本发明涉及中药领域,具体涉及一种骨刺片及其制备方法。
背景技术
骨刺片散风邪,祛寒湿,舒筋活血,通络止痛。用于颈椎、胸推、腰椎、跟骨等骨关节增生性疾病,对风湿、类风湿性关节炎有一定疗效市。售骨刺片由于组方和工艺原因,疗效不甚理想,制剂采用传统工艺制备,存在崩解迟缓、疗效低等不足。

发明实施方案如下:
取肉豆蔻77g,佩兰66g,阿魏22g,骨碎补100g,党参44g,桂枝20g,威灵仙111g,牡蛎(煅)50g,杜仲叶500g,鸡血藤111g,制川乌33g,延胡索(制)35g,三七18g,马钱子粉44g,粉碎成60目粗粉,采用二氧化碳超临界萃取法提取,萃取压力22~42Mpa,萃取温度23~44℃,分离器压力13~20Mpa,分离器温度40~51℃,分离时间2.7~3.9小时,二氧化碳流量每小时27~34L,得提取液;取提取液64~78℃减压干燥,得干膏;取干膏加入枸橼酸80~120g,采用高能纳米冲击磨粉碎成粒径200~300nm的混合干膏粉;取混合干膏粉,微晶纤维素35~45g,羟丙纤维素35~45g,交联羧甲基纤维素钠35~45g,硅酸镁铝15~25g,氯化钠5~15g,甘露醇4~6g,麦芽糊精4~6g,混合均匀,用50~70%乙醇湿法制粒,60℃~80℃干燥,外加海藻酸钠7~9g,硼酸1~3g,整粒,压片,制得骨刺片1300片。

肉豆蔻:中国药典2005年版一部标准。本品又名豆蔻、肉果、玉果。为肉豆蔻科植物肉豆蔻MyristicafragransHoutt的种仁。主产于马来西亚、印度尼西亚、斯里兰卡等国,热带地区广泛栽培。我国台湾、广东、云南等地有引种栽培。夏季采摘成熟果实,除去果皮,剥去假种皮,低温慢慢烘干。
佩兰:中国药典2005年版一部标准。本品为菊科植物佩兰EupatoriumfortuneiTurcz.的干燥地上部分。夏、秋二季分两次采割,除去杂质,晒干。
阿魏:中国药典2005年版一部标准。本品为伞形科植物新疆阿魏FerulaSINKIANGENSISK.M.Shen或阜康阿魏FerulafukanensisK.M.Shen的树脂。春末夏初盛花期至初果期,分次由茎上部往下斜割.收集渗出的乳状树脂,阴干。
骨碎补:中国药典2005年版一部标准。本品为水龙骨科植物槲蕨Drynariafortunei(Kunze)J.Sm.的干燥根茎。全年均可采挖,除去泥沙,干燥.或再燎去茸毛(鳞片)。
党参:中国药典2005年版一部标准。本品为桔梗科植物党参Codonopsispilosula(Franch.)Nannf.、素花党参CodonopsispilosulaNannf.var.modesta(Nannf.)LT.Shen或川党参CodonopsistangshenOliv.的干燥根。秋季采挖,洗净,晒干。
桂枝:中国药典2005年版一部标准。本品为樟科植物肉桂CinnamomumcassiaPresl的干燥嫩枝。春、夏二季采收,除去叶,晒干,或切片晒干。
威灵仙:中国药典2005年版一部标准。本品为毛茛科植物威灵仙ClematischinensisOsbeck、棉团铁线莲ClematishexapetalaPall.或东北铁线莲ClematismanshuricaRupr.的干燥根及根茎。秋季采挖.除去泥沙.晒干。
牡蛎:中国药典2005年版一部标准。本品为牡蛎科动物长牡蛎OstreagigasThunberg、大连湾牡蛎OstreatalienwhanensisCrosse或近江牡蛎Ostrearriv-ularisGould的贝壳。全年均可采收,去肉,洗净,晒干。
杜仲叶:中国药典2005年版一部标准。本品为杜仲科植物杜仲EucommiaulmoidesOliv.的干燥叶。夏、秋二季枝叶茂盛时采收,晒干或低温烘干。鸡血藤:中国药典2005年版一部标准。本品为豆科植物密花豆SpatholobussuberectusDunn的干燥藤茎。秋、冬二季采收,除去枝叶,切片,晒干。
制川乌:中国药典2005年版一部标准。本品为川乌的炮制加工品。
延胡索:中国药典2005年版一部标准。本品为罂粟科植物延胡索CorydalisyanhusnoW.T.Wang的干燥块茎。夏初茎叶枯萎时采挖,除去须根,洗净,置沸水中煮至恰无白心时,取出,晒干。
三七:中国药典2005年版一部标准。本品为五加科植物三七Punaxnotoginseng(Burk.)F.H.Chen的干燥根及根茎。秋季花开前采挖,洗净,分开主根、支根及根茎,干燥。支根习称“筋条”,根茎习称“剪口”。
马钱子粉:中国药典2005年版一部标准。本品为马钱子的炮制加工品。
石榴皮:中国药典2005年版一部标准。本品为石榴科植物石榴PunicagranatumL.的干燥果皮。秋季果实成熟后收集果皮,晒干。
枸橼酸:中国药典2010年版二部标准。
微晶纤维素:中国药典2010年版二部标准。
羟丙纤维素:中国药典2010年版二部标准。
交联羧甲基纤维素钠:中国药典2010年版二部标准。
硅酸镁铝:中国药典2010年版二部标准。

依据中药新药治疗临床研究指导原则中医证候疗效判定标准:
临床痊愈:中医临床症状、体征消失或基本消失,证候积分减少≥95%。
显效:中医临床症状、体征明显改善,证候积分减少≥70%。
有效:中医临床症状、体征均有好转,证候积分减少≥30%。
无效:中医临床症状、体征均均无明显改善或者加重,证候积分减少<30%。
计算公式:[(治疗前积分-治疗后积分)÷治疗前积分]×100%。

2.根据权利要求1所述中药的制备方法,其特征是取肉豆蔻77g,佩兰66g,阿魏22g,骨碎补100g,党参44g,桂枝20g,威灵仙111g,牡蛎(煅)50g,杜仲叶500g,鸡血藤111g,制川乌33g,延胡索(制)35g,三七18g,马钱子粉44g,粉碎成60目粗粉,采用二氧化碳超临界萃取法提取,萃取压力22~42Mpa,萃取温度23~44℃,分离器压力13~20Mpa,分离器温度40~51℃,分离时间2.7~3.9小时,二氧化碳流量每小时27~34L,得提取液;取提取液64~78℃减压干燥,得干膏;取干膏加入枸橼酸80~120g,采用高能纳米冲击磨粉碎成粒径200~300nm的混合干膏粉;取混合干膏粉,微晶纤维素35~45g,羟丙纤维素35~45g,交联羧甲基纤维素钠35~45g,硅酸镁铝15~25g,氯化钠5~15g,甘露醇4~6g,麦芽糊精4~6g,混合均匀,用50~70%乙醇湿法制粒,60℃~80℃干燥,外加海藻酸钠7~9g,硼酸1~3g,整粒,压片,制得骨刺片。
3.根据权利要求1所述中药的制备方法,其特征是取肉豆蔻77g,佩兰66g,阿魏22g,骨碎补100g,党参44g,桂枝20g,威灵仙111g,牡蛎(煅)50g,杜仲叶500g,鸡血藤111g,制川乌33g,延胡索(制)35g,三七18g,马钱子粉44g,粉碎成60目粗粉,采用二氧化碳超临界萃取法提取,萃取压力32Mpa,萃取温度34℃,分离器压力17Mpa,分离器温度45℃,分离时间3.3小时,二氧化碳流量每小时31L,得提取液;取提取液71℃减压干燥,得干膏;取干膏加入枸橼酸100g,采用高能纳米冲击磨粉碎成粒径200~300nm的混合干膏粉;取混合干膏粉,微晶纤维素40g,羟丙纤维素40g,交联羧甲基纤维素钠40g,硅酸镁铝20g,氯化钠10g,甘露醇5g,麦芽糊精5g,混合均匀,用60%乙醇湿法制粒,70℃干燥,外加海藻酸钠8g,硼酸2g,整粒,压片,制得骨刺片。
