Abstract

本发明涉及一种番石榴多酚的制备方法,属于农业加工技术领域;该方法具体为:将新鲜的番石榴洗净后进行打浆,准确称取一定质量的番石榴浆加入浸提剂进行浸泡、回流浸提后离心,分离出上清液后,真空浓缩,最后采用冷冻干燥的方法制得番石榴多酚;该方法所得的具体参数为用体积分数70%的乙醇溶液,按1:11的料液比在pH3,70℃条件下回流浸提30min,浸提2次,本方法所得的番石榴多酚浸提率最高。

技术领域
本发明涉及一种多酚的制备方法,具体涉及一种番石榴多酚的制备方法,属于农业加工技术领域。
背景技术
番石榴是亚热带水果,性喜湿热,畏寒,在-1℃以下幼树冻死,成年树冻伤。番石榴适栽于海南及广西、广东、福建、云南南部。番石榴果实营养丰富,是保健饮料的重要原料。番石榴是夏秋季提供VC最廉价、最易得的大众化果品,有“热带苹果”之称。果实每千克含VC1250mg~2460mg,是柑枯的9倍,比香蕉、菠萝多30~80倍。鲜食营养易被人体吸收;加工成饮料,风味不变,VC大部分仍保留。目前两广、福建生产的番石榴汁,已远销西欧,很受欢迎。据相关报道,番石榴中也含有一定的酚类化合物。
酚类化合物是一类具有大而复杂基团的化合物,从化学上讲,酚是苯环(又称芳香环)上连有一个或多个羟基的化合物。多酚物质(po1ypheno1s)是含有酚官能基团的物质,是构成植物固体部分的主要物质,主要由无色多酚和色素两类物质组成。多酚物质具有特殊的生物活性,在医疗、保健等方面有重要的作用和利用价值,它一方面具有VP的某些性质及一定的杀菌能力,是强的抗自由基防衰老物质,其活性相当于VE的25倍;另一方面可防止视网膜黄斑病变的发生和发展。此外,多酚物质还是氧化还原的贮藏器———褐色物质的来源,可用于制作除臭剂、防锈剂、废水处理剂等。酚类化合物广泛存在于植物食品中,由于其羟基取代的高反应性和吞噬自由基的能力而有很好的抗氧化活性。此外,酚类化合物还具有预防高血压、抗动脉硬化、抗血栓、抗胃溃疡、抗菌、抗炎、抗肿瘤、抗突变的生物活性及皮肤保健及美容等功能。因此,提取番石榴中的多酚物质会在医药、保健品、食品和化妆品领域都具有广阔的应用前景。
多酚提取分离工艺有离子沉淀法、溶剂萃取法、吸附提取分离法、微波提取法、纸层析、薄层层析、柱层析、SFE、HPLC(高效液相色谱法)、GC、HPLC-MS、LC-ESI-MS等。离子沉淀法在茶多酚的提取分离上应用较多,有机溶剂萃取是利用多酚物质在不同溶剂中溶解度不同,将多酚与其他物质分离。
本发明以番石榴为原料,采用有机溶剂萃取的方法对其多酚类物质的提取条件进行了研究,为番石榴的开发利用提供新的价值。国内尚未见这方面的报道。
发明内容
本发明的目的在于提供一种番石榴多酚的制备方法,以番石榴为原料,采用有机溶剂萃取的方法对其多酚类物质的提取条件进行研究,为番石榴的开发利用提供新的价值。
为了达到上述目的,本发明所采用的技术方案是:
一种番石榴多酚的制备方法,具体步骤包括:
(1)原料的预处理:将新鲜无病虫害的番石榴洗净后进行打浆;
(2)溶剂浸泡:用70%的乙醇溶液进行浸泡;
(3)回流浸提:按1:11的料液比在pH3,70℃条件下回流浸提30min,浸提2次;
(4)离心:将浸提液置于离心机中3000r/min,离心20min,弃渣,收集上清液备用;
(5)真空浓缩:将上述上清液置于旋转蒸发仪中,设置真空度82.7~90.6KPa,温度45~55℃,将乙醇全部除去,水分去除60%~70%;
(6)冷冻干燥:将上述浓缩液进行预冷后,置于冷冻干燥机中真空冷冻干燥;
(7)包装、冷藏:将冷冻干燥所得番石榴多酚粉末采用避光材料进行密闭包装后,低温冷藏。
本发明具有如下优点:
(1)以番石榴为原料,采用有机溶剂萃取的方法对其多酚类物质的提取条件进行研究,为番石榴的开发利用提供新的价值。
(2)本方法不受无机物存在的影响,再生方便,解吸条件温和,而且操作简单,回收率高,安全可靠,对环境无污染,因此比较适合大规模的工业化生产。
具体实施方式
下面通过实施例对本发明做进一步详细说明,这些实施例仅用来说明本发明,并不限制本发明的范围。
实施例1
一种番石榴多酚的制备方法,具体步骤包括:
(1)原料的预处理:将新鲜无病虫害的番石榴洗净后进行打浆;
(2)溶剂浸泡:用70%的乙醇溶液进行浸泡;
(3)回流浸提:按1:11的料液比在pH3,70℃条件下回流浸提30min,浸提2次;
(4)离心:将浸提液置于离心机中3000r/min,离心20min,弃渣,收集上清液备用;
(5)真空浓缩:将上述上清液置于旋转蒸发仪中,设置真空度83KPa,温度45℃,将乙醇全部除去,水分去除60%;
(6)冷冻干燥:将上述浓缩液进行预冷后,置于冷冻干燥机中真空冷冻干燥;
(7)包装、冷藏:将冷冻干燥所得番石榴多酚粉末采用避光材料进行密闭包装后,低温冷藏。
实施例2
一种番石榴多酚的制备方法,其特征在于,具体步骤包括:
(1)原料的预处理:将新鲜无病虫害的番石榴洗净后进行打浆;
(2)溶剂浸泡:用70%的乙醇溶液进行浸泡;

(7)包装、冷藏:将冷冻干燥所得番石榴多酚粉末采用避光材料进行密闭包装后,低温冷藏。
实施例3
一种番石榴多酚的制备方法,其特征在于,具体步骤包括:
(1)原料的预处理:将新鲜无病虫害的番石榴洗净后进行打浆;
(2)溶剂浸泡:用70%的乙醇溶液进行浸泡;
(3)回流浸提:按1:11的料液比在pH3,70℃条件下回流浸提30min,浸提2次;

(7)包装、冷藏:将冷冻干燥所得番石榴多酚粉末采用避光材料进行密闭包装后,低温冷藏。
实施例4
一种番石榴多酚的制备方法,其特征在于,具体步骤包括:

(6)冷冻干燥:将上述浓缩液进行预冷后,置于冷冻干燥机中真空冷冻干燥;
(7)包装、冷藏:将冷冻干燥所得番石榴多酚粉末采用避光材料进行密闭包装后,低温冷藏。
