Abstract

本发明公开了一种冠心宁注射液及其制备方法,由丹参、川芎、焦亚硫酸钠、依地酸钙钠、HS-15、注射用水配制而成。冠心宁注射液制备方法:将丹参、川芎加水煎煮,煎液滤过,浓缩,用乙醇沉淀处理2次,滤过,浓缩滤液,用注射用水稀释,用稀盐酸调节pH值,冷藏,滤过,用氢氧化钠溶液调节pH值,浓缩滤液,冷藏,滤过得溶液1;取注射用水100ml、加入依地酸钙钠、焦亚硫酸钠搅拌至溶解,活性炭过滤得溶液2;将HS-15加入到100ml注射用水搅匀得溶液3;将3种溶液合并,过滤,调pH值,补注射用水至全量,灌封,即得。本发明采用了新的处方和工艺,其突出的优势是极大地提高了冠心宁注射液的用药安全性。

一种冠心宁注射液及其制备方法
技术领域
本发明涉及医药制剂工艺领域,特别涉及一种冠心宁注射液及其制备方法。
背景技术
冠心宁注射液是含有丹参和川芎有效提取物的中药制剂,在国内上市已有30多年的历史,临床上常用于心脑血管疾病的治疗,具有活血化瘀 、通脉养心的作用。
冠心宁注射液是临床常用药物之一,说明书中记载偶见荨麻疹、风团,血管性神经水肿、过敏性哮喘等不良反应,其中过敏反应是冠心宁最为突出的不良反应,临床有过敏史者都禁用本药。经调查,现行市售冠心宁注射剂其助溶剂多为聚三梨酯80(吐温-80),相关药理研究发现:2%吐温-80给豚鼠1m l/只静脉注射后出现身体歪斜,嗜睡等明显症状,5%吐温-80豚鼠均出现耳朵发紫、步态不稳、身体歪斜,侧卧、嗜睡等非常明显的症状;Beagle犬静脉缓注吐温-80,剂量降低至0.02%吐温-80,以4ml/体积给予时,动物仍然会出现很显的过敏反应症状(孙立,刘晓萌等,J Toxicol August 2007 Vo1.21 No.4),其它文献资料也确切证明吐温-80是导致其临床用药时发生不良反应(过敏反应)的主要原因。
冠心宁注射液因其在治疗冠心病及缺血性脑血管疾病方面疗效确切,被临床广泛应利用,成为当今高发的心脑血管疾病人群健康的强有力保障,但其越发突出的不良反应(过敏反应)严重制约着冠心宁注射液在临床上的大面积应用,故开发安全性高的冠心宁注射制剂就成为时代所需。
发明内容
[0005] 本发明目的在于提供一种全新处方和制备工艺的冠心宁注射液、使冠心宁注射液不仅质量稳定而且还具有较高的临床用药安全性。为了达上上述目的,本发明采用如下技术方案:由丹参2000.0g、川芎2000.0g、1.0g焦亚硫酸钠、2.0g依地酸钙钠、3.0g~20.0gHS-15、注射用水配制而成。
冠心宁注射液制备方法,取丹参2000.0g、川芎2000.0g 加水煎煮3次,第二、三次各1.5小时,合并煎液,滤过,滤液浓缩至约2000ml,用乙醇沉淀处理二次,第一次使含醇量达75%,第二次为85%,每次加乙醇后均冷藏放置,滤过,合并滤液,浓缩至约700ml,用注射用水稀释至4000ml,用稀盐酸调节pH值至3.2;冷藏,滤过,用10%氢氧化钠溶液调节pH值至6.8,浓缩至700ml,冷藏,滤过,得溶液1;取注射用水100ml、加入依地酸钙钠2.0g、1.0g焦亚硫酸钠搅拌使其溶解,加入活性炭2.0g,煮沸30min,过滤得溶液2;取3.0g~20.0gHS-15加入到30℃~40℃100ml注射用水均匀搅拌得溶液3;将3种溶液加入一起中,过滤,滤液调节pH值至6.8,加注射用水定容为1000.0ml,灌封,即得。
本发明中的制备方法全过程在无菌条件下进行,采用非最终灭菌工艺。安全性实验证明:本发明在安全性方面(过敏反应)明显优于市售冠心宁注射液。
具体实施方式
实施例1  冠心宁注射液,由丹参2000.0g、川芎2000.0g、1.0g焦亚硫酸钠、2.0g依地酸钙钠、3.0g~20.0gHS-15、注射用水配制而成。

实施例3  冠心宁注射液,由丹参2000.0g、川芎2000.0g、1.0g焦亚硫酸钠、2.0g依地酸钙钠、3.0g~20.0gHS-15、注射用水配制而成。

Claims (6)

1.一种冠心宁注射液,由活性成分丹参提取物、川芎提取物、增溶剂、螯合剂、抗氧剂及注射用水组成, PH值为6.8。
2.如权利要求1所述的冠心宁注射液,其特征在于增溶剂采用HS-15(15-羟基硬脂酸聚乙二醇酯)。
3.如权利要求1所述的冠心宁注射液,其特征在于螯合剂采用依地酸钙钠。
4.如权利要求1所述的冠心宁注射液,其特征在于抗氧化剂采用焦亚硫酸钠。
5.如权利要求1-4所述的冠心宁注射液,其特征在于由丹参2000.0g、川芎2000.0g、1.0g焦亚硫酸钠、2.0g依地酸钙钠、3.0g~20.0gHS-15、注射用水配制而成,PH值为6.8,过滤至澄明度合格后灌封。
6.如权利要求1-5所述的冠心宁注射液,其特征在于采用如下制备工艺:
a..取丹参2000.0g、川芎2000.0g 加水煎煮3次,第二、三次各1.5小时,合并煎液,滤过,滤液浓缩至约2000ml;
b.用乙醇沉淀处理二次,第一次使含醇量达75%,第二次为85%,每次加乙醇后均冷藏放置,滤过,合并滤液,浓缩至约700ml;
c.用注射用水稀释至4000ml,用稀盐酸调节pH值至3.2;冷藏,滤过,用10%氢氧化钠溶液调节pH值至6.8,浓缩至700ml,冷藏,过滤,得溶液1;
d.取注射用水100ml、加入依地酸钙钠2.0g、1.0g焦亚硫酸钠搅拌使其溶解,加入活性炭2.0g,煮沸30min,过滤得溶液2;
e.取3.0g~20.0gHS-15加入到30℃~40℃100ml注射用水均匀搅拌得溶液3;
f..将3种溶液加入一起中,过滤,滤液调节pH值至6.8,加注射用水定容为1000.0ml,灌封,即得,本品全过程在无菌条件下进行,采用非最终灭菌工艺。
