oral care product for children.
The carrier component is a foaming agent which is any one or the combination of two or more than two of lauryl sodium sulfate, sodium lauroyl sarcosinate or cocamidopropyl betaine in any proportion; the amount of the foaming agent is 0.4-10% of the total weight of the oral care product for children, and the preferable amount is 1-5% of the total weight of the oral care product for children.
The carrier component is an abrasive which is calcium carbonate or silicon dioxide; the dosage of the abrasive is 5-60% of the total weight of the oral care product for children, and the preferred dosage is 15-40% of the total weight of the oral care product for children.
The carrier component is a thickening agent which is any one or a combination of two or more than two of sodium carboxymethylcellulose, xanthan gum or carrageenan in any proportion; the amount of the thickener is 0.1-5% of the total weight of the oral care product for children, and the preferable amount is 0.1-3% of the total weight of the oral care product for children.
The carrier component is a flavoring agent which is any one or the combination of two or more than two of essence, natural menthol or cooling agent in any proportion; the amount of the flavoring agent is 0.05-5% of the total weight of the oral care product for children, and the preferable amount is 0.5-2% of the total weight of the oral care product for children.
The carrier component is a nutrient substance which is any one or the combination of two or more than two of sodium ascorbyl phosphate, vitamin A palmitate, tocopherol acetate and vitamin B3 in any proportion; the dosage of the nutrient substances is 0.05-5% of the total weight of the oral care product for children, and the preferred dosage is 0.1-1% of the total weight of the oral care product for children.
Compared with the prior art, the invention has the following beneficial effects:
1. the inventor discovers that children are used to eat frequently, the pH value in the oral cavity can be reduced after organic acid enters the oral cavity, and teeth are corroded by acid, and the xylitol in the children oral care composition can promote salivary secretion, reduce bacteria on the teeth while rinsing the oral cavity, increase the concentration of basic amino acid and ammonia at saliva and decayed tooth spots, and simultaneously slow down the reduction of the pH value in the oral cavity to promote the remineralization of the teeth, so that the children are prevented from being corroded by acid from the source; the calcium-containing compound can supplement the calcium ion concentration in saliva in time, prevent tooth demineralization and promote tooth remineralization, the calcium-containing compound plays a role in inhibiting demineralization and promoting remineralization in the whole process of preventing decayed teeth, and in addition, the probiotics can also have self-aggregation and copolymerization effects with harmful bacteria such as decayed tooth bacteria, periodontal bacteria, oral odor bacteria and the like to form a biological membrane which is taken out of the oral cavity after being wrapped, so that the growth of pathogenic bacteria is inhibited, a barrier for preventing the colonization and infection of the pathogenic bacteria is formed, the oral pathogenic bacteria are reduced fundamentally, and the decayed teeth are prevented at the terminal.
2. In the children oral care composition, the xylitol, the calcium-containing compound and the probiotics act synergistically to prevent decayed teeth in different links, and the decayed teeth prevention capability of the children oral care composition is obviously superior to that of a single active substance, so that the children oral care composition can inhibit oral bacteria superior to that of the single active substance, can maintain normal oral microbial flora, guarantee oral health, and finally achieve the effects of removing oral peculiar smell, preventing decayed teeth, promoting tooth calcification, firming teeth, nourishing gums and healthy periodontium;
3. the inventor finds that different amounts of xylitol determine the secretion amount of different saliva in the oral cavity, and the calcium-containing compound can supplement calcium ions in the saliva in time and promote tooth remineralization; when saliva washes the oral cavity, probiotics are continuously washed, effective aggregation and adhesion effects of the probiotics are limited, and a certain amount of probiotics is needed to supplement and balance, so that the inventor respectively optimizes the dosage of xylitol, calcium-containing compounds and xylitol and probiotics in the oral care composition, and discovers that the oral care effect is not obvious if the dosage proportion is lower than the optimized range, and the appearance and stability of toothpaste are seriously affected if the dosage proportion is higher than the optimized range through optimization tests, so that finally, the dosage of every two components in the children oral care composition is mutually restricted, the effect and synergistic effect of each component are exerted to the maximum extent, and the final realization of the invention destination is ensured.

EXAMPLE 1 toothpaste for children

The preparation method of the children toothpaste comprises the following steps:

Adding xylitol, calcium lactate, Lactobacillus salivarius lyophilized powder, saccharin sodium, sodium benzoate, sodium ascorbyl phosphate, vitamin B3, and edible pigment into deionized water according to formula dosage, dissolving, adding sorbitol, glycerol, and polyethylene glycol-400, and stirring to obtain mixture 1;
uniformly mixing and dispersing silicon dioxide, sodium carboxymethylcellulose and xanthan gum to obtain a mixture 2;
adding vitamin A palmitate and tocopherol acetate into the essence, and dissolving in a water bath to obtain a mixture 3;

Example 3 toothpaste for children

The children oral care composition comprises xylitol, calcium lactate and lactobacillus salivarius lyophilized powder; the weight ratio of the xylitol to the calcium lactate is 3:1, and the weight ratio of the xylitol to the lactobacillus salivarius freeze-dried powder is 3: 2.
The preparation method of this example is the same as example 1.
Example 4 stability testing of children's toothpaste
The children's toothpaste of examples 1-3 was subjected to appearance and quality stability tests, specifically, the children's toothpaste of examples was aged at a high temperature of 45 ℃ for three months and then tested for stability, and the test results are shown in table 1.

EXAMPLE 5 bacteriostatic test of children's toothpaste
In this embodiment, the children toothpaste in embodiment 2 is used as an experimental sample to perform a bacteriostatic test, specifically a bacteriostatic test on streptococcus mutans and fusobacterium nucleatum.
The specific experimental method is that the toothpaste stock solution of the example 2 with the adjusted absorbance and the pathogenic bacterium solution in the logarithmic growth phase are mixed in equal amount, the growth curve of pathogenic bacteria is measured by an absorbance method at 0h, 1.5h, 3h, 4.5h, 6h and 7.5h respectively, and the smaller the △ OD is, the stronger the inhibition effect is.
Negative control: a toothpaste having the same formulation and preparation method as in example 2, except that the children's oral care composition of the present invention was not added.
The result of the inhibition effect of the toothpaste on the growth OD of the streptococcus mutans is shown in figure 1; the results of the inhibition of growth OD of F.nucleatum are shown in FIG. 2; in fig. 1 and 2, the left side of each growth time point is the result of the negative control, and the right side is the result of the children's toothpaste of example 2.
As can be seen from FIGS. 1 and 2, the oral care composition for children of the present invention has a significant inhibitory effect on Streptococcus mutans and Fusobacterium nucleatum, and the toothpaste containing the composition of the present invention has a significantly enhanced inhibitory effect on Streptococcus mutans and Fusobacterium nucleatum.
Variations and modifications to the above-described embodiments may occur to those skilled in the art, which fall within the scope and spirit of the above description.
