本发明涉及一株绿色木霉菌(Trichoderma viride)3838菌株及其应用,属于应用微生物学领域。该菌株已于2016年4月19日保藏在中国微生物菌种保藏管理委员会普通微生物中心;保藏单位地址:北京市朝阳区北辰西路1号院3号,中国科学院微生物研究所;保藏号:CGMCC No.12372。本发明的绿色木霉菌(Trichoderma viride)3838菌株在制备快速促进根结线虫卵块孵化菌剂中的应用。本发明的有益效果在于:用绿色木霉菌(Trichoderma viride)3838菌株制备的菌剂能够促进根结线虫卵块快速孵化出J2,孵化速率能够提高200%以上,具有快速、高效的优点;在有关使用J2的实验研究中具有潜在和实际的应用价值。

技术领域
本发明涉及一株绿色木霉菌(Trichoderma viride)3838菌株及其应用,属于应用微生物学领域。
背景技术
植物寄生线虫是对农作物危害很大的一类害虫,仅根结线虫(Meloidogyne spp.)每年在全球造成的损失就高达数百亿美元。在我国,植物寄生线虫危害水稻、花生、烟草、茶叶、果树、蔬菜、花卉、药材等,其中以茄科、葫芦科、十字花科等植物受害尤为严重,严重危害农业生产的一类植物病原物。目前根结线虫、孢囊线虫、松材线虫和茎线虫是主要植物寄生线虫,其中根结线虫(Meloidogyne spp.)寄主植物就超过3000多种,但是就目前的研究背景下,管理和控制根结线虫危害的方法非常有限。
根结线虫卵的孵化是其侵染作物的必要步骤,二代线虫(J2)是重要的防治对象。但是由于根结线虫的生活史必须经历根内寄生阶段,所以J2的获得并不容易,大大制约了实验室研究进度。
本发明提供的绿色木霉菌(Trichoderma viride)3838菌株及其应用,经文献检索,未发现与本发明内容相同文献的公开报道。
发明内容
本发明的目的在于克服现有技术的不足,提供一种绿色木霉菌(Trichodermaviride)3838菌株及其应用。
本发明具体是这样实现的:
发明人在前期研究中,筛选获得了具有快速促进南方根结线虫卵块孵化功能的一株绿色木霉菌(Trichoderma viride)3838菌株,该菌株已于2016年4月19日保藏在中国微生物菌种保藏管理委员会普通微生物中心;保藏单位地址:北京市朝阳区北辰西路1号院3号中国科学院微生物研究所;保藏号:CGMCC No.12372。
本发明的绿色木霉菌(Trichoderma viride)3838菌株在制备快速促进根结线虫卵块孵化菌剂中的应用。
本发明的有益效果在于:用绿色木霉菌(Trichoderma viride)3838菌株制备的菌剂能够促进根结线虫卵块快速孵化出J2,孵化速率能够提高200%以上,具有快速、高效的优点;在有关使用J2的实验研究中具有潜在和实际的应用价值。
具体实施方式
本发明菌剂的发酵生产方法(常规方法):
1、将保藏号为CGMCC No.12372的绿色木霉菌(Trichoderma viride)3838菌株的菌丝体接种到试管琼脂培养基上,培养基配方为PDA培养基,即马铃薯200g;葡萄糖20g;琼脂20g;水1000mL。121℃灭菌20分钟。25-28℃培养3-5天,获得试管种。
2、再将试管种接种到500mL三角瓶(每瓶装350mL)液体培养基中,培养基配方为PDB培养基,即马铃薯200g;葡萄糖20g;水1000mL,pH自然。121℃灭菌20分钟。然后在25-28℃下,摇床转速以180-210rpm/min于摇床上发酵5-8天得到发酵液,备用。
本发明菌剂对南方根结线虫卵块的孵化试验:
1、南方根结线虫卵块(Meloidogyne incognita)的培养
将温室培养的带有南方根结线虫的番茄根用无菌水洗净,解剖针挑取大小均一的根结线虫卵块置于0.1%的NaClO溶液中消毒3分钟,再用28℃无菌水多次冲洗卵块,富集备用。
2、防效试验方法
(1)试验所用菌剂及处理
A、液体菌剂:将原液发酵液(28℃)依次稀释为1×、10×、100×的菌剂;将发酵液原液加热70℃的基础上,依次稀释为1×、10×、100×的菌剂;将发酵液原液加热100℃的基础上,依次稀释为1×、10×、100×的菌剂;
B、空白对照:无菌水;
(2)试验方法
在3cm平板中分别加入2mL不同处理的菌剂,以加2mL无菌水作为对照,然后分别向处理和对照中各加入1个卵块,放入28℃恒温箱中培养,每隔6小时观察一次并记录卵块孵化出的J2条数。每个处理重复3次。
实施例1:用不同浓度菌剂促进南方根结线虫卵块孵化的试验
按上述试验方法进行菌剂促进南方根结线虫卵块孵化的试验。列表所示为孵化量数据。

结果表明,该菌剂在10×浓度下对J2的孵化效果最好。
实施例2:不同时间下菌剂促进南方根结线虫卵块孵化的试验
实施例2方法同实施例1。试验结果见表2。

结果表明,该菌剂对J2的孵化数量随着时间的增加而增加,比对照有显著提高,持续有效。
实施例3:用不同温度加热后菌剂促进南方根结线虫卵块孵化的试验
实施例3方法同实施例1。试验结果见表3。
表3不同温度加热后的菌剂在稀释1×,10×浓度下对J2的孵化量(6h)
结果表明,该菌剂在加热100℃后,稀释10×浓度下对J2的孵化效果最好。
