Abstract
本发明公开了一种含蒜素的复方制品及其制备方法,主要原料为大蒜、人参、山楂、枸杞子、猴头菇和茯苓。先利用纤维素酶分解及软化纤维素,破坏细胞壁,促使有效成分充分释放;然后采用乙醇和水提取;最后将滤液经系列工艺制成复方制品。本发明产品选用传统天然植物进行配伍,无毒副作用,长期服用具有良好的提高免疫功能和消除自由基作用;本方法集酶解、醇提和水提于一体,比传统的溶剂提取方法提取率更高,比单一的提取方法成分更全面,制成的含蒜素的复方制品营养更充分、食用更便捷。

技术领域
本发明属于保健食品和饮料领域,具体涉及一种含蒜素的复方制品及其制备方法。
背景技术
大蒜(学名Allium sativum L.):大蒜素是以大蒜Allium Sativum L.鳞茎为原料提取的产品。是从葱科葱属植物(Allium Sativum)的鳞茎(大蒜头)中提取的一种有机硫化合物,学名二烯丙基硫代亚磺酸酯。大蒜素 Allicin;Diallyldisulfid-S-oxide;Diallydisulfide。别名:蒜素、蒜辣素、大蒜辣素,分子式:C6H10S2O,分子量:162,化学名:二烯丙基二硫醚(CH2=CH-CH2-S(O)-S-CH2-CH=CH2)。大蒜具有强烈的大蒜臭,味辣。大蒜素抗菌作用:可抑制痢疾杆菌、伤寒杆菌繁殖,对葡萄球菌、肺炎球菌等有明显的抑制灭杀作用。临床上口服大蒜素可治疗动物肠炎、下痢、食欲不振等。大蒜素具有降低胃内亚硝酸盐含量和抑制硝酸盐还原菌的作用。用大蒜素治疗萎缩性胃炎患者,服药1 个月后胃液pH值降低,酸度增加,且降低胃液内亚硝酸盐含量。大蒜素用于治疗慢性胃病,对胃区不适、饱胀、隐痛、返酸、嗳气、烧灼及食欲不振等症状者有明显改善,提示大蒜素对慢性胃病有一定治疗作用,大蒜素对心血管的作用是通过降低血浆总胆固醇、降血压、抑制血小板活性、降低红细胞压积、降低血液粘度等作用。有实验证实大蒜素具有预防胃癌发生的作用,其对胃液分离出的硝酸盐还原菌的生长,及其产生亚硝酸盐的能力均有明显的抑制作用,可降低人体胃液中亚硝酸盐含量,从而降低患胃癌的风险。主要作用如下:1、抑菌杀菌。对大肠杆菌、沙门氏菌、金黄色葡萄球菌、痢疾杆菌、伤寒杆菌、肺炎球菌,链球菌等有害菌有明显抑制和杀灭作用,对有益菌如干酪乳杆菌则无抑制作用。2、诱食增食。散发独特的大蒜香味,使动物产生食欲感,使之迅速摄食;同时摄取后可增强胃液分泌和胃肠蠕动,促进消化,从而促进动物生长,显著提高饲料报酬。3、解毒保健。可显著降低汞、氰化物、亚硝酸盐等有害物的毒性。促进体质健壮,增强抗病力,具有理想的保健功能。4、对各种感污引起、赤皮、肠炎、出血等疾病的治疗有特效。5、降低胆固醇。降低7a-胆固醇羟化酶的活性,使血清、蛋黄和肝中的胆固醇含量下降。6、无毒、无副作用,无药物残留,无耐药性,是替代抗生素最佳物质。
人参(学名:PanaxginsengC.A.Mey.)对强身健体、益寿延年的具有效果。经现代医学研究及化验分析表明,人参内含有一种叫人参皂甙的化学物质,它对调节人的中枢神经系统、强心、抗疲劳、调节物质代谢等有明显功效,所以对治疗神经系统.心血管系统、内分泌系统及生殖系统的多种疾病有很好的治疗作用。
山楂(学名:Crataegus pinnatifida.)的主要成分是黄酮类物质,对心血管系统有明显的药理作用。目前,从山楂中分离的黄酮成分有30 余种,主要有含碳键的黄酮苷类、黄酮醇及其苷类、双氧黄酮苷类、聚合黄酮类。另一类较为重要的成分是三萜类物质,有强心、增加冠脉血流、改善血流循环等重要作用。
枸杞子(学名:LyciumbarbarumL.):果实含枸杞多糖,有丰富的胡萝卜素、维生素A、B1、B2、C和钙、铁等眼睛保健的必需营养,故擅长明目。枸杞有提高机体免疫力的作用,可以补气强精、滋补肝肾、抗衰老、强身健体的作用。枸杞具有降低血压,血脂和血糖的作用,能防止动脉结业证样硬化,保护肝脏,抑制脂肪肝,促进肝细胞再生。
猴头菇(学名:Hericium erinaceus):猴头菌的营养成分很高,每百克含蛋白质26.3克,是香菇的二倍。它含有氨基酸多大17种,其中人体所需的占8种。每百克猴头含脂肪4.2克,是名副其实的高蛋白、低脂肪食品,另外还富含各种维生素和无机盐。猴头菇有增进食欲,增强胃粘膜屏障机能,提高淋巴细胞转化率,提升白细胞等作用。故可以使人体提高对疾病的免疫能力。
茯苓(学名:Wolfiporia cocos):茯苓中的主要成分为茯苓聚糖,含量很高。对多种细菌有抑制作用;能降胃酸,对消化道溃疡有预防效果;对肝损伤有明显的保护作用;能多方面对免疫功能进行调节;能使化疗所致白细胞减少加速回升能增强机体免疫功能,茯苓多糖有明显的抗肿瘤作用;有利尿作用;并有镇静的作用。
随着科学技术的发展与创新,为克服原药材利用率低、用量大等缺点,传统技术常用醇提、水提、浓缩等技术进行制备,但提取之后大量的药渣基本被弃用,既浪费资源,又污染环境。同时,大部分中草药的有效成分都被包裹在植物细胞壁内,用一般的溶剂提取难以破坏细胞壁,提取率低。国外20世纪80年代已有利用酶法处理提取天然产物的报道,利用酶具有的特性,选择相应的酶,在中药提取分离中,分解、破坏细胞壁结构,有效成分溶解或助溶于溶剂中,从而达到在传统的溶剂提取方法基础上,加速有效成分释放,从而提高提取率,提高原料的利用价值。研究表明,纤维素酶具有分解纤维素、破坏细胞壁、增加植物细胞内容物溶出量的作用。本发明利用纤维素酶将中草药进行酶解,提取天然植物和中草药的活性物质,使人体更加容易吸收的多糖类、皂甙类、黄酮类物质等有效成分,使资源充分得到利用。
发明内容
本发明的目的在于提供一种含蒜素的复方制品及其制备方法,产品选用传统天然植物进行配伍,无毒副作用,长期服用具有良好的提高免疫功能和消除自由基作用;本方法集酶解、醇提和水提于一体,比传统的溶剂提取方法提取率更高,比单一的提取方法成分更全面,制成的含蒜素的复方制品营养更充分、食用更便捷。
为实现上述目的,本发明采用如下技术方案:
一种含蒜素的复方制品的主要原料为大蒜、人参、山楂、枸杞子、猴头菇和茯苓。
主要原料按重量份数计:大蒜2000-2500份、人参1000-1250份、山楂1000-1250份、枸杞子1000-1250份、猴头菇800-1000份、茯苓800-1000份。
主要原料按重量份数计:大蒜1800-2300份、人参800-1000份、山楂800-1000份、枸杞子800-1000份、猴头菇800-1000份、茯苓800-1000份。
各原料经称量配料、混合粉碎,采用中国药典标准药筛20-50目过筛后直接分装得到产品。
主要原料经酶解、醇提和水提集成方法得到的提取物加入药用辅料制成粉剂、胶囊剂、片剂、丸剂或颗粒剂。
主要原料和辅料按重量份数计:主要原料:大蒜2000-2500份、人参1000-1250份、山楂1000-1250份、枸杞子1000-1250份、猴头菇800-1000份、茯苓800-1000份;辅料:糊精200-1000份、葡萄糖300-500份。
主要原料和辅料按重量份数计:主要原料:大蒜1800-2300份、人参900-1150份、山楂800-1000份、枸杞子800-1000份、猴头菇800-1000份、茯苓800-1000份;辅料:糊精300-800份、木糖醇200-300份。
主要原料和辅料按重量份数计:主要原料:大蒜2250份、人参1125份、山楂1000份、枸杞子1000份、猴头菇1000份、茯苓1000份;辅料:糊精500份、葡萄糖300份、木糖醇200份。
制备方法包括如下步骤:
(1)将合格原料称量,混合,粉碎,过5-20目筛;
(2)用蒸馏水浸泡,料液比为1:3-20,调节pH为4.0-6.0,温度为50-60℃,用原料总重量0.1-1.0%的纤维素酶进行酶解,时间为1-4小时,然后过滤,得滤液A和滤渣B;
(3)将滤渣B用体积分数为70-95%的乙醇溶液提取,料液比为1:5-10,加热至微沸,回流1-2小时,提取2次,得滤渣C;合并两次滤液,回收乙醇,60℃浓缩至相对密度为1.05-1.15,得浓缩液D,采用膜过滤,目数为200-300目;
(4)将滤渣C用饮用水提取,料液比为1:5-20,加热至微沸,时间为1-2小时,提取2次;合并两次滤液,得滤液E;将滤液A、E合并,60℃浓缩至相对密度为1.05-1.15,得到浓缩液F,采用膜过滤,目数为200-300目;
(5)将浓缩液D、F合并,然后加入辅料混匀,冷冻干燥至含水量在5%以下。
本发明的显著优点在于:产品选用传统天然植物进行配伍,无毒副作用,长期服用具有良好的提高免疫功能和消除自由基作用;本方法集酶解、醇提和水提于一体,比传统的溶剂提取方法提取率更高,比单一的提取方法成分更全面,制成的含蒜素的复方制品营养更充分、食用更便捷。
具体实施方式
实施例1:速溶粉剂
原料:大蒜2000g、人参1000g、山楂1000g、枸杞子1000g、猴头菇800g、茯苓800g
辅料:糊精500g、木糖醇300g
制备方法:
1、原料前处理:将合格原料按配方称量,然后混合、粉碎,过筛目数为10目。
2、酶解:将过筛后的药材用蒸馏水浸泡,料液比为1:10,调节PH为5.0,温度为50℃,用原料总量的0.5%的纤维素酶进行酶解,时间为4小时,然后过滤,收集滤液A备用,得滤渣B。
3、醇提:将滤渣B用体积分数为80%的乙醇溶液提取,料液比为1:10,加热至微沸,回流提取2小时,然后过滤;用滤渣按上法条件再次提取,过滤,滤渣为C;合并两次滤液,回收乙醇,浓缩至相对密度为1.05-1.15(60℃),得到浓缩液D。
4、水提:将滤渣C用饮用水提取,料液比为1:8,加热至微沸,煎煮提取2小时,然后过滤;用滤渣按上法条件再次提取,过滤,合并两次滤液,得滤液E备用,余下滤渣弃去。将滤液A、E合并,浓缩至相对密度为1.05-1.15(60℃),得到浓缩液F。上述所述的膜过滤均采用目数为300目。
5、添加辅料、干燥:将浓缩液D、F合并,然后加入辅料混匀,采用冷冻干燥,控制水分5%以下,收集得提取粉制品。
实施例2:颗粒剂
原料:大蒜2300g、人参1150g、山楂1000g、枸杞子1000g、猴头菇800g、茯苓800g
辅料:糊精550g、葡萄糖500g、
制备方法同实施例1中。
实施例3:片剂
原料:大蒜2500g、人参1250g、山楂1250g、枸杞子1250g、猴头菇1000g、茯苓1000g
辅料:糊精500g、葡萄糖300g、木糖醇200g
制备方法:
1、原料前处理:将合格原料按配方称量,然后混合、粉碎,过筛目数为20目。
2、酶解:将过筛原料用蒸馏水浸泡,料液比为1:15,调节PH为4.8,温度为50℃,用原料总量的0.6%的纤维素酶进行酶解,时间为3小时,然后过滤,收集滤液A备用,得滤渣B。
3、醇提:将滤渣B用体积分数为75%的乙醇溶液提取,料液比为1:15,加热至微沸,回流提取2小时,然后过滤;用滤渣按上法条件再次提取,过滤,滤渣为C;合并两次滤液,回收乙醇,浓缩至相对密度为1.05-1.15(60℃),得到浓缩液D。
4、水提:将滤渣C用饮用水提取,料液比为1:10,加热至微沸,煎煮提取2小时,然后过滤;用滤渣按上法条件再次提取,过滤,合并两次滤液,得滤液E备用,余下滤渣弃去。将滤液A、E合并,浓缩至相对密度为1.05-1.15(60℃),得到浓缩液F。
5、添加辅料、干燥:将浓缩液D、F合并,然后加入辅料混匀,采用冷冻干燥,控制水分5%以下,收集冷冻干粉。
6、将收集所得干燥制品经过常规工艺混合制粒压片成型。
实施例4:胶囊剂
原料:大蒜1800g、人参900g、山楂800g、枸杞子800g、猴头菇800g、茯苓800g
辅料:糊精500g。
制备方法同实施例1,然后再经常规工艺制粒之后填充胶囊。
含蒜素的复方制品的主要成分(g/100g)
【含量测定】
大蒜素含量检测:按照气相色谱法(中国药典2000年版二部附录Ⅴ E)测定。
人参皂甙的含量检测:应用液相色谱-紫外检测法测定 GB/T22996-2008。
粗多糖的含量检测:应用硫酸-苯酚比色法测食品中的粗多糖的含量和NY/T1676-2008 食用菌中粗多糖含量的测定。
以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所做的均等变化与修饰,皆应属本发明的涵盖范围。
Claims (8)
1.一种含蒜素的复方制品,其特征在于:主要原料按重量份数计:大蒜2000-2500份、人参1000-1250份、山楂1000-1250份、枸杞子1000-1250份、猴头菇800-1000份、茯苓800-1000份。
2.一种含蒜素的复方制品,其特征在于:主要原料按重量份数计:大蒜1800-2300份、人参800-1000份、山楂800-1000份、枸杞子800-1000份、猴头菇800-1000份、茯苓800-1000份。
3.根据权利要求1或2所述的含蒜素的复方制品,其特征在于:各原料经称量配料、混合粉碎,采用中国药典标准药筛20-50目过筛后直接分装得到产品。
4.根据权利要求1或2所述的含蒜素的复方制品,其特征在于:主要原料经酶解、醇提和水提集成方法得到的提取物加入药用辅料制成粉剂、胶囊剂、片剂、丸剂或颗粒剂。
5.根据权利要求1所述的含蒜素的复方制品,其特征在于:主要原料和辅料按重量份数计:
主要原料:大蒜2000-2500份、人参1000-1250份、山楂1000-1250份、枸杞子1000-1250份、猴头菇800-1000份、茯苓800-1000份;
辅料:糊精200-1000份、葡萄糖300-500份。
6.根据权利要求2所述的含蒜素的复方制品,其特征在于:主要原料和辅料按重量份数计:
主要原料:大蒜1800-2300份、人参900-1150份、山楂800-1000份、枸杞子800-1000份、猴头菇800-1000份、茯苓800-1000份;
辅料:糊精300-800份、木糖醇200-300份。
7.根据权利要求1所述的含蒜素的复方制品,其特征在于:主要原料和辅料按重量份数计:主要原料:大蒜2250份、人参1125份、山楂1000份、枸杞子1000份、猴头菇1000份、茯苓1000份;
辅料:糊精500份、葡萄糖300份、木糖醇200份。
8.一种制备如权利要求4所述的含蒜素的复方制品的方法,其特征在于:包括如下步骤:
(1)将合格原料称量,混合,粉碎,过5-20目筛;
(2)用蒸馏水浸泡,料液比为1:3-20,调节pH为4.0-6.0,温度为50-60℃,用原料总重量0.1-1.0%的纤维素酶进行酶解,时间为1-4小时,然后过滤,得滤液A和滤渣B;
(3)将滤渣B用体积分数为70-95%的乙醇溶液提取,料液比为1:5-10,加热至微沸,回流1-2小时,提取2次,得滤渣C;合并两次滤液,回收乙醇,60℃浓缩至相对密度为1.05-1.15,得浓缩液D,采用膜过滤,目数为200-300目;
(4)将滤渣C用饮用水提取,料液比为1:5-20,加热至微沸,时间为1-2小时,提取2次;合并两次滤液,得滤液E;将滤液A、E合并,60℃浓缩至相对密度为1.05-1.15,得到浓缩液F,采用膜过滤,目数为200-300目;
(5)将浓缩液D、F合并,然后加入辅料混匀,冷冻干燥至含水量在5%以下。
