本发明涉及一种中药制剂的制备方法,具体是红景天注射液在配制过程中加入活性炭吸附除去杂质和热原,同时提高成品的灯检合格率。配制过程采用低温除炭过滤方法,使大株红景天注射液中的杂质和热原更加有效的去除,明显提高产品灯检合格率。

红景天注射液低温除炭制备方法
技术领域
本发明涉及一种中药制剂的制备方法,具体是红景天注射液在配制过程中加入活性炭吸附除去杂质和热原,同时提高成品的灯检合格率。
背景技术
在已有技术中,大株红景天注射液是通化玉圣药业有限公司独家生产的专利产品,国药准字Z20060361。性状:本品为淡黄色至棕黄色的澄明液体。 成份:大株红景天。适应症状:活血化瘀。冠心病稳定型劳累性心绞痛。症见:胸部刺痛,绞痛,固定不移,痛引肩背及臂内侧,胸闷,心悸不宁,唇舌紫暗,脉细涩。用法用量:静脉滴注。一次10ml加入5%葡萄糖注射液中,一日一次,10天为一疗程。
制备方法:取大株红景天1500g,加水提取三次,每次加水6倍,提取2小时,合并提取液,滤过,滤液浓缩至相对密度为1.15~1.20(50℃)的清膏,加乙醇使含醇量达65%,搅匀,冷藏 24小时(5~10℃),滤过,回收乙醇并浓缩至相对密度为1.15~1.20(50℃)的清膏,再加乙醇使含醇量达85%,搅匀,冷藏24小时(5~10℃),滤过,滤液回收乙醇并浓缩至相对密度为1.20~1.30(50℃)的清膏,加入注射用水1000ml,调pH值至7.0,加热煮沸30分钟,冷藏72小时(5~10℃),滤过,滤液以截留分子量为10000的超滤柱超滤,超滤液经微孔滤膜过滤,灌封,灭菌,即得。
中药注射剂的传统工艺中活性炭在40~60℃吸附性最好,但是在大株红景天注射液配制过程中冷除炭(0~10℃)和热除炭进行对比,最终产品的灯检收率有明显的不同,低温除炭工艺灯检收率明显提高。
发明内容
本发明的目的是针对大株红景天注射液灯检收率偏低,提高灯检收率,降低成本,增加经济效益的情况下,对红景天配制工艺进行实验分析,采用冷除炭(5~10℃)和热除炭进行对比,明显提高产品灯检收率。节约成本。
本发明的技术解决方案是:红景天注射液低温除炭制备方法,其步骤如下:取大株红景天1500g,加水提取三次,每次加水6倍,提取2小时,合并提取液,滤过,滤液浓缩至相对密度为1.15~1.20的清膏, 温度50℃;加乙醇使含醇量达65%,搅匀,在温度5~10℃冷藏24小时,滤过,回收乙醇并浓缩至相对密度为1.15~1.20的清膏,温度50℃;再加乙醇使含醇量达85%,搅匀,在温度5~10℃冷藏24小时,滤过,滤液回收乙醇并浓缩至相对密度为1.20~1.30的清膏,温度50℃;加入注射用水1000ml,加入3‰的活性炭,加热煮沸30分钟,在温度0~10℃冷藏72小时,保持0~10℃低温除炭滤过滤,滤液以截留分子量为10000的超滤柱超滤,超滤液经微孔滤膜过滤,灌封,灭菌,即得。

本发明的优点是:配制过程采用低温除炭过滤方法,使大株红景天注射液中的杂质和热原更加有效的去除,明显提高产品灯检合格率。

具体实施方式
实施例1
红景天注射液微高温水中投料提取方法:取大株红景天1500g,加水提取三次,每次加水6倍,提取2小时,合并提取液,滤过,滤液浓缩至相对密度为1.15~1.20(50℃)的清膏,加乙醇使含醇量达65%,搅匀,冷藏 24小时(5~10℃),滤过,回收乙醇并浓缩至相对密度为1.15~1.20(50℃)的清膏,再加乙醇使含醇量达85%,搅匀,冷藏24小时(5~10℃),滤过,滤液回收乙醇并浓缩至相对密度为1.20~1.30(50℃)的清膏,加入注射用水1000ml,加入3‰的活性炭,加热煮沸30分钟,冷藏72小时(0~10℃),保持0~10℃低温除炭滤过,滤液以截留分子量为10000的超滤柱超滤,超滤液经微孔滤膜过滤,灌封,灭菌,即得。
实施例2

对配制药液冷热除炭进行对比结果见表1
注:灯检采用日本卫材全自动灯检机(AIM288/ B833)同一参数灯检。
可见,大株红景天注射液配制过程低温除炭最终产品灯检收率明显提高。

1.一种红景天注射液低温除炭制备方法,其特征在于步骤如下:取大株红景天1500g,加水提取三次,每次加水6倍,提取2小时,合并提取液,滤过,滤液浓缩至相对密度为1.15~1.20的清膏, 温度50℃;加乙醇使含醇量达65%,搅匀,在温度5~10℃冷藏 24小时,滤过,回收乙醇并浓缩至相对密度为1.15~1.20的清膏,温度50℃;再加乙醇使含醇量达85%,搅匀,在温度5~10℃冷藏24小时,滤过,滤液回收乙醇并浓缩至相对密度为1.20~1.30的清膏,温度50℃;加入注射用水1000ml,加入3‰的活性炭,加热煮沸30分钟,在温度0~10℃冷藏72小时,保持0~10℃低温除炭滤过,滤液以截留分子量为10000的超滤柱超滤,超滤液经微孔滤膜过滤,灌封,灭菌,即得。
