Abstract

本发明公开了鸡爪槭赤枫简易组培繁殖工艺,它包括以下步骤:S1、外植体灭菌,采集无病害的鸡爪槭赤枫当年生枝条,去掉多余叶片,切成带芽茎段进行灭菌;S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,诱导腋芽萌发;S3、生根培养,切割直接诱导的腋芽,接种于生根培养基上,诱导根产生。本发明的有益效果是:本发明建立了赤枫的组培繁殖体系,实现了赤枫繁殖体系的突破,获得了简易的组培繁殖技术,这种技术不经过愈伤组织阶段,能较快的获得组培苗,并且繁殖后能迅速的移栽成活;生产成本低廉,得到的组培苗遗传状一致,且繁殖系数高,繁殖周期短,是鸡爪槭赤枫快繁的有效途径,且避免了病毒积累。

技术领域
本发明涉及植物组织培养技术领域,特别是鸡爪槭赤枫简易组培繁殖工艺。
背景技术
槭树科(Aceraceae)槭树属(Acer)通常泛称槭树,习惯又称“枫”或“枫树”。全世界槭树共有200多种,分布于亚洲、欧洲、北美洲以及非洲北部。我国有160余种,占全世界总数的75%以上,全国各地均有分布,主产长江流域及其以南的各省区。槭树可用作建筑用材、造纸原料、药物提取、工业用油和蜜源植物。有些槭树树形丰富、姿态潇洒、叶形丰富、叶色多变,誉为世界著名的观叶植物。鸡爪槭(Acer palmatum)又名鸡爪枫,属槭树科、槭树属落叶小乔木或乔木。树姿优美秀丽,叶形美观,叶色多变,是著名的观赏树种。世界各国早已引种栽培,变种和变型很多。日本鸡爪槭目前培育出450余个赏叶品种。
我国槭树观赏资源的开发利用与国外相比有很大差异。槭树作为重要的观叶树种,栽培苗木供不应求。其繁殖目前主要采用扦插和播种繁殖方法。虽然,槭树科植物在大田里的生长有着很大的容量,但目前进行组培繁育的品种较少。
综上所述,国内外对槭树科植物的组织培养研究较少,仅有少数几个种建立了较为完善的叶腋增殖组织培养再生体系,如大槭树、美洲糖槭、尖尾槭、茶条槭、青榨槭、复叶槭。而对于扦插生根困难的鸡爪槭及其品种鲜有报道。
鸡爪槭赤枫(Acer palmatum ‘Sangokaku’)为纵多鸡爪槭栽培变异品种之一,其枝条赤红,幼叶淡黄色,秋季叶片鲜黄色或橙红色,颇具观赏价值。是现今市场上较为普遍及珍贵的观赏品种,但对于赤枫的组培繁殖方法还从未有过报道。
发明内容
本发明的目的在于克服现有技术的缺点,提供一种繁殖率高、繁殖速度快、操作简便的鸡爪槭赤枫简易组培繁殖工艺。
本发明的目的通过以下技术方案来实现:鸡爪槭赤枫简易组培繁殖工艺,它包括以下步骤:
S1、外植体灭菌,采集无病害的鸡爪槭赤枫当年生枝条,去掉多余叶片,切成带芽茎段进行灭菌,灭菌采用酒精加升汞的组合灭菌方式,先用酒精灭菌30~60s,再用升汞灭菌3~5min,然后用无菌水清洗4~6遍;
S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,所述的培养基为MS+ IAA0.5mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养2~3周,诱导腋芽萌发;
S3、生根培养,切割直接诱导的腋芽,接种于生根培养基上,生根培养基为1/2MS+NAA0.2mg/L+IAA0.1mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养2~3周,诱导根产生。
本发明具有以下优点:
本发明为采集赤枫茎段外植体,在初代培养基上诱导腋芽发生,然后直接将腋芽切入生根培养基,诱导生根。
本发明建立了赤枫的组培繁殖体系,实现了赤枫繁殖体系的突破,获得了简易的组培繁殖技术,这种技术不经过愈伤组织阶段,能较快的获得组培苗,并且繁殖后能迅速的移栽成活。
本发明生产成本低廉,得到的组培苗遗传状一致,且繁殖系数高,繁殖周期短,是鸡爪槭赤枫快繁的有效途径,且避免了病毒积累。
本发明培养基配方简单,培养流程简捷,操作简便,培养效率高,能够比较容易的获得鸡爪槭赤枫无菌苗,炼苗易成活,省时省力。

具体实施方式
下面结合附图及实施例对本发明做进一步的描述,本发明的保护范围不局限于以下所述:
实施例1:
鸡爪槭赤枫简易组培繁殖工艺,它包括以下步骤:
S1、外植体灭菌,在晴天中午或下午采集无病害的鸡爪槭赤枫当年生枝条,去掉多余叶片,切成带芽茎段进行灭菌,灭菌采用酒精加升汞的组合灭菌方式,先用酒精灭菌30s,再用升汞灭菌5min,然后用无菌水清洗4~6遍;
S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,所述的培养基为MS+ IAA0.5mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养15天,诱导腋芽萌发,如图1所示;
S3、生根培养,切割直接诱导的腋芽,接种于生根培养基上,如图2所示,生根培养基为1/2MS+NAA0.2mg/L+IAA0.1mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养15天,诱导根产生,如图3所示。
实施例2:
鸡爪槭赤枫简易组培繁殖工艺,它包括以下步骤:
S1、外植体灭菌,在晴天中午或下午采集无病害的鸡爪槭赤枫当年生枝条,去掉多余叶片,切成带芽茎段进行灭菌,灭菌采用酒精加升汞的组合灭菌方式,先用酒精灭菌50s,再用升汞灭菌4min,然后用无菌水清洗4~6遍;
S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,所述的培养基为MS+ IAA0.5mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养2周,诱导腋芽萌发,如图1所示;
S3、生根培养,切割直接诱导的腋芽,接种于生根培养基上,如图2所示,生根培养基为1/2MS+NAA0.2mg/L+IAA0.1mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养2周,诱导根产生,如图3所示。
实施例3:
鸡爪槭赤枫简易组培繁殖工艺,它包括以下步骤:

S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,所述的培养基为MS+ IAA0.5mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养3周,诱导腋芽萌发,如图1所示;

1.鸡爪槭赤枫简易组培繁殖工艺,其特征在于:它包括以下步骤:
S1、外植体灭菌,采集无病害的鸡爪槭赤枫当年生枝条,去掉多余叶片,切成带芽茎段进行灭菌,灭菌采用酒精加升汞的组合灭菌方式,先用酒精灭菌30~60s,再用升汞灭菌3~5min,然后用无菌水清洗4~6遍;
S2、接种及繁殖,将灭菌后带芽茎段接种于培养基上生长,所述的培养基为MS+ IAA0.5mg/L,并将接种后的培养材料置于光照强度为2000~3000LX、光照时间为16h/d、温度为24~26℃的培养室内培养,培养2~3周,诱导腋芽萌发;
