本发明涉及系统治疗鱼类寄生虫病的药用饲料。
在水产馆和池塘中照料、养殖和保持淡水鱼和海水鱼,在很大程度上使用很多鱼场和水产养殖设备,在这种情况下,危害和侵袭鱼类的寄生虫病会导致严重的经济损失。经常使鱼类严重发生疾病的是下列病原虫:双鞭甲虫:Oodinium pillularis(淡水)和Oodinium ocellatum(海水);鞭毛虫:Costia necatrix和Hexamite symphysodonis;纤毛原虫:Ichthyophthirius multifiliis(淡水)、厚壳桂虫(海水)、斜管虫属和车轮虫属(淡水);单殖亚纲:表皮和鳃吸虫,例如三代虫属和指环虫属。
治疗外寄生虫病即治疗生活在鱼表皮内或表皮上的寄生虫病现仅有所谓浴池疗法,即将适当杀生物浓度的抗寄生虫活性物质加到浴池的水中。
通常只有自由游动阶段的寄生虫即所谓游虫由此而被杀伤,但这个阶段的寄生虫并不一定在鱼的表皮中或表皮内。
因此,整个治疗的基础是消灭游虫,从而防止新的感染或重新感染,以及使鱼的免疫系统增加或专一地防止各个阶段的寄生病原虫的感染。
根据有经验的专家们的意见,外寄生虫病只能进行外部治疗,而不能通过系统施用治疗剂(例如药用饲料)进行治疗。
另一方面,根据文献介绍(参阅N.Herwig,“Handbook of Drugsand Chemicals used in the Treatment of Fish Diseases”,pp.128和136,pub.Charles C.Thomas,Springfield,Illinois,U.S.A.;E.Amlacher,“Taschenbuch der Fischkrankheiten”,pub.GustavFischer Verlag,Stuttgart,1981;L.Zemer,Das Aquarium,220,515/1987)可以理解为,内寄生虫病(例如肠寄生虫)几乎只有通过系统施用治疗剂(例如药用饲料)才能成功地进行治疗。
鱼类(观赏鱼和有用鱼)的外寄生虫病的浴池疗法经常使用杀生物活性染料或防腐活性染料,例如三苯甲烷和吖啶类衍生物。这些化合物包括:N-〔4-〔〔4-二乙基氨基)-苯基〕-苯基亚甲基〕-2,5-亚环己烯-1-基〕-N-乙基-硫酸乙铵(亮绿);N-〔4-〔〔4-(二甲基氨基)-苯基〕-苯基亚甲基〕-2,5-亚环己烯-1-基〕-N-甲基-氯化甲铵和草酸甲铵(孔雀绿);N-〔4-〔二-〔4-(二甲基氨基)-苯基〕-亚甲基〕-2,5-亚环己烯-1-基〕-N-甲基-氯化甲铵(龙胆紫);与3,6-二氨基吖啶混合的3,6-二氨基-10-氯化甲基吖啶(吖啶黄素);9-氨基吖啶和2-乙氧基-6,9-乳酸二氨基吖啶一水合物(利凡诺)。
另外,还使用其他类化合物的抗寄生虫活性物质,例如喹啉衍生物,如奎宁;季铵盐;银盐和铜盐;金属胶体;硝基呋喃,例如nifur-pirinol;以及硝基噻唑,例如2-氨基-5-硝基噻唑。
但是使用所述抗寄生虫活性物质的浴池疗法存在许多缺点:对鱼表皮内或表皮下的寄生虫大多数都无效;全部养殖用水(水产馆,池塘或大量的养殖设备)都要经常处理;另外,需要使用大量活性物质。在处理期间由于分解过程(化学的、生物的和光化学的)和表面吸附作用,活性物质的损耗不能预先计算;所以其浓度也很不稳定。由于在很大程度上所用的物质都是染料,使水带很浓的颜色,并且浴池溶液使各种物质染上颜色,造成污染。还由于对整个系统都进行治疗性处理,杀生物活性物质的存在产生了不希望有的、在某些环境条件下甚至严重的副作用,例如伤害了其他重要的水生生物,如水生植物和浮游生物,在过滤系统中对生物群体的伤害,从而对生物的降解效力产生不良影响。上述治疗活性化合物对鱼还有严重毒性,特别是对鱼鳃具有毒性,致使治疗的安全性受到限制。
因此,本发明的任务在于提供治疗鱼类外寄生虫病和外细菌病并且又能避免上述各种缺点的方法。
本发明提供了鱼药用饲料,其含有三苯甲烷衍生物或各种三苯甲烷衍生物的结合物或其与作为活性物质的至少一种化合物相结合的结合物,它们选自硫堇、硝基噻唑、硝基呋喃、喹啉和吖啶衍生物、能分解为醛类的化合物、过渡金属衍生物、金属胶体、六亚甲基四胺和/或季铵盐。
本发明还涉及该药用饲料在系统治疗外寄生虫病和外细菌病中的应用。本发明药用饲料中活性物质的浓度优选为0.005-10(重量)百分比,尤以0.05-3(重量)%为佳。
由于使用本发明的药用饲料,上述浴池疗法的种种缺点可完全避免,其在系统治疗观赏鱼和有用鱼的外寄生虫病和外细菌病中获得的效果使专家们完全感到出乎意料之外。
令人惊奇地发现,本发明的具有系统疗效的抗外寄生虫和外细菌的药用饲料的效果是新的,出乎意料的,而且是专家们未能预料到的,而这些寄生虫和细菌是在鱼的表皮上和/或表皮下侵袭鱼的。与现今采用的浴池疗法相比,本发明的新治疗法具有明显的优点,例如可以通过鱼的口服施用药用饲料。在鱼表皮中或表皮下的寄生虫或细菌受到令人惊奇的治疗性攻击。由于用水量很少,可以从药用饲料中提取活性物质。另外由于施用活性物质的绝对量很低,整个系统只需装载可略量的活性物质,因此对其他水生生物的毒性实际上降低到零,过滤器的降解作用也不受影响,不再看到水带颜色,对鳃的毒性作用消除了,因此处理期间当然不再发生大量活性物质的损耗。与此同时,所需的副作用可能是防治内寄生虫。
通过选择适当的活性物质,可将其与新的药用饲料相组合。在观赏鱼和有用鱼中,现列举或前已叙述过的最重要的病原虫有:双鞭甲虫,如Oodinium;鞭毛虫:如Costia(Hexamita);纤毛原虫:如Ichthyophthirius;厚壳桂虫,斜管虫属和车轮虫属;以及单殖亚纲:表皮和鳃吸虫,例如三代虫属和指环虫属。
出乎意料的是证明了还能防治外细菌病,例如由于革兰氏阴性菌和革兰氏阳性菌(如气单胞菌、假单胞菌和屈挠细菌)引起的所有表皮和鳃的感染均能防治。
在系统治疗外寄生虫病和外细菌病的情况下,使用的活性物质具有适当的抗菌谱活性时,其副作用当然也同时能治疗由寄生虫和细菌引起的所有观赏鱼和有用鱼的系统性体内各种疾病。
意想不到的是在一开始时就可将前面已经提到的仅用于浴池疗法的所有各种活性物质用于系统治疗外寄生虫病和外细菌病,在特殊的情况下也可使用肠用药物组合物但仅用于治疗内寄生虫病,尤其是肠寄生虫病。
仅用于浴池疗法的可使用三苯甲烷衍生物,例如孔雀绿(其盐例如氯化物、草酸盐和硫酸盐);孔雀绿甲醇碱;亮绿(各种盐);结晶紫(各种盐);龙胆紫(各种盐);副品红(各种盐、碱);品红(啬薇苯胺)(各种盐);以及所有阳离子染料的无色化合物。除此之外,还有硫堇衍生物(除亚甲兰B外);含低分子量的N-甲基化衍生物的硫堇衍生物;以及硫堇衍生物的无色化合物;还有硝基噻唑衍生物,例如2-氨基-5-硝基噻唑;以及过渡金属衍生物,例如铜盐和银盐以及它们的复合物,铜金属胶体和银金属胶体;以及所有抗菌素季铵盐。
此外,有些活性化合物可用于所述口服治疗的药物组合物,但决不能用于治疗外寄生虫病,因其用法还未知道。这类化合物有吖啶衍生物,例如吖啶黄素,9-氨基吖啶,疟涤平,利凡诺,以及所有吖啶衍生物的还原前体;亚甲兰B;硝基呋喃衍生物,例如nifurpirinol;喹啉衍生物,例如喹宁(各种盐、碱);醛类,例如甲醛及其衍生物,如六亚甲基四胺;enheptine和N-乙酰衍生物acimitrazole;以及盐酸3,6-二氨基-10-甲基吖啶和吖啶黄素。
用作上述抗外寄生虫和抗外细菌活性物质的基质或载体,通常可将下列各种形式的载体或饲料类型用于观赏鱼和有用鱼,即薄膜状饲料,毛屑状饲料,碎片状饲料,压榨饲料,膨胀饲料,颗料饲料,碎屑饲料,片状饲料,饲料糊和饲料悬浮液,饲料胶和肉冻状饲料或富含吸引剂或诱饵的其他载体。
将治疗活性物质加入到在工艺上适宜的生产过程的预备阶段,例如加入到原料粉末混合物及其含水工艺阶段,从而将活性物质分布在已经尽可能研磨成均匀的细粉末状的粉末中。
我们业已发现,上述大多数活性物质尤其以阳离子形式存在时(例如孔雀绿,吖啶盐,季铵盐等),通过载体物质或饲料能够产生非常强的吸附结合。这种活性物质固定的结果可不必再用水浸提,并且使用者的手也不需要脱色。这种方法在使用时非常安全,即使在使用对人体有毒害或危险的情况下也很安全。因此,进行生产、包装、贮藏和治疗时不需要进行安全测量。使用阳离子染料(例如无色化合物或不溶于水的其他衍生物)的还原前体时也可获得类似的优点。

本发明的各种药用饲料都能治疗外寄生虫病和外细菌病,由于各种类型的观赏鱼(淡水和海水)和有用鱼(淡水和海水)都是处在常规的封闭和非封闭的养殖系统中,所以治疗的副作用也能系统治疗由内寄生虫和内细菌引起的各种系统疾病。甚至处于天然环境中的鱼,通过喂给该饲料也能进行治疗。

药用饲料的生产:本发明的药用饲料可采用专业人员熟悉的方法进行生产,即在最终制成毛屑状、碎片状、片状、胶囊状或挤压物饲料前,按所需量的活性物质与饲料混合物混合,或将活性物质的溶液喷在最终的饲料上,最后进行干燥。
活性试验是将含有0.2(重量)%孔雀绿的观赏鱼用毛屑状饲料喂给患有外寄生虫病的观赏鱼,不加药用饲料的作为对照,结果如下:
1.Ichthyophthirius的治疗。
30条大泥鳅(Botia macracautha)和30条thread鱼(Trichogastertrichopteri)经Ichthyophthirius严重感染后,每天一次喂给含孔雀绿的药用饲料。与未处理的对照相比,5天内治疗成功率达100%。
2.Costia和Chilodonella的治疗

3.其他外寄生虫病的治疗
按类似上述实施例的方法,喂给所述含孔雀绿的药用饲料,能被完全治愈的外寄生虫病有Oodinium pillulare,Trichodina spec.和Gyrodactylus。
用上述其他药用饲料也相应获得成功的治疗。
用所述药用饲料治疗外寄生虫病的特殊优点在于鱼完全没有受到伤害:在治疗期间和治疗后,受过治疗的鱼完全没有患小病或受伤害的征状。
此外,三苯甲烷染料(例如亮绿和孔雀绿)抗革兰氏阳性菌的效果还使本发明药用组合物中这些活性物质有可能治疗革兰氏阳性病原菌引起的内外细菌病,例如棒状杆菌,诺卡氏菌和分枝杆菌。
已知结晶紫和龙胆紫抗人体和哺乳动物的内蠕虫病的效果也可将其用于防治鱼的内、外蠕虫病,这些病例如是由单殖亚纲(例如三代虫属和指环虫属)、多节绦虫亚纲、吸虫纲(例如复口吸虫属)和线虫纲(例如毛线虫属)引起的。
另一个综合治疗的基本方法在于将本发明的药用饲料与治疗剂浴池结合使用,在这种情况下,将有效治疗量的类似或其他抗寄生虫或抗细菌活性物质加到水中。这种结合疗法也有可能用于防治非寄生虫,自由游动的或牢固固定在植物,石头或任何表面上的不同阶段的病原菌。由此产生的优点则使系统部分或完全消除感染,降低由自由游动阶段的病原菌引起的重新感染率,以及使用药物组合物从体内和使用治疗浴池从体外防治鱼上的寄生虫病原菌。
最后,必须看到,在通过水另外再施用活性物质时,则本文开始时所述的浴池疗法的基本缺点仍然存在。

Claims (4)

1.一种用于制备含有至少一种活性物质或活性物质结合物和一种载体或饲料混合物的药用饲料的方法,其中或者
(A)将所述活性物质或活性物质结合物与所述载体或饲料混合物混合后,最终以所需量加工成毛屑状、碎片状、片状、胶囊状、压榨物等,或
(B)将活性物质或活性物质结合物的溶液喷在最终形成的饲料产品上,然后进行干燥;
其特征在于,所述活性物质或活性物质结合物包含浓度为0.005-10%(重量)的至少一种下列物质本身,或与其他活性物质相结合:孔雀绿、亮绿、龙胆紫、结晶紫、副品红、品红、利凡诺、奎宁、六亚甲基四胺、过渡金属化合物和/或季铵盐,并且所述活性物质或活性物质结合物与载体或饲料物质的结合方式使得其不会被水冲掉。
2.权利要求1所述的方法,其特征在于所述活性物质的量的比例为0.05-3%(重量)。
3.权利要求1或2所述的方法,其特征在于所述活性物质为孔雀绿或其适宜的盐或衍生物。
4.权利要求1或2所述的方法,其特征在于所述活性物质为奎宁或其适宜的盐或衍生物。
