本发明公开的是一种维生素C方格星虫蛋白酶解蛋白泡腾片。由方格星虫酶解蛋白粉、维生素C、糖粉、柠檬酸、安赛蜜、碳酸氢钠、硬脂酸镁及绿茶香精为原料制备。方格星虫酶解蛋白粉主要成份是多肽类物质及氨基酸,它与维生素C这两种成份被人体吸收后能提高人体免疫力及抗病能力,两者配伍后,维生素C 能活化方格星虫酶解蛋白粉中脯氨酸羟化酶和赖氨酸羟化酶,促进脯氨酸和赖氨酸向羟脯氨酸和羟赖氨酸的转化,进而促进蛋白质的吸收利用,提高生物利用度。本发明还具有以下特点:故崩解作用强大,可以使药物迅速崩解成细小颗粒而迅速溶解。在人体内吸收良好,尤其适用于儿童服用。药液清凉可口,口感好,人们易于接受。

背景技术
方格星虫 (Sipunculus nudus) 属于星虫动物门,星虫科,方格星虫属动物,俗称沙虫。个体较大,肉嫩味美,是我国著名的海珍品之一,深受消费者欢迎。方格星虫含有丰富的营养成分。我国多种本草中记载了其食用和药用价值,东南沿海民间称其为“海洋冬虫夏草”,将其作为一种高级补品。现代医药研究表明方格星虫含有多种活性物质,能够调节机体多种功能,并含有丰富的蛋白质、微量元素等,具有显著的提高免疫力、延缓衰老、抗氧化、抗疲劳、耐缺氧、耐高温等功效,对心血管系统具有明显的保护作用。目前有关方格星虫的成品制剂并不多见。
泡腾片是一种放入水中后能够迅速产生气泡 (即泡腾) 而使片剂迅速崩解的,供内服或外用的固体制剂,其最大特点为崩解剂是以一种或数种有机酸 (酸剂) 和一种或数种遇酸可产生二氧化碳的无机盐 (碱剂) 共同组成,遇水发生酸碱中和反应放出二氧化碳使片剂崩解,使固体剂型迅速变为液体剂型,这样的变化带来的效果是:既具有固体制剂剂量准确、质量稳定、生产自动化程度高、成本低、易携带的特点。又具有液体制剂易于吞服、生物利用度高,对胃肠道刺激性小等特点。因此,尤其适用于儿童,儿童在身体发育时期,补充蛋白质类、多肽类物质是很有必要的,而方格星虫的蛋白质酶解成多肽后,制备成一种易于人体吸收的食品,能增强人体的免疫力,提高人体的抗病能力,但多肽类物质进入人体后存在着如何提高生物利用度的问题。因此,提高方格星虫多肽在人体内的生物利用度就是本发明要致力解决的问题。
发明内容
本发明的目的是提供一种维生素C方格星虫酶解蛋白泡腾片,克服现有方格星虫产品味道不好,吸收不好,生物利用度不高的不足。
为实现上述目的,本发明采取了下述具体技术措施,一种维生素C方格星虫酶解蛋白泡腾片,主要由下述质量百分比的原料制备:
方格星虫酶解蛋白粉20-23%,维生素C 1-2%,糖粉8-12%,柠檬酸25-28%,安赛蜜1-1.5%,碳酸氢钠30-35%,硬脂酸镁1-1.2%,绿茶香精0.3-0.5%;
本发明维生素C方格星虫酶解蛋白泡腾片通过以下方法制备
(1) 经过冷冻干燥得到的方格星虫酶解蛋白粉密封保存备用;将称取的维生素C粉碎过100目筛密封保存备用;将柠檬酸和食用碳酸氢钠,分别于105℃干燥2 h,粉碎过100目筛,密封保存备用;糖粉粉碎过100目筛,密封保存备用;安赛蜜和绿茶香精,粉碎过100目筛;硬脂酸镁粉碎过100目筛,置于干燥器中保存备用;制备过程中控制环境温度及湿度分别在25℃、40% 以下;
(2) 将方格星虫酶解蛋白粉,维生素C,糖粉及安赛蜜是分别平均分成两份,分别与柠檬酸和食用碳酸氢钠混合制备颗粒,制备颗粒时用 95% 乙醇,得到酸颗粒和碱颗粒,两种颗粒于室温条件下减压干燥12 h,不能加热,干燥结束后整粒;
(3) 压片前,将酸颗料和碱颗粒均匀混合后,加入硬脂酸镁和绿茶香精混合均匀;
(4) 将步骤(3)中充分混匀的颗粒压片,得到维生素C方格星虫酶解蛋白泡腾片,设定片重为1 g。
上述制备方法中所用的方格星虫酶解蛋白粉通过以下方法制备:
(1) 将方格星虫洗净除去杂质,加适量水打浆,加至5倍量水,加入活性是1000 U/g的胃蛋白酶,按照酶与底物比为2%-8%,加入酶活为1000 U/g的胃蛋白酶,调整温度至37℃,pH是3.0,边搅拌边酶解120 min;再按照酶与底物比为3%-8%,加入活性是300 U/g的胰蛋白酶,调整温度至55℃,pH是8.0,边搅拌边酶解60 min,100℃加热10 min灭酶,4000转/分离心15 min,取上清液;
(2) 采用分子量为3000 Da的超滤膜将酶解液过滤,收集超滤分子量小于3000 Da的部分,再采用分子量为150的纳滤膜,进行纳滤处理脱盐,纳滤条件:分子量为150截留膜,进膜压力11 Bar,出膜压力11Bar,最后将蛋白酶解产物进行冷冻干燥,冷冻干燥条件:真空度为40 pa,共熔点温度-30℃,-30℃升至0℃,0-10 min ;10―30 min,0℃升至30℃;30―360 min,30℃;360―480 min,30℃升至40℃;489―720 min,40℃;720―840 min,40℃降至20℃;最终得到方格星虫酶解蛋白粉。
采取上述措施的本发明,主要以方格星虫酶解蛋白粉和维生素C为原料,方格星虫酶解蛋白粉主要成份是多肽类物质及氨基酸,这两种成份被人体吸收后能提高人体免疫力及抗病能力。方格星虫酶解蛋白粉与维生素C配伍后,维生素C 能活化方格星虫酶解蛋白粉中脯氨酸羟化酶和赖氨酸羟化酶,促进脯氨酸和赖氨酸向羟脯氨酸和羟赖氨酸的转化,进而促进蛋白质的吸收利用,提高生物利用度。本发明使用时将所得泡腾片2片投入150 mL温开水中,其悬浮或落于瓶底,并立即放出大量气泡,片剂随之迅速溶解,在1 min内形成淡黄色透明溶液,且口感良好新奇。本发明还具有以下特点:
(1) 其生产、运输、贮藏是固体形式,
(2) 其崩解剂由酸剂和碱剂构成,故崩解作用强大,可以使药物迅速崩解成细小颗粒而迅速溶解。
(3) 本发明在人体内吸收良好,尤其适用于儿童服用。
(4) 药液清凉可口,可以作为清凉饮料,口感好,人们易于接受。
具体实施方式
实施例1
(一)方格星虫酶解蛋白粉制备:
(1) 将1000克方格星虫洗净除去杂质,加适量水打浆,再加水至5000克继续打浆至均匀,再按照酶与底物比为2%,加入活性是1000 U/g的胃蛋白酶,调整温度至37℃,pH是3.0,边搅拌边酶解120 min;再按照酶与底物比为3%,再加入活性是300 U/g的胰蛋白酶,调整温度至55℃,pH是8.0,边搅拌边酶解60 min,100℃加热10 min灭酶,4000转/分离心15 min,取上清液;
(2) 采用分子量为3000 Da的超滤膜将酶解液过滤,收集超滤分子量小于3000 Da的部分,再采用分子量为150的纳滤膜,进行纳滤处理脱盐,纳滤条件:分子量为150截留膜,进膜压力11 Bar,出膜压力11Bar,最后将蛋白酶解产物进行冷冻干燥,冷冻干燥条件:真空度为40 pa,共熔点温度-30℃,—30℃升至0℃,0-10 min ;10―30 min,0℃升至30℃;30―360 min,30℃;360―480 min,30℃升至40℃;489―720 min,40℃;720―840 min,40℃降至20℃;最终得到方格星虫酶解蛋白粉23克;
(二)维生素C方格星虫酶解蛋白泡腾片制备:
(1) 经过冷冻干燥得到的方格星虫酶解蛋白粉密封保存备用;将称取的维生素C粉碎过100目筛密封保存备用;将柠檬酸和食用碳酸氢钠,分别于105℃干燥2 h,粉碎过100目筛,密封保存备用;糖粉粉碎过100目筛,密封保存备用;安赛蜜和绿茶香精,粉碎过100目筛;硬脂酸镁粉碎过100目筛,置于干燥器中保存备用;制备过程中控制环境温度及湿度分别在25℃、40% 以下;
(2)取方格星虫酶解蛋白粉20克,维生素C 1克,糖粉8克,柠檬酸25克,安赛蜜1克,碳酸氢钠30克,硬脂酸镁1克,绿茶香精0.3克;
将方格星虫酶解蛋白粉,维生素C,糖粉及安赛蜜是分别平均分成两份,分别与柠檬酸和食用碳酸氢钠混合制备颗粒,制备颗粒时用 95% 乙醇,得到酸颗粒和碱颗粒,两种颗粒于室温条件下减压干燥12 h,不能加热,干燥结束后整粒;
(3) 压片前,将酸颗料和碱颗粒均匀混合后,加入硬脂酸镁和绿茶香精混合均匀;
(4) 将步骤(3)中充分混匀的颗粒压片,得到维生素C方格星虫酶解蛋白泡腾片,设定片重为1 g。
实施例2
(一)方格星虫酶解蛋白粉制备:

(二)维生素C方格星虫酶解蛋白泡腾片制备:
(1) 经过冷冻干燥得到的方格星虫酶解蛋白粉密封保存备用;将称取的维生素C粉碎过100目筛密封保存备用;将柠檬酸和食用碳酸氢钠,分别于105℃干燥2 h,粉碎过100目筛,密封保存备用;糖粉粉碎过100目筛,密封保存备用;安赛蜜和绿茶香精,粉碎过100目筛;硬脂酸镁粉碎过100目筛,置于干燥器中保存备用;制备过程中控制环境温度及湿度分别在25℃、40% 以下;
(2) 取方格星虫酶解蛋白粉22克,维生素1.5克,糖粉10克,柠檬酸26克,安赛蜜1.3克,碳酸氢钠33克,硬脂酸镁1.1克,绿茶香精0.4;
将方格星虫酶解蛋白粉,维生素C,糖粉及安赛蜜是分别平均分成两份,分别与柠檬酸和食用碳酸氢钠混合制备颗粒,制备颗粒时用 95% 乙醇,得到酸颗粒和碱颗粒,两种颗粒于室温条件下减压干燥12 h,不能加热,干燥结束后整粒。
(3) 压片前,将酸颗料和碱颗粒均匀混合后,加入硬脂酸镁和绿茶香精混合均匀;
(4) 将步骤(3)中充分混匀的颗粒压片,得到维生素C方格星虫酶解蛋白泡腾片,设定片重为1 g。
实施例3
(一)方格星虫酶解蛋白粉制备:
(1) 将1000克方格星虫洗净除去杂质,加适量水打浆,再加水至5000克继续打浆至均匀,再按照酶与底物比为4%,加入活性是1000 U/g的胃蛋白酶,调整温度至37℃,pH是3.0,边搅拌边酶解120 min;再按照酶与底物比为5%,再加入活性是300 U/g的胰蛋白酶,调整温度至55℃,pH是8.0,边搅拌边酶解60 min,100℃加热10 min灭酶,4000转/分离心15 min,取上清液;

(1) 经过冷冻干燥得到的方格星虫酶解蛋白粉密封保存备用;将称取的维生素C粉碎过100目筛密封保存备用;将柠檬酸和食用碳酸氢钠,分别于105℃干燥2 h,粉碎过100目筛,密封保存备用;糖粉粉碎过100目筛,密封保存备用;安赛蜜和绿茶香精,粉碎过100目筛;硬脂酸镁粉碎过100目筛,置于干燥器中保存备用;制备过程中控制环境温度及湿度分别在25℃、40% 以下;
(2) 取方格星虫酶解蛋白粉23克,维生素C 2克,糖粉12克,柠檬酸28克,安赛蜜1.5克,碳酸氢钠35克,硬脂酸镁1.2克,绿茶香精0.5克;
将方格星虫酶解蛋白粉,维生素C,糖粉及安赛蜜是分别平均分成两份,分别与柠檬酸和食用碳酸氢钠混合制备颗粒,制备颗粒时用 95% 乙醇,得到酸颗粒和碱颗粒,两种颗粒于室温条件下减压干燥12 h,不能加热,干燥结束后整粒。
(3) 压片前,将酸颗料和碱颗粒均匀混合后,加入硬脂酸镁和绿茶香精混合均匀;
(4) 将步骤(3)中充分混匀的颗粒压片,得到维生素C方格星虫酶解蛋白泡腾片,设定片重为1 g。
Claims (2)
1.一种维生素C方格星虫酶解蛋白泡腾片,其特征在于所述泡腾片主要由下述质量百分比的原料制备:
方格星虫酶解蛋白粉20-23%,维生素C 1-2%,糖粉8-12%,柠檬酸25-28%,安赛蜜1-1.5%,碳酸氢钠30-35%,硬脂酸镁1-1.2%,绿茶香精0.3-0.5%;
所述维生素C方格星虫酶解蛋白泡腾片通过以下方法制备:
(1) 经过冷冻干燥得到的方格星虫蛋白酶解物密封保存备用;将称取的维生素C粉碎过100目筛密封保存备用;将柠檬酸和食用碳酸氢钠,分别于105℃干燥2 h,粉碎过100目筛,密封保存备用;糖粉粉碎过100目筛,密封保存备用;安赛蜜和绿茶香精,粉碎过100目筛;硬脂酸镁粉碎过100目筛,置于干燥器中保存备用;制备过程中控制环境温度及湿度分别在25℃、40% 以下;
(2) 将方格星虫蛋白酶解产物,维生素C,糖粉及安赛蜜是分别平均分成两份,分别与柠檬酸和食用碳酸氢钠混合制备颗粒,制备颗粒时用 95% 乙醇,得到酸颗粒和碱颗粒,两种颗粒于室温条件下减压干燥12 h,不能加热,干燥结束后整粒;
(3) 压片前,将酸颗料和碱颗粒均匀混合后,加入硬脂酸镁和绿茶香精混合均匀;
(4) 将步骤(3)中充分混匀的颗粒压片,得到维生素C方格星虫酶解蛋白泡腾片,设定片重为1 g。
2.根据权利要求1所述的维生素C方格星虫酶解蛋白泡腾片,其特征在于
方格星虫酶解蛋白粉通过以下方法制备:
(1) 将方格星虫洗净除去杂质,加适量水打浆,加至5倍量水,加入活性是1000 U/g的胃蛋白酶,按照酶与底物比为2%-8%,加入酶活为1000 U/g的胃蛋白酶,调整温度至37℃,pH是3.0,边搅拌边酶解120 min;再按照酶与底物比为3%-8%,加入活性是300 U/g的胰蛋白酶,调整温度至55℃,pH是8.0,边搅拌边酶解60 min,100℃加热10 min灭酶,4000转/分离心15 min,取上清液;
(2) 采用分子量为3000 Da的超滤膜将酶解液过滤,收集超滤分子量小于3000 Da的部分,再采用分子量为150的纳滤膜,进行纳滤处理脱盐,纳滤条件:分子量为150截留膜,进膜压力11 Bar,出膜压力11Bar,最后将蛋白酶解产物进行冷冻干燥,冷冻干燥条件:真空度为40 pa,共熔点温度-30℃,-30℃升至0℃,0-10 min ;10―30 min,0℃升至30℃;30―360 min,30℃;360―480 min,30℃升至40℃;489―720 min,40℃;720―840 min,40℃降至20℃;最终得到方格星虫酶解蛋白粉。
