本发明涉及一种植物源农用杀菌剂及其配制方法。该杀菌剂是由吴茱萸果实、果柄、果壳、叶经提取加工而成。其中主要活性成分的重量百分比例为:羟基吴茱萸碱6~10%、吴茱萸碱5~10%、吴茱萸次碱5~10%、去氢吴茱萸碱6~10%、双吴茱萸碱1~5%、吴茱萸庚碱1~5%、其他生物碱成分6~40%。将其加工制作成水乳剂,稀释后可用于防治多种植物病害。本发明的优点在于:原料易得、工艺简单、成本低,对油菜菌核病、小麦赤霉病、葡萄炭疽病等多种植物病害均有非常好的防治效果,且对作物安全,不污染环境,是一种适合于我国无公害农产品生产要求的生物农药制剂。

一、技术领域
本发明涉及一种吴茱萸杀菌剂及其配制方法。
二、背景技术
有关专家认为,中国应大力发展植物源农药。目前中国使用的农药80%以上为有机合成农药,大面积使用高毒、高残留的有机合成农药导致了严重的环境污染、生态平衡破坏、病原物抗药性等问题。随着人们崇尚自然、保护环境、关注食品安全的意识日渐增强,无公害生物农药产业及生物防治研究领域获得了难得的发展机遇。因此,开发植物源农药,已成为中国农药和中草药发展的一条新途径。用传统方法开发一个新农药,需筛选数万个新化合物甚至更多,耗资巨大、研发周期长。从生物体,尤其是野生植物中分离筛选强活性先导化合物,具有来源广泛、研发周期短、成本低的优点,且开发出的植物源农药具有环境友好、作用机制特异、人、畜以及其他非靶标生物相对安全、不易产生抗药性、在自然环境中易降解、无残留等优点,是发展有机农业、促进农业可持续发展的理想对象。
三、发明内容
本发明旨在提供一种吴茱萸杀菌剂及其配制方法。
本发明公开了一种吴茱萸杀菌剂,其特征在于,所述杀菌剂以吴茱萸活性总生物碱提取物为主要有效成分,吴茱萸活性总生物碱提取物中总生物碱含量为30~90%,所述吴茱萸活性总生物碱提取物包含:羟基吴茱萸碱、吴茱萸碱、吴茱萸次碱、去氢吴茱萸碱和其他生物碱成分;
所述杀菌剂按重量百分比计包括如下组分:
羟基吴茱萸碱 6~10%
吴茱萸碱 5~10%
吴茱萸次碱 5~10%
去氢吴茱萸碱 6~10%
双吴茱萸碱 1-5%
吴茱萸庚碱 1-5%
其他生物碱成分 6~40%
水 5~50%
表面活性剂 5~10%
所述吴茱萸活性总生物碱提取物杀菌剂由如下制备方法获得:
(1)将吴茱萸果实、果柄、果壳、叶置于提取釜中,加入3~10倍生药体积的低级醇,低级醇为C1~C5醇类,其浓度为50~95%,提取温度为30~80℃,过滤得到滤液,并反复提取1~4次,合并滤液得到粗提取液;
(2)粗提液浓缩至原来的1/3后,利用大孔吸附树脂吸附(型号为D101或HP-20或AB-8),用1倍柱体积的水洗去水溶性杂质,例如糖等,改用30%~80%乙醇对其进行梯度洗脱,收集洗脱液;
(3)将洗脱液减压浓缩至无有机溶剂味的流浸膏,得到纯度50~90%的总酚酸提取物。
(4) 配制过程是:先将表面活性剂、吴茱萸总生物碱提取物在匀质机中混合,在30~50℃温度下,边搅拌边将水加入匀质机中,搅拌30分钟,冷却至常温,形成水乳剂。使用时可以根据需要,加水稀释成不同浓度后喷洒使用。
本发明的优点在于:1、室内和温室试验结果表明,该产品对油菜菌核病菌、小麦赤霉病菌和葡萄炭疽病菌等多种植物病原菌均有非常好的抑制效果。2、该发明产品原料易得、加工简单、对设备要求低、使用经济。本发明涉及的植物芸香科(Rutaceae)吴茱萸属(Evodia)全世界约有150种,分布于亚洲、非洲东部和大洋洲。我国有20种5变种,其中多数为民间常用中草药,早在《神农本草经》中就有吴茱萸的记载,应用历史悠久,临床效果显著。吴茱萸属中的吴茱萸(E. rutaecarpa (Juss.) Benth)、石虎(E. rutaecarpa (Juss.)Benth var. officinalis (Dode) Huang) 及疏毛吴茱萸(E. rutaecarpa (Juss.) Benthvar. bodinieri (Dode) Huang)已作为法定中药列入我国药典2005年版中,具有散寒止痛、降逆止呕、助阳止泻、疏肝解郁、行气止痛之功效,颇受人们的关注。吴茱萸属的近代研究自1929年朝比奈等从吴茱萸中分离出主要成分为吲哚喹唑啉(indoloquinazoline)骨架的吴茱萸碱、吴茱萸次碱并确定了结构以来,已有很大的进展,经过大量前期试验,对活性成分提取溶剂筛选发现,以乙醇-水、甲醇-水、丙酮-水作溶剂提取吴茱萸中杀菌活性成分较好,考虑到加工成本及设备要求,选用相对便宜的乙醇-水作为提取溶剂。该产品加工不需要特殊设备,主要加工过程包括提取、浓缩和混合,一般农药企业均可加工。3、该发明产品对环境安全。该杀菌剂主要原料来源于植物,对环境及非靶标生物安全,是一种适合于我国无公害农产品生产要求的生物农药制剂。

五、具体实施方式:
结合具体实施方式对本发明作进一步详细描述,但本发明的内容并不仅仅限于所列举的实施方式。
实施例1
吴茱萸果实、果柄、果壳5 kg,用75%乙醇60℃回流提取3次,得总浸膏,总浸膏用A8大孔树脂吸附,分别用水、30%、50%、75%乙醇洗脱,收集大孔树脂30%、50%以及75%洗脱部位,浓缩洗脱液至1.5 kg流浸膏,得吴茱萸总生物碱提取物。将0.5 kg表面活性剂(如聚山梨醇酯-80)和吴茱萸总生物碱提取物1.5 kg流浸膏置于匀质机中,加热至30℃,边搅拌边将1kg水加入匀质机中,搅拌30分钟,充分混匀后,冷却至常温,形成水乳剂,获得吴茱萸杀菌剂。

吴茱萸叶50 kg,用95%乙醇60℃回流提取3次,得总浸膏,总浸膏用D101大孔树脂吸附,分别用水、30%、50%、80%乙醇洗脱,收集大孔树脂30%、50%以及80%洗脱部位,浓缩洗脱液至1.5 kg流浸膏,得吴茱萸总生物碱提取物。将0.5 kg表面活性剂(如聚氧化乙烯类非离子型表面活性剂:OP-10)和吴茱萸总生物碱提取物1.5 kg流浸膏置于匀质机中,加热至50℃,边搅拌边将1 kg水加入匀质机中,搅拌30分钟,充分混匀后,冷却至常温,形成水乳剂,获得吴茱萸杀菌剂。
实施例3
按实施例1配制的吴茱萸杀菌剂室内毒力测定:
室内毒力测定供试植物病原菌包括油菜菌核病菌(Sclerotinia sclerotiorum)、小麦赤霉病菌(Fusarium graminearum)、黄瓜灰霉病菌(Botrytis cinerea)、葡萄炭疽病菌(Colletotrichum gloeosporioides)、水稻纹枯病菌(Rhizoctonia solani)、玉米小斑病菌(Helminthosporium maydis)、草莓灰霉病菌(Botrytis cinerea)、水稻恶苗病菌(Fusarium moniliforme)、小麦纹枯病菌(Rhizoctonia cerealis)、番茄早疫病菌(Alternaria solani)、棉花黄萎病菌(Verticillium dahliae)和稻瘟病菌(Magnaporthe oryzae)等12种常见植物病原菌。所有供试病原菌均为田间采集分离菌株。
采用菌丝生长速率法对12种植物病原菌进行室内毒力测定。将各菌株在PDA平板上活化培养,然后用5mm口径的打孔器在菌落边缘打取菌碟。将供试杀菌剂用无菌水制成母液,然后使用倍比稀释法制成5个浓度梯度的PDA含药平板,以无菌水为对照,每个处理3个重复。将各病原菌菌碟接种至各处理平板上,置于25℃培养箱培养。待对照接近长满板,十字交叉法测定菌落直径,并计算抑制率。
试验结果
按实施例1配制的吴茱萸杀菌剂稀释浓度为500倍、1000倍、2000倍、4000倍和8000倍,对各植物病原菌的抑制活性见表1。从表中结果可以看出,按本发明技术生产的吴茱萸杀菌剂对供试病原菌均有较好的药效。

实施例4
按实施例2配制的吴茱萸杀菌剂对几种植物病原真菌的活体药效试验:
试验在江苏省中国科学院植物研究所温室水泥池进行,供试作物为油菜、小麦和水稻,水泥池内未使用过任何化学杀菌剂,测试对象为油菜菌核病菌、小麦赤霉病菌和稻瘟病菌。
实验方法:
供试药剂为按照实施例2配制的吴茱萸杀菌剂,供试浓度为750倍、1500倍和3000倍稀释,以清水为空白对照,所有药剂均使用背负式喷雾器进行喷雾。在用药前一天分别使用病原菌的菌碟或者孢子悬浮液进行人工接种,用药后7天进行第二次喷药,待对照充分发病后,调查发病情况并计算防效。
测试结果表明:在供试剂量处理下,各浓度杀菌剂对作物均无不良影响,生长正常。按实施例2配制的水乳剂对油菜菌核病、小麦赤霉病和稻瘟病防治效果良好。在750倍稀释使用剂量下对三种病害的防治效果均为85%以上;在1500倍稀释使用剂量下防治效果均为60%以上。
Claims (2)
1.一种吴茱萸杀菌剂,其特征在于,所述杀菌剂以吴茱萸活性总生物碱提取物为主要有效成分,吴茱萸活性总生物碱提取物中总生物碱含量为30~90%,所述吴茱萸活性总生物碱提取物包含:羟基吴茱萸碱、吴茱萸碱、吴茱萸次碱、去氢吴茱萸碱和其他生物碱成分;
所述杀菌剂按重量百分比计包括如下组分:
羟基吴茱萸碱 6~20%
吴茱萸碱 6~10%
吴茱萸次碱 6~10%
去氢吴茱萸碱 6~10%
其他生物碱成分 6~40%
水 5~50%
表面活性剂 5~10%
所述吴茱萸活性总生物碱提取物杀菌剂由如下制备方法获得:
a. 将吴茱萸果实、果柄、果壳、叶置于提取釜中,加入3~10倍生药体积的低级醇,低级醇为C1~C5醇类,其浓度为50~95%,提取温度为30~80℃,过滤得到滤液,并反复提取1~4次,合并滤液得到粗提取液;
b. 粗提液浓缩至原来的1/3后,利用大孔吸附树脂吸附,用1倍柱体积的水洗去水溶性杂质后,改用30%~80%乙醇对其进行梯度洗脱,收集洗脱液,所述的大孔树脂为型号D101、HP-20或AB-8;
c. 将洗脱液减压浓缩至无有机溶剂味的流浸膏,得到纯度50~90%的活性总生物碱提取物;
d. 先将表面活性剂、吴茱萸总生物碱提取物在匀质机中混合,在30~50℃温度下,边搅拌边将水加入匀质机中,搅拌30分钟,冷却至常温,形成水乳剂,即得吴茱萸活性总生物碱提取物杀菌剂;
其中,羟基吴茱萸碱、吴茱萸碱、吴茱萸次碱和去氢吴茱萸碱的结构如下:

2.如权利要求1所述的吴茱萸杀菌剂,其特征在于在防治油菜菌核病、小麦赤霉病、黄瓜灰霉病、葡萄炭疽病、水稻纹枯病、玉米小斑病、草莓灰霉病、水稻恶苗病、小麦纹枯病、番茄早疫病、棉花黄萎病、稻瘟病中的应用。
