技术领域
本发明属于生物制剂技术领域,特别涉及一种减缓老年痴呆的蛹虫草葡萄酵素的制作方法。
背景技术
酵素又称为“酶”,是具有生物催化功能的高分子物质。几乎所有的细胞活动进程都需要酵素的参与。现代社会,随着人们生活节奏的加快和环境污染的加剧,人们体内酵素的含量日益减少,进而降低了人体免疫力和代谢速率,使人们患病的几率大增。目前,日常食品中的酵素含量太低,无法满足人们的正常需要。为了符合现代人对营养需要的多样性、平衡性和易于吸收性,高效的酵素产品的生产迫在眉睫。
蛹虫草具有和冬虫夏草相似的活性成分和药理作用,被称为可人工培养的“冬虫夏草”。2009年,蛹虫草被批准为新资源食品,且其价格远低于冬虫夏草,因而越来越受到广大消费者的青睐。中医认为,蛹虫草入肺肾二经,既补肺阴,又补肾阳,功能主治为补肺益肾,止咳化痰。现代医学研究其具有细胞免疫和体液免疫调节作用,还具有抗疲劳作用。因此,若能开发以蛹虫草为主要原料的产品,不仅能推动蛹虫草产业的进一步发展,更能为广大人民群众的健康带来福祉。尽管市场上已有蛹虫草相关的产品,但市场规模小,附加值低,功效不明确,亟需开发高效的、高附加值的蛹虫草系列产品。并且,现有的酵素减缓老年痴呆效果差。
发明内容
针对现有技术的不足,本发明提供一种减缓老年痴呆的蛹虫草葡萄酵素的制作方法。
本发明的技术方案是这样实现的:该方法包括如下步骤:
步骤1:制备酵母菌种液
种液罐扩大培养基:YPD液体培养基,其质量百分数为1%酵母提取物,2%蛋白胨,2%葡萄糖,2%琼脂,其余为水,将YPD液体培养基放入0.5M3种子罐内,在115℃下高温灭菌30-50分钟;
无菌操作条件下将1.5公斤酵母菌干菌粉溶于3倍的无菌水中,形成菌液,将所述的菌液装入血清瓶并接种到0.5M3种子罐,在28-30℃下通入无菌压缩空气,通气量为0.2-0.3立方米/分钟,培养20-30h,待菌体生长至对数期末期时停止培养,重复上述步骤可制得所需质量的酵母菌液的种液。
步骤2.制备乳酸菌种液
种液罐扩大培养基:麦芽复合汁增菌培养基,其10%麦芽汁,0.5%大豆蛋白胨,0.5%酵母膏,1%牛肉膏,0.2%K2HPO4,其余为水,将麦芽复合汁增菌培养基放入0.5M3种子罐内,在115℃下高温灭菌30-50分钟;
无菌操作条件下将1.5公斤乳酸菌干菌粉溶于3倍的无菌水中,菌液装入血清瓶并接种到0.5M3种子罐,37℃恒温静置培养24-36h,待菌体生长至对数期末期时停止培养,可重复上述步骤可制得所需乳酸菌的种液;
步骤3.发酵培养
将质量比为1.5:1:10的葡萄、苹果和水混合打碎后过滤得到滤液,用所述滤液制成培养基,在115℃下高温灭菌30-50分钟,将所述培养基加入发酵罐中,然后按质量为所述滤液的5%-10%的接种量接种步骤1制得的酵母菌的种液,在28℃的温度下培养48h,所述的发酵罐的体积为V立方米,通气量为(0.4-0.6)V立方米/分钟,搅拌转速100-120rpm/min;然后按质量为所述滤液的5%-10%的接种量接种步骤2制得的乳酸菌种液,在37℃的温度下继续培养5-7d至发酵结束,得到发酵液;
步骤4.离心处理,获取酵素原液
发酵液导入离心机,经6000rpm离心10min,所得上清液即为酵素原液;
步骤5、向酵素原液加入虫草素提取物,按照酵素原液与虫草素提取物质量比为1000:50,在115℃下高温灭菌30-50分钟,进行分装。
所述的乳酸菌为地衣芽孢杆菌,其质量为1.5公斤。

本发明延缓老年痴呆通过实验获得效果:
参加蛹虫草酵素服用的老年痴呆病患者有34人,每人每天早晚各服1次,每次200ml,90天为一个评测周期。使用本发明的酵素后,有效人数25人,均有不同程度的延缓效果,症状不再发展。其中7人,有一定恢复,且维持在较好状态,保持不变。

1.一种减缓老年痴呆的蛹虫草葡萄酵素的制作方法,其特征在于:包括如下步骤:
步骤1:制备酵母菌种液
种液罐扩大培养基:YPD液体培养基,其质量百分数为1%酵母提取物,2%蛋白胨,2%葡萄糖,2%琼脂,其余为水,将YPD液体培养基放入0.5M3种子罐内,在115℃下高温灭菌30-50分钟;
无菌操作条件下将1.5公斤酵母菌干菌粉溶于3倍的无菌水中,形成菌液,将所述的菌液装入血清瓶并接种到0.5M3种子罐,在28-30℃下通入无菌压缩空气,通气量为0.2-0.3立方米/分钟,培养20-30h,待菌体生长至对数期末期时停止培养,重复上述步骤可制得所需质量的酵母菌液的种液;
步骤2.制备乳酸菌种液
