Abstract

本发明涉及用于促进试剂(agent)(例如核酸)进入细胞的方法和反应试剂(reagent)的领域。
核酸和其类似物在基础研究以及预防性和治疗性干预中有许多应用。然而,原核和真核细胞对于核酸/类似物通常是不可渗透的。通常核酸/类似物的摄取需要病毒或非病毒递送载体,以激发它们的潜能(potential)。为了进行得有效,递送载体/核酸或类似物的组合或复合必须克服许多挑战,例如细胞外降解、细胞膜屏障的穿透(cell membrane barrier penetration)和胞内释放,同时使细胞毒性和抗原性最小化。许多原本很有用的多肽(例如蛋白质、肽)和其它生物活性分子也是细胞不可渗透的。例如许多化合物的活性在生化分析中比在细胞分析或活体内的活性要高。细胞活性的下降被认为是主要是由于向细胞内的递送不良。
脂质转染胺(lipofectamine)是广泛使用的用于促进试剂(例如核酸)进入细胞的试剂的实例。然而,脂质转染胺被认为对特别是哺乳动物细胞有毒性作用,这可以例如使体外实验难以评价且活体内实验难以执行。
其它被提出的促入剂(entry-promoting agent)描述在例如WO2009/015143(Biodegradable cationic polymer gene transfer compositions andmethods of use);WO02/22174(作为生物相容性基因递送剂的阳离子脂质聚合物(cationic lipopolymer as biocompatible gene delivery agent));WO2010/086406(非病毒转染剂);US5,958,894(两亲性双胍衍生物)中。
需要替代性的促进进入的试剂(促入剂),用于例如核酸和其它物质,当活性受限于较差的细胞进入时。本发明识别出了促进进入的试剂和方法。这样的促进进入的试剂和方法被认为是有益于例如为特别是真核细胞提供良好的进入促进、较弱的抗原性、低成本和/或低毒性。这样的促进进入的试剂和方法可能例如在诸如以下等领域是有用的:核酸/类似物的转染,例如功能的研究;稳定细胞系的产生;基因沉默;和DNA疫苗。如本领域普通技术人员将容易理解的,这样的促入剂和方法可能在相关的RNA干扰(RNAi)或其它反义技术中是有用的。递送反应试剂或药物至细胞中的的挑战并不仅仅局限于核酸。蛋白质和肽通常也很难进入细胞,促入技术也是有用的。而且,许多被描述为小于1000克/摩尔的小分子也难以进入细胞,而通过改进的细胞递送技术将会提高它们作为试剂或药物的效用。
PHMB(聚六亚甲基双胍)是已知的作为安全有效的杀生物剂(biocidalagent),并被用作消毒剂(sanitiser)和防腐剂:US7897553、US4758595、US2008261841;US20040009144。本申请的发明者已经出人意料地发现PHMB和相关的分子是有用的促入剂。出人意料地观察到PHMB(举例)本身进入多种细胞,包括细菌、真菌和哺乳动物细胞。更出人意料地是,PHMB(举例)能够与多种分子形成纳米颗粒并将这些分子递送至这些细胞内。最后发现所递送的从核酸至小分子的分子在细胞内是能发挥作用的。

本发明的进一步的方面提供了用于制备复合物的方法,所述复合物包括如以上所定义的促入剂(例如PHMB)和被引剂,所述方法包括在复合(complexation)缓冲液中温育所述促入剂和被引剂,例如在高pH,例如在10-13.5的pH。认为形成的纳米颗粒包括所述促入剂(例如PHMB)和所述被引剂,例如寡核苷酸聚合物(DNA,PNA,siRNA),蛋白质、肽和小分子。具体地,在与细胞一起使用之前,纳米颗粒的形成可以通过在合适的缓冲液中温育PHMB和如以上所描述的类似的分子与寡核苷酸、蛋白质、肽和小分子来实现。合适的温育缓冲液可以包括水、PBS和在实验室中常用的其它缓冲液。以上论述了高pH缓冲液。最佳的缓冲液可以取决于所述促入剂和所述递送的分子的具体的特性,如对本领域技术人员来说将是显而易见的。纳米颗粒的形成和细胞递送通常通过在混合所述两种成分之前在复合缓冲液中稀释配偶体分子(partner molecules)来实现。而且,混合所述两种组分通常在结合其它赋形剂或其它活性成分以及应用于细胞或在活体内使用之前进行。认为高效的纳米颗粒的形成发生在几秒或几分钟内,但所述程序可以进行几个小时。用于高效的纳米颗粒形成的合适的比例随着不同的搭档组合物而变化。例如,1-20:1(wt:wt)的PHMB:质粒DNA提供高效的纳米颗粒的形成。以上给出了导致纳米颗粒形成的PHMB;DNA组合(combinations)的实例。当使用不同的配偶体分子比例时,本领域技术人员能够评估纳米颗粒的形成和递送效率。可以用许多方式评估纳米颗粒的形成。例如,本领域技术人员个人将能够利用动态光散射(DLS)和显微镜方法来评估纳米颗粒的形成。
本发明的进一步的方面提供了包含促入剂(例如PHMB)和如以上所定义的被引剂的复合物,其中,通过包括在例如高pH(例如10-13.5的pH)的复合缓冲液中温育所述促入剂和所述被引剂的方法,所述复合物是可获得的。

实施例1:PHMB和相关的阳离子聚合物作为载体用于递送到细菌、真菌和哺乳动物细胞中。
我们已经评估了PHMB和相关分子作为载体用于体素到原核和真核细胞。还没有出现一种技术作为实用性的基因转移方法用于临床。载体的理想的特性包括:
·它应该能够与生物分子相互作用
·自身以及在转载有生物分子时进入
·一旦进入就释放生物分子的能力
·低细胞毒性且适于活体内应用。
·瞬时转染的高转染效率是用于研究的重要特征,如低细胞毒性、和用于难以转染细胞的效率。
我们认为PHMB和相关的分子可以能够充当有效的载体。聚六亚甲基双胍(PHMB)是为广谱抗菌剂且对哺乳动物细胞具有更小的毒性的阳离子聚合物(VantocilTM Arch biocides limited,英国)。在递送试剂中,低毒性是不寻常的且是非常有益的性质,且在广泛的应用中,这些化合物的所观察到的低毒性和安全记录是该技术的特征。
n=2to40,Bx529VANTOCIL100的重均分子量(Mw)和多分散性分别是3035和1.9。这样的Mw对应约14(更精确地13.8)的“n”值。端基:胺、胍和氰基胍。聚六亚甲基单胍(PHMG)是具有抗菌性质的PHMB的类似物(Zhou等,2010)。
PHMB的当前用途:
·用作消毒剂,手术伤口敷料的建议的选择(Stephen Gilliver2009)。
·存在于消毒剂、游泳池消毒剂、固体表面清洁剂、口腔清洗剂和隐形眼镜洗液(Lucas等,2009)
·用于处理种蛋(hatching eggs)、作为化妆品和纺织品中的除臭剂和防腐剂、和用于处理冷却系统以防止军团菌等
我们认为PHMB作为载体是有用的,该载体要么作为共价结合物(covalent conjugate)要么作为非共价复合物,可能具有疏水和/或静电相互作用。如果是这种情况,PHMB的其它性能可以增强它的作为载体的用途。已经很好地研究了PHMB的毒性特征(toxicity profile)(Muller等,2008;Kathryn V.Montague,2004),
且根据在人类患者中的试验,认为PHMB是弱的过敏原(理想的性质)(Schnuch等,2007)。抗菌性能也可以有利于减少细胞培养中的污染问题。

PHMB和类似物能够递送蛋白质分子(例如R-藻红蛋白或结合IgG抗体的alexa488)到诸如海拉细胞等哺乳动物细胞中,参见图35。
实施例4.PHMB与核酸分子快速地形成复合物,且高pH提高转染效率。
我们的方法显示了当载体和装载物在具有高pH(13.5-10)的缓冲液中复合时,可以实现更高的转染效率。我们测试了用于PHMB和核酸的复合物的形成所需的时间量。我们的结果显示仅仅5分钟已足够于形成能够进入哺乳动物细胞的复合物,或甚至允许所述复合物在室温下保持几小时也不会损害所述转染能力。参见图34。

Claims (28)

G1和G2独立地表示包括双胍或胍的阳离子基团,其中,L1和L2直接连接到所述胍的N原子;
L1和L2是所述聚合物中的G1和G2阳离子基团之间的连接基团,且独立地表示包含C1-C140碳原子的脂肪族基团,例如烷基基团,诸如:亚甲基、亚乙基、亚丙基、C4、C5、C6、C7、C8、C9或C10;C1-C10、-C20、-C30、-C40、-C50、-C60、-C70、-C80、-C90、-C100、-C110、-C120、-C130或-C140烷基;或C1-C140(例如,C1、C2、C3、C4、C5、C6、C7、C8、C9或C10;C1-C10、-C20、-C30、-C40、-C50-C60、-C70、-C80、-C90、-C100、-C110、-C120、-C130或-C140)、脂环基、杂环基、芳香族基、芳基、烷基芳基、芳基烷基或氧化烯基;或聚亚烷基基团,所述聚亚烷基基团可选地由一个或多个的优选为一个的氧、氮或硫原子、官能团或饱和或不饱和的环部分中断;
N和G3为可选择的端基;
X可以是存在或不存在的;
L3、L4和X是所述聚合物中的G4和G5阳离子基团之间的连接基团,且独立地表示含有C1-C140碳原子的脂肪族基团,例如烷基基团,诸如:亚甲基、亚乙基、亚丙基、C4、C5、C6、C7、C8、C9或C10;C1-C10、-C20、-C30、-C40、-C50、-C60、-C70、-C80、-C90、-C100、-C110、-C120、-C130或-C140烷基;或L3和L4和X可以独立地是C1-C140(例如,C1、C2、C3、C4、C5、C6、C7、C8、C9或C10;C1-C10、-C20、-C30、-C40、-C50、-C60、-C70、-C80、-C90、-C100、-C110、-C120、-C130或-C140)、脂环基、杂环基、芳香族基、芳基、烷基芳基、芳基烷基、氧化烯基;或聚亚烷基基团,所述聚亚烷基基团可选地由一个或多个的优选为一个的氧、氮或硫原子、官能团以及饱和或不饱和的环部分中断;
“G4”和“G5”是阳离子部分且可以是相同或不同的,且它们中的至少一个是双胍部分或氨基甲酰基胍,且另一部分可以是双胍或氨基甲酰基胍或胺;
且阳离子部分G4和G5不包含单个的胍基团。
2.如权利要求1所述的方法,其中所述式1a的促入剂包括下述一种或多种或由下述一种或多种组成
或所述式1b的促入剂包括或由以下物质中的一种或多种组成:聚(烯丙基双胍基-共-烯丙胺)、聚(N-乙烯基双胍)、聚(烯丙基氨基甲酰基胍基-共-烯丙胺)或聚烯丙基双胍。
3.如权利要求1所述的方法,其中,所述细胞是原核细胞。
4.如权利要求1所述的方法,其中,所述细胞是真核细胞,所述真核细胞可选地是在组织或器官中的真核细胞。
5.如权利要求1、2或4中任一项所述的方法,其中,所述细胞是哺乳动物细胞。
6.如前述权利要求中任一项所述的方法,其中,所述被引剂包括核酸或核酸类似物。
7.如权利要求6所述的方法,其中,所述被引剂包括质粒DNA。
8.如权利要求6所述的方法,其中,所述被引剂包括RNAi分子。
9.如权利要求1-5中任一项所述的方法,其中,所述被引剂包括多肽、可选地包括肽或蛋白质。
10.如权利要求1-5中任一项所述的方法,其中,所述被引剂包括小分子药物、生物活性试剂或细胞成像探针/造影剂。
11.如前述权利要求中任一项所述的方法,其中,所述被引剂和所述促入剂是共价地连接的。
12.如权利要求1-10中任一项所述的方法,其中,所述被引剂和所述促入剂作为制剂提供,例如作为非共价复合物提供,可选地作为纳米颗粒提供。
13.如权利要求1-12中任一项所述的方法,其中,所述方法在体外执行。
14.如权利要求1-12中任一项所述的方法,其中,所述方法在活体内执行。
15.如权利要求1-13中任一项所述的方法,其中,所述方法在先体外后体内执行。
16.如权利要求12所述的方法,其中,所述方法用被引剂相对于促入剂过量达100倍摩尔比至促入剂相对于被引剂过量达1000倍摩尔比来执行。
17.用于制造目标多肽的方法,所述方法包括从宿主细胞的细胞培养来制备目标多肽的步骤,其中,所述宿主细胞是已经通过外源核酸分子转化的宿主细胞(或它的祖细胞已经被转化),以使所述细胞合成所述目标多肽,其中,所述转染包括在如前述权利要求的任一项中所定义的促入剂存在下,将所述宿主细胞暴露于所述外源核酸分子。
18.如前述权利要求的任一项中所定义的促入剂和如前述权利要求的任一项中所定义的被引剂,用于对需要所述被引剂的对象进行治疗。
19.如前述权利要求的任一项中所定义的促入剂和如前述权利要求的任一项中所定义的被引剂在制造用于治疗需要所述被引剂的对象的药物中的用途。
20.治疗需要如前述权利要求的任一项中所定义的被引剂的对象的方法,所述方法包括用如前述权利要求的任一项中所定义的促入剂和所述被引剂治疗所述对象的步骤。
21.如前述权利要求的任一项中所定义的促入剂和如前述权利要求的任一项中所定义的被引剂在细胞内分子的检测中的用途,可选地通过原位杂交检测细胞内分子,可选地在所述递送的核酸是标记有荧光团或其它可检测的分子的情况下。
22.组分试剂盒(a kit of parts)或组合物,所述组分试剂盒或组合物包含如前述权利要求的任一项中所定义的促入剂和被引剂,所述组分试剂盒或组合物可选地包含复合物,其中,所述复合物是通过包括在复合缓冲液中混合或温育所述促入剂和所述被引剂的方法可获得的(或是通过该方法获得的),例如是在高pH下,所述高pH例如为10-13.5的pH。
23.本发明的组合物或组分试剂盒,所述组合物或组分试剂盒用于在成像或诊断方法中使用。
24.药物组合物,所述药物组合物包括如前述权利要求的任一项中所定义的促入剂和被引剂。
25.试剂组合物,所述试剂组合物包括如前述权利要求的任一项中所定义的促入剂和被引剂。
26.权利要求1或2任一项中所定义的促入剂用于促进生物活性物质的细胞递送的用途。
27.如权利要求1-16中任一项所述的方法,其中,所述促入剂和递送的试剂分子在具有高pH的缓冲液中已经混合或温育或一起提供,所述高pH可选地为10-13.5的pH。
28.用于制备复合物的方法,所述复合物包括如前述权利要求的任一项中所定义的促入剂(例如PHMB)和如前述权利要求的任一项中所定义的被引剂,所述方法包括在复合缓冲液中混合或温育所述促入剂和所述被引剂,可选地在高pH下,例如10-13.5的pH下。
