技术领域

背景技术
众所周知,蛋白质在体内通过多种机制被清除,包括通过血清蛋白酶降解、通过肾脏消除、或通过受体清除。因此,已经进行了多种尝试来避免这样的蛋白质清除机制以增加生理活性的蛋白质的半衰期,从而增强它们的治疗效果。具体而言,为了增加蛋白质的半衰期,已经对于包括连接至蛋白质的聚乙二醇聚合物(PEG)、白蛋白、脂肪酸或抗体Fc片段(恒定区)的蛋白质缀合物进行了研究。这种研究旨在将这种物质共价地连接至生理活性的蛋白质,从而增加生理活性的蛋白质的血清半衰期并缩短药物施用的间隔,从而增加患者的便利。具体而言,为了稳定蛋白质并抑制它们与蛋白酶的接触和它们通过肾脏的消除,使用了将高度可溶的聚合物比如PEG化学地附着至蛋白质药物的表面的方法。在这种方法的情况下,众所周知,聚合物非特异性地结合至靶蛋白的特异性位点或各个位点来增加蛋白质的溶解度,稳定蛋白质并阻止蛋白质的水解,并且进一步地,不引起具体的副作用(Sada等,J.Fermentation Bioengineering 71:137-139,1991)。然而,这种方法存在一些问题,因为即使连接至生理活性的蛋白质的PEG可以增加蛋白质的稳定性,但是它显著降低了蛋白质的效价,并且随着PEG分子量增加,其与蛋白质的反应性降低,导致产量降低。此外,当蛋白质的特定氨基酸残基利用脂肪酸进行修饰时,修饰的脂肪酸可逆地结合至血清白蛋白以增加蛋白质的血清半衰期,但是半衰期为大约1天至1周,这表明半衰期的增加不是非常显著的。此外,存在缺点,因为与白蛋白可逆地解离的生理活性的蛋白质轻易地通过肾脏被消除。
由于这些原因,已经进行了努力以使用免疫球蛋白片段来增加包括蛋白质在内的生理活性物质的半衰期。具体而言,已经积极地进行了研究以通过将治疗性蛋白质与这种免疫球蛋白Fc片段融合来增加治疗性蛋白质的稳定性。
在哺乳动物中通过基因重组方法将干扰素(韩国专利特开公布号2003-9464)、白介素-4受体、白介素-7受体或红细胞生成素受体(韩国专利号249572)作为与免疫球蛋白Fc片段的融合进行表达是已知的。而且,国际专利公布号WO 01/03737公开了融合蛋白,其包括经由寡肽连接体连接至免疫球蛋白Fc片段的细胞因子或生长因子。而且,美国专利号5,116,964公开了融合蛋白,其包括通过基因重组方法融合至免疫球蛋白Fc片段的氨基或羧基末端的LHR(淋巴细胞表面糖蛋白)或CD4蛋白,并且美国专利号5,349,053公开了IL-2与免疫球蛋白Fc片段的融合蛋白。此外,通过基因重组方法制备的Fc融合蛋白的实例包括干扰素-β或其衍生物与免疫球蛋白Fc片段的融合蛋白(国际专利公布号WO 00/23472)、IL-5受体与免疫球蛋白Fc片段的融合蛋白(美国专利号5,712,121)、干扰素-α与免疫球蛋白G4Fc片段的融合蛋白(美国专利号5,723,125)和CD4蛋白与免疫球蛋白G2Fc片段的融合蛋白(美国专利号6,451,313)。此外,美国专利号5,605,690涉及对免疫球蛋白Fc片段中的氨基酸残基进行修饰并公开了使用通过修饰免疫球蛋白Fc片段中具体的补体结合位点或受体结合位点的氨基酸残基获得的Fc片段通过基因重组方法制备的TNFR-IgG1Fc融合蛋白。而且,使用如上所述修饰的免疫球蛋白Fc区通过基因重组方法制备融合蛋白的方法也在美国专利号6,277,375、6,410,008和6,444,792中公开。但是,具有与其融合的免疫球蛋白Fc片段的生物药需要克服由Fc片段固有的效应子功能引起的细胞毒性问题。
发明内容
技术问题
本发明人已经进行了大量的努力来制备具有免疫球蛋白Fc片段到生理活性的多肽造成的增加的血清体内半衰期的缀合物。结果,本发明人已经发现,当生理活性的多肽以单体形式连接至免疫球蛋白Fc片段时,与当生理活性的多肽以多聚体形式存在时相比,甚至在大鼠动物模型中,它显示了显著降低的受体介导的清除率并且还显示了长的半衰期,从而完成本发明。
技术方案

有益效果
如上所述,本发明的缀合物——其包括连接至免疫球蛋白Fc缀合物的生理活性的多肽单体——显示了显著降低的受体介导的内化或受体介导的清除率,并且因此具有增加的血清半衰期。因此,本发明的缀合物可以提供具有增加的血清半衰期和治疗优越性的药物。

具体而言,因为与多聚体缀合物——其包括连接至免疫球蛋白Fc片段的两分子或更多分子的生理活性的多肽——相比,根据本发明的单体缀合物由生理活性的多肽的受体介导的内化降低,因此具有长的体内作用持续时间。特别地,已知,肾脏消除——其是决定分子半衰期的另一机制——取决于分子的分子量。因此,如果肾脏消除是决定半衰期的重要变量,则随着具有更高分子量的多聚体缀合物的比率增加,半衰期将增加。然而,在本发明中,已经发现,单体缀合物显示了更长的体内作用持续时间。就此而言,可见受体介导的内化在增加根据本发明的缀合物的体内半衰期中是重要的因素。这样的结果可归因于单体缀合物的有益效果(例如,减小的位阻),该有益效果不能在多聚体缀合物中发现。
因此,与包括生理活性的多肽多聚体-免疫球蛋白Fc片段缀合物或包括框内连接至免疫球蛋白Fc的生理活性的多肽二聚体的缀合物的药物组合物相比,本发明的长效药物组合物可显示更长的体内作用持续时间。具体而言,在本发明的药物组合物中,单体缀合物——其包括连接至单个免疫球蛋白Fc片段的一分子的生理活性的多肽——相对于多聚体缀合物——其包括连接至免疫球蛋白Fc片段的两分子或更多分子的相同的生理活性的多肽——可以以至少19:1,优选地至少99:1更优选地至少500:1的摩尔比存在,并且在这种情况下,包括单体缀合物和多聚体缀合物的组合物因为组合物的体内半衰期没有降低,因此提供了优异的持久的制剂。
如本文所使用,术语“生理活性的多肽-免疫球蛋白Fc片段缀合物”指的是包括缀合至免疫球蛋白Fc片段的生理活性的多肽的缀合物。优选地,缀合物可包括插入生理活性的多肽和免疫球蛋白Fc片段之间以将生理活性的多肽连接至免疫球蛋白Fc片段的非肽基连接体。

同时,多聚体缀合物——其包括连接至免疫球蛋白Fc片段的两分子或更多分子的生理活性的多肽——包括如此形式:其中两分子或更多分子的生理活性的多肽连接至单个免疫球蛋白Fc片段(例如,由通过二硫键等彼此连接的两个Fc链组成)。
具体而言,生理活性的多肽二聚体-免疫球蛋白Fc片段缀合物包括如此形式:其中两分子的生理活性的多肽连接至单个免疫球蛋白Fc片段(例如,由通过二硫键等彼此连接的两个Fc链组成)。在此,生理活性的多肽可以经由非肽基连接体连接至组成免疫球蛋白Fc片段的两条链的每条,但不限于此。这种构型在图1(B)中显示。
此外,框内缀合物包括如此形式:其中生理活性的多肽多聚体,特别是生理活性的多肽二聚体,框内连接至单个免疫球蛋白Fc片段,并且也形成多聚体,其中包括生理活性的多肽的两个或更多个融合多肽与免疫球蛋白Fc片段融合。

如本文所使用,术语“非肽基连接体”指的是由彼此连接的两个或更多个重复单元组成的生物相容的聚合物,其中重复单元通过任何非肽共价键彼此连接。这种非肽基连接体可具有两个末端或三个末端。
本发明中使用的非肽基连接体可选自聚乙二醇、聚丙二醇、乙二醇与丙二醇的共聚物、聚氧乙烯化的多元醇、聚乙烯醇、多糖、葡聚糖、聚乙烯乙醚、生物可降解的聚合物比如聚乳酸(PLA)和聚乳酸-乙醇酸(PLGA)、脂质聚合物、甲壳质、透明质酸和其组合,但不限于此。优选地,其为聚乙二醇。
此外,本领域中已知的它们的衍生物和可以容易地以现有技术制备的衍生物也落入本发明的范围之内。
在通过常规框内融合方法制备的融合蛋白中使用的肽基连接体具有缺点,因为它容易地被蛋白酶体内裂解,并且因此也不能获得通过载体增加活性药物的血清半衰期的预期效果。为此,除了肽基连接体之外,非肽基连接体也可用于本发明来制备缀合物。非肽基连接体可以是对蛋白酶具有抗性以维持肽的血清半衰期——与载体的血清半衰期相似——的聚合物。因此,任何非肽基连接体可用于本发明而没有任何限制,只要它由具有上述功能的聚合物——即对蛋白酶具有体内抗性的聚合物——制成。非肽基连接体具有1-100kDa的分子量,优选地1-20kDa的分子量,但不限于此。
此外,本发明中连接至免疫球蛋白Fc片段的非肽基连接体可以不仅由一种聚合物制成,而且可以由不同种类的聚合物的组合制成。
用于本发明的非肽基连接体具有能够结合至免疫球蛋白Fc片段和蛋白药物的活性基团。
非肽基聚合物两端处的活性基团优选地选自活性醛基、丙醛基、丁醛基、马来酰亚胺基和琥珀酰亚胺衍生物。在此,琥珀酰亚胺衍生物可以是琥珀酰亚胺基丙酸酯、羟基琥珀酰亚胺基、琥珀酰亚胺基羧甲基或琥珀酰亚胺基碳酸酯。具体而言,当非肽基聚合物在其两端处具有活性醛基时,生理活性的多肽和免疫球蛋白分别有效地结合至非肽基连接体的两端,同时使非特异性反应最小化。经由醛键通过还原烷基化生成的最终产物比经由酰胺键连接生成的最终产物更稳定得多。醛活性基团可以在低pH下选择性地结合至N末端并且可以在高pH——例如9.0的pH——下与赖氨酸残基形成共价键。
非肽基连接体两端处的活性基团可以是相同的或不同的。例如,非肽基连接体的一端可具有马来酰亚胺基,和另一端可具有醛基、丙醛基、或丁醛。当在两端处具有羟基活性基团的聚乙二醇被用作非肽基连接体时,羟基可以通过已知的化学反应被活化成为多种活性基团。可选地,具有改性的活性基团的市售的聚乙二醇可以被用于制备本发明的缀合物。
如本文所使用,术语“生理活性的多肽”共同地指在体内具有任何生理活性的多肽,其一般具有多肽结构并且具有多种生理活性。生理活性的多肽包括起调节基因表达和生理功能以及纠正由缺乏或过多分泌参与调节体内功能的物质而引起的异常状况的功能。生理活性的多肽可包括一般的蛋白质治疗剂。
在本发明的缀合物中,生理活性的多肽的种类和大小没有被明确地限定,只要它可以以单体形式连接至免疫球蛋白Fc片段,以与当它以多聚体形式连接至免疫球蛋白Fc片段时相比,显示降低的受体介导的内化或受体介导的清除率。进一步地,它更优选为生理活性的多肽,其受体介导的内化和受体介导的清除率是主要的体内蛋白清除机制。

(1H-咪唑-4-基)-乙酰基-1(GEGTFTSDL SKQMEEEAVR LFIEWLKNGGPSSGAPPPS(Bachem))被用于制备本发明的实施例中的GLP-1激动剂-免疫球蛋白Fc片段。
在本文中,胃泌酸调节素衍生物的实例包括韩国专利特开公布号10-2012-0137271中公开的所有那些,并且胰岛素释放肽衍生物的实例包括韩国专利特开公布号10-2009-0008151中公开的那些,但不限于此。
由于免疫球蛋白Fc区是在体内被代谢的生物可降解的多肽,将其用作药物载体是安全的。而且,由于免疫球蛋白Fc区具有低于整个免疫球蛋白分子的分子量,因此它在缀合物的制备、纯化和产率方面是有益的。此外,由于Fab区——由于抗体之间的氨基酸序列的区别,其展示高的非同质性——被去除,因此Fc区具有大大增加的物质同质性和低的诱发血清抗原性的可能性。

在本发明中,免疫球蛋白Fc片段意欲不仅包括天然氨基酸序列,而且包括其序列突变体。如本文所使用,术语“氨基酸序列突变体”指的是由于一个或多个氨基酸残基的缺失、插入、非保守或保守置换或其组合而与天然氨基酸序列不同的序列。例如,在IgG Fc的情况下,在位置214到238、297到299、318到322或327到331处的氨基酸残基——其已知为在结合中是重要的——可被用作用于修饰的适合靶标。
此外,各种突变体也是可能的,包括具有能够形成二硫键的区域的缺失、天然Fc的N末端处的数个氨基酸的缺失、或甲硫氨酸残基添加至天然Fc的N末端的突变体。此外,为了消除效应子功能,可以去除补体结合位点,例如,C1q结合位点,并且还可以去除抗体依赖细胞介导的细胞毒作用(ADCC)位点。制备这样的免疫球蛋白Fc片段的序列衍生物的技术在国际专利公布号WO 97/34631和WO 96/32478中公开。
蛋白质和肽中的氨基酸交换——其通常不改变分子的活性——在本领域中是已知的(H.Neurath,R.L.Hill,The Proteins,Academic Press,New York,1979)。最常发生的交换——以两个方向——是Ala/Ser、Val/Ile、Asp/Glu、Thr/Ser、Ala/Gly、Ala/Thr、Ser/Asn、Ala/Val、Ser/Gly、Thy/Phe、Ala/Pro、Lys/Arg、Asp/Asn、Leu/Ile、Leu/Val、Ala/Glu、Asp/Gly。
在一些情况下,免疫球蛋白Fc片段也可以通过例如磷酸化、硫酸化、丙烯酰化(acrylation)反应、糖基化、甲基化、法尼基化、乙酰化或酰胺化修饰。
上述的Fc突变体是显示与本发明的Fc区的生理活性相同的生理活性,但是对热、pH等具有提高的结构稳定性的突变体。
此外,这些Fc片段可以从由人和动物——包括牛、山羊、猪、小鼠、兔、仓鼠、大鼠和豚鼠——分离的天然形式获得,或者可以是从转化的动物细胞或微生物获得的其重组形式或衍生物。在本文中,可以通过将完整的免疫球蛋白从人或动物的活体分离并用蛋白酶处理它,从天然免疫球蛋白获得Fc片段。当完整的免疫球蛋白用木瓜蛋白酶处理时,它被裂解为Fab和Fc,和当它用胃蛋白酶处理时,它被裂解为pF’c和F(ab)2。例如,这些片段可以经历排阻色谱以分离Fc或pF’c。
优选地,它是使用人Fc区从微生物获得的重组免疫球蛋白Fc区。
此外,免疫球蛋白Fc区可以是如此形式:具有天然糖链、与天然形式相比增加的糖链或与天然形式相比减少的糖链,或者可以是去糖基化的形式。免疫球蛋白Fc糖链的增加、减少或去除可以使用常规的方法进行,比如化学方法、酶法和使用微生物的基因工程方法。通过从Fc去除糖链而获得的免疫球蛋白Fc区显示了对补体(c1q)结合亲和力的急剧降低和抗体依赖细胞介导的细胞毒作用或补体依赖细胞毒性的降低或丧失,并且因此不诱发不必要的体内免疫应答。就此而言,去糖基化或无糖基化(aglycosylated)形式的免疫球蛋白Fc片段可以更适合用作药物载体。
如本文所使用,术语“去糖基化”意思是从Fc区酶促地去除糖部分,并且术语“无糖基化”意思是在原核细胞,优选地大肠杆菌中产生的未糖基化的Fc片段。
同时,免疫球蛋白Fc区可以源于人或动物,比如牛、山羊、猪、小鼠、兔、仓鼠、大鼠或豚鼠。优选地,它是人源的。此外,免疫球蛋白Fc片段可以是源自IgG、IgA、IgD、IgE和IgM、其组合、或其杂化物的Fc片段。优选地,它源自IgG或IgM,其是人血液中最丰富的蛋白质,并且最优选地,它源自已知增强配体结合蛋白的半衰期的IgG。
术语“组合”,如本文所使用,意思是将编码相同来源的单链免疫球蛋白Fc片段的多肽连接至不同来源的单链多肽以形成二聚体或多聚体。换句话说,二聚体或多聚体可以由选自IgG Fc、IgA Fc、IgM Fc、IgD Fc和IgE Fc片段的两个或更多个片段形成。
如本文所使用,术语“杂化物(hybrid)”意思是编码不同来源的两个或更多个免疫球蛋白Fc片段的序列存在于单链免疫球蛋白Fc片段中。在本发明中,各种类型的杂化物是可能的。换句话说,结构域杂化物可以由选自IgG Fc、IgM Fc、IgA Fc、IgE Fc和IgD Fc的CH1、CH2、CH3和CH4的1到4个结构域构成,并且可包括铰链区。
另一方面,IgG可以被分为IgG1、IgG2、IgG3和IgG4亚类,并且其组合或杂化物可用于本发明。优选的为IgG2和IgG4亚类,和最优选的为很少具有效应子功能比如CDC(补体依赖细胞毒性)的IgG4的Fc片段。换句话说,用作本发明中的药物载体的最优选的免疫球蛋白Fc片段是人IgG4衍生的未糖基化的Fc片段。人Fc片段比非人Fc片段是更优选的,非人Fc片段可在人体内充当抗原并且引起不期望的免疫应答,比如针对抗原产生新抗体。
在本发明的一个实例中,通过将生理活性的多肽单体经由非肽基聚合物连接至免疫球蛋白Fc片段(实施例)来制备缀合物,并且已经发现,与以二聚体形式存在的生理活性的多肽中的缀合物相比,制备的缀合物显示了降低的受体介导的内化,并且因此具有增加的体内半衰期(图2和3)。
在又另一方面,本发明提供了生理活性的多肽单体-免疫球蛋白Fc片段缀合物,其包括经由非肽基连接体连接至免疫球蛋白Fc片段的生理活性的多肽单体,其中生理活性的多肽以单体形式经由非肽基连接体连接至免疫球蛋白Fc片段,该缀合物与二聚体缀合物——其包括经由非肽基连接体连接至单个免疫球蛋白Fc片段的两分子的生理活性的多肽,或缀合物——其包括框内连接至免疫球蛋白Fc片段的生理活性的多肽二聚体——相比,显示了降低的受体介导的内化或受体介导的清除率。

在又另一方面,本发明提供了用于制备长效药物组合物的方法,该方法包括:(a)将生理活性的多肽连接至免疫球蛋白Fc片段以制备生理活性的多肽-免疫球蛋白Fc片段缀合物的混合物;和(b)自该混合物分离生理活性的多肽单体-免疫球蛋白Fc片段缀合物,该生理活性的多肽单体-免疫球蛋白Fc片段缀合物包括连接至单个免疫球蛋白Fc片段的一分子的生理活性的多肽。
在此,生理活性的多肽、免疫球蛋白Fc片段、非肽基连接体和长效药物组合物如上面所描述。
本发明的方法中的步骤(a)是将生理活性的多肽经由非肽基连接体共价地连接至免疫球蛋白Fc片段的步骤。步骤(a)可包括以下步骤:(i)将生理活性的多肽和免疫球蛋白Fc片段的任一个连接至非肽基连接体的一端处的活性基团;和(ii)将剩下的一个连接至非肽基连接体的另一端处的活性基团。步骤(a)可进一步在步骤(i)和(ii)之间包括分离连接至非肽基连接体的一端的生理活性的多肽或免疫球蛋白Fc片段的步骤。为了制备该缀合物,韩国专利号10-0725315的公开内容可以通过引用并入本文。
当通过该过程制备缀合物时,除了包括连接至免疫球蛋白Fc片段的生理活性的单体的缀合物之外,还可以作为副产物生成包括二聚体或多聚体形式的生理活性的多肽的缀合物。
因此,本发明的方法可进一步包括,在制备缀合物的混合物的步骤(a)之后,自该混合物分离生理活性的多肽单体-免疫球蛋白Fc片段缀合物的步骤(b),该生理活性的多肽单体-免疫球蛋白Fc片段缀合物包括连接至免疫球蛋白Fc片段的生理活性的多肽单体。
步骤(b)中的分离条件可根据使用的非肽基连接体、生理活性的多肽等的种类而变化。
在下文中,本发明将参照实施例进一步详细地描述。但是,应当理解,这些实施例仅是说明性的目的并且不意欲限制本发明的范围。
实施例:制备GLP-1激动剂-免疫球蛋白Fc片段缀合物
将3.4-kDa二乙酮(propion)-ALD2PEG(IDB,韩国)与GLP-1激动剂的赖氨酸残基位点特异性地反应。为了获得如此缀合物:在该缀合物中,PEG和GLP-1激动剂以1:1的比例彼此连接,然后使反应混合物经历阳离子交换柱层析以纯化单-聚乙二醇化(mono-PEGylated)的GLP-1激动剂。为了制备GLP-1激动剂-免疫球蛋白Fc片段缀合物——其包括特异性地连接至免疫球蛋白Fc片段的N末端的单-聚乙二醇化的GLP-1,在5.0-8.2的pH下进行反应。偶联反应之后,使用疏水柱和阴离子交换柱进行两步纯化过程,从而获得包括位点特异性地连接至免疫球蛋白Fc片段的GLP-1激动剂的GLP-1激动剂-免疫球蛋白Fc片段缀合物。
分析通过该过程制备的GLP-1激动剂-免疫球蛋白Fc片段缀合物,结果,发现,仅GLP-1激动剂单体-免疫球蛋白Fc片段缀合物在制备的缀合物中存在,或者包括痕量的GLP-1激动剂二聚体-免疫球蛋白Fc片段缀合物的多聚体缀合物以基于制备的缀合物的总重量的5%(w/w)或更低的量存在。
比较实施例:制备GLP-1激动剂二聚体-免疫球蛋白Fc片段缀合物
将包括连接至免疫球蛋白Fc片段的GLP-1激动剂的DNA序列克隆入动物细胞表达载体中。使用转染溶液(FreeStyleTM MAX Reagent,Invitrogen)将编码重组蛋白的DNA转染入动物细胞系293-F(Freestyle 293-F细胞,Invitrogen),然后将细胞培养48小时并且收获培养物。使用亲和柱从培养物中纯化表达的GLP-1激动剂二聚体-免疫球蛋白Fc片段缀合物。
实验实施例1:受体内化的评估
使用eXpress Activated GPCR Internalization Assay评估受体内化。具体而言,将表达人GLP-1受体的U2OS细胞接种入白色的96孔板中并培养24-48小时。实施例的GLP-1激动剂-免疫球蛋白Fc片段缀合物从3μM被连续地4倍稀释,并且比较实施例的框内GLP-1激动剂-免疫球蛋白Fc片段缀合物从15μM被连续地4倍稀释,并将每个稀释物加入到孔板中。然后,在37℃下的CO2培养箱中培养3小时诱发每种缀合物的受体内化。然后,将用于检测内吞受体的底物加入到孔板中并且使得在室温下反应60分钟,并利用发光板阅读器测量板的发光。测量的结果在图2中示出。

实验实施例2:GLP-1激动剂-免疫球蛋白片段缀合物之间的体内药代动力学的比
较的测试
为了比较实施例的GLP-1激动剂-免疫球蛋白Fc片段缀合物和GLP-1激动剂-免疫球蛋白Fc片段缀合物之间的体内药代动力学,使用正常的SD大鼠分析缀合物的血清浓度的改变。
具体而言,将GLP-1激动剂-免疫球蛋白Fc片段缀合物(400mcg/kg)和GLP-1激动剂-免疫球蛋白Fc片段缀合物(400mcg/kg)的每个在生理盐水中稀释并以2mL/kg的剂量皮下施用给动物。在施用测试物质4、8、24、48、72、96、120、144、168、192、216、240、288、312和336小时之后,从大鼠的颈静脉采集血液,并从血液中分离血清。接下来,每个血清样品中的药物浓度通过酶联免疫吸附试验量化,并且量化结果在图3中示出。
结果,实施例的GLP-1激动剂-免疫球蛋白Fc片段缀合物和比较实施例的GLP-1激动剂-免疫球蛋白Fc片段缀合物的血清半衰期分别为40.9小时和28小时,并且缀合物的最大血清浓度分别为1758.6ng/mL和742.7ng/mL。换句话说,当药物以相同的剂量皮下施用给正常的大鼠时,显示了与二聚体GLP-1激动剂-免疫球蛋白Fc片段缀合物(图3)相比,本发明的单体GLP-1激动剂-免疫球蛋白Fc片段缀合物在体内吸收和半衰期方面是优异的。

Claims (10)
1.一种长效药物组合物,其包括缀合物,所述缀合物包括连接至免疫球蛋白Fc片段的生理活性的多肽,
其中所述组合物包括单体缀合物,所述单体缀合物包括经由具有两个末端的非肽基连接体连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的一个免疫球蛋白Fc链的一分子的所述生理活性的多肽,和二聚体缀合物,所述二聚体缀合物包括连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的两分子的相同的生理活性的多肽,条件是所述组合物中所述单体缀合物与所述二聚体缀合物的摩尔比为至少19;
其中二聚体缀合物中的两个免疫球蛋白Fc链的每个经由具有两个末端的非肽基连接体连接至一分子的生理活性多肽;
其中与二聚体缀合物——其包括经由非肽基连接体连接至单个免疫球蛋白Fc片段的两分子的所述生理活性的多肽,或缀合物——其包括框内连接至所述免疫球蛋白Fc片段的生理活性的多肽二聚体——相比,所述单体缀合物显示了降低的受体介导的内化或受体介导的清除率;和
其中所述生理活性的多肽为胰高血糖素样肽-1(GLP-1)或GLP-1激动剂;并且
其中所述非肽基连接体是聚乙二醇。
2.权利要求1所述的长效药物组合物,其中所述非肽基连接体具有从1到100kDa范围内的分子量。
3.权利要求1所述的长效药物组合物,其中所述免疫球蛋白Fc片段由选自CH1、CH2、CH3和CH4结构域的1到4个结构域构成。
4.权利要求1所述的长效药物组合物,其中所述免疫球蛋白Fc片段进一步包括铰链区。
5.权利要求1所述的长效药物组合物,其中所述免疫球蛋白Fc片段选自IgG、IgA、IgD、IgE和IgM。
6.权利要求1所述的长效药物组合物,其中所述免疫球蛋白Fc片段是IgG4 Fc片段。
7.权利要求1所述的长效药物组合物,其中所述免疫球蛋白Fc片段是非糖基化的。
8.一种制备权利要求1所述的长效药物组合物的方法,其包括:
(a)经由具有两个末端的非肽基连接体将生理活性的多肽连接至具有两个免疫球蛋白Fc链的免疫球蛋白Fc片段以制备生理活性的多肽-免疫球蛋白Fc片段缀合物的混合物;和
(b)自所述混合物分离生理活性的多肽单体-免疫球蛋白Fc片段缀合物,所述生理活性的多肽单体-免疫球蛋白Fc片段缀合物包括经由非肽基连接体连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的一个免疫球蛋白Fc链的一分子的所述生理活性的多肽。
9.权利要求8所述的方法,其中与二聚体缀合物——其包括经由所述非肽基连接体连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的两分子的所述生理活性的多肽,或经由所述非肽基连接体框内连接至具有两个免疫球蛋白Fc链的所述免疫球蛋白Fc片段的生理活性的多肽二聚体相比,所述长效药物组合物的缀合物显示了降低的受体介导的内化或受体介导的清除率,所述长效药物组合物的缀合物包括经由所述非肽基连接体连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的一个免疫球蛋白Fc链的一分子的所述生理活性的多肽。
10.单体缀合物用于制备长效药物组合物的用途,所述长效药物组合物显示降低的受体介导的内化或受体介导的清除率,
其中所述组合物包括单体缀合物,所述单体缀合物包括经由具有两个末端的非肽基连接体连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的一个免疫球蛋白Fc链的一分子的所述生理活性的多肽,和二聚体缀合物,所述二聚体缀合物包括连接至具有两个免疫球蛋白Fc链的单个免疫球蛋白Fc片段的两分子的相同的生理活性的多肽,条件是所述组合物中所述单体缀合物与所述二聚体缀合物的摩尔比为至少19;
其中二聚体缀合物中的两个免疫球蛋白Fc链的每个经由具有两个末端的非肽基连接体连接至一分子的生理活性多肽;
其中与二聚体缀合物——其包括经由非肽基连接体连接至单个免疫球蛋白Fc片段的两分子的所述生理活性的多肽,或缀合物——其包括框内连接至所述免疫球蛋白Fc片段的生理活性的多肽二聚体——相比,所述单体缀合物显示了降低的受体介导的内化或受体介导的清除率;并且
其中所述生理活性的多肽为胰高血糖素样肽-1(GLP-1)或GLP-1激动剂;并且
其中所述非肽基连接体是聚乙二醇。
