技术领域
本发明属于中药材制备领域。
背景技术
杜仲是杜仲科植物杜仲的干燥树皮,始载于《神农本草经》,将其列为上品。杜仲性味甘温,归肝肾经。具有补肝肾、强筋骨、安胎、降血压之功效。
杜仲炮制后入药的最早记载是雷敩的《雷公炮炙论》:“凡使,削取粗皮”,此后历代医书、本草纲目均强调去粗皮而延续至今,历版《中国药典》和各省市炮制规范对杜仲的炮制亦要求“刮去残留粗皮,洗净”。唐代《新修本草》也记载:“用之削薄,去上皮,横理切,令断丝也”。杜仲姜制首见于宋代《太平惠民和剂局方》:“令无丝为度,或只锉碎以姜汁拌炒,令丝绝亦得”,作为杜仲姜制与熟制方法和标准相传。陈嘉谟《本草蒙筌》也有“刮净粗皮,咀成薄片,姜汁润透,连炒去丝”、“姜汁和酒炙,去丝”等相关论述,但都没有记载具体工艺参数。纵观古今文献,杜仲炮制多采用姜汁炒、盐水炒、酒炒、糯米炒、蜜炙等加辅料的炮制方法,其炮制程度各不相同,但去丝或断丝是一致的。由此看来,杜仲炒断丝是古今炮制的主要目的,盐制法是目前炮制杜仲的主流方法,现行版《中国药典》、教科书及各地炮制规范均收载了盐杜仲的炮制方法。2015年版《中国药典》一部记载:“取杜仲块或丝,照盐水炙法,炒至断丝、表面焦黑色”。
杜仲含松脂醇二葡萄糖苷,杜仲胶,木质素及其苷类,环烯醚萜类,还含绿原酸、咖啡酸、熊果酸等有机酸类,以及17种游离氨基酸和15种无机元素等。现代药理实验已证实,杜仲具有降压、镇静、镇痛、增强免疫、抗炎、抗应激、利尿、延缓衰老等作用。松脂醇二葡萄糖苷为杜仲降血压的主要有效成分。现行版中国药典把松脂醇二葡萄糖苷作为指标性成分载入药典,其含量的高低直接影响杜仲临床疗效。炮制对杜仲理化性质和药理作用均有一定的影响。盐味咸性寒,盐制杜仲能引药入肾,直达下焦,补肝肾、强筋骨、安胎的作用增强,常用于肾虚腰痛,筋骨无力,妊娠漏血,胎动不安和高血压等。而生姜味辛性温,能温中、和胃、止呕。用生姜炮制可增强杜仲温补肾阳、补中强志、补肾壮骨之功。主要用于治疗腰部以下沉重凉痛,腰不能直,生活不能自理;舌苔白厚腻、舌质淡白、舌体胖大,脉沉迟软等症的虚寒性腰痛。
发明内容
本发明的目的是提出一种杜仲的炮制方法。
本发明是通过以下技术方案实现的。
本发明所述的一种杜仲的炮制方法,包括如下步骤。
(1)净选:取原药材,拣去药材中的杂质、异物、非药用部位,其余放置在干净的容器内,杂质量≤2.0%。
(2)闷润:按杜仲与生姜的质量比100:12-20的比例,先将生姜压榨得到的姜汁再加水稀释至原来的1.5-2.5倍的质量,然后,将姜汁加入到杜仲中,拌匀,充分润透,未润透的杜仲≤10%。
(3)炒炙:待姜汁被吸尽后,置炒制容器内,用武火150-250℃炒15-25min或至规定程度时,取出。
(4)干燥:晾干或放入电热恒温鼓风干燥箱中,于60-70℃温度干燥。
(5)过筛:用药典筛筛去碎末,碎末含量≤3.0%。
本发明步骤(2)所述的杜仲与生姜的质量比优选100:15。
本发明步骤(3)所述的武火炒优选160-170℃。
本发明用HPLC法测定了生杜仲、姜杜仲中松脂醇二葡萄糖苷含量,结果显示,姜杜仲中松脂醇二葡萄糖苷及浸出物含量均符合药典标准,可达到温补肾阳、补脾胃壮骨之功。
本发明所述的杜仲中松脂醇二葡萄糖苷的含量测定方法如下。
色谱条件:色谱柱:Diamonsil C18色谱柱(250×4.6mm,5μm);流动相:甲醇-水(25:75);柱温:30℃;检测波长:277nm;流速:1.0 mL/min;进样量:10μL。
对照品溶液的制备:取松脂醇二葡萄糖苷对照品适量,精密称定,加甲醇制成每1mL含松脂醇二葡萄糖苷0.1mg的溶液,即得。
供试品溶液的制备:取本品约3g,剪成碎片,揉成絮状,取约2g,精密称定,置索氏提取器中,加入三氯甲烷适量,加热回流6小时,弃去三氯甲烷液,药渣挥去三氯甲烷,再置索氏提取器中,加入甲醇适量,加热回流6小时,提取液回收甲醇至适量,转移至10mL量瓶中,加甲醇至刻度,摇匀,滤过,取续滤液,即得。
具体实施方式
本发明将通过以下实施例作进一步说明。
实施例1。
(1)净选:取原药材,拣去药材中的杂质、异物、非药用部位,其余放置在干净的容器内,杂质量≤2.0%。
(2)闷润:将1.5kg的生姜压榨得到的姜汁再加水稀释至1.8倍,将姜汁加入到10kg的杜仲中,拌匀,充分润透,未润透的杜仲≤10%。
(3)炒炙:待姜汁被吸尽后,置炒制容器内,用武火150℃炒25min,取出。
(4)干燥:晾干或放入电热恒温鼓风干燥箱中,于60℃温度干燥。
(5)过筛:用药典筛筛去碎末,碎末含量为2.2%。
(6)含量测定:在上述条件下检测姜杜仲中松脂醇二葡萄糖苷含量为:0.08%。浸出物含量为:13.5%。
实施例2。
(1)净选:取原药材,拣去药材中的杂质、异物、非药用部位,其余放置在干净的容器内,杂质量≤2.0%。
(2)闷润:将7.5kg的生姜压榨得到的姜汁再加水稀释至2.2倍,将姜汁加入到50kg的杜仲中,拌匀,充分润透,未润透的杜仲≤10%。
(3)炒炙:待姜汁被吸尽后,置炒制容器内,用武火250℃炒15min,取出。
(4)干燥:晾干或放入电热恒温鼓风干燥箱中,于70℃温度干燥。
(5)过筛:用药典筛筛去碎末,碎末含量为2.0%。
(6)含量测定:在上述条件下检测姜杜仲中松脂醇二葡萄糖苷含量为:0.08%。浸出物含量为:12.8%。
实施例3。
(1)净选:取原药材,拣去药材中的杂质、异物、非药用部位,其余放置在干净的容器内,杂质量≤2.0%。
(2)闷润:将7.5kg的生姜压榨得到的姜汁再加水稀释至2.5倍,将姜汁加入到50kg的杜仲中,拌匀,充分润透,未润透的杜仲≤10%。
(3)炒炙:待姜汁被吸尽后,置炒制容器内,用武火200℃炒20min,取出。
(4)干燥:晾干或放入电热恒温鼓风干燥箱中,于65℃温度干燥。
(5)过筛:用药典筛筛去碎末,碎末含量为1.5%。
(6)含量测定:在上述条件下检测姜杜仲中松脂醇二葡萄糖苷含量为:0.09%。浸出物含量为:14.2%。

(2)按杜仲与生姜的质量比100:12-20的比例,先将生姜压榨得到的姜汁再加水稀释至原来的1.5-2.5倍的质量,然后,将姜汁加入到杜仲中,拌匀,充分润透,未润透的杜仲≤10%;
