本发明公开了一种健骨注射液及其制备方法,取战骨(茎)片2500g,5.0g-15.0gHS-15,苯甲醇10mL和注射用水组成。健骨注射液制备方法:取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上,取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过,滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时,滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入5.0g-15.0gHS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得。全过程在无菌条件下进行,采用非最终灭菌工艺。与市售健骨注射液相比较,本发明采用了新的处方和工艺,制成的产品符合药典标准,其突出的优势是极大地提高了健骨注射液的临床用药安全性。

一种健骨注射液及其制备方法
技术领域
本发明涉及医药制剂工艺领域,特别涉及一种健骨注射液及其制备方法。
技术背景
战骨是一种常用的壮族药,植物名为黄毛豆腐柴,系马鞭草科豆腐柴属植物(Premna fulva Craib),又名土霸王、穿云箭,具有活血散淤、强筋健骨、祛风止痛之功效,广西民间壮族常用于治疗腰腿痛、跌打扭伤、风湿性关节炎和类风湿性关节炎、肝区疼痛等,为广西道地药材。战骨茎的提取液(商品名健骨注射液)具有抗炎消肿、镇痛作用,用于治疗肥大性脊椎炎(即腰椎骨质增生症),总有效率为97.37%,显效率为62.7%。健骨注射液是由战骨(茎)提取而制成的中药注射剂,具有活血散瘀,强筋健骨,驱风止痛。用于脊椎骨质增生症,对风湿性关节痛亦有疗效。
健骨注射液是临床常用药物之一。经调查,现行市售健骨注射剂其增溶剂为聚三梨酯80(吐温-80),相关药理研究发现:2%吐温-80给豚鼠1m l/只静脉注射后出现生身体歪斜,嗜睡等明显症状,5%吐温-80豚鼠均出现耳朵发紫、步态不稳、身体歪斜,侧卧、嗜睡等非常明显的症状;Beagle犬静脉缓注吐温-80,剂量降低至0.02%吐温-80,以4ml/体积给予时,动物仍然会出现很显的过敏反应症状(孙立,刘晓萌等,J Toxicol August 2007 Vo1.21 No.4),其它文献资料也确切证明吐温-80是导致临床用药时发生不良反应(过敏反应)的主要原因之一,为了进一步提高健骨注射液临床用药的安全性,本发明选择了比吐温-80安全性更高的药用辅料。
发明内容
本发明目的在于提供一种全新处方和制备工艺的健骨注射液、使健骨注射液不仅质量稳定而且还具有比市售更高的临床用药安全性。为了达上上述目的,本发明采用如下技术方案:战骨(茎)片2500g,5.0g-15.0g HS-15,苯甲醇10 mL和注射用水组成。
健骨注射液具体制备方法,取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上,取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过,滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时,滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入5.0g-15.0g HS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得。
本发明中的制备方法全过程在无菌条件下进行,采用非最终灭菌工艺。
安全性实验证明:本发明在安全性方面(过敏反应)明显优于市售健骨注射液。
具体实施方式
实施例1  健骨注射液,由战骨(茎)片2500g,5.0g HS-15,苯甲醇10 mL和注射用水组成。
实施例1  取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上,取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过,滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时,滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入5.0g HS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得。
实施例2  健骨注射液,由战骨(茎)片2500g,10.0g HS-15,苯甲醇10 mL和注射用水组成。
实施例2  取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上,取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过,滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时,滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入10.0g HS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得。
实施例3  健骨注射液,由战骨(茎)片2500g,15.0g HS-15,苯甲醇10 mL和注射用水组成。
实施例3  取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上,取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过,滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时,滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入15.0g HS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得。

Claims (5)

1.一种健骨注射液,由战骨(茎)片活性提取物、增溶剂、抗氧剂、及注射用水组成。
2.如权利要求1所述的健骨注射液,其特征在于增溶剂采用HS-15(15-羟基硬脂酸聚乙二醇酯)。
3.如权利要求1所述的健骨注射液,其特征在于抗氧剂采用苯甲醇。
4.如权利要求1-3所述的健骨注射液,其特征在于由战骨(茎)片2500g,5.0g-15.0g HS-15,苯甲醇10 mL和注射用水组成,过滤至澄明度合格后灌封,全过程在无菌条件下进行,采用非最终灭菌工艺。
5.如权利要求1-4所述的健骨注射液,其特征在于采用如下制备工艺:
a..取战骨(茎)片2500g,洗净,用20%乙醇浸泡72小时以上;
b.取上清液,滤过,滤液浓缩至为投料量的8倍,加乙醇使含醇量达80%,搅拌均匀,放置12小时,滤过;
c.滤液用20%氢氧化钠溶液调节pH值至7.0~7.5,放置24小时;
d.放置的滤液再次滤过,回收乙醇后,加适量注射用水及活性炭,放置过夜,滤过,滤液加入5.0g-15.0g HS-15和苯甲醇10ml,用20%盐酸溶液调节pH值至7.0~7.5,加注射用水至1000ml,滤过,灌封,即得,本品全过程在无菌条件下进行,采用非最终灭菌工艺。
