背景技术
干粉吸入(DPI)药物疗法多年来已经用于治疗呼吸病症,诸如哮喘、慢性阻塞性肺疾病(COPD)和变应性鼻炎。
意图作为干粉吸入的药物应当以微粉化的颗粒的形式使用,所述颗粒通常通过研磨或通过其它技术(诸如喷雾干燥)得到。
通常如下制备意图吸入的干粉制剂:混合微粉化的药物和粗载体颗粒,产生有序的混合物,其中微粉化的活性颗粒附着于载体颗粒表面上,同时在吸入器装置中。
所述载体会使得微粉化的粉末具有更低的粘合性,并改善它的流动性,从而使得在生产过程(倾倒、填充等)中处理粉末更容易。
但是,已知的是,干粉倾向于变成带静电荷。在药用粉末中的摩擦生电是一个非常复杂的且没有很好理解的过程,尽管已经证实它受许多因素的影响。
在各种生产操作(研磨、混合、运输和填充)过程中,粉末会由于微粒间的碰撞和与固体表面(例如容器壁)的接触而积累静电荷。
已经在经由分离、附着和团聚形成的药物损失机理中鉴别出这种接触和摩擦诱导的起电的过程。此外,在过程中包含的能量越多,材料积累显著水平的静电荷的倾向越大。

粉末混合物的净静电荷高度依赖于生产过程中颗粒-基质和颗粒-颗粒碰撞的频率,所述碰撞常常在粉末样品上产生净电荷,所述电荷可以是正电荷、负电荷或两种电荷。
WO 01/78693和WO 01/78695公开了包含粗颗粒级分和细颗粒级分(作为载体)和添加剂(诸如硬脂酸镁或亮氨酸)的干粉制剂,及其制备方法。
所述的制剂可以以简单的方式生产,是化学和物理稳定的,并具有良好的吸入性能。
但是,所述文件没有提供关于静电荷的任何信息。
另一方面,静电荷电率(electrostatic chargeability)的减小可以改善在运行生产工艺(筛分、倾倒)过程中和在填充吸入器过程中的流动性质。
这又会导致制剂中的活性成分的提高的同质性,并因此导致递送剂量和细颗粒剂量的提高的再现性和准确度。
考虑到上面方面,非常有利的是,提供一种用于制备粉末制剂的方法,所述粉末制剂诸如在WO 01/78693和WO 01/78695中描述的那些,其能够减少静电荷,并因此改善它们的性能特征。
发明内容
本发明涉及一种制备吸入用干粉制剂所用的载体颗粒的方法,所述载体颗粒包括:i)共微粉化的颗粒级分,所述颗粒由赋形剂和添加剂的混合物制成,所述混合物具有低于20微米的质量中值直径(MMD);ii)粗赋形剂颗粒级分,所述颗粒具有等于或高于80微米的MMD,所述方法包括下述步骤:
a)共微粉化所述赋形剂颗粒和添加剂颗粒;
b)添加并混合得到的共微粉化的颗粒和所述粗赋形剂颗粒;其特征在于,首先通过在室温暴露于50-75%的相对湿度24-60小时的时间,调节步骤a)的共微粉化的颗粒。
在第二个方面,本发明涉及一种制备吸入用干粉制剂的方法,所述方法包括下述步骤:混合上述载体颗粒和一种或多种活性成分。
在第三个方面,本发明涉及用于吸入用干粉制剂中的共微粉化的颗粒的混合物,所述颗粒由赋形剂和添加剂制成,所述混合物具有-9×10-10至-5×10-8nC/g的质量电荷密度,所述混合物可通过特定方法得到,所述方法包括:通过在室温暴露于50-75%的相对湿度24-60小时的时间,进行调节。

定义

体积直径(VD)通过颗粒密度与质量直径(MD)相关联(假定大小独立于颗粒密度)。

本发明涉及一种制备吸入用干粉制剂所用的载体颗粒的方法,所述载体颗粒包括:i)共微粉化的颗粒级分,所述颗粒由赋形剂和添加剂的混合物制成;ii)粗赋形剂颗粒级分和一种或多种活性成分,所述方法包括下述步骤:
a)共微粉化所述赋形剂颗粒和添加剂颗粒;

作为调节步骤的结果,减少了共微粉化的颗粒的电荷获得,并因此减少了所有载体颗粒的电荷获得。与包含含有未调节的共微粉化的颗粒的载体的那些粉末制剂相比,包含所述载体颗粒的对应粉末制剂表现出更好的流动性质。
此外,与没有经过调节的制剂相比,包含通过本发明的方法得到的载体颗粒的制剂表现出提高的活性成分同质性以及更好的递送剂量准确度和更好的细颗粒剂量再现性。
甚至当它包含低剂量强度活性成分时,对于包含通过本发明的方法得到的载体颗粒的制剂而言,递送剂量的准确度通常优于±5%、优选地优于±2.5%。
令人惊奇地,在调节以后,共微粉化的颗粒级分也显示出颗粒间粘合相互作用的减少,这表现为:基本流动能(basic flow energy)和克服所述物质的流态化阻力所需的能量(通过流态化能(fluidisation energy)来测量)的减少。
作为所有这些优点的后果,也证实会稍微提高有关制剂的可吸入分数。
在调节以后,在微粉化步骤中产生的无定形物质也显著减少,这提示,所述步骤会诱导赋形剂颗粒的有效重结晶。
另一方面,鉴别的暴露条件不会以显著的方式影响共微粉化的颗粒的粒度和水含量。
后一方面有益于在有关制剂中的活性成分的稳定性,因为已知,水分吸收的增加可以影响它们的理化稳定性,尤其是吸湿的和/或亲水的活性成分。
必须如下调节所述共微粉化的颗粒:在室温,暴露于50-75%的相对湿度6-60小时的时间。
所述室温与22±2℃(优选地±1℃)的温度相对应。
有利地,在55-70%的相对湿度暴露12-48小时、优选地24-48小时、更优选地48小时的时间。在一个优选的实施方案中,在55%的相对湿度暴露24小时,而在其它优选的实施方案中,在75%的相对湿度暴露24小时。在其它优选的实施方案中,在至少55%的相对湿度暴露48小时,因为已经观察到,从所述相对湿度值开始,所述共微粉化的颗粒的表面能的降低更大,并持续更长的时间。
所述相对湿度值可以变化±5%。
不受理论的限制,可以假定,表面能越高,物质的反应性越高,因此形成静电荷的可能性越高。
有利地,所述细和粗赋形剂颗粒可以由任意药理学上可接受的惰性物质或其组合构成;优选的赋形剂是由结晶性的糖类、尤其是乳糖制成的那些;最优选的赋形剂是由α-乳糖一水合物制成的那些。
优选地,所述粗赋形剂颗粒和所述细赋形剂颗粒由相同的生理上可接受的药理学上惰性的物质构成。
由赋形剂和添加剂的混合物制成的共微粉化的颗粒级分必须具有低于20微米(有利地等于或低于15微米、优选地等于或低于10微米、甚至更优选地等于或低于6微米)的MMD。
有利地,90%的颗粒的质量直径低于35微米,更有利地低于25微米,优选地低于15微米,甚至更优选地低于10微米。
在微粉化的颗粒级分内的赋形剂和添加剂之间的比例,将随制剂的组成和添加剂材料的本性和性质而变化。
有利地,所述共微粉化的颗粒级分由90-99.5重量%的赋形剂和0.5-10重量%的添加剂材料(优选地95-99%的赋形剂和1-5%的添加剂)组成。优选的比例是98%的赋形剂和2%的添加剂。
有利地,所述添加剂材料可以包括一种或多种润滑剂或由其组成,所述润滑剂选自:硬脂酸及其盐诸如硬脂酸镁、月桂基硫酸钠、硬脂酰富马酸钠、硬脂醇、单棕榈酸蔗糖。
优选地,所述润滑剂是硬脂酸镁。
或者,所述添加剂材料可以是抗粘附材料诸如氨基酸,其优选地选自:亮氨酸、异亮氨酸、赖氨酸、缬氨酸、蛋氨酸、苯丙氨酸。所述添加剂可以是氨基酸的衍生物的盐,例如阿司帕坦或乙酰舒泛K。
所述添加剂材料还可以包括一种或多种水溶性的表面活性物质(例如卵磷脂,尤其是大豆卵磷脂)或由其组成。
其它可能的添加剂材料包括滑石、二氧化钛、二氧化铝和二氧化硅。
有利地,至少90重量%的添加剂颗粒具有不超过35微米的起始质量直径和不超过15微米(优选地不超过10微米)的MMD。
通过研磨(有利地,在球磨机中),使构成微粉化的颗粒级分的赋形剂颗粒和添加剂颗粒共微粉化,在某些情况下,可以发现共微粉化至少2小时是有利的,尽管应当理解,处理时间通常取决于赋形剂颗粒的起始粒度和要实现的希望的尺寸减小。
在本发明的一个优选实施方案中,使用气流粉碎机,优选地在惰性气氛下,例如在氮下,从具有小于250微米的质量直径的赋形剂颗粒和具有小于35微米的质量直径的添加剂开始,共微粉化所述颗粒。
作为一个实例,可以使用可商业得到的α-乳糖一水合物诸如MeggleD 30或Spherolac 100(Meggle,Wasserburg,Germany)作为起始赋形剂。
本发明的方法的粗赋形剂颗粒必须具有至少80微米、更有利地大于90微米、优选地大于100微米、更优选地大于175微米的MMD。
有利地,所有粗颗粒具有在50-1000微米(优选地60-500微米)范围内的质量直径。
在本发明的某些实施方案中,所述粗颗粒的质量直径可以是80-200微米,优选地90-150微米,而在另一个实施方案中,所述质量直径可以是200-400微米,优选地210-355微米。
一般而言,本领域技术人员会通过筛分(使用适当的分级器)来选择最合适的粗赋形剂颗粒尺寸。
当所述粗颗粒的质量直径是200-400微米时,粗赋形剂颗粒优选地具有相对较多裂缝的表面,也就是说,在它们上面存在裂缝和缝隙和其它凹陷区域,这些在本文中统称为裂隙。可以以在WO 01/78695和WO01/78693(通过引用并入本文)中描述的裂隙指数或粗糙性系数的方式,定义“相对较多裂缝的”粗颗粒,且可以根据其中报道的描述来表征它们。还可以以振实密度或总侵入体积(instrusion volume)(如在WO 01/78695中所报道地进行测量)的方式,表征所述的粗颗粒。
所述粗颗粒的振实密度有利地小于0.8g/cm3,优选地在0.8-0.5g/cm3之间。所述总侵入体积是至少0.8cm3,优选至少0.9cm3。
微粉化的颗粒级分和粗颗粒级分之间的比例是1:99至40:60重量%,优选地2:98至30:70重量%,甚至更优选地5:95至20:80重量%。在一个优选的实施方案中,所述比例是10:90至15:85重量%。
所述混合粗赋形剂颗粒和微粉化的颗粒级分的步骤,通常在合适的混合机(例如转鼓混合机诸如Turbula、旋转混合机或即时混合机诸如Diosna)中进行至少5分钟、优选地至少30分钟、更优选地至少2小时。一般而言,本领域技术人员会调节混合时间和混合机的旋转速度,以得到同质混合物。
当需要球形化的粗赋形剂颗粒以便得到硬弹丸时,所述混合步骤通常进行至少4小时。
在一个优选的实施方案中,本发明涉及一种制备吸入用干粉制剂所用的载体颗粒的方法,所述载体颗粒包括:i)共微粉化的颗粒级分,所述颗粒具有等于或低于10微米的MMD,其由98-99重量%的α-乳糖一水合物和1-2重量%的硬脂酸镁的混合物制成;ii)由α-乳糖一水合物制成的粗颗粒级分,所述颗粒具有212-355微米的质量直径,所述共微粉化的颗粒和所述粗颗粒之间的比例是10:90至15:85重量%,所述方法包括下述步骤:
a)共微粉化所述α-乳糖一水合物颗粒和所述硬脂酸镁颗粒;
b)添加并混合得到的共微粉化的颗粒和所述粗颗粒;其特征在于,通过在室温暴露于55-75%的相对湿度24-48小时的时间,调节步骤a)的共微粉化的颗粒。
本发明也涉及一种制备吸入用干粉制剂的方法,所述方法包括下述步骤:混合通过要求保护的方法可得到的载体颗粒和一种或多种活性成分。
有利地,至少90%的药物(活性成分)颗粒具有小于10微米、优选地小于8微米、更优选地小于6微米的粒度。
在本发明的某些实施方案中,具体地,当使用低剂量强度活性成分时,不超过50%的颗粒具有低于1.7微米的体积直径;且至少90%的颗粒具有低于8微米的体积直径。
通过在合适的混合机(如上面报道的那些)中混合所述组分,制备所述载体颗粒与所述活性成分颗粒的混合物。
任选地,当使用至少2种活性成分时,首先将一种活性成分与一部分载体颗粒相混合,并迫使得到的混合物穿过筛子,然后,将其它活性成分和剩余部分的载体颗粒与筛过的混合物相混合;最后,通过筛子筛分得到的混合物,并再次混合。
技术人员会选择筛子的筛目尺寸,这取决于粗赋形剂颗粒的粒度。
载体颗粒和活性成分之间的比例取决于使用的吸入器装置的类型和需要的剂量。
活性成分的量应当能够允许将治疗有效剂量递送进肺中。
合适的活性剂可以是用于治疗性的和/或预防性的用途的药物。可以包含在所述制剂中的活性剂包括这样的产品:其通常通过吸入而口服给药,用于治疗诸如呼吸性疾病等疾病。

可以用于提供吸入来递送的其它活性剂包括:抗心律不齐药、安神药、他汀类、强心苷类、激素类、抗高血压药、抗糖尿病药、抗寄生虫药和抗癌药、镇静剂和镇痛药、抗生素、抗风湿药、免疫疗法、抗真菌药和抗低血压药、疫苗、抗病毒药、蛋白、多肽和肽(例如肽激素和生长因子)、多肽疫苗、酶、内啡肽类、参与血液凝固级联的脂蛋白和多肽、维生素和其它,例如细胞表面受体阻滞剂、抗氧化剂和自由基清除剂。这些化合物中的几种可以以药理学上可接受的酯、缩醛、盐、溶剂化物(诸如水合物)、或这样的酯或盐的溶剂化物(如果存在的话)的形式施用。上述化合物的外消旋混合物以及一种或多种光学异构体是在本发明的范围内。

包含用于吸入的β2-激动剂、抗胆碱能药或皮质类固醇(单独的,或以它们的任意组合)的制剂,构成本发明的具体实施方案。
优选的组合包括:富马酸福莫特罗二水合物/二丙酸倍氯米松、维兰特罗/糠酸氟替卡松、昔萘酸沙美特罗/丙酸氟替卡松、富马酸福莫特罗脱水物/环索奈德、富马酸福莫特罗脱水物/糠酸莫米松、富马酸福莫特罗脱水物/布地奈德、富马酸福莫特罗脱水物/丙酸氟替卡松、富马酸福莫特罗脱水物/噻托溴铵、富马酸福莫特罗二水合物/格隆溴铵、富马酸福莫特罗二水合物/格隆溴铵/二丙酸倍氯米松、富马酸福莫特罗二水合物/噻托溴铵/二丙酸倍氯米松。
包含富马酸福莫特罗二水合物、二丙酸倍氯米松和任选的抗胆碱能支气管扩张剂(诸如噻托溴铵或格隆溴铵)的组合是特别优选的。
本发明也涉及共微粉化的颗粒的混合物,所述颗粒由赋形剂和添加剂制成,且具有非常低的负静电荷残余,所述混合物可通过特定方法得到,所述方法包括:通过在室温暴露于50-75%的相对湿度24-60小时的时间,进行调节。质量电荷密度应当是-9×10-10至-5×10-8nC/g,优选-9×10-9至-1×10-9。
使用在实施例2中描述的法拉第笼子(Faraday cage),测定质量电荷密度。
还通过改善的流态化性质来表征要求保护的混合物,所述改善的流态化性质表现为:与未调节的混合物相比,它们的基本流动能(BFE)和它们的流态化能显著更低。
所述BFE有利地是15-30mJ、优选地18-26mJ,而所述流态化能有利地是5-15mJ、优选地8-12mJ。
在调节以后,无定形(amorphous)物质的量有利地小于5%w/w、优选地小于3%w/w、更优选地小于2%w/w、甚至更优选地等于或小于1%w/w。通过已知的方法,可以测定无定形物质的量。
例如,如在实施例4中所报道的,通过包含H/D交换和FT-Raman光谱法的光谱法方案,可以测定它。否则,通过动态蒸汽吸附(DVS)实验(使用例如Hiden Igasorb测湿度天平),或通过等温气体灌注量热法(IGPC)(使用例如2277热活度监测热量计(TA Instrument Ltd)),可以测定它。
一般而言,添加剂的量应当不超过10重量%,这基于所述共微粉化的颗粒的混合物的总重量。
但是,认为就大多数添加剂而言,添加剂材料的量应当不超过5%、优选地不超过2%、或甚至不超过1重量%、或不超过0.5%,这基于所述混合物的总重量。一般而言,添加剂材料的量是至少0.01重量%,这基于所述混合物的总重量。
在本发明的优选实施方案之一中,所述赋形剂是α-乳糖一水合物,且所述添加剂材料是硬脂酸镁,其以0.5-2%、优选地2重量%的量存在,这基于所述混合物的总重量。
所述添加剂可以形成围绕赋形剂颗粒表面的包衣,或可以形成不连续的包衣,如在WO 96/23485中所报道的。
如果使用硬脂酸镁,所述添加剂会以这样的方式包被赋形剂颗粒表面:表面包被程度是至少5%、优选地超过10%、更优选地超过15%、甚至更优选地等于或超过35%。
可以如下测定表面包被程度(其指示被硬脂酸镁包被的赋形剂颗粒的总表面的百分比):测量水接触角,然后应用在文献中已知的Cassie和Baxter的方程式,所述方程式引用在Colombo I等人Il Farmaco 1984,39(10),328-341的第338页,并报告在下面。

为了本发明的目的,使用基本上基于角度测定测量(goniometricmeasurement)的方法,测定接触角。这些暗示在固体基质和受测液体之间形成的角的直接观察。因此实现是非常简单的,唯一的限制与源自操作人员内的变异性的可能偏倚有关。但是,应当强调,通过采用完全自动化的操作,诸如计算机辅助的图像分析,可以克服该缺陷。一个特别有用的方案是停滴法或静滴法,其通常如下实现:将液滴放在通过压实得到的圆盘形式的粉末表面上(压实的粉末圆盘方法)。
还可以通过扫描电子显微术(SEM,一种众所周知的通用分析技术),测定硬脂酸镁包被乳糖颗粒表面的程度。
这样的显微术可以配备EDX分析仪(电子分散的X-射线分析仪),其可以产生对某些类型的原子(例如镁原子)选择性的图像。以此方式,可能获得关于硬脂酸镁在载体颗粒表面上的分布的清楚数据集。
或者,根据已知的操作,可以将SEM与IR或Raman光谱法相组合,用于测定包被程度。
可以有利地使用的另一种分析技术是X-射线光电子光谱法(XPS),通过该方法,已经可能计算包围乳糖颗粒的硬脂酸镁膜的包被程度和深度。
要求保护的共微粉化的颗粒的混合物可以用于任意吸入用干粉制剂中。
优选地,它用于这样的干粉制剂中:所述干粉制剂另外包含上述的粗赋形剂颗粒和一种或多种选自上述类别的活性成分。
所述的干粉制剂可以与任意干粉吸入器一起使用。
干粉吸入器可以分成2个基本类型:
i)单次剂量吸入器,用于施用单个细分剂量的活性化合物,每个单次剂量通常填充在胶囊中;
ii)多次剂量吸入器,其预装载足够更长的治疗周期的量的活性物质。
所述的吸入用干粉制剂特别适用于包含贮库的多次剂量干粉吸入器中,通过所述装置(例如在WO 2004/012801中描述的装置)的启动,可以根据需要从所述贮库取出单个治疗剂量。可以使用的其它多次剂量装置是,例如,GlaxoSmithKline的DISKUSTM,AstraZeneca的TURBOHALERTM,Schering的TWISTHALERTM和Innovata的CLICKHALERTM。作为面市的单次剂量装置的实例,可以提及GlaxoSmithKline的ROTOHALERTM和Boehringer Ingelheim的HANDIHALERTM。
下述实施例例证了本发明的方法。
实施例
实施例1-由赋形剂和添加剂制成的共微粉化的颗粒的制备
制备了约40kg共微粉化的颗粒。
通过在运行于氮下的气流粉碎机中研磨,将比例为98:2重量%的具有小于250微米粒度的α-乳糖一水合物颗粒(Meggle D 30,Meggle)和具有小于35微米粒度的硬脂酸镁颗粒共微粉化,以得到共微粉化的颗粒级分。
在处理结束时,所述共微粉化的颗粒具有约6微米的质量中值直径(MMD)。
此后,单独保存该批的一部分作为对照,在22±1℃的温度,在表1报道的不同的相对湿度和时间条件,对剩余部分进行调节。
所述相对湿度值可以变化±5%。
所有样品贮存在聚乙烯袋中。

实施例2-静电荷和流态化性质的测定
使用NanoerTM技术(Nanopharm Ltd,Bath,UK),进行测量。
使用与静电计相连的法拉第桶(Faraday Pail),测量微粉化的颗粒的静电荷。所述静电计与计算机相连,用于数据获取。将10g材料放入法拉第笼子中,然后如下得到荷质比:将在静电计上测得的净电荷除以进入法拉第笼子中的材料的质量。
如下表征微粉化的颗粒:使用FT4粉末流变仪(FreemanTechnologies,Welland,UK)来测定对通气的阻力,后者被量化为不同粉末的流态化能。在每种情况下,在25mm内径圆筒中分析10ml样品粉末。调节所述样品,以使用23.5mm刀片去除包装历史,所述刀片以20mm/s向下经过螺旋状路径。在记录粉末床的质量、体积、高度和经历的施加的力的同时,也测定各个粉末的堆密度。
在表2中报告了静电荷的测量结果。

所述值指示,进行调节的样品会表现出一些非常低的残余负电性电荷,而未经过调节的样品表现出双极电荷。

在调节以后,在共微粉化的颗粒内的粘合相互作用发生降低。这通过基本流动能(粉末的流动性能的量度)和流态化能(克服流态化阻力所需的能量)的降低来证实。
可能注意到,BFE随着相对湿度百分比的增加而降低。
实施例3-表面能的测定
通过反相气相色谱法(IGC),测量表面能。
使用macros的SMS-iGC 2000和SMS-iGC v1.3标准分析套件和SMS-iGC v1.21高级分析套件,进行所有分析。使用火焰离子化检测器(FID)来测定保留时间。
在IGC上运行之前,在冷的(约5℃)干燥的环境中保藏所述样品。对于所有实验,使用SMS柱填充附件,将粉末填充进硅烷化的玻璃柱(300mm长×4mm直径)中。在预调节以后,顺序地分析所有柱3次,以检查不可逆的化学吸附效应和平衡。
在该研究中,在氦载气中在25℃和0%相对湿度(RH)预处理所述柱2小时,以调节样品。然后,在25℃进行表面能测量(顺序地进行3次,运行之间有2-小时调节)。对于所有洗脱液,在10sccm的氦总流速和0.03P/0的注射蒸汽浓度,进行所有实验。
结果报告在图1中。
图1显示了每种调节过的样品以及Meggle D30和硬脂酸镁(MgSt)参照物的分散表面能(dispersive surface energy)。该图表明,与MaggleD30相比,每种调节过的样品发生分散表面能的增加,这证实,所述微粉化过程会诱导乳糖的表面能的增加。
检查揭示,加工过的Maggie D30-MgSt混合物的分散表面能随它们的贮存条件而变化。在55%RH,观察到贮存24小时(48.7mJm-2)和48小时(49.5mJm-2)的微粉化的混合物的分散表面能的微小变化。但是,在60%RH,观察到贮存24和48小时的微粉化的混合物之间的显著变化(分别是48.3和42.6mJm-2)。在60%RH时观察到的分散表面能的降低,提示所述样品更易于从周围环境中吸收水分。在该更高的%RH,在Maggle D30-MgSt混合物上存在的高能量位点可能已经被水分猝灭,这可能启动无定形乳糖区域的重结晶。这得到在60%RH放置48小时的混合物的分散表面能与Meggle D30参照物的分散表面能的相似性(42.6mJm-2相对于41.8mJm-2)的支持。
令人感兴趣地,在75%RH放置24小时的微粉化的混合物表现出比放置24小时的其它混合物更低的表面能(46.0mJm-2相对于48.7mJm-2和48.3mJm-2)。这进一步证实,湿度的增加是降低微粉化的Meggle D30-MgSt混合物的分散能的突出因素。但是,在75%RH放置24小时的样品的表面能仍然大于在60%RH放置48小时的混合物,这证实,这些混合物的分散表面能的降低似乎取决于时间和相对湿度。
乳糖(41.8mJm-2)和硬脂酸镁(42.1mJm-2)的分散表面能都与在文献中报道的值(例如乳糖的41mJm-2和硬脂酸镁的41mJm-2)比较一致。
实施例4-无定形含量的测定
使用包含H/D交换和FT-Raman光谱法的光谱方案来探测微粉化的颗粒的无定形含量。所述方法利用下述事实:在无定形乳糖中的羟基在氧化氘蒸汽中易于氘化,而结晶性的乳糖则不会。无定形相的氘化导致强度从OH-伸长区(3400-3150cm-1)向OD-伸长区(2600-2300cm-1)迁移。所述OD-伸长带然后可以用作无定形含量水平的直接指示。

与未调节的和未微粉化的参照样品相比,分析了5个共微粉化的颗粒样品(#1、#2、#3、#4和#7)。
图2显示了在暴露于D2O蒸汽(25%相对湿度,超过12小时)以后进行调节的共微粉化的颗粒的样品的OD伸长带。
结果指示,与没有调节过的样品相比,在调节过的样品中的所有批次含有显著微小量的无定形物质。
这提示,采用的调节过程已经有效地重新结晶显著量的无定形物质,所述无定形物质存在于预调节过的样品中。
实施例5-载体的制备
以90:10重量%的比例,将实施例1的每种共微粉化的颗粒样品与α-乳糖一水合物的裂隙粗颗粒(其具有212-355微米的质量直径,并通过筛分得到)相混合。
所述混合在Turbula混合机中进行4小时。
分析得到的颗粒混合物(在下文中称作载体(CARRIER))的粒度(使用筛分系统)和流动性。
通过筛分,测定粒度。
根据在欧洲药典中描述的方法,测试流动性质。
简而言之,将粉末混合物(约110g)倒入干燥的漏斗中,所述漏斗配有合适直径的孔口,所述孔口被合适的装置封闭。打开漏斗的底部开口,并记录整个样品流出漏斗所需的时间。对于100g样品,以秒和十分之几秒表达流动性。
尽管密度和粒度不受调节的影响,在包含经过调节的共微粉化的颗粒的载体中,流动性降低。
对于所述的样品,穿过4mm直径的流速被证实是在136至134s/100g之间,而包含未调节的共微粉化的颗粒的载体的流速被证实是约142s/100g。
实施例6-干粉制剂的制备
使用包含未调节的共微粉化的颗粒、共微粉化的颗粒样品#2和样品#8的载体颗粒。
在Turbula混合机中,在32r.p.m.,将在实施例5中得到的每种载体的一部分与微粉化的富马酸福莫特罗二水合物(FF)一起混合30分钟,迫使得到的混合物穿过具有0.3mm(300微米)筛目尺寸的筛子。
在Turbula混合机中,在32r.p.m.,将微粉化的二丙酸倍氯米松(BDP)和剩余部分的载体与筛过的混合物一起混合60分钟,以得到最终制剂。
所述活性成分与10mg载体的比例是,6微克FF二水合物(理论的递送剂量4.5微克)和100微克BDP。
在生产过程中,没有观察到团聚物。
在将它装载进WO 2004/012801所述的多次剂量干粉吸入器中以后,以活性成分的分布均匀度和气溶胶性能的方式,表征粉末制剂。
通过从混合物的不同部分取出20个样品,评价活性成分的分布均匀度,并通过HPLC进行评价。
根据在欧洲药典2008年第6版段落2.9.18、第293-295页中报道的条件,使用Andersen级联冲击器(装置D),进行气溶胶性能评价。
在气雾化10个剂量以后,拆卸ACI装置,并通过用溶剂混合物洗涤,回收在各阶(stages)中沉积的药物的量,然后通过高效液相色谱法(HPLC)进行定量。计算下述参数:i)递送剂量,这是从冲击器中回收的装置所递送的药物的量;ii)细颗粒剂量(FPD),这是从具有等于或低于5.0微米的粒度的S3-AF各阶回收的递送剂量的量;iii)细颗粒分数(FPF),这是细颗粒剂量的百分比;iv)MMAD。
分别在表4和5中,报道了以分布均匀度和气溶胶性能(平均值±S.D)的方式的结果。

从表4的数据可以看出,与包含未调节的共微粉化的颗粒的制剂相比,使用经过调节的共微粉化的颗粒制备的制剂表现出两种活性成分的增加的分布均匀度。
从表5的数据也可以看出,使用经过调节的共微粉化的颗粒制备的制剂会提供更准确的FF递送剂量,所述活性成分以更低的剂量存在。此外,使用经过调节的共微粉化的颗粒制备的制剂表现出两种活性成分的提高的可吸入分数的趋势。
Claims (10)
1.一种制备吸入用干粉制剂所用的载体颗粒的方法,所述载体颗粒包括:i)共微粉化的颗粒级分,所述颗粒由95-99重量%乳糖和1-5重量%硬脂酸镁的混合物制成,所述混合物具有低于20微米的质量中值直径;ii)粗乳糖颗粒级分,所述颗粒具有等于或高于80微米的质量中值直径,且相对于所述级分i)的比例是95:5至80:20重量%,所述方法包括下述步骤:
a)共微粉化所述乳糖颗粒和硬脂酸镁颗粒;
b)添加并混合得到的共微粉化的颗粒和所述粗乳糖颗粒;其特征在于,首先通过在室温暴露于50-75%的相对湿度24-60小时的时间,调节步骤a)的共微粉化的颗粒。
2.根据权利要求1所述的方法,其中通过暴露于55-70%的相对湿度48小时的时间,调节所述共微粉化的颗粒。
3.根据权利要求1-2中任一项所述的方法,其中所述乳糖是α-乳糖一水合物。
4.根据权利要求中1-2中任一项所述的方法,其中所述粗乳糖颗粒的质量直径是212-355微米。
5.根据权利要求中1所述的方法,其中所述级分i)的混合物由98重量%的乳糖和2重量%的硬脂酸镁组成。
6.根据权利要求中1所述的方法,其中所述级分ii)相对于级分i)的比例是90:10重量%。
7.一种制备吸入用干粉制剂的方法,所述方法包括下述步骤:混合根据权利要求1-6中任一项所述的载体颗粒和一种或多种活性成分。
8.根据权利要求7所述的方法,其中所述活性成分是β2-肾上腺素受体激动剂,其选自:沙丁胺醇、特布他林、非诺特罗、沙美特罗、福莫特罗、茚达特罗、维兰特罗和米维特罗。
9.根据权利要求7所述的方法,其中所述活性成分是皮质类固醇,其选自:布地奈德、丙酸氟替卡松、糠酸氟替卡松、糠酸莫米松、二丙酸倍氯米松和环索奈德。
10.根据权利要求7所述的方法,其中所述活性成分是抗胆碱能支气管扩张剂,其选自:异丙托溴铵、噻托溴铵、氧托溴铵和格隆溴铵。
