Abstract

本发明公开了一种富含β-乙氧基芸香糖胶囊的生产方法。具体方法包括:经酶转化的高β-乙氧基芸香糖含量的苦荞麦原料,经乙醇提取,色谱柱层析分离,真空浓缩,喷雾干燥,造粒,灌制胶囊工序。所得胶囊产品中β-乙氧基芸香糖的含量大于20%,并具有降低血糖的作用。

Description

一种富含β-乙氧基芸香糖胶囊的生产方法
技术领域
本发明涉及食品、保健食品生产技术领域,尤其涉及到一种富含β-乙氧基芸香糖胶囊的生产方法。 
背景技术
苦荞麦(Fagopyrum tataricum(Linn)Gaertn)为蓼科荞麦属植物苦荞麦的种子,起源于中国,具有降血糖、降血压、降血脂的功能,被认为是三降食品。 
从我们的研究中发现,苦荞麦中降血糖的活性成分主要归结于β-乙氧基芸香糖(Ethanol -β-rutinoside)的降血糖作用。该物质在苦荞麦中的含量相对是低的,在苦荞麦中天然的含量不足千分之一,要实现产业化有较大难度。深入研究发现β-乙氧基芸香糖在酶的作用下可以由芦丁转化而来,在最佳条件下β-乙氧基芸香糖的含量能提高10多倍,达到1%的含量,再经提取富集使β-乙氧基芸香糖的含量提高到20%,生产出以β-乙氧基芸香糖为功能成分的降血糖胶囊产品,为苦荞麦的深加工奠定基础。 
发明内容
本发明针对上述现状,将经酶转化的高β-乙氧基芸香糖含量的原料,通过提取、层析分离,喷雾干燥,造粒、灌装生产出β-乙氧基芸香糖含量大于20%的胶囊产品。为达上述目的,本发明采用如下的技术方案: 
1、β-乙氧基芸香糖的提取与富集:将经酶转化的高β-乙氧基芸香糖含量的苦荞麦原料,按料液比1∶10(W/V)加入65-70%的乙醇,室温下提取2小时,过滤,渣再重复提取一次,过滤,合并二次的滤液。减压浓缩至1/10体积,浓缩液加入一倍体积的去离子水后,过大孔树脂柱AB-8或X-5,用水,5%,10%,90%乙醇洗脱,收集10%乙醇洗脱液并减压浓缩至1/10体积,将该浓缩液上MCI柱层析,分别用水,10%,20%,90%乙醇洗脱,用薄层层析法检测,收集含β-乙氧基芸香糖的洗脱液。 
2、浓缩:将含β-乙氧基芸香糖的洗脱液减压浓缩至比重为1.2左右,取出得β-乙氧基芸香糖浓缩物。 
3、调配:按β-乙氧基芸香糖浓缩物中干物质量的10%加入木糖醇,按β-乙氧基芸香糖浓缩物中干物质量的10%加入糊精或麦芽糊精,均质乳化30分钟并保持60℃。 
4、喷雾干燥:将调配好的物料在进风温度160-170℃,出风温度83-85℃条件下喷雾干燥,控制含水量小于5%,得β-乙氧基芸香糖干燥粉。 
5、造粒:按β-乙氧基芸香糖干燥粉30%的量加入95%乙醇,搅拌均云后,湿法造粒。60-65℃真空干燥至含水量小于5%。取出后先过30目,取筛下物,再过80目取筛上物。 
6、胶囊填充:取80目筛上物灌入胶囊中同,每粒胶囊干物质量350mg。 
具体实施方式
下面结合实施例对本发明作进一步详细的描述,但本发明的实施方式不限于此。 
实施例1: 
将5kg经酶转化的高β-乙氧基芸香糖含量的苦荞麦原料,置于100升提取罐中,加50升70%乙醇,室温提取2小时,残渣再提取一次。合并乙醇提取液,得滤液97升,浓缩到10升,再加入10升去离子水,然后通过大孔树脂柱AB-8(15×180cm)柱,先用10升去离子水洗脱,再用10升5%乙醇洗脱,然后用10升10%乙醇洗脱、后用90%乙醇洗脱。收集10%乙醇洗脱液并减压浓缩,得浓缩物约2升。将2升浓缩物上MCI柱层析(5×150cm),分别用5升去离子水洗脱、5升10%、5升20%乙醇洗脱,用薄层层析法检测,收集含β-乙氧基芸香糖的洗脱液,浓缩至比重为1.2左右,取出加入木糖醇25克,糊精或麦芽糊精25克,在进风温度168℃,出风温度85℃条件下喷雾干燥,得β-乙氧基芸香糖干粉287克。在β-乙氧基芸香糖干粉中加入86克95%乙醇,搅拌均匀后,湿法造粒。65℃真空干燥至含水量小于5%,取出后先过30目,取筛下物,再过80目,取筛上物,得β-乙氧基芸香糖提取物247克,灌入胶囊。 
以上较佳实施例仅用于说明本发明的内容,除此之外,本发明还有其他实施方式,但凡本领域技术人员因本发明所涉及之技术启示,而采用等同替换或等效变形方式形成的技术方案均落在本发明的保护范围内。 

Claims (7)

1.一种富含β-乙氧基芸香糖胶囊的生产方法,包括乙醇提取,层析分离,浓缩,喷雾干燥,造粒,灌制胶囊工序。
2.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:将经酶转化的高β-乙氧基芸香糖含量的苦荞麦原料,按料液比1∶10(W/V)加入65-70%的乙醇,室温下提取2小时,过滤,渣再重复提取一次,过滤,合并二次的滤液。减压浓缩至1/10体积,浓缩液加入一倍体积的去离子水后,过大孔树脂柱AB-8或X-5,用水,5%,10%,90%乙醇洗脱,收集10%乙醇洗脱液并减压浓缩至1/10体积,将该浓缩液上MCI柱层析,分别用水,10%,20%,90%乙醇洗脱,用薄层层析法检测,收集含β-乙氧基芸香糖的洗脱液。
3.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:将含β-乙氧基芸香糖的洗脱液减压浓缩至比重为1.2左右,取出得β-乙氧基芸香糖浓缩物。
4.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:按β-乙氧基芸香糖浓缩物中干物质量的10%加入木糖醇,按β-乙氧基芸香糖浓缩物中干物质量的10%加入糊精或麦芽糊精,均质乳化30分钟并保持60℃。
5.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:将调配好的物料在进风温度160-170℃,出风温度83-85℃条件下喷雾干燥,控制含水量小于5%,得β-乙氧基芸香糖干燥粉。
6.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:按β-乙氧基芸香糖干燥粉30%的量加入95%乙醇,搅拌均云后,湿法造粒。60-65℃真空干燥至含水量小于5%。取出后先过30目,取筛下物,再过80目取筛上物。
7.根据权利要求1所述的富含β-乙氧基芸香糖胶囊的生产方法,其特征在于:取80目筛上物灌入胶囊中同,每粒胶囊干物质量350mg。
