技术领域
本发明涉及一种中药保健品组合药物,更具体的说涉及一种抗辐射桑杞枣组合物、制备方法及其应用。
技术背景
随着生活节奏的加快,很多长期遭受电磁辐射的人群,如医院放射科工作人员,机场、车站、码头的安检人员、高铁乘务员、高度依赖电脑工作的白领、用眼过度的学生、长期伏案的文字工作者等都处于亚健康状态,出现如脸色晦暗、皮肤干燥、毛发稀疏早白、黑眼圈、腰膝酸软、肢寒畏冷、夜尿频多、失眠不安、眩晕健忘、潮热盗汗等症状。因此,寻找和开发一些安全性高、具有抗氧化、抗辐射等保健功能的医药健康产品能顺应大多数人群的需求,具有广阔的市场。
桑椹、枸杞、大枣均为卫生部批准的药食同源的中药材。
桑椹俗称桑椹子、葚、桑枣等,是多年生木本植物桑的成熟果实。成熟的桑椹味美多汁、甘甜可口、风格独特,富含人体必需氨基酸、维生素、硒、钾、钙、镁、磷等矿物质、糖、微量元素、有机酸等营养成分和色素、花青素、白藜芦醇、没食子酸、黄酮类等多种功能性成分。具有补肝益肾、增强人体免疫力、抗氧化抗衰老、防癌抗诱变、降低血糖血脂、防止血管硬化、排毒养颜、乌发美容等防病保健功能。这些保健功能和药理作用与桑椹中主要活性成分有关,其中抗坏血酸和花青素具有很强的抗氧化、抗辐射作用,可改善中老年人、工作一族尤其是女性腰膝酸软、易疲劳、食欲不振等亚健康状态,不同品种桑椹中,以黑桑椹的抗氧化能力和DPPH自由基清除能力较强。目前,在食品工业中已研究出的桑椹产品有桑椹复合饮品、桑椹果酒、桑椹果醋等。
枸杞属茄科植物,其成熟果实亦称枸杞子,是我国传统名贵中药材,素有“红宝”美称,具有多种保健功效。枸杞含有人体必需的多种营养成分如氨基酸、脂肪、糖、无机盐、多种微量元素和维生素等。此外,枸杞含有多种生物活性成分,包括类黄酮、枸杞多糖、枸杞子油、类胡萝卜素、玉蜀黄素、酸浆果红素、甜菜碱等,因此具有多种保健功效,现代医学发现枸杞子具有补气强精、滋补肝肾、抗衰老、止消渴、暖身体、保护细胞、神经和血管、降低血压、血脂和血糖、抗脂肪肝、抗肿瘤的功效。枸杞子作为一种保健食品,已经广泛应用于国内外市场中。其中,枸杞多糖是诸多药理功效的主要活性成分。枸杞多糖是从枸杞中提取而得的一种水溶性多糖,具有增强机体免疫力、降血糖、降血脂、抗衰老、抗肿瘤和保肝护肝等功效,可改善老年人、工作一族和学生一族人群易疲劳、食欲不振和视力模糊等的亚健康状态,并具有抗氧化、抗衰老等作用。
大枣自古以来是重要的滋补品,是我国传统的药食两用果品之一,具有补中益气、养血安神等功效,其味甘香甜、营养丰富,现代药理学和食品化学与营养学研究表明,大枣富含多糖、蛋白质、维生素C、黄酮类化合物、三萜酸、环磷酸腺苷等多种生理活性物质,具有补脾胃、养血分、护肝脏、抗衰老、降低血清胆固醇、防治高血压、抑制癌细胞增值等功效。其中红枣多糖为主要活性成分,已证实红枣多糖具有抗氧化、抗疲劳、调节免疫、抑制癌细胞增殖、改善糖尿病症状、保肝护肝、抗凝血等多种生理活性。大枣中的活性多糖由于含量多、药用价值高而受到广泛关注,具有多种生理功效与保健功能。
发明内容
针对背景技术中存在的问题,本发明提供了一种抗辐射桑杞枣组合物及其制备方法,所述组合物由鲜桑葚、枸杞和大枣的提取物组成。
所述组合物由桑葚汁水的干燥物、脱脂后的枸杞提取物和大枣粗多糖组成。
所述组合物中桑葚、枸杞和大枣的重量比为桑椹20-30:12-18:5-10。
所述组合物中桑葚、枸杞和大枣的重量为50:30:15。
抗辐射桑杞枣组合物的制备方法,包括以下步骤:
1)根据规定的重量比称量鲜活桑椹、枸杞和大枣;
2)选拣新鲜的黑桑葚并用pH5-6稀酸清洗,将清洗后桑椹用50-60℃的水漂烫3-5min,对桑椹榨汁离心,得到澄清的桑椹汁水;
3)加入桑椹汁水重量15-25%的麦芽糊精作为助干剂,采用喷雾干燥制备得到桑椹提取物干粉;
4)将枸杞的拣选去除果梗及霉烂果实,清水漂洗;在洗净后的枸杞加6倍量石油醚浸提1h,脱脂过滤,得到枸杞果渣;将枸杞果渣再加入3-6倍量水浸泡1-2h后,煎煮0.5-1h,过滤,得到枸杞汁,再在枸杞果渣中补加2-8倍量水,煎煮30min,过滤,合并枸杞汁,真空干燥,得到枸杞提取物;
5)将大枣清洗和去核,将去核的大枣切块于干燥箱中60℃烘干,粉碎过40目筛,得到大枣粗粉;将大枣粗粉采取3-6倍量石油醚回流提取1-2h去除脂肪,抽滤后烘干得到脱脂大枣;
6)将脱脂大枣加入5-8倍量水,于70-80℃条件下浸提2-5次,每次1-2h,过滤、离心收集上清液,得到大枣水提液;
7)将大枣水提液真空浓缩后加入乙醇边加边搅拌,至乙醇终浓度为70%(V:V)以上,并于2-5℃放置过夜充分沉淀,除去上清液得沉淀,再将沉淀复溶于少量蒸馏水中,乙醇沉淀,沉淀复溶后用依次用无水乙醇、丙酮、乙醚洗涤,真空干燥得到红枣粗多糖。复溶后以木瓜蛋白酶和Sevage结合法去除游离蛋白质,蒸馏水透析4d,浓缩后醇沉,减压真空干燥即得红枣粗多糖提取物;
8)将上述的桑椹提取物干粉、枸杞提取物、红枣粗多糖提取物分别进行粉碎、过100目筛后,将三者混合得到桑杞枣组合物;
9)将桑杞枣组合物添加辅料制成粉剂、颗粒剂、胶囊剂、速溶片或泡腾片剂。
上述所述的抗辐射桑杞枣组合物在制备抗辐射药物中的应用。
本发明的有益技术效果是:本发明提供的桑杞枣组合物在抗氧化和抗辐射方面有良好的应用;其活性优于桑葚与枸杞的组合。
具体实施方式
实施例1供试样品的制备
1桑杞枣组合物的制备1
1.1取新鲜采摘的黑色的桑椹50kg,去除果梗及霉烂、损伤的果实,pH5-6稀酸清洗浸泡、清水漂洗。将清洗干净的桑椹用50-60℃的热水漂烫3-5分钟,采用榨汁机榨汁,离心将果汁和果渣分离,得到澄清桑椹原果汁(即桑葚汁水)。
加入澄清桑椹原果汁重量15-25%用量的助干剂麦芽糊精,采用喷雾干燥法干燥(干燥条件:物料进料速度35-45r/min,进风温度170-185℃,出口温度80-85℃,雾化器转速35000r/min)或冷冻干燥法干燥(条件:预冻-40℃4h,-10℃8h,0℃3h,10℃4h,30℃3h,30℃干燥10h),得到富含花青素的紫红色的桑椹提取物干粉。
1.2称取干燥枸杞36kg,将枸杞的拣选去除果梗及霉烂果实,清水漂洗;在洗净后的枸杞加6倍量石油醚浸提1h,脱脂过滤,得到枸杞果渣。将枸杞果渣再加入3-6倍量水浸泡1-2h后,煎煮0.5-1h,过滤,得到枸杞汁,再在枸杞果渣中补加8倍量水,煎煮30min,过滤,合并枸杞汁,真空干燥,得到富含粗多糖的淡黄棕色的枸杞提取物。
1.3挑选干燥、成熟、饱满,无霉烂的大枣,称取大枣15kg,去核,流水清洗。将去核的大枣切块于干燥箱中60℃烘干,粉碎过40目筛,得到大枣粗粉。大枣粗粉采取3-6倍量石油醚回流提取2h去除脂肪,抽滤后烘干。将脱脂后的大枣粗粉加入5-8倍量水,于70-80℃条件下浸提2-5次,每次1-2h,过滤、离心收集上清液,真空浓缩后加入4倍体积无水乙醇边加边搅拌,至乙醇终浓度为70%(V:V)以上,并于2-5℃放置过夜充分沉淀,除去上清液得沉淀,再将沉淀复溶于少量蒸馏水中,乙醇沉淀,沉淀复溶后用依次用无水乙醇、丙酮、乙醚洗涤,真空干燥得到红枣粗多糖。复溶后以木瓜蛋白酶和Sevage结合法去除游离蛋白质,蒸馏水透析4d,浓缩后醇沉,减压真空干燥即得红枣粗多糖提取物。
1.4将上述1.1-1.3得到的桑椹提取物干粉、枸杞提取物和红枣粗多糖提取物三者混合得到桑杞枣组合物A组。
1.5将桑杞枣组合物加入适宜的辅料或附加剂,通过适宜制备方法分别制成粉剂、颗粒剂、胶囊剂、速溶片、泡腾片等剂型以满足不同人群需要。
1.6将上述1.1、1.2得到的桑椹提取物干粉和枸杞提取物分别进行粉碎、过100目筛后,得到桑杞组合物对照组。
1.7分别称取鲜活桑椹40kg、枸杞24kg、大枣10kg,根据上述制备方法制备得到桑杞枣组合物B组。
1.8分别称取鲜活桑椹60kg、枸杞36kg、大枣20kg,根据上述制备方法制备得到桑杞枣组合物C组。
1.9分别称取鲜活桑椹50kg、枸杞36kg、大枣15kg,清洗,混合后用5倍药材重量的水回流提取3次,每次1h,合并提取液,加入助干剂麦芽糊精,干燥,粉碎制备得到桑杞枣组合物对照组。

实施例2桑杞枣组合物的抗辐射活性考察
1实验过程
准备SPF级昆明种雄性小鼠300只,体重为18~22g,分为5批,每批60只动物,并分别给名为外周血白细胞计数实验批、骨髓有核细胞实验批、小鼠骨髓细胞微核实验批、血液中SOD活性实验批和血清溶血素实验批。每批动物分为6组,分别给名为:模型对照组、桑杞枣组合物A组、桑杞枣组合物B组、桑杞枣组合物C组、桑杞枣组合物对照组和桑杞组合物对照组,模型对照组给药蒸馏水,其他各组对应给药实施例1中制备得到的桑杞枣组合物A组、桑杞枣组合物B组、桑杞枣组合物C组、桑杞枣组合物对照组和桑杞组合物对照组提取物。给药剂量为将提取物配置成为50mg/mL浓度溶液的水溶液,每天灌胃1次,灌胃体积为0.1mL/10g Bw,对照组给予等体积的蒸馏水。连续灌胃21天后剂量组与辐射模型对照组均以同一剂量γ射线全身照射1次,根据不同指标选择不同的照射剂量。照射后继续灌胃至每项实验结束。
2实验数据分析
2.1外周血白细胞计数实验批的各组动物均以3Gy剂量的γ射线全身照射1次,分别于照射前、照射后第3天、第14天3次采尾尖血,计算白细胞数。具体见表1所示:

2.2骨髓有核细胞实验批于照射后第3天,颈椎脱臼处死小白鼠,分离股骨,用1mL注射器共吸取5mL Hank’S液,冲出股骨中的全部骨髓细胞,细胞悬液通过4号针头的注射器,使细胞在悬液中分散,镜下计数,计算每lmL骨髓细胞中的有核细胞数。具体如表2所示:

2.3小鼠骨髓细胞微核实验批采用上述相同的照射方法于照射后第3天,颈椎脱臼处死小白鼠,取出胸骨,用止血钳挤出骨髓液与小牛血清混匀,常规涂片,自然干燥后甲醇固定10min,Giemsa染色法染色,蒸馏水冲洗,晾干。光学显微镜下,每只动物计数10000个嗜多染红细胞(PCE)微核细胞数,微核率以千分率表示。具体见表3所示:

2.4血中SOD活性实验批的射线照射剂量为6Gy。于照射后第7天,摘眼球采血,离心,取血清,按试剂盒说明书测定血清SOD活性。

2.5血清溶血素实验批的射线照射剂量为2Gy。于照射第14天,进行血清半数溶血值(HC50)测定。取羊血,用生理盐水洗涤3次,每只鼠经腹腔注射2%(v/v,用生理盐水配制)压积RBC 0.2mL进行免疫。5天后,摘除眼球取血,离心,收集血清。按文献中的方法,测定和计算溶血素值,后者以半数溶血值(HC50)表示。具体见表5所示:

实施例3桑杞枣组合物的抗氧化活性考察
精密称取49.3mg DPPH用甲醇定容至250mL,得5×10-4mol/L的DPPH溶液。将桑杞枣组合物A组、桑杞枣组合物B组、桑杞枣组合物C组和桑杞枣组合物对照组溶解于甲醇中,制得质量浓度为0.01、0.02、0.04、0.08、0.12、0.16、0.20mg/mL的样品溶液。添加1.0mL 5×10-4mol/L的DPPH溶液至1.0mL各样品溶液中,用甲醇定容至5mL。25℃条件下静置30min,于517nm波长处测定吸光度。根据式(1)计算样品对DPPH自由基的清除率。对DPPH自由基的清除效果具体参见附图1所示。
其中,S为DPPH自由基清除率/%;A0为未加样的DPPH溶液(2.0mL DPPH+2.0mL甲醇)的吸光度;A1为样品与DPPH反应后的吸光度;A2为样品空白(2.0mL+2.0mL甲醇)的吸光度。
用70%乙醇配制质量浓度分别为0.20~1.0mg/mL的样品溶液(桑杞枣组合物A组、桑杞枣组合物B组、桑杞枣组合物C组和桑杞枣组合物对照组)1mL。取2.5mL于10支10mL试管中,分别加入2.5mL 1g/100mL铁氰化钾,混合均匀,于50℃水浴中反应20min。取出试管,再加入2.5mL 10g/100mL三氯乙酸,混合后以3000r/min离心10min。取上清液2.5mL,加入蒸馏水2.5mL、0.1g/100mL氯化铁0.5mL,混合均匀,静置10min后,测定波长700nm处的吸光度,所有测定结果均为3次平均值。绘制A700nm与样品质量浓度关系图,对比样品的还原力。具体见附图2所示。
抑制脂质过氧化活性的测定为:取0.50mL卵磷脂溶液,加入pH 7.4PBS缓冲液1.00mL,将桑杞枣组合物A组、桑杞枣组合物B组、桑杞枣组合物C组和桑杞枣组合物对照组不同质量浓度的样品溶液1.00mL,2.5mmol/L EDTA-Fe(Ⅱ)1.00mL,混匀后37℃水浴中反应45min,再加入28g/100mL三氯乙酸2.00mL,1g/100mL的硫代巴比妥酸1.00mL,混匀后置于100℃沸水浴中加热10min,冷却后在532nm波长处测定吸光度。用PBS缓冲液调零,空白管用PBS缓冲液代替样品测吸光度A0。每个试样作3个平行样,取其平均值。按式(2)计算脂质过氧化抑制率。具体见附图3所示。
式中:S为抑制率/%;A0为不含样品的吸光度;A1为含样品的吸光度。
实施例4桑杞枣组合物制剂的研究
1.桑杞枣组合物粉剂的制备:
【处方】桑椹、枸杞、大枣粗提取物混合物 3g(2-5g)
【制备】将桑椹、枸杞、大枣粗提取物分别进行粉碎、过100目筛、混合、分装得到桑杞枣组合物粉剂。
【使用】该粉剂不添加任何辅料和附加剂,临用时除去包装直接用100mL左右温开水进行冲饮,具有桑椹、枸杞、大枣的特异性香气、口感酸甜。
2.桑杞枣组合物颗粒剂的制备:
【处方】桑椹、枸杞、大枣粗提取物混合物 5份(5-10份)
糖粉 10份(10-20份)
糊精 10份(5-20份)
【制备】将桑椹、枸杞、大枣粗提取物及糖粉、糊精分别进行粉碎、过筛、混合、加入适量70%乙醇制软材、制粒、干燥、整理粒、分装、密封、包装得到桑杞枣组合物颗粒剂。
【使用】临用时取1袋桑杞枣组合物颗粒剂除去包装直接用100mL左右温开水溶解后直接饮用,具有桑椹、枸杞、大枣的特异性香气、口感酸甜。
3.桑杞枣组合物胶囊剂的制备:
【处方】桑椹、枸杞、大枣粗提取物混合物 5份(5-10份)
糖粉 10份(10-20份)
糊精 10份(5-20份)
【制备】将桑椹、枸杞、大枣粗提取物及糖粉、糊精分别进行粉碎、过筛、混合、加入适量70%乙醇制软材、制粒、干燥、整粒、分装、密封、包装填充胶囊得到桑杞枣组合物胶囊剂。
【使用】用温开水送服,一日2次,一次1粒。
4.桑杞枣组合物速溶片的制备:
【制备】将处方中桑椹、枸杞、大枣粗提取物和各种辅料分别进行粉碎、过筛、混合后按粉末直接压片法制备得到桑杞枣组合物速溶片。
【使用】临用时取1片桑杞枣组合物速溶片直接用服用,在无水的情况下在口腔中崩解迅速,口感良好,具有桑椹、枸杞、大枣的特异性香气。该速溶片特别适合特殊野外作业人群使用。
5、桑杞枣组合物泡腾片的制备
【制备】将处方中桑椹、枸杞、大枣粗提取物和各种辅料分别进行粉碎、过筛、混合后按常规泡腾片制备方法进行制备。
【使用】临用时取1片桑杞枣组合物泡腾片除去包装直接用100mL左右温开水溶解后直接饮用,具有桑椹、枸杞、大枣的特异性香气、口感酸甜。
Claims (7)
1.一种抗辐射桑杞枣组合物,其特征在于:所述组合物由鲜桑葚、枸杞和大枣的提取组成。
2.根据权利要求1所述的抗辐射桑杞枣组合物,其特征在于:所述组合物由桑葚汁水的干燥物、脱脂后的枸杞提取物和大枣粗多糖提取物组成。
3.根据权利要求2所述的抗辐射桑杞枣组合物,其特征在于:所述组合物中桑葚、枸杞和大枣的重量比为桑椹20-30:12-18:5-10。
4.根据权利要求3所述的抗辐射桑杞枣组合物,其特征在于:所述组合物中桑葚、枸杞和大枣的重量为50:30:15。
5.一种抗辐射桑杞枣组合物的制备方法,其特征在于:包括以下步骤:
1)根据规定的重量比称量鲜桑椹、枸杞和大枣;
2)选拣新鲜的黑桑葚并用pH5-6稀酸清洗,将清洗后桑椹用50-60℃的水漂烫3-5min,对桑椹榨汁离心,得到澄清的桑椹汁水;
3)加入桑椹汁水重量15-25%的麦芽糊精作为助干剂,采用喷雾干燥制备得到桑椹提取物干粉;
4)将枸杞拣选去除果梗及霉烂果实,清水漂洗;在洗净后的枸杞加 6倍量石油醚浸提1h,脱脂过滤,得到枸杞果渣;将枸杞果渣再加入3-6倍量水浸泡1-2h后,煎煮0.5-1 h,过滤,得到枸杞汁,再在枸杞果渣中补加2-8倍量水,煎煮30 min,过滤,合并枸杞汁,真空干燥,得到枸杞提取物;
5)将大枣清洗和去核,将去核的大枣切块于干燥箱中 60℃烘干,粉碎过 40 目筛,得到大枣粗粉;将大枣粗粉采取3-6倍量石油醚回流提取1-2 h 去除脂肪,抽滤后烘干得到脱脂大枣;
6)将脱脂大枣加入5-8倍量水,于70- 80 ℃条件下浸提 2-5 次,每次1-2h,过滤、离心收集上清液,得到大枣水提液;
7)将大枣水提液真空浓缩后加入乙醇边加边搅拌,至乙醇终浓度为 70%(V:V)以上,并于2-5℃放置过夜充分沉淀,除去上清液得沉淀,再将沉淀复溶于少量蒸馏水中,乙醇沉淀,沉淀复溶后用依次用无水乙醇、丙酮、乙醚洗涤,真空干燥得到红枣粗多糖;复溶后以木瓜蛋白酶和Sevage结合法去除游离蛋白质,蒸馏水透析4d,浓缩后醇沉,减压真空干燥即得红枣粗多糖提取物;
8)将上述的桑椹提取物干粉、枸杞提取物、红枣粗多糖提取物分别进行粉碎、过100目筛后,将三者混合得到桑杞枣组合物;
9)将桑杞枣组合物添加辅料制成粉剂、颗粒剂、胶囊剂、速溶片或泡腾片剂。
6.权利要求1-4任一项所述的抗辐射桑杞枣组合物在制备抗辐射药物或保健品中的应用。
7.权利要求1-4任一项所述的抗辐射桑杞枣组合物在制备抗氧化药物和保健品中的应用。
