Abstract

本发明涉及分子生物学领域,公开了一种粪便样品稳定液,稳定液中成分如下:0.02M‑4M硫氰酸胍(Guanidine thiocyanate)、pH=5‑10且浓度为2‑20M的Tris‑HCI、0.05‑0.1mM NaCI、1‑15mM EDTA。本发明可有效保持粪便样品中被检测成分的稳定性,使病人在家中自助样品提取成为可能,该粪便稳定液不会有碍下游检测(核酸提取和检测),粪便和稳定液的混合物可以直接进入检测流程,极大地方便了粪便样品的后续处理流程灵活性,有助于消化系统疾病的检测和治疗。

粪便样品稳定液及制备方法与应用
技术领域
本发明涉及分子生物学领域,尤其涉及了一种粪便样品稳定液。
背景技术
目前,医院或体检中心的粪便化验,样品因不稳定而不能保存,必须立即送检。这给病人造成了一定的不便,给医护人员增加了工作负担,同时,也给检测实验室也增加了运作和及时提供结果的难度。因此病人配合提供样品的比例下降,使后续相关的医疗手段的有效性也受到影响。
发明内容
本发明的目的是提供一种能够有效保持粪便样品中被检测成分的稳定性,无毒或腐蚀成分,不妨碍下游检测,操作安全方便的粪便样品稳定液。
为了解决上述技术问题,本发明通过下述技术方案得以解决:
一种粪便样品稳定液,所述的稳定液由Tris(三羟甲基氨基甲烷)、硫氰酸胍(Guanidine thiocyanate)、乙二胺四乙酸(EDTA)、氯化钠及水组成;所述硫氰酸胍的终浓度为0.02M-4M,Tris的终浓度为2-20mM,氯化钠的终浓度为0.05-0.1mM、乙二胺四乙酸的终浓度为1-15mM。
上述粪便样品稳定液的制备方法如下:
将Tris溶于水中,用盐酸调节pH为5-10,制得Tris-HCI溶液;将乙二胺四乙酸溶于水,用盐酸调节pH为5-10,制得EDTA溶液;将氯化钠溶于水中,并加入硫氰酸胍、Tris-HCI溶液、EDTA溶液混合后,杀菌,得到粪便样品稳定液。
取按照上述配比配制的粪便样品稳定液,加入与稳定液等体积的粪便样品,混匀;控制温度在50摄氏度以内,保存。
本发明由于采用了以上技术方案,具有显著的技术效果:
本发明可有效保持粪便样品中被检测成分的稳定性,使病人在家中自助样品提取成为可能,该粪便稳定液不会有碍下游检测(核酸提取和检测),粪便和稳定液的混合物可以直接进入检测流程,极大地方便了粪便样品的后续处理流程灵活性,有助于消化系统疾病的检测和治疗。

具体实施方式
下面结合附图1与实施例对本发明作进一步详细描述:
实施例1
配制粪便样品稳定液,配方如下:
0.02-4M硫氰酸胍(Guanidine thiocyanate),2-20mM Tris-HCI(pH=5-10),0.05-0.1mMNaCI,1-15mM EDTA。
其制备方法如下:
将Tris溶于水中,用盐酸调节pH为5-10,制得Tris-HCI溶液;将乙二胺四乙酸溶于水,用盐酸调节pH为5-10,制得EDTA溶液;将氯化钠溶于水中,并加入硫氰酸胍、Tris-HCI溶液、EDTA溶液混合后,杀菌,得到粪便样品稳定液。
取配制好的粪便样品稳定液,加入与稳定液等体积的粪便样品,混匀;控制温度在50摄氏度以内,保存。
对比实验:不添加本发明粪便样品稳定液,室温条件下,不同保存天数,检测所提取的DNA含量,见下表:

由上表可见,不加粪便样品保存液的粪便样品,在室温下保存0天,1天,3天,5天用同样方法用等量粪便提取总DNA,体积一样,测DNA含量,在常温下保存三天后DNA提取量出现明显下降。
本发明说明书附图1显示一份粪便样品经收集并加入本发明稳定液后,在摄氏50度放置,于第3,5,7天提取核酸,然后用荧光实时PCR检测其中肌动蛋白基因(Actin)的结果。其中呈弧形的曲线在大约相同位置跨过绿色水平基本底线,表明样品中的肌动蛋白基因一直稳定,检测结果等同,结果表明,本发明稳定液可有效长期保持粪便样品中被检测成分的稳定性。
总之,以上所述仅为本发明的较佳实施例,凡依本发明申请专利范围所作的均等变化与修饰,皆应属本发明专利的涵盖范围。

Claims (3)

1.一种粪便样品稳定液,其特征在于:所述的稳定液由Tris、硫氰酸胍、乙二胺四乙酸、氯化钠及水组成;所述硫氰酸胍的终浓度为0.02M-4M,Tris的终浓度为2-20mM,氯化钠的终浓度为0.05-0.1mM、乙二胺四乙酸的终浓度为1-15mM。
2.一种如权利要求1所述的粪便样品稳定液的制备方法,其特征在于:
将Tris溶于水中,用盐酸调节pH为5-10,制得Tris-HCI溶液;将乙二胺四乙酸溶于水,用盐酸调节pH为5-10,制得EDTA溶液;将氯化钠溶于水中,并加入硫氰酸胍、Tris-HCI溶液、EDTA溶液混合后,杀菌,得到粪便样品稳定液。
3.一种如权利要求1所述的粪便样品稳定液的应用,其特征在于:取配制好的粪便样品稳定液,加入与稳定液等体积的粪便样品,混匀;控制温度在50摄氏度以内,保存。
