本发明公开了千层金组培快繁方法,它包括以下步骤:S1、外植体灭菌,S2、启动培养,将灭菌后的外植体切割至2~3cm幼嫩茎段,接种于启动培养基中培养;S3、初代培养,将腋芽切下转移至初代培养基上进行培养;S4、继代培养,将腋芽切下转移至继代培养基中进行培养;S5、炼苗移栽,将生根健壮的千层金组培苗,洗掉培养基后,直接移栽进温室大棚苗床中。本发明的有益效果是:本发明利用组织培养技术,获得千层金组培苗,培养基配方简单,培养流程简捷,操作简便,繁殖系数较高,能够比较容易的获得千层金无菌苗,炼苗易成活;省时省力,为千层金工厂化育苗及其深加工提供了技术支持。

技术领域
本发明涉及植物组织培养技术领域,特别是千层金组培快繁方法。
背景技术
千层金(Melaleuca bracteata)为桃金娘科白千层属常绿乔木,别名黄金宝树、黄金香柳、黄金串钱柳,是20世纪90年代初选育的变异新树种,1999年由新西兰首次引入中国。此树树型优美,主杆直立,嫩枝红色,易修剪成型,生长快,叶面全年金黄色或鹅黄色,气味芳香,可耐-4℃的低温和42℃的高温,既抗旱又抗涝,适宜水边生长,能抗盐碱、抗强风,是目前世界上最流行、视觉效果最好的彩叶乔木新树种之一,可用于庭院、池畔、道路中央隔离带造景,公园绿化、城市景观等。千层金除极具观赏价值外,其枝叶富含挥发油,是提取珍贵植物精油—茶树油的主要树种之一,茶树油具有光谱抗微生物、环保、温和天然清香、无腐蚀等特性,被广泛应用于医药、化妆品、日用化工等行业。因此栽培千层金具有很高的园林观赏价值和良好的经济效益。
扦插繁殖千层金可以采用嫩枝扦插、高空压条法进行繁殖,但通常以嫩枝扦插比较多,一般在4至8月雨水多且夜温不会太低的时候进行,由于生根期间湿度比较大,容易出现叶子及茎干腐烂。千层金自然繁殖率低,扦插繁殖受季节限制,后期管理流程繁琐,扦插苗成活率低,难以规模化生产。
发明内容
本发明的目的在于克服现有技术的缺点,提供一种繁殖率高、繁殖速度快、操作简便的千层金组培快繁方法。
本发明的目的通过以下技术方案来实现:千层金组培快繁方法,它包括以下步骤:
S1、外植体灭菌,采集无病虫害的千层金幼嫩茎段,去掉多余叶片,于流水中冲洗半个小时以上,再用洗涤剂浸泡8~12min,最后用流水冲洗,直至无泡沫出现,再放入超净工作台上,用酒精处理15~25s,无菌水清洗1遍,升汞处理3~7min,无菌水冲洗4~6遍;
S2、启动培养,将灭菌后的外植体切割至2~3cm幼嫩茎段,接种于启动培养基中,启动培养基为MS+6-BA 0.5mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间15~17h/d,温度22~26℃的培养室中培养2~3周,生长出腋芽;
S3、初代培养,将腋芽切下转移至初代培养基上进行培养,初代培养基为MS+6-BA 0.5mg/L+NAA 0.2mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间15~17h/d,温度22~26℃的培养室中培养3~4周,植株生长并生根,在叶腋处重新生长出腋芽;
S4、继代培养,将腋芽切下转移至继代培养基中,继代培养基与启动培养基相同,培养3~4周后,在植株的叶腋处又重新分化出新的腋芽;
S5、炼苗移栽,将生根健壮的千层金组培苗,洗掉培养基后,直接移栽进温室大棚苗床中,移栽基质为草炭土混合蛭石,草炭土与蛭石的混合比例1~3:1。
本发明具有以下优点:
本方法是一种从千层金茎段直接诱导腋芽增殖,再使腋芽生根的快速繁殖方法,利用组织培养技术,获得了千层金组培苗。本发明培养基配方简单,培养流程简捷,操作简便,繁殖系数较高,能够比较容易的获得千层金无菌苗,炼苗易成活;省时省力,为千层金工厂化育苗及其深加工提供了技术支持。

具体实施方式
下面结合附图及实施例对本发明做进一步的描述,本发明的保护范围不局限于以下所述:
实施例1:
千层金组培快繁方法,它包括以下步骤:
S1、外植体灭菌,采集无病虫害的千层金幼嫩茎段,去掉多余叶片,于流水中冲洗半个小时以上,再用洗涤剂浸泡10min,最后用流水冲洗,直至无泡沫出现,再放入超净工作台上,用酒精处理20s,无菌水清洗1遍,升汞处理5min,无菌水冲洗5遍;
S2、启动培养,将灭菌后的外植体切割至2~3cm幼嫩茎段,接种于启动培养基中,启动培养基为MS+6-BA 0.5mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间16h/d,温度24℃的培养室中培养,培养2~3周后,腋芽生长2~4cm,如图1所示;
S3、初代培养,将腋芽切下转移至初代培养基上进行培养,初代培养基为MS+6-BA 0.5mg/L+NAA 0.2mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间16h/d,温度24℃的培养室中培养,培养3~4周后,植株生长6~7cm,开始生根,根系发达,生根数为2~4根,根长3~5cm,在叶腋处重新生长出腋芽,增殖系数在8~10左右,如图2所示;
S4、继代培养,将腋芽切下转移至继代培养基中,继代培养基与启动培养基相同,培养3~4周后,在植株的叶腋处又重新分化出新的腋芽,如图3所示;
S5、炼苗移栽,将生根健壮的千层金组培苗,如图4、图5所示,洗掉培养基后,直接移栽进温室大棚苗床中,如图6所示,移栽基质为草炭土混合蛭石,混合比例2:1,注意遮光降温保湿,成活率高,可达100%。
实施例2:
千层金组培快繁方法,它包括以下步骤:
S1、外植体灭菌,采集无病虫害的千层金幼嫩茎段,去掉多余叶片,于流水中冲洗半个小时以上,再用洗涤剂浸泡8min,最后用流水冲洗,直至无泡沫出现,再放入超净工作台上,用酒精处理15s,无菌水清洗1遍,升汞处理7min,无菌水冲洗6遍;
S2、启动培养,将灭菌后的外植体切割至2~3cm幼嫩茎段,接种于启动培养基中,启动培养基为MS+6-BA 0.5mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间15h/d,温度26℃的培养室中培养,培养2~3周后,腋芽生长2~4cm,如图1所示;
S3、初代培养,将腋芽切下转移至初代培养基上进行培养,初代培养基为MS+6-BA 0.5mg/L+NAA 0.2mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间15h/d,温度26℃的培养室中培养,培养3~4周后,植株生长6~7cm,开始生根,根系发达,生根数为2~4根,根长3~5cm,在叶腋处重新生长出腋芽,增殖系数在8~10左右,如图2所示;

S2、启动培养,将灭菌后的外植体切割至2~3cm幼嫩茎段,接种于启动培养基中,启动培养基为MS+6-BA 0.5mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间17h/d,温度22℃的培养室中培养,培养2~3周后,腋芽生长2~4cm,如图1所示;
S3、初代培养,将腋芽切下转移至初代培养基上进行培养,初代培养基为MS+6-BA 0.5mg/L+NAA 0.2mg/L+蔗糖30g/L+琼脂5g/L,PH调整至5.8~6.2,于光照强度为3000LX,光照时间17h/d,温度22℃的培养室中培养,培养3~4周后,植株生长6~7cm,开始生根,根系发达,生根数为2~4根,根长3~5cm,在叶腋处重新生长出腋芽,增殖系数在8~10左右,如图2所示;

1.千层金组培快繁方法,其特征在于:它包括以下步骤:
S1、外植体灭菌,采集无病虫害的千层金幼嫩茎段,去掉多余叶片,于流水中冲洗半个小时以上,再用洗涤剂浸泡8~12min,最后用流水冲洗,直至无泡沫出现,再放入超净工作台上,用酒精处理15~25s,无菌水清洗1遍,升汞处理3~7min,无菌水冲洗4~6遍;
