目前,无论是大人还是孩子生病时抗生素类药物成为医生和患者的首选,而大量使用抗生素会带来较强毒副作用,直接伤害身体,尤其是对儿童听力。抗生素的毒副反应最严重的是过敏反应。研究表明,每种抗生素对人体均有不同程度的伤害。比如链霉素、卡那霉素可引起眩晕、耳鸣、耳聋;庆大霉素、卡那霉素、万古霉素可损害肾脏等等。而耳朵对抗生素的副作用最为敏感,比如链霉素、庆大霉素、卡那霉素最易影响耳朵毛细胞,而使听力下降;抗生素用多了会使细菌产生耐药性,使抗生素药物效果变差,甚至无效;抗生素用得太多,也会让杀灭的细菌产生耐药性。而且,细菌的这种耐药性也是可以相互传播的,细菌对某种抗生素耐药,同时亦可对其他抗生素耐药,而且耐药性还可以在不同的细菌、人体正常菌群的细菌与致病菌之间,通过耐药基因相互传播,使细菌耐药性复杂化;抗生素用得过多过滥,会大量杀灭体内正常细菌,让致病菌乘虚而入,可以造成人的死亡。比如说人体肠道细菌,按一定的比例组合,各菌间互相制约,互相依存,在质和量上形成一种生态平衡,长期应用广谱抗生素,敏感肠菌被抑制,未被抑制的细菌乘机繁殖,从而引起菌群失调,可以引起一些维生素的缺乏,使身体抵抗力下降。
益生菌是一类通过改善宿主肠道微生物菌群的平衡而发挥作用的活性微生物,具有改善肠道菌群结构,抑制病原菌,消除致癌因子,提高机体免疫力,降低胆固醇等重要生理功效。现有市场上销售的益生菌相关产品种类比较繁多,但普遍存在质量不稳定、活菌数低,暴露在空气后很快失活,药用作用短。
发明内容
有鉴于现有技术的上述缺陷,本发明提供一种益生菌口服制剂及其制备方法,将益生菌包埋在微孔淀粉的空腔内,产品活菌数高,产品质量稳定,保存期长,药效持久,适应症效果显著。
为实现上述目的,本发明提供了一种益生菌口服制剂,各组份及重量百分比为:婴儿双岐杆菌菌粉3%~6%,嗜酸乳杆菌菌粉3%~6%,粪肠球菌菌粉3%~6%,蜡样芽孢杆菌菌粉3%~6%,凹土3%~6%,鲜橙浓缩粉10%~20%,乳酸菌素喷干粉10%~20%,多孔淀粉30%~60%。
优选的是,各组份最佳重量百分比为:婴儿双岐杆菌菌粉6%,嗜酸乳杆菌菌粉6%,粪肠球菌菌粉6%,蜡样芽孢杆菌菌粉6%,凹土6%,鲜橙浓缩粉20%,乳酸菌素喷干粉20%,多孔淀粉30%。
本发明还提供一种益生菌口服制剂的制备方法,其特征在于,包括以下步骤:
步骤(1):纯化凹土,将凹土置于去离子水中浸泡,20~28h,采用超声波超声40~60min,静置分层,取中间层凹土悬液,真空冷冻干燥后,研磨,过筛,备用;
步骤(2):制备多孔淀粉,医用淀粉在35℃~45℃的条件下干燥,粉碎,得到预糊化淀粉;把所述预糊化淀粉制备成乳浊液,然后用超声波在温度20℃~25℃、频率为15kHz~30kHz的条件下处理乳浊液,最后加入水解酶液进行反应,恒温水浴振荡24h后停止酶反应,加入NaOH溶液终止反应,抽滤水洗得湿淀粉样品,置烘箱内于20℃~60℃条件下烘干至恒重,用粉碎机粉碎获得多孔淀粉;
步骤(3):按照重量比分别称取婴儿双岐杆菌菌粉,嗜酸乳杆菌菌粉,粪肠球菌菌粉,蜡样芽孢杆菌菌粉,凹土,鲜橙浓缩粉,乳酸菌素喷干粉,粉碎成300~400目后,置于同一容器内,搅拌机搅拌均匀得到益生菌混合物;
步骤(4):按照重量比分别称取步骤(1)得到的纯化凹土和步骤(2)得到的多孔淀粉、步骤(3)得到的益生菌混合物,将三者混合,搅拌机100r/min搅拌20-40min,直至益生菌混合物和纯化凹土均包被在多孔淀粉的空腔内;
步骤(5):填袋、包装。优选的是,所述步骤(1)的真空冷冻温度为-10℃~-50℃。
婴儿双岐杆菌对婴幼儿有许多好处,如营养、免疫及抗感染作用,并且还具有抗过敏、抗肿瘤、调整肠道功能及改善营养的作用等,在临床上,双歧杆菌具有调整肠道功能紊乱的作用,可以预防腹泻,减少便秘,即双向调节,这种调节能起到预防和治疗各种肠道疾病的效果。
嗜酸乳杆菌调整肠道菌群平衡,抑制肠道不良微生物的增殖。嗜酸乳杆菌对致病微生物具有拈抗作用。嗜酸乳杆菌能分泌抗生物素类物质,对肠道致病菌产生的拮抗作用。嗜酸乳杆菌在人体整个胃肠道中,能够释放有益双歧杆菌等其他益生菌生长的物质,增加大肠内益生菌的数量、增强它们的生命力。
粪肠球菌是人和动物肠道内主要菌群之一,其能产生天然抗生素,有利于机体健康;同时还能产生细菌素等抑菌物质,抑制大肠杆菌和沙门氏菌等病原菌的生长,改善肠道微环境;还能抑制肠道内产尿素酶细菌和腐败菌的繁殖,减少肠道尿素酶和内毒素的含量,使血液中氨和内毒素的含量下降。
蜡样芽孢杆菌适用于婴幼儿腹泻,慢性腹泻,肠功能紊乱及肠炎的治疗,蜡样芽孢杆菌进入肠道后对葡萄球菌的生长有拮抗和抑制作用,对双岐杆菌,乳酸杆菌,消化链球菌等人体有益菌的生长具有促进作用,故能改善人体微生态环境。
鲜橙浓缩粉由橙汁浓缩制得,提高身体抵挡细菌侵害的能力,减少胆结石的发病率,不仅含有丰富的维生素C还含有维生素A、B、D及柠檬酸、苹果酸、果胶等成分,能增强毛细血管韧性;助尽快排泄脂类及胆固醇,并减少外源性胆固醇的吸收,故具有降低血脂的作用。具有行气化痰,健脾温胃,助消化,增食欲等药用功效,可作为健胃剂、芳香调味剂,基于上述的一出,本发明采用鲜橙浓缩粉还可以改善口感。
乳酸菌素用于消化不良、肠炎、小儿腹泻等病症的治疗。可在肠道内壁表面形成保护层,有效地阻止病原菌与病毒的侵袭;可刺激肠道产生抗体,从而提高肠道自身的免疫功能;能够保护并促进肠道内有益菌的生长,可选择性地杀死致病菌;能调节肠道内水和电解质的平衡;可促进胃酸的分泌,增强消化功能。
凹凸棒石粘土简称凹土,是一种天然非金属粘土矿物,具有特殊纤维晶体形态结构。由于其本身独特的微观结构和性质,赋予凹土许多优良性能,如吸附性、胶体性、粘结性、催化性等。凹土富含生物活性物质,这些物质在动物胃肠酸性或偏酸性环境下,吸水后溶散,其中的Ca2+、Mg2+、Na+、K+、Fe3+、Al3+等20多种矿物元素游离出来,凹土本身可作为益生菌载体制备益生菌制剂。
多孔淀粉是由具有生淀粉酶活力的酶在低于淀粉糊化温度下作用于生淀粉颗粒后形成的一种蜂窝状多孔变性淀粉。其表面的淀粉空腔可作为一个容器包埋益生菌和凹土,减轻机械挤压、胃酸等对益生菌的影响,使其顺利到达肠道发挥益生作用。
本发明的有益效果:本发明提供了一种益生菌口服制剂及其制备方法,各组份及重量百分比为:婴儿双岐杆菌菌粉3%~6%,嗜酸乳杆菌菌粉3%~6%,粪肠球菌菌粉3%~6%,蜡样芽孢杆菌菌粉3%~6%,凹土3%~6%,鲜橙浓缩粉10%~20%,乳酸菌素喷干粉10%~20%,多孔淀粉30%~60%,将益生菌包埋在微孔淀粉的空腔内,产品活菌数高,产品质量稳定,保存期长,药效持久,适应症效果显著。
具体实施方式
为了更好地理解与实施本发明,下面结合具体实施的例子对本发明做进一步详细的描述。
实施例1:
为实现上述目的,本发明提供了一种益生菌口服制剂,各组份重量百分比为:婴儿双岐杆菌菌粉6%,嗜酸乳杆菌菌粉6%,粪肠球菌菌粉6%,蜡样芽孢杆菌菌粉6%,凹土6%,鲜橙浓缩粉20%,乳酸菌素喷干粉20%,多孔淀粉30%。
本发明还提供一种益生菌口服制剂的制备方法,包括以下步骤:
步骤(1):纯化凹土,将凹土置于去离子水中浸泡,20h,采用超声波超声40min,静置分层,取中间层凹土悬液,真空冷冻干燥,温度为-30℃,研磨,过筛,备用;
步骤(2):制备多孔淀粉,医用淀粉在35℃的条件下干燥,粉碎,得到预糊化淀粉;把所述预糊化淀粉制备成乳浊液,然后用超声波在温度20℃、频率为15kHz的条件下处理乳浊液,最后加入水解酶液进行反应,恒温水浴振荡24h后停止酶反应,加入NaOH溶液终止反应,抽滤水洗得湿淀粉样品,置烘箱内于20℃条件下烘干至恒重,用粉碎机粉碎获得多孔淀粉;
步骤(3):按照重量比分别称取婴儿双岐杆菌菌粉,嗜酸乳杆菌菌粉,粪肠球菌菌粉,蜡样芽孢杆菌菌粉,凹土,鲜橙浓缩粉,乳酸菌素喷干粉,粉碎成300目后,置于同一容器内,搅拌机搅拌均匀得到益生菌混合物;
步骤(4):按照重量比分别称取步骤(1)得到的纯化凹土和步骤(2)得到的多孔淀粉、步骤(3)得到的益生菌混合物,将三者混合,搅拌机100r/min搅拌20min,直至益生菌混合物和纯化凹土均包被在多孔淀粉的空腔内;
步骤(5):填袋、包装。
实施例2:
为实现上述目的,本发明提供了一种益生菌口服制剂,
各组份重量百分比为:婴儿双岐杆菌菌粉4%,嗜酸乳杆菌菌粉4%,粪肠球菌菌粉4%,蜡样芽孢杆菌菌粉4%,凹土6%,鲜橙浓缩粉20%,乳酸菌素喷干粉20%,多孔淀粉38%。
本发明还提供一种益生菌口服制剂的制备方法,包括以下步骤:
步骤(1):纯化凹土,将凹土置于去离子水中浸泡,28h,采用超声波超声60min,静置分层,取中间层凹土悬液,真空冷冻干燥,温度为-40℃,研磨,过筛,备用;
步骤(2):制备多孔淀粉,医用淀粉在45℃的条件下干燥,粉碎,得到预糊化淀粉;把所述预糊化淀粉制备成乳浊液,然后用超声波在温度25℃、频率为30kHz的条件下处理乳浊液,最后加入水解酶液进行反应,恒温水浴振荡24h后停止酶反应,加入NaOH溶液终止反应,抽滤水洗得湿淀粉样品,置烘箱内于60℃条件下烘干至恒重,用粉碎机粉碎获得多孔淀粉;
步骤(3):按照重量比分别称取婴儿双岐杆菌菌粉,嗜酸乳杆菌菌粉,粪肠球菌菌粉,蜡样芽孢杆菌菌粉,凹土,鲜橙浓缩粉,乳酸菌素喷干粉,粉碎成400目后,置于同一容器内,搅拌机搅拌均匀得到益生菌混合物;
步骤(4):按照重量比分别称取步骤(1)得到的纯化凹土和步骤(2)得到的多孔淀粉、步骤(3)得到的益生菌混合物,将三者混合,搅拌机100r/min搅拌40min,直至益生菌混合物和纯化凹土均包被在多孔淀粉的空腔内;
步骤(5):填袋、包装。
实施例3:
为实现上述目的,本发明提供了一种益生菌口服制剂,各组份及重量百分比为:婴儿双岐杆菌菌粉3%,嗜酸乳杆菌菌粉3%,粪肠球菌菌粉3%,蜡样芽孢杆菌菌粉3%,凹土6%,鲜橙浓缩粉20%,乳酸菌素喷干粉20%,多孔淀粉42%。
本发明还提供一种益生菌口服制剂的制备方法,包括以下步骤:
步骤(1):纯化凹土,将凹土置于去离子水中浸泡,25h,采用超声波超声50in,静置分层,取中间层凹土悬液,真空冷冻干燥,干燥温度为-25℃,研磨,过筛,备用;
步骤(2):制备多孔淀粉,医用淀粉在40℃的条件下干燥,粉碎,得到预糊化淀粉;把所述预糊化淀粉制备成乳浊液,然后用超声波在温度22℃、频率为25kHz的条件下处理乳浊液,最后加入水解酶液进行反应,恒温水浴振荡24h后停止酶反应,加入NaOH溶液终止反应,抽滤水洗得湿淀粉样品,置烘箱内于40℃条件下烘干至恒重,用粉碎机粉碎获得多孔淀粉;
步骤(3):按照重量比分别称取婴儿双岐杆菌菌粉,嗜酸乳杆菌菌粉,粪肠球菌菌粉,蜡样芽孢杆菌菌粉,凹土,鲜橙浓缩粉,乳酸菌素喷干粉,粉碎成300目后,置于同一容器内,搅拌机搅拌均匀得到益生菌混合物;
步骤(4):按照重量比分别称取步骤(1)得到的纯化凹土和步骤(2)得到的多孔淀粉、步骤(3)得到的益生菌混合物,将三者混合,搅拌机100r/min搅拌30min,直至益生菌混合物和纯化凹土均包被在多孔淀粉的空腔内;
步骤(5):填袋、包装。

1.一种益生菌口服制剂,其特征在于,各组份及重量百分比为:婴儿双岐杆菌菌粉3%~6%,嗜酸乳杆菌菌粉3%~6%,粪肠球菌菌粉3%~6%,蜡样芽孢杆菌菌粉3%~6%,凹土3%~6%,鲜橙浓缩粉10%~20%,乳酸菌素喷干粉10%~20%,多孔淀粉30%~60%。
2.如权利要求1所述的一种益生菌口服制剂,其特征在于,各组份最佳重量百分比为:婴儿双岐杆菌菌粉6%,嗜酸乳杆菌菌粉6%,粪肠球菌菌粉6%,蜡样芽孢杆菌菌粉6%,凹土6%,鲜橙浓缩粉20%,乳酸菌素喷干粉20%,多孔淀粉30%。
3.一种益生菌口服制剂的制备方法,其特征在于,包括以下步骤:
步骤(1):纯化凹土,将凹土置于去离子水中浸泡,20~28h,采用超声波超声40~60min,静置分层,取中间层凹土悬液,真空冷冻干燥后,研磨,过筛,备用;

步骤(3):按照重量比分别称取婴儿双岐杆菌菌粉,嗜酸乳杆菌菌粉,粪肠球菌菌粉,蜡样芽孢杆菌菌粉,凹土,鲜橙浓缩粉,乳酸菌素喷干粉,粉碎成300~400目后,置于同一容器内,搅拌机搅拌均匀得到益生菌混合物;
步骤(4):按照重量比分别称取步骤(1)得到的纯化凹土和步骤(2)得到的多孔淀粉、步骤(3)得到的益生菌混合物,将三者混合,搅拌机100r/min搅拌20-40min,直至益生菌混合物和纯化凹土均包被在多孔淀粉的空腔内;
步骤(5):填袋、包装。
4.如权利要求3所述的一种益生菌口服制剂的制备方法,其特征在于,所述步骤(1)的真空冷冻温度为-10℃~-50℃。
