本发明涉及一种具有抗肿瘤提高免疫功能的复合营养食品及其生产方法,属于营养食品加工领域。以甲鱼、灵芝、姬松茸、银耳、香菇、猴头菇、玉米胚芽、柠檬、红枣为原料,经超低温冷冻、破碎、变温变相复合浸提、分离纯化、杀菌等处理制备的营养食品。产品具有良好的适口性及特殊的营养作用,富含维生素、矿物质、食用菌活性多糖、多不饱和脂肪酸等功能成分,具有提高免疫功能、抗肿瘤的作用。特别适用肿瘤患者术后及放化疗患者的康复。本发明采用甲鱼全利用技术,辅以食用菌、玉米胚芽等原料,生产中无废弃物产生,原料利用率高,产品不含任何香精、色素、防腐剂、稳定剂等添加剂,无毒副作用,食用安全。

背景技术
肿瘤是一类严重危害人类生命健康的常见病,给人类带来了极大的痛苦和困扰。根据肿瘤对人体危害的大小及其生长特性分为良性肿瘤和恶性肿瘤两类,各种癌症即是恶性肿瘤,肺癌居全国恶性肿瘤发病率之首。中国肿瘤发病率多年持续上升,已成为一个必需高度重视的公共卫生问题甚至社会问题。根据全国肿瘤登记中心发布的《2012中国肿瘤登记年报》,每分钟就有6人确诊为癌症。我国居民因癌症死亡率为12.94%。肿瘤发病原因内、外因皆有。内因如精神因素、内分泌失调、免疫缺陷与遗传因素等。先天性免疫缺陷或长期应用免疫抑制药的人群中,肿瘤的发病率较高。外因有化学、物理、生物、营养等多种,外界致癌因素是引起癌症的重要刺激因素,大约80%~90%的癌症是由环境因素引起的。例如吸烟与被动吸烟可诱发肺癌;长期接触煤焦油或芳香胺或偶氮染料或亚硝胺类化合物等可诱发皮肤、泌尿道、呼吸道等部位癌变;长期放射线及紫外线可诱发皮肤癌或恶性黑色素瘤;饮食营养是维持生命、保持健康体魄的物质基础,在很大程度上饮食对机体的机能和状态有重要的影响,不科学不合理膳食可诱发肿瘤,核酸代谢异常、遗传物质发生突变往往是在正常细胞转化为肿瘤细胞的开始,而代谢作用的正常与否与饮食营养有密切关系,与饮食直接有关的肿瘤有胃癌、食管癌、肝癌、肠癌、乳腺癌,以上各种癌症患者死亡率约占全部恶性肿瘤45%。寄生虫与病毒也会诱发动物肿瘤;另外某些药物如各种抗肿瘤药特别是烷化剂,本身也具有致癌性;某些解热镇痛药、抗癫痫药、抗组胺药、激素类等也与癌症的病因有关。一般认为75%~90%的肿瘤是由环境因素所引起。
为治愈肿瘤患者,西医采用手术治疗、化学治疗、放射治疗、生物学治疗、基因治疗、介入治疗、细胞免疫疗法等多种方法进行直接治疗,各种方法优缺点并存。中医理论认为肿瘤发生发展的重要原因和病机是体质虚弱、正气不足、气血虚弱,导致脏腑功能失调,进而出现气滞、血瘀、湿聚、痰结等一系列病理变化,最终形成肿瘤。中医预防、治疗肿瘤的特色和优势是扶正培本、祛邪。采用具有滋阴、补气、补阳、养血、清热解毒、活血化瘀、软坚散结、化痰祛湿等功能的食材或药材进行辨证治疗,调整病人的阴阳平衡、提高病人的免疫能力,起到治疗肿瘤的效果。
免疫功能(immunologic function;immune function;immunological function;immunity)是指机体对疾病的抵抗能力。免疫功能是免疫系统根据免疫识别而发挥的作用。如果免疫功能低下,机体难以抵抗细菌、病毒、真菌等的致病微生物侵袭;不能及时清除体内衰老的死亡细胞;不能及时发现和消灭体内发生变异的细胞,无法防止恶性肿瘤的发生。有关研究表明甲鱼、灵芝、姬松茸、银耳、香菇、猴头菇、玉米胚芽、柠檬、红枣都有明显增强人体免疫力的作用。
发明内容
本发明提供一种具有抗肿瘤提高免疫功能的复合营养食品及其生产方法。本产品不添加任何食品添加剂;不含香精、色素、甜味剂及防腐剂。产品富含氨基酸、维生素、矿物质以及类黄酮、皂甙、生物碱、植物甾醇等功能成分及生物活性成分,可改善和提高机体免疫及抗肿瘤功能。产品生产过程无污染,无废渣、废汽及有害物质产生,实现绿色生产;产品风味纯正、品质优良,食用方便。
本发明采取的技术方案是,包括下列质量份的原料和步骤:
甲鱼颗粒料100份,红枣30~40份,香菇25~40份,银耳20~25份,姬松茸20~25份,猴头菇15~20份,蜂巢蜜10~30份,玉米胚芽10~18份,灵芝3~10份,柠檬片4~8份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于20~25℃纯净水中空腹饲养3~7d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的4~5倍,然后取出,保证甲鱼具有生命体征,活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍2~3min,然后将甲鱼置于质量浓度为55~65%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-60℃~-80℃超低温冰柜中冷冻处理3~5d,然后取出置于-2℃~0℃洁净条件下进行缓化松弛处理2~3d,然后对甲鱼胴体进行破解,摘除泄殖腔,-2℃~0℃条件下破碎成粒度为0.5mm~1mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-3℃~-5℃条件下分别干法破碎成粒度为60~80目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入8~18倍的纯净水,继续混合均匀,送入辊式研磨罐,于-2℃~0℃、24~48h的条件下进行动态研磨亲水相成分提取,研磨辊转速15~30转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在3℃~8℃、4~8h的条件下进行动态研磨亲脂相成分提取,研磨辊转速10~15转/min,得到的混合物3℃~8℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为3℃~5℃、10000~15000转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口。
本发明优点:本发明产品在生产过程中不添加任何食品添加剂;产品不含香精、色素、甜味剂及防腐剂。本发明甲鱼及其他原辅料的处理均避免高温操作,最大限度保留原料中热敏性功效成分及生物酶的生理活性。尤其对甲鱼的预处理具有独特的优势:空腹饲养的甲鱼采用食用酒精杀菌避免高温漂烫;空腹饲养的甲鱼采用酒精溶液闷灌,避免常规宰杀放血及甲鱼血的收集带来的繁琐,杀死甲鱼同时使其消化道被酒精充满,对产生异味的甲鱼消化道进行杀菌和除异味;甲鱼胴体进行超低温冷冻处理,然后在-2℃~0℃进行缓化松弛处理,较常规冷冻处理可进一步对其进行深层冷冻杀菌,同时避免酶、维生素、氨基酸等功效成分被破坏,在-2℃~0℃、2~3d的缓化松弛过程中促进自然酶促反应,使甲鱼中功效成分处于活跃状态,有利于浸提。
在功效成分浸提过程中利用原料的亲水性及亲油性调整浸提体系的极性,实现水溶性成分及油溶性成分的高效提取,避免采用化学试剂实现溶液极性的转变和调整,提高产物的食用安全性。
所用原料均为具有提高机体免疫力的作用的药食兼用原料,可改善和提高机体免疫及抗肿瘤功能,长期食用无任何毒副作用。
本发明产品经过长期食用证明具有显著的抗肿瘤提高免疫功能的功效。
所用原辅料具体如下:
甲鱼:学名鳖(AmydaSincnsis)。俗称水鱼、团鱼和王八等,卵生爬行水陆两栖变温动物,用肺呼吸,生活在江河、湖泊、池塘中。中国现存主要有中华鳖、山瑞鳖、斑鳖、鼋。而以中华鳖最为常见,除新疆、西藏和青海外,其它各省均产,以湖南、湖北、江西、安徽、江苏等省产量较高。中华鳖被列入《国家保护的有益的或者有重要经济、科学研究价值的陆生野生动物名录》。自古以来甲鱼就被人们视为滋补佳品。《随患居饮食谱》记载甲鱼甘平,滋肝肾之阴,清虚劳之热,宜蒸煮食之。《名医别录》记载甲鱼的肉具有补中益气之功效。《本草纲目》记载甲鱼肉有滋阴补肾,清热消淤,健脾健胃、疗痨伤、壮阳气等多种功效,可治虚劳盗汗,阴虚阳亢,腰酸腿疼,久病泄泻,小儿惊痫,妇女闭经、难产等症。《日用本草》认为,甲鱼的血外敷能治面神经,可除中风口渴,虚劳潮热,并可治疗骨结核。
相关研究表明,甲鱼富含蛋白质、无机盐、维生素A、维生素B1、维生素B2、烟酸、碳水化合物、脂肪,以及胶原、肽类、多种酶类、微量元素等人体必需的多种营养素。据分析,每百克鲜甲鱼肉含水分73~83g、蛋白质15.3~17.3g、脂肪0.1~3.5g、碳水化合物1.6~1.49g、灰分0.9~1g、镁3.9mg、钙1~107mg、铁1.4~4.3mg、磷0.54~430mg、维生素A 13~20国际单位、维生素B 10.02mg、维生素B2 0.037~0.047mg、尼克酸3.7~7mg、硫胺素0.62mg、核黄素0.37mg、热量288~744千焦耳。另外甲鱼的脂肪以不饱和脂肪酸为主,约占总脂肪的75.43%,其中多不饱和脂肪酸占32.4%,是牛肉的6.54倍,铁等微量元素是其他食品的几倍甚至几十倍。
甲鱼的头、甲、骨、肉、卵、胆、脂肪均可入药。甲鱼肉不但是美味佳肴,而且是一种用途很广的滋补药品和中药材料,甲鱼肉性平、味甘,归肝经,具有滋阴凉血、补益调中、补肾健骨、清热养阴、平肝熄风、软坚散结的功效。常食甲鱼能够增强机体抗病能力、调节内分泌、防治身虚体弱、肝脾肿大、肺结核等症。甲鱼的背甲称为鳖甲,味咸、性寒,含骨胶原、氨基酸、微量元素等成分,具有滋阴清热、潜阳熄风、软坚散结等功能,主治阴虚发热、劳热骨蒸、胁下坚硬、腰痛、经闭症、惊痫等病症。现代医学研究表明鳖甲具有抑制结缔组织的增生、提高血浆蛋白、散结消肿的作用,可提高抗肿瘤的免疫能力;鳖甲熬制的鳖甲胶,具有滋阴益肾、散消痞、强筋健骨的功效,能够散淤血、消脾肿、除痨、调月经,可防治肾亏虚弱、头晕遗精等症。甲鱼血含有动物胶、角蛋白、碘和维生素D等成份,味甘、咸,性平,归肝经,具有滋阴清热、活血通络、补血、消肿、平肝火功效,主治虚劳潮热、阴虚低热、胁痛、口眼歪斜、脱肛、肝硬化、肝脾肿大、、闭经、经漏和小儿尺癫等症。甲鱼的胆可治痔漏。甲鱼卵味咸,性寒,主治久泻久痢。另外,甲鱼卵盐藏煨食,止小儿下痢。甲鱼卵盐淹煮吞,补阴虚。甲鱼头味甘、微涩,性平,入脾、大肠经。甲鱼头焙干研末,黄酒冲服,治久痢脱肛、产后子宫下垂、阴疮。甲鱼脂肪味甘、咸,性平,归肝、肾经,可滋阴养阳,主治体弱虚赢、须发早白。现代科学认为,甲鱼富含维生素A、维生素E、胶原蛋白和多种氨基酸、不饱和脂肪酸、微量元素,具有填精补肾、提高人体免疫功能,促进新陈代谢,增强人体的抗病能力,养颜美容,延缓衰老等的作用。尤其对癌症术后或放化疗后患者具有解毒、扶正、抗癌功效。
红枣:红枣又称大枣、干枣、枣子等。红枣味甘性温、归脾胃经,有补中益气、养血安神、生津解药毒、缓和药性、促进机体复原、降低胆固醇、升高白血球、抗过敏等的功能。自古以来红枣就作为一种营养佳品深受人们喜爱。红枣含有蛋白质、脂肪、糖类、有机酸、维生素A、维生素C、维生素B1、维生素B2、维生素P(芦丁)等多种维生素和胡萝卜素、类胡萝卜素,以及钙、磷、铁、镁等多种矿物质和丰富氨基酸、环磷酸腺苷等的营养成分。其中维生素C的含量是葡萄、苹果的70~80倍,被称为“天然VC丸”。相关研究结果已证明维生素C及维生素P具有预防癌症和高血压、高血脂的作用。现代营养学及医学研究结果表明红枣提取物具有能提高人体免疫力、增强抗病能力、抑制癌细胞、促进白细胞的生成、降低血清胆固醇、提高血清白蛋白、保护肝脏等作用。动物实验结果表欧明红枣多糖能显著提高免疫抑制小鼠免疫功能,升高血清中促进造血及兴奋免疫相关细胞因子的水平,改善胸腺及脾脏组织的组织形态、超微结构和能量代谢水平,使胸腺、脾脏、骨髓中兴奋造血与免疫的细胞因子基因表达上调、抑制免疫的细胞因子基因表达下调。红枣中含有的三萜类化合物及环磷酸腺苷,具有较强的抑癌、抗过敏作用。
香菇(Lentinula edodes Pegle或Lentinula edodes Sing):又名香蕈、冬菇,是著名的食药用菌之一。传统医学认为香菇味甘、性平。具有补气健脾、和胃益肾、助食之功效。香菇每100克食用部分中含水13克、脂肪1.8克、碳水化合物54克、粗纤维7.8克、灰分4.9克、钙124毫克、磷415毫克、铁25.3毫克、维生素B1 0.07毫克、维生素B2 1.13毫克、尼克酸18.9毫克。香菇含丰富的维生素D原。相关研究结果表明香菇中含有的香菇多糖及糖肽具有促进淋巴细胞产生活化因子、增强腹腔巨噬细胞吞噬率、提高机体免疫力、抗病毒、抗癌抑癌、保肝、促进病体康复等功效。有关研究结果表明香菇多糖能促进辅助性T淋巴细胞的产生,并提高T淋巴细胞的杀伤活性,增强人体体液免疫功能。香菇多糖还可提高小鼠腹腔巨噬细胞的吞噬功能。香菇含有双链结构的核糖核酸,进入人体后,会产生具有抗癌作用的干扰素,具有抗病毒能力。香菇对小白鼠肉瘤180的抑制率为97.5%,对艾氏癌的抑制率为80%。
银耳:别名白木耳、白耳、桑鹅、白耳子。银耳性平,味甘、淡,无毒。归肺、胃、肾经。能养肺阴、济肾燥、提神益气。具有滋补生津、强精补肾、养阴清热润肺、养胃清肠润燥、补气和血、强心壮身、补脑提神、美容嫩肤等功效。主治虚劳咳嗽、痰中带血、津少口渴、病后体虚、气短乏力等症。不但能增强机体抗肿瘤的免疫能力,还能增强肿瘤患者对放疗、化疗的耐受力。银耳还具有缓解肌肉疲劳、防止放射性损伤、增加机体免疫力等作用。
银耳蛋白质中含有17种氨基酸,人体所必需的氨基酸中的3/4银耳都能提供。银耳还含有多种矿物质,如钙、磷、铁、钾、钠、镁、硫等,其中钙、铁的含量很高,在每百克银耳中,含钙643毫克,铁30.4毫克。此外,银耳中还含有海藻糖、多缩戊糖、甘露糖醇等肝糖。银耳富含的天然植物性胶质,对消化道中异物具有较强的吸附作用。银耳中的酸性多糖类物质,能增强人体的免疫力,调动淋巴细胞,加强白细胞的吞噬能力,兴奋骨髓造血功能,具有抗肿瘤作用。银耳中的膳食纤维可助胃肠蠕动,减少脂肪吸收,从而达到减肥的效果。银耳还能增强肿瘤患者对放疗、化疗的耐受力。
姬松茸:姬松茸(Agaricus blazei murrill,又名巴西蘑菇),原产巴西、秘鲁。可食部分每100克干品中含粗蛋白40~45克、可溶性糖类38~45克、粗纤维6~8克、脂肪3~4克、灰分5~7克。姬松茸不但含有丰富的蛋白质,而且含有多种维生素及钾、磷、镁、钙、钠、铜、硼、锌、硒、铁、锰、钼、锗等多种矿质元素和麦角甾醇。姬松茸蛋白质组成中含18种氨基酸,包括人体所需8种必需氨基酸。姬松茸所含的甘露聚糖对抑制肿瘤(尤其是腹水癌)、增强精力、防治心血管病等都有疗效。姬松茸含丰富硒、核酸、外源凝集素、甾醇和脂肪酸和活性多糖等抗肿瘤成分。姬松茸含丰富多糖类和固醇类成分能有效抑制肿瘤细胞的生长,具有抗肿瘤作用,对致癌物质有吸收、排泄作用。尤其是松茸活性多糖体含量高达6.55g/100g,是灵芝的5倍。独特的β-(1-6)-D-葡萄糖结构直接作用激活机体免疫功能、提高免疫力,可快速提升自然免疫细胞的生长活性,辅助提高治疗效果。在增强免疫系统功能、抑制肿瘤效果方面姬松茸位居各药用真菌之首。有关研究表明结果姬松茸性平、味甘,归心、肺、肝、肾经,具有健脑、消炎、益肾、降血糖、改善糖尿病、降胆固醇、增强精力、改善动脉硬化、防治心血管病等功效。近来研究结果发现姬松茸还可促进人体骨髓的造血功能,干扰白血病细胞增殖,通过改善化疗对骨髓造血功能的抑制,使外周血红蛋白浓度、血小板及白细胞总数趋向正常值;有力抵抗滤过性病毒,防止病毒和有害物体进入人体危害脆弱的组织,大幅度提高人体免疫功能;阻挠和干扰病毒生长;保护肝、肾的正常生理功能。
猴头菇:又叫猴头菌,只因外形酷似猴头而得名,学名Hericium erinaceus,英文名Lion's Mane Mushroom、Bearded Tooth Mushroom。猴头菇为齿菌科植物猴头的子实体,猴头菌的营养价值很高,干品中每百克含蛋白质26.3克,脂肪4.2克,及各种维生素和无机盐。猴头菇含有17种氨基酸,包括人体所需8种必需氨基酸。传统医学认为猴头菇性平、味甘。具有利五脏、助消化、健胃、补虚、抗癌、益肾精之功效。主治食少便溏、胃及十二指肠溃疡、浅表性胃炎、神经衰弱、食道癌、胃癌、眩晕等病症,对肠癌有辅助治疗作用,对皮肤及肌肉癌肿抗癌功效明显。
猴头菇含挥发油、蛋白质、多糖类、氨基酸等成分。动物实验结果表明猴头菇提取物对小鼠肉瘤180有明显抑制作用。体外对艾氏腹水癌细胞有抑制作用,能抑制其脱氧核酸的合成,阻止胸腺嘧啶脱氧核糖核苷和尿嘧啶核苷酸的掺入。
研究表明猴头菇有增进食欲,增强胃粘膜屏障机能,提高淋巴细胞转化率,提升白细胞等作用,可以提高人体对疾病的抵抗能力。
蜂巢蜜:俗称“蜂窝”,是经蜜蜂酿制成熟的蜂蜜并封上蜡盖的蜂巢,营养成份和活性物质比普通蜂蜜高。由于蜂巢蜜未经人为加工,羟甲基糠醛、重金属含量低,含有丰富的生物酶、维生素、多种微量元素,同时集蜂胶,蜂王浆,花粉,蜂蜜,幼蜂为一体,具有更好的保健治病功效。蜂巢蜜不含王浆,所以不含激素。相关研究结果表明巢蜜具有高热能、抗菌消炎、养肺润肠、解毒护肝、益脾养肾、造血、改善睡眠、促进组织再生、促进儿童生长发育、提升人体免疫能力等功效。
灵芝:学名Ganoderma Lucidum Karst,为多孔蓖科真菌灵芝的子实体。灵芝药用在我国已有2000多年的历史,被历代医药家视为滋补强壮、扶正固本的神奇珍品,具有补气安神,止咳平喘的功效,用于眩晕不眠,心悸气短,虚劳咳喘。《神农本草经》记载“赤芝,味苦平。主胸中结,益心气,补中,增慧智,不忘。久食,轻身不老,延年神仙。一名丹芝。”
主要含有氨基酸、多肽、蛋白质、真菌溶菌酶(fungal lysozyme),以及糖类(还原糖和多糖)、麦角甾醇、三萜类、香豆精甙、挥发油、硬脂酸、苯甲酸、生物碱、维生素B2及C等。灵芝孢子还含甘露醇、海藻糖(trehalose)。经过大量临床研究,灵芝对神经衰弱、高脂血症、冠心病心绞痛、心律失常、克山病、高原不适症、肝炎、出血热、消化不良、气管炎等具有疗效。现代医药学研究结果表明灵芝对多种理化及生物因素引起的肝损伤有保护作用,保肝解毒;灵芝子实体提取物主要为多糖和蛋白质,对小鼠S180抑制率达87.6%,具有抗肿瘤作用;由灵芝分离出的蛋白多糖腹腔注射可使小鼠腹腔渗出液中的细胞、巨噬细胞、多形核白细胞增加,表明其有免疫加强作用。对绵羊红细胞(SRBC)免疫小鼠的脾脏溶血空斑形成细胞数明显增加,说明其免疫调节作用。
玉米胚芽:玉米胚芽含丰富蛋白质、脂肪、维生素、植物甾醇等营养物质,其中蛋白质含量在15%左右,而且主要为水溶性蛋白质,氨基酸组成于鸡蛋白蛋白质组成接近,是营养价值较高的优质蛋白。玉米胚芽含脂肪脂肪在45%以上,其中85%是以油酸和亚油酸为主的不饱和脂肪酸。其中亚油酸是人体必需脂肪酸,是构成人体细胞的组成部分,在人体内可与胆固醇相结合,呈流动性正常代谢,有防治动脉粥样硬化等心血管疾病的功效。具有防治干眼病、夜盲症、皮炎、支气管扩张等多种功能,并具有一定的抗癌作用。
柠檬:芸香科柑桔属的常绿小乔木柠檬(Citrun Limonia Osbeck)的果实。柠檬富含柠檬酸、苹果酸、糖类、橙皮甙、柚皮甙、维生素C、维生素B1、维生素B2、烟酸、钙、磷、铁等多种营养成分。柠檬味酸、微甘,性平微寒,入肝、胃经。具有化痰止咳,生津,健脾、利肝、抗硬化、抗坏血、抗菌杀菌、增加免疫力等功效。主治支气管炎、维生素C缺乏症、烦渴、食欲不振、纳减等症。
现代医学、营养学研究表明柠檬是高度碱性食品,具有很强的抗氧化作用,具有促进机体的新陈代谢、促进肝排毒、延缓衰老、清除自由基、抑制皮肤色素沉着等作用。

具体实施方式
实施例1
包括下列质量份的原料和步骤:
甲鱼颗粒料100份,红枣30份,香菇25份,银耳20份,姬松茸20份,猴头菇15份,蜂巢蜜10份,玉米胚芽10份,灵芝3份,柠檬片4份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于20℃纯净水中空腹饲养3d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的4倍,然后取出,保证甲鱼具有生命体征,死亡甲鱼不可应用。活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍2min,进行表皮杀菌处理,注意保持甲鱼头在酒精溶液外面;然后将甲鱼置于质量浓度为55%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-60℃超低温冰柜中冷冻处理3d,然后取出置于-2℃洁净条件下进行缓化松弛处理2d,然后对甲鱼胴体进行破解,摘除泄殖腔,-2℃条件下破碎成粒度为0.5mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-3℃条件下分别干法破碎成粒度为60目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入8倍的纯净水,继续混合均匀,送入辊式研磨罐,于-2℃、24h的条件下进行动态研磨亲水相成分提取,研磨辊转速15转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在3℃、4h的条件下进行动态研磨亲脂相成分提取,研磨辊转速10转/min,得到的混合物3℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为3℃、10000转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口,即为具有抗肿瘤提高免疫功能的营养食品。
实施例2
包括下列质量份的原料和步骤:
甲鱼颗粒料100份,红枣40份,香菇40份,银耳25份,姬松茸25份,猴头菇20份,蜂巢蜜30份,玉米胚芽18份,灵芝10份,柠檬片8份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于25℃纯净水中空腹饲养7d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的5倍,然后取出,保证甲鱼具有生命体征,死亡甲鱼不可应用,活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍3min,进行表皮杀菌处理,注意保持甲鱼头在酒精溶液外面。然后将甲鱼置于质量浓度为65%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-80℃超低温冰柜中冷冻处理5d,然后取出置于0℃洁净条件下进行缓化松弛处理3d,然后对甲鱼胴体进行破解,摘除泄殖腔,0℃条件下破碎成粒度为1mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-5℃条件下分别干法破碎成粒度为80目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入18倍的纯净水,继续混合均匀,送入辊式研磨罐,于0℃、48h的条件下进行动态研磨亲水相成分提取,研磨辊转速30转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在8℃、8h的条件下进行动态研磨亲脂相成分提取,研磨辊转速15转/min,得到的混合物8℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为5℃、15000转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口,即为具有抗肿瘤提高免疫功能的营养食品。
实施例3
包括下列质量份的原料和步骤:
甲鱼颗粒料100份,红枣35份,香菇32.5份,银耳22.5份,姬松茸22.5份,猴头菇17.5份,蜂巢蜜20份,玉米胚芽14份,灵芝6.5份,柠檬片6份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于22.5℃纯净水中空腹饲养5d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的4.5倍,然后取出,保证甲鱼具有生命体征,死亡甲鱼不可应用,活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍2.5min,进行表皮杀菌处理,注意保持甲鱼头在酒精溶液外面。然后将甲鱼置于质量浓度为60%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-70℃超低温冰柜中冷冻处理4d,然后取出置于-1℃洁净条件下进行缓化松弛处理2.5d,然后对甲鱼胴体进行破解,摘除泄殖腔,-1℃条件下破碎成粒度为0.8mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-4℃条件下分别干法破碎成粒度为80目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入13倍的纯净水,继续混合均匀,送入辊式研磨罐,于-1℃、36h的条件下进行动态研磨亲水相成分提取,研磨辊转速22转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在5.5℃、6h的条件下进行动态研磨亲脂相成分提取,研磨辊转速12转/min,得到的混合物5.5℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为4℃、12500转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口,即为具有抗肿瘤提高免疫功能的营养食品。
下边通过实验进一步说明本发明产品具有显著的抗肿瘤、提高免疫功能的功效。
实验例1、本发明复合营养食品的抗肿瘤作用研究,
1、以16~19g的雄性小鼠为实验对象,分别接种肉瘤S180瘤株和艾氏腹水癌瘤株,实验组饲以本发明复合营养食品,以基础饲料组为阴性对照组,结果发现本发明复合营养食品对肉瘤S180瘤株引发的实体瘤,艾氏腹水癌瘤株引发的实体瘤和腹水瘤均有抑制作用。具体实验如下:
(1)本发明复合营养食品对小鼠艾氏腹水癌的抑制实验:将雄性昆明种小鼠36只,体重16~19g,随机分为3组,每组12只,阴性对照组、本发明复合营养食品实验组0.6ml/只·d剂量组(实验组A)及0.3ml/只·d剂量组(实验组B)。在接种瘤株之前,实验组动物以本发明复合营养食品灌胃3天。艾氏腹水癌瘤株以每只鼠5xl06个瘤细胞接种于右前肢腋下。第2d开始,阴性对照组继续自由摄食摄水,实验动物组自由摄食摄水的同时继续按上述剂量以本发明复合营养食品灌胃喂饲。实验时间为12天。第12天停止供食供水,称体重,按常规方法处死,取肿瘤并称重,计算肿瘤抑制率。
参照我国抗肿瘤药物的评价标准“当肿瘤抑制率>30%,经统计学处理有显著性差异(P<0.05),则认为该药物对肿瘤有一定疗效”的依据。本发明复合营养食品对肿瘤抑制率在30%以上,与阴性对照组比较,有极显著性差异(P<0.01)。说明本发明复合营养食品对艾氏腹水癌瘤株有抑制作用。

自然杀伤细胞(natural killer cell,NK)是机体重要的免疫细胞,与抗肿瘤、抗病毒感染和免疫调节有关,可抗病毒性感染,非特异性地杀伤肿瘤细胞,对T淋巴细胞、B淋巴细胞和骨髓干细胞等功能具有调节作用,还可通过释放淋巴因子对免疫应答进行调节。白细胞介素(interleukin,IL)是由多种细胞产生并作用于多种细胞的一类细胞因子。它和血细胞生长因子相互协调,相互作用,共同完成造血和免疫调节功能。IL-2属于淋巴细胞活素,可用于病毒感染、免疫缺陷病及自身免疫病的治疗,具有促进T淋巴细胞分化、增殖,增强其杀伤作用,促进NK细胞增殖,增强NK细胞活性,促进B淋巴细胞和ADCC(antibody-dependent cell-mediated cytotoxicity,抗体依赖的细胞介导细胞毒性作用)细胞的增殖及多种免疫功能。
(1)方法
Wistar雄性高龄大鼠36只,随机分成3组,每组12只,对照组、实验B组(每天灌胃饲喂本发明复合营养食品2ml/只鼠剂量组)、实验C组(每天灌胃饲喂本发明复合营养食品1ml/只鼠剂量组)。各组大鼠单笼饲养,实验时间60d,第61d按常规方法处死,无菌条件下,取大鼠脾脏、分离单个核细胞。采用乳酸脱氢酶法和MTT比色法(MTT学名3-(4,5-二甲基噻唑-2)-2,5-二苯基四氮唑溴盐,商品名噻唑蓝。)分别测定大鼠脾细胞NK细胞活性和IL-2的诱生能力。
(2)结果
实验结果表明本发明复合营养食品对大鼠细胞NK细胞活性明显高于对照组(P<0.01),有增强机体NK细胞活性的作用。

表4结果表明,实验A组(高剂量组)大鼠的IL—2活性明显高于对照组(P<0.05),实验B组(低剂量组)大鼠的IL—2活性无明显变化,表明一定剂量的本发明复合营养食品能促进机体IL—2的诱生能力。

实验结果表明饲以本发明复合营养食品的大鼠与机体免疫力关系密切的自然杀伤(NK)细胞活性和白细胞介素2(IL—2)的诱生能力明显高于对照组。
Claims (2)
1.一种具有抗肿瘤提高免疫功能的复合营养食品,其特征在于是由下列质量份的原料和步骤制成的:
甲鱼颗粒料100份,红枣30~40份,香菇25~40份,银耳20~25份,姬松茸20~25份,猴头菇15~20份,蜂巢蜜10~30份,玉米胚芽10~18份,灵芝3~10份,柠檬片4~8份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于20~25℃纯净水中空腹饲养3~7d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的4~5倍,然后取出,保证甲鱼具有生命体征,活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍2~3min,然后将甲鱼置于质量浓度为55~65%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-60℃~-80℃超低温冰柜中冷冻处理3~5d,然后取出置于-2℃~0℃洁净条件下进行缓化松弛处理2~3d,然后对甲鱼胴体进行破解,摘除泄殖腔,-2℃~0℃条件下破碎成粒度为0.5mm~1mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-3℃~-5℃条件下分别干法破碎成粒度为60~80目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入8~18倍的纯净水,继续混合均匀,送入辊式研磨罐,于-2℃~0℃、24~48h的条件下进行动态研磨亲水相成分提取,研磨辊转速15~30转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在3℃~8℃、4~8h的条件下进行动态研磨亲脂相成分提取,研磨辊转速10~15转/min,得到的混合物3℃~8℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为3℃~5℃、10000~15000转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口。
2.如权利要求1所述的一种具有抗肿瘤提高免疫功能的复合营养食品的生产方法,其特征在于是由下列质量份的原料和步骤制成的:
甲鱼颗粒料100份,红枣30~40份,香菇25~40份,银耳20~25份,姬松茸20~25份,猴头菇15~20份,蜂巢蜜10~30份,玉米胚芽10~18份,灵芝3~10份,柠檬片4~8份;
所述甲鱼颗粒料的制备方法如下:将鲜活甲鱼置于20~25℃纯净水中空腹饲养3~7d,水需保持清澈洁净,每天更换一次,用水量为甲鱼质量的4~5倍,然后取出,保证甲鱼具有生命体征,活甲鱼用洁净水喷淋洗净表面,悬挂,自颈部以下浸入质量浓度为75%的食用酒精溶液中浸渍2~3min,然后将甲鱼置于质量浓度为55~65%的食用酒精溶液中,在酒精液面加盖密闭,直至甲鱼被闷灌死亡,将死亡的甲鱼取出,置于-60℃~-80℃超低温冰柜中冷冻处理3~5d,然后取出置于-2℃~0℃洁净条件下进行缓化松弛处理2~3d,然后对甲鱼胴体进行破解,摘除泄殖腔,-2℃~0℃条件下破碎成粒度为0.5mm~1mm的甲鱼颗粒料;
步骤如下:
(一)食用菌、玉米胚芽、柠檬片、红枣的低温破碎
按配方比例进行准确称量:干燥的、去根、免洗免摘的洁净的灵芝、姬松茸、银耳、香菇、猴头菇以及干燥的玉米胚芽、柠檬片、红枣,并于-3℃~-5℃条件下分别干法破碎成粒度为60~80目的粉末,备用;
(二)变温变相复合浸提
将除玉米胚芽粉外的所有原料混合均匀,按其质量比加入8~18倍的纯净水,继续混合均匀,送入辊式研磨罐,于-2℃~0℃、24~48h的条件下进行动态研磨亲水相成分提取,研磨辊转速15~30转/min,然后加入玉米胚芽粉,利用玉米胚芽中的脂肪调整浸提体系溶解极性,在3℃~8℃、4~8h的条件下进行动态研磨亲脂相成分提取,研磨辊转速10~15转/min,得到的混合物3℃~8℃无菌条件下贮存备用;
(三)分离纯化、杀菌
将经步骤(二)得到的混合物进行高速低温分离纯化,分离条件为3℃~5℃、10000~15000转/min,分离液在135℃、4s条件下进行超高温瞬时杀菌,控制物料出口温度35℃,进行无菌罐装封口。
