睾酮是雄激素的一种。雄激素是一类含19个碳原子的类固醇激素,体内主要有睾酮,双氢睾酮,雄烯二酮,去氢表雄酮,去氢表雄酮硫酸盐等。由于睾酮的量远大于双氢睾酮,其生物效能又明显大于其它代谢物,所以,睾酮常被作为雄性激素的代表。睾酮除维持男子性功能和副性特征以及雄性攻击意识外,还有刺激组织摄取氨基酸,促进RNA、DNA和蛋白质合成,促进骨骼肌和骨骼生长,刺激红细胞生成,增加肌糖原储备,降低脂肪含量等作用,因此,睾酮与人体的运动能力、肌肉力量的增长、疲劳的消除等都有密切的关系。
玛卡,学名:Lepidium meyenii Walp.。是原产南美洲的一种十字花科植物。叶子椭圆,根茎形似小圆萝卜,可食用,是一种纯天然食品,营养成份丰富,有“南美人参”之誉。玛卡的下胚轴可能呈金色或者淡黄色、红色、紫色、蓝色、黑色或者绿色。淡黄色的根最常见,形状、味道也最好。
玛卡是一种非常独特的高原农作物,含有丰富的营养成分和药用成分,具有强身健体、增强人体免疫力、提高生育力,快速恢复体力、抗疲劳等功效,而且无任何毒副作用,因而其在当地民间有“秘鲁人参”和“南美人参”的
美誉。以玛卡为主要原料生产的各类保健品近年来在欧美、日本等市场上已得到越来越多消费者的青睐。
本发明提供了一种纳米级东革阿里和玛卡混合提取物,采用超临界萃取和超细研磨技术,得到纳米级提取物,更有利于人体吸收,能有效促进男性睾酮分泌。
发明内容
针对现有技术中存在的上述不足,本发明所要解决的技术问题之一是提供一种纳米级东革阿里和玛卡混合提取物的制备方法。
本发明所要解决的技术问题之二是提供上述纳米级东革阿里和玛卡混合提取物促进睾酮分泌的应用。
本发明目的是通过如下技术方案实现的:
一种纳米级东革阿里和玛卡混合提取物的制备方法,包括以下步骤:将东革阿里和玛卡分别粉碎,按质量比为(1-3):(1-3)混合,再加入到酶液中混合均匀,经超声浸提、离心分离、过滤、浓缩、干燥、超细研磨得到纳米级东革阿里和玛卡混合提取物。
优选地,所述的酶液由下述重量份的原料组成:0.1-0.9份酶、0.01-0.09份酶稳定剂、95-105份水组成;所述酶由枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶混合而成,所述枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶的质量比为(1-3):(1-3):(1-3)。
优选地,所述的酶稳定剂由D-山梨醇、木三糖、木二糖混合而成,所述D-山梨醇、木三糖、木二糖的质量比为(1-3):(1-3):(1-3)。
优选地,所述的东革阿里、玛卡分别粉碎至5-50目,所述超声浸提中酶液与东革阿里的质量比为10:1-50:1,超声浸提的超声波功率为200-500W,超声浸提的温度为15-45℃,超声浸提的时间为15-45min。
优选地,所述的过滤为0.1-0.5μm的聚偏二氟乙烯超滤膜过滤。
优选地,所述的浓缩为孔径为0.0001μm的反渗透膜浓缩。
优选地,所述的干燥为离心式喷雾干燥、压力式喷雾干燥、冷冻干燥、真空干燥中的一种或多种。
本发明还提供了上述方法制备的纳米级东革阿里和玛卡混合提取物促进睾酮分泌的应用,直接加水冲泡饮用或用于增强性功能的保健制剂,所述保健制剂包括片剂、胶囊、滴丸、注射液、冻干粉针、汤剂、缓释制剂、散剂、颗粒剂、栓剂、气雾剂。
即本发明提供了将上述方法制备的纳米级东革阿里和玛卡混合提取物用于制备增强性功能的保健制剂的应用。
具体的,在本发明中:
枯草杆菌蛋白酶,CAS号:9014-01-1。
木瓜蛋白酶,CAS号:9001-73-4。
单宁酶,CAS号:9025-71-2。
D-山梨醇,CAS号:50-70-4。
木三糖,CAS号:47592-59-6。
木二糖,CAS号:6860-47-5。
本发明纳米级东革阿里和玛卡混合提取物的制备方法,采用超临界萃取和超细研磨技术,得到纳米级提取物,活性成分提取率高,更有利于人体吸收,东革阿里和玛卡中含有的有效生物活性成分具有增强人体自身免疫力的功能,促进自身睾酮素的分泌,保持旺盛精力,减缓压力,清除肾脏的毒素,强效补肾,加强人体血液循环及新陈代谢,抗氧化、抗衰老,功效显著。

下面结合实施例对本发明做进一步的说明,以下所述,仅是对本发明的较佳实施例而已,并非对本发明做其他形式的限制,任何熟悉本专业的技术人员可能利用上述揭示的技术内容加以变更为同等变化的等效实施例。凡是未脱离本发明方案内容,依据本发明的技术实质对以下实施例所做的任何简单修改或等同变化,均落在本发明的保护范围内。
实施例中原料介绍:
东革阿里采用广州市礼顺堂贸易有限公司提供的野生东革阿里。
玛卡采用云南丽江黑玛卡。
实施例1
纳米级东革阿里和玛卡混合提取物的制备方法,包括以下步骤:
将东革阿里和玛卡分别粉碎至10目,将粉碎后东革阿里和粉碎后的玛卡按质量比为2:1混合,得到混合物;再按酶液与混合物的质量比为20:1(即每1g混合物用20g酶液)混合均匀,采用超声浸提,超声浸提的超声波功率300W,超声浸提的温度为30℃,超声浸提的时间为30分钟,得到浸提液;将浸提液通过离心机离心去除悬浮物,得到离心液,离心液经过孔径为0.1μm的聚偏二氟乙烯超滤膜过滤,得到过滤液,过滤液再经孔径为0.0001μm的反渗透膜浓缩,得浓缩液,经-30℃冻干成粉,最后采用超细研磨机(超细研磨机为无锡市飞鸣机械有限公司生产的型号为CXM的超细研磨机)进行超细研磨至500nm,得到实施例1的纳米级东革阿里和玛卡混合提取物。
所述的酶液由0.2重量份枯草杆菌蛋白酶、0.2重量份木瓜蛋白酶、0.2重量份单宁酶、0.02重量份D-山梨醇、0.02重量份木三糖、0.02重量份木二糖、100重量份水搅拌混合均匀得到。
实施例2
与实施例1基本相同,区别仅在于:所述的酶液由0.3重量份枯草杆菌蛋白酶、0.3重量份木瓜蛋白酶、0.02重量份D-山梨醇、0.02重量份木三糖、0.02重量份木二糖、100重量份水搅拌混合均匀得到。得到实施例2的纳米级东革阿里和玛卡混合提取物。
实施例3
与实施例1基本相同,区别仅在于:所述的酶液由0.3重量份枯草杆菌蛋白酶、0.3重量份单宁酶、0.02重量份D-山梨醇、0.02重量份木三糖、0.02重量份木二糖、100重量份水搅拌混合均匀得到。得到实施例3的纳米级东革阿里和玛卡混合提取物。
实施例4
与实施例1基本相同,区别仅在于:所述的酶液由0.3重量份木瓜蛋白酶、0.3重量份单宁酶、0.02重量份D-山梨醇、0.02重量份木三糖、0.02重量份木二糖、100重量份水搅拌混合均匀得到。得到实施例4的纳米级东革阿里和玛卡混合提取物。
实施例5
与实施例1基本相同,区别仅在于:所述的酶液由0.2重量份枯草杆菌蛋白酶、0.2重量份木瓜蛋白酶、0.2重量份单宁酶、0.03重量份D-山梨醇、0.03重量份木三糖、100重量份水搅拌混合均匀得到。得到实施例5的纳米级东革阿里和玛卡混合提取物。
实施例6
与实施例1基本相同,区别仅在于:所述的酶液由0.2重量份枯草杆菌蛋白酶、0.2重量份木瓜蛋白酶、0.2重量份单宁酶、0.03重量份D-山梨醇0.03重量份木二糖、100重量份水搅拌混合均匀得到。得到实施例6的纳米级东革阿里和玛卡混合提取物。
实施例7
与实施例1基本相同,区别仅在于:所述的酶液由0.2重量份枯草杆菌蛋白酶、0.2重量份木瓜蛋白酶、0.2重量份单宁酶、0.03重量份木三糖、0.03重量份木二糖、100重量份水搅拌混合均匀得到。得到实施例7的纳米级东革阿里和玛卡混合提取物。
实施例8
上述实施例1制备的纳米级东革阿里和玛卡混合提取物促进睾酮分泌的应用,取0.3g加温度为80℃的开水300mL冲泡后饮用。能够增强人体自身免疫力的功能,促进自身睾酮素的分泌。
测试例1
对实施例1-7制得的纳米级东革阿里和玛卡混合提取物中主要成分进行浸提率测试。
采用高效液相色谱法测试,具体结果如表1所示。

比较实施例1与实施例2-4,实施例1(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶复配)主要成分浸提率明显高于实施例2-4(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶任意二者复配);比较实施例1与实施例5-7,实施例1(D-山梨醇、木三糖、木二糖复配)主要成分浸提率明显高于实施例5-7(D-山梨醇、木三糖、木二糖任意二者复配)。
测试例2
对实施例1-7制得的纳米级东革阿里和玛卡混合提取物进行促睾酮分泌测试。
试验对象:SPF级雄性SD大鼠(北京大学医学部实验动物中心提供),8周龄,体重(178.81±6.63)g,自然采光,自由饮食,环境温度为(23±2)℃,湿度为40%~60%。
分为7组,每组10只SPF级雄性SD大鼠,实施例1-7制得的纳米级东革阿里和玛卡混合提取物,取0.3g加温度为80℃的开水300mL冲泡,每天灌胃给药一次,于下午进行,给药剂量为20g/kg体重。
2周后进行血清睾酮含量测试,差异具有显著性(P<0.05),具体结果如表2所示。

比较实施例1与实施例2-4,实施例1(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶复配)血清睾酮含量明显低于实施例2-4(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶任意二者复配);比较实施例1与实施例5-7,实施例1(D-山梨醇、木三糖、木二糖复配)血清睾酮含量明显低于实施例5-7(D-山梨醇、木三糖、木二糖任意二者复配)。
测试例3
将实施例1-7制得的纳米级东革阿里和玛卡混合提取物,置于25℃,相对湿度85%环境下保藏半年,然后进行大肠杆菌(ATYCC 25922)菌落总数测试,参照GBT 4789.2-2008食品卫生微生物学检验菌落总数测定,具体结果见表3。

发明人意外的发现,比较实施例1与实施例2-4,实施例1(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶复配)防腐性能明显优于实施例2-4(枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶任意二者复配);比较实施例1与实施例5-7,实施例1(D-山梨醇、木三糖、木二糖复配)防腐性能明显优于实施例5-7(D-山梨醇、木三糖、木二糖任意二者复配);其协同增效的机理有待发明人进一步研究探索。
Claims (8)
1.一种纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,包括以下步骤:将东革阿里和玛卡分别粉碎,按质量比为(1-3):(1-3)混合,再加入到酶液中混合均匀,经超声浸提、离心分离、过滤、浓缩、干燥、超细研磨得到纳米级东革阿里和玛卡混合提取物。
2.如权利要求1所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的酶液由下述重量份的原料组成:0.1-0.9份酶、0.01-0.09份酶稳定剂、95-105份水组成;所述酶由枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶混合而成,所述枯草杆菌蛋白酶、木瓜蛋白酶、单宁酶的质量比为(1-3):(1-3):(1-3)。
3.如权利要求2所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的酶稳定剂由D-山梨醇、木三糖、木二糖混合而成,所述D-山梨醇、木三糖、木二糖的质量比为(1-3):(1-3):(1-3)。
4.如权利要求1-3中任一项所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的东革阿里、玛卡分别粉碎至5-50目,所述超声浸提中酶液与东革阿里的质量比为10:1-50:1,超声浸提的超声波功率为200-500W,超声浸提的温度为15-45℃,超声浸提的时间为15-45min。
5.如权利要求1-3中任一项所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的过滤为0.1-0.5μm的聚偏二氟乙烯超滤膜过滤。
6.如权利要求1-3中任一项所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的浓缩为孔径为0.0001μm的反渗透膜浓缩。
7.如权利要求1-3中任一项所述的纳米级东革阿里和玛卡混合提取物的制备方法,其特征在于,所述的干燥为离心式喷雾干燥、压力式喷雾干燥、冷冻干燥、真空干燥中的一种或多种。
8.如权利要求1-7中任一项所述方法制备的纳米级东革阿里和玛卡混合提取物促进睾酮分泌的应用,直接加水冲泡饮用或用于增强性功能的保健制剂,所述保健制剂包括片剂、胶囊、滴丸、注射液、冻干粉针、汤剂、缓释制剂、散剂、颗粒剂、栓剂、气雾剂。
