发明内容
本发明提供一种健脾补肾龟苓膏,包括以下重量份的成分:
其中山药用纤维素酶酶解处理,黑豆用纤维素酶和蛋白酶酶解处理,其它7种原料用煎煮法处理。
本发明还提供一种健脾补肾龟苓膏的制备工艺,包括以下步骤:
1.将龟甲洗净、破碎后,投入水中浸泡2-4小时;
2.按5-10万U/g龟甲的量加入蛋白酶,搅拌混匀,酶解1-4小时后,加热使蛋白酶灭活,然后趁热过滤,得到酶解龟甲胶原蛋白肽;
3.将黑豆洗净、破碎后投入水中浸泡1-2小时;
4.按4-10万U/g黑豆的量加入纤维素酶和蛋白酶,酶解2-4小时,加热使纤维素酶和蛋白酶灭活,得到酶解黑豆蛋白肽和水溶性黑豆花色素;
5.将干片山药破碎,投入水中浸泡2-4小时;
6.按4-10万U/g山药的量加入纤维素酶,酶解2-4小时,加热使纤维素酶灭活,得到酶解山药提取物;
7.称取仙草、茯苓、金银花、蒲公英、甘草、益智仁、枸杞七味原料,投入水中,煎煮1-2次,每次煎煮1-2小时;合并煎煮液,离心过滤,得出煎煮提取液;
8.合并酶解龟甲蛋肽、黑豆蛋白肽、水溶性黑豆花色素、山药提取物和煎煮提取液,浓缩后加入矫味剂、稳定剂,加热搅拌至糊化后,分装、灭菌,得出龟苓膏成品。
优选的,步骤1)、步骤3)和步骤5)中将龟甲、黑豆、山药洗净破碎至细度小于2×2mm,水的用量为龟甲、黑豆和山药重量的5-10倍。
优选的,所述蛋白酶为木瓜蛋白酶和/或菠萝蛋白酶。

实施例1
1)将1kg龟甲洗净,破碎至细度小于2×2mm,然后投入5L的水中浸泡2小时;
2)缓慢加入木瓜蛋白酶(80万U/g)0.25kg和菠萝蛋白酶(80万U/g)0.25kg,边加边搅拌混匀;
3)调pH至5.0,升温至50℃,保温3小时后,加热至100℃使酶灭活,然后趁热过滤,得到滤液,即酶解龟甲胶原蛋白肽;
4)将1.5kg黑豆洗净,破碎至细度小于2×2mm,然后投入8L的水中浸泡2小时;
5)缓慢加入纤维素酶(20万U/g)1.25kg和木瓜蛋白酶(80万U/g)0.5kg,边加边搅拌混匀;
6)调节pH至5.0,升温至50℃,保温2.5小时后,加热至100℃使酶灭活,然后趁热过滤,得到滤液,即是黑豆花色素和黑豆蛋白肽;
7)将1kg山药洗净,破碎至细度小于2×2mm,然后投入5L的水中浸泡2小时;
8)缓慢加入纤维素酶(20万U/g)1.0kg,边加边搅拌混匀;
9)调节pH至5.0,升温至50℃,保温2.5小时后,加热至100℃使酶灭活,然后趁热过滤,得到滤液,即是山药提取物;
10)取仙草5kg、茯苓4kg、金银花2kg、蒲公英2kg、益智仁0.7kg,枸杞0.35kg、甘草0.5kg,置于多功能提取罐中煎煮二次,每次加120L的水、煎煮1小时;合并煎煮液,离心过滤,得到滤液,即煎煮提取液;
11)合并酶解龟甲胶原蛋白肽、黑豆花色素、黑豆蛋白肽山药提取物和煎煮提取液,加入80kg冰糖、20kg淀粉,定容至500L,边加热边搅拌,待糊化后,分装,然后经高温灭菌,冷却后即得到龟苓膏成品。
实施例2

11)合并酶解龟甲胶原蛋白肽、黑豆花色素、黑豆蛋白肽、山药提取物和煎煮提取液,加入80kg冰糖、20kg淀粉,定容至500L,边加热边搅拌,待糊化后,分装,然后经高温灭菌,冷却后即得到龟苓膏成品。人体试食试验:
试验对象:排除标准:①近两个月接受过五脏治疗者;②近一个月服用过与受试功能有关的药品或保健品者;③嗜酒、吸毒者;④有严重的肾功能患者;⑤有保健品过敏史及其他严重过敏史者。
试验分组:20名志愿者(女8人,男12人),年龄30-55岁,其中有6名男性表现为尿频、四肢不温;6名男性表现为尿频、失眠、潮热盗汗;4名女性表现月性过多,四肢不温、经常盗汗;4名女性表现为尿频、腰酸、四肢发冷、畏寒;按各种症状分为受试者15名和对照组15名。受试者口服本实施例1龟苓膏产品,每日2盒(每盒200±5g),共90天;对照组试验期间不服用任何调经类的相关产品或药品。所有志愿者在试验期间正常饮食,不改变原来的饮食习惯。
测试方法:观察调查试验前后各试验者的身体情况变化。
对试食前后身体情况变化进行对比,结果受试者中7人认为小便减少、盗汗情况好转、四肢没有以前那么冷,说明整体情况开始好转,2人认为小便次数减少,1人认为腰酸现象,盗汗情况好转;对照组中3人认为小便次数减少,其它情况无好转,2人认为月经量时多时正常,经期期间腰酸、腿软,另外5人无变化。试验结果表明,本发明的龟苓膏具有健脾补肾的作用。
Claims (7)
1.一种健脾补肾龟苓膏,由以下方法制备而成:按重量份计,
1)将龟甲5-15份洗净、破碎后,投入水中浸泡2-4小时;
2)按5-10万U/g龟甲的量加入蛋白酶,搅拌混匀,酶解1-4小时后,加热使蛋白酶灭活,然后趁热过滤,得到酶解龟甲胶原蛋白肽;
3)将黑豆5-20份洗净、破碎后投入水中浸泡1-2小时;
4)按4-10万U/g黑豆的量加入纤维素酶和蛋白酶,酶解2-4小时,加热使纤维素酶和蛋白酶灭活,得到酶解黑豆蛋白肽和水溶性黑豆花色素;
5)将干片山药5-15份破碎,投入水中浸泡2-4小时;
6)按4-10万U/g山药的量加入纤维素酶,酶解2-4小时,加热使纤维素酶灭活,得到酶解山药提取物;
7)称取仙草90-98份、茯苓40-60份、金银花12-16份、蒲公英12-16份、甘草8-15份、益智仁3-5份、枸杞1-5份七味原料,投入水中,煎煮1-2次,每次煎煮1-2小时;合并煎煮液,离心过滤,得出煎煮提取液;
8)合并酶解龟甲胶原蛋白肽、黑豆蛋白肽、水溶性黑豆花色素、山药提取物和煎煮提取液,浓缩后加入矫味剂、稳定剂,加热搅拌至糊化后,分装、灭菌,得到龟苓膏成品;
所述蛋白酶为木瓜蛋白酶和/或菠萝蛋白酶。
2.一种根据权利要求1所述的健脾补肾龟苓膏的制备工艺,包括以下步骤:
1) 将龟甲洗净、破碎后,投入水中浸泡2-4小时;
2) 按5-10万U/g龟甲的量加入蛋白酶,搅拌混匀,酶解1-4小时后,加热使蛋白酶灭活,然后趁热过滤,得到酶解龟甲胶原蛋白肽;
3) 将黑豆洗净、破碎后投入水中浸泡1-2小时;
4) 按4-10万U/g黑豆的量加入纤维素酶和蛋白酶,酶解2-4小时,加热使纤维素酶和蛋白酶灭活,得到酶解黑豆蛋白肽和水溶性黑豆花色素;
5) 将干片山药破碎,投入水中浸泡2-4小时;
6) 按4-10万U/g山药的量加入纤维素酶,酶解2-4小时,加热使纤维素酶灭活,得到酶解山药提取物;
7) 称取仙草、茯苓、金银花、蒲公英、甘草、益智仁、枸杞七味原料,投入水中,煎煮1-2次,每次煎煮1-2小时;合并煎煮液,离心过滤,得出煎煮提取液;
8)合并酶解龟甲胶原蛋白肽、黑豆蛋白肽、水溶性黑豆花色素、山药提取物和煎煮提取液,浓缩后加入矫味剂、稳定剂,加热搅拌至糊化后,分装、灭菌,得到龟苓膏成品。
3.根据权利要求2所述的制备工艺,其特征在于:所述步骤1)、步骤3)和步骤5)中将龟甲、黑豆、山药洗净破碎至细度小于2×2mm,水的用量为龟甲、黑豆和山药重量的5-10倍。
4.根据权利要求2或3所述的制备工艺,其特征在于:所述木瓜蛋白酶和菠萝蛋白酶的用量为:使其酶活比值为1:1-5。
5.根据权利要求2或3所述的制备工艺,其特征在于:酶解时,pH值为4.0-6.0,温度为40-55℃。
6.根据权利要求4所述的制备工艺,其特征在于:酶解时,pH值为4.0-6.0,温度为40-55℃。
7.根据权利要求2或3所述的制备工艺,其特征在于:步骤7)水的用量为七味原料总重量的8-10倍。
