Abstract

本发明涉及一种皮肤活性物质检测用试剂盒及利用该试剂盒检测皮肤活性物质的方法,具体地,涉及利用含有以长寿基因著称的klotho基因的检测试剂盒,从皮肤细胞中将诱导klotho基因表达的候选物质分离、精制,从而检测抗衰老及美白物质等皮肤活性物质的皮肤活性物质检测用试剂盒及利用该试剂盒检测皮肤活性物质的方法。

检测皮肤活性物质的检测试剂盒和检测皮肤活性物质的方法
技术领域
本发明涉及一种皮肤活性物质检测用试剂盒及利用该试剂盒检测皮肤活性物质的方法,具体地,涉及利用含有以长寿基因著称的klotho基因的检测试剂盒,从皮肤细胞中将诱导klotho基因表达的候选物质分离、精制,从而检测抗衰老及美白物质等皮肤活性物质的皮肤活性物质检测用试剂盒及利用该试剂盒检测皮肤活性物质的方法。
背景技术
皮肤随着年龄的增长或者暴露在太阳光下而老化。随着老化,表皮的角质层脱落并更新所需要的时间延长,角质层增厚,真皮由于弹力纤维和保湿成分的含量下降或排列变化而产生皱纹。迄今为止,为了阻断引起皮肤衰老的原因中的一种光老化,人们对预防紫外线照射引起的皮肤变化进行了各种研究。另外,人们还研究了防止随着年龄增长的老化,即由遗传调节的老化的各种方法。这些研究都集中在寻找统筹调节老化的机制上。
对klotho基因的研究就是其中之一。敲除(knock-out)klotho基因的小鼠出现伴随1,25-(OH)2D3浓度上升出现动脉硬化、血管钙化、管组织钙化、肺气肿、活动减退、生殖腺形成异常、不孕、皮肤萎缩、运动失调症、低血糖症及高磷酸血症(Mutation of the mouse klotho gene leads to a syndromeresembling ageing.Nature 1997;390,45-51)。与此相反,klotho蛋白的表达增加时,出现寿命增加、胰岛素抗性增加及IGF-1抗性增加(Kurosu等,2005)。在人体中,也有报道称klotho蛋白的单核酸多态性与寿命减少、骨质疏松、中风及冠状动脉疾病有关(Arking等,2002;Kawano等,2002;Mullin等,2005;Ogata等,2002;Yamada等,2005)。
根据之前的研究,klotho主要在人的肾脏表达,在胎盘、前列腺及小肠中也有表达。但在klotho越来越重要的情况下,迄今为止未见klotho在皮肤细胞,即角质形成细胞、成纤维细胞或黑色素细胞中表达的报道。因此,也没有对增加klotho的表达的物质的研究的报道。
发明内容
发明目的
本发明的目的在于,制备一种能够在皮肤细胞中增加klotho蛋白表达的试剂盒,并利用该试剂盒测定在角质形成细胞、成纤维细胞及黑色素细胞等皮肤细胞中的klotho蛋白表达,发现能够检测抗老化物质及美白物质等皮肤活性物质,从而完成了本发明。
因此,本发明的目的在于提供一种通过在皮肤细胞中的klotho基因的表达与否,从而筛选皮肤活性物质的检测试剂盒及利用该试剂盒检测皮肤活性物质的方法。
技术方案
为了达到上述目的,本发明提供一种检测皮肤活性物质的检测试剂盒,该试剂盒含有klotho蛋白基因、用于扩增klotho蛋白基因的引物、探针和皮肤细胞,所述用于扩增klotho蛋白基因的引物具有SEQ ID No:1和SEQ IDNo:2所示的序列,所述探针具有SEQ ID No:3所示的序列,在所述皮肤细胞中能够筛选能够增加klotho蛋白表达量的物质,从而检测皮肤活性物质。
此外,本发明还提供利用所述检测试剂盒检测活性物质的方法。
有益效果
根据本发明的皮肤活性物质检测试剂盒,通过在皮肤细胞中的klotho基因的表达与否,可迅速简便地筛选如抗老化物质及美白物质等皮肤活性物质。
附图说明
图1a是在人胚胎肾脏细胞(HEK293)、角质形成细胞(NHK)及成纤维细胞(NHF)中测定klotho蛋白的RT-PCR扩增结果;图1b是在人黑色素细胞中测定klotho蛋白表达的RT-PCR扩增结果;此时,NHM-增强黑(NHM-promoblack)及NHM-DP是源自黑人的黑色素细胞,NHM-MP是源自亚洲人的黑色素细胞。
图2是在人胚胎肾脏细胞(HEK293)、角质形成细胞(NHK)及成纤维细胞(NHF)中检测klotho蛋白表达量的图。
图3是角质形成细胞经视黄酸(RA:retinoic acid)及视黄醇(ROL:retinol)处理后,在第2天及第8天检测klotho蛋白表达量的图。
具体实施方式
本发明涉及一种在皮肤细胞中检测klotho蛋白表达量的方法,通过此在皮肤细胞中筛选能够诱导长寿基因klotho蛋白表达的物质,从而检测皮肤活性物质。所述皮肤活性物质可包括改善皮肤皱纹或弹力或者改善粗糙皮肤的抗老化物质;及提亮肤色或改善由于痣、雀斑、皮肤杂质及太阳光线引起的皮肤晒黑等皮肤色素沉着现象的美白物质。
在皮肤细胞中能够检测klotho蛋白表达量的本发明的检测试剂盒含有klotho蛋白基因、用于扩增klotho蛋白的引物、探针(参照表1)和皮肤细胞,所述用于扩增klotho蛋白基因的引物具有SEQ ID No:1和SEQ ID No:2(序列号1和2)所示的序列,所述探针具有SEQ ID No:3(序列号3)所示的序列,在所述皮肤细胞中能够筛选能够增加klotho蛋白表达量的物质,从而检测抗老化物质。

根据本发明的一种实施方式,所述皮肤细胞可以是人角质形成细胞、成纤维细胞或黑色素细胞,但不限于此。
此外,本发明涉及利用所述检测试剂盒,将皮肤细胞用候选物质处理后,通过测定klotho蛋白表达与否的步骤及将能够增加klotho蛋白表达的物质分离及精制的步骤,从而检测皮肤活性物质的方法。
根据本发明的另一种实施方式,利用所述检测试剂盒,测定已知为抗氧化物质的视黄酸及视黄醇能够增加klotho蛋白表达,由此测定根据本发明的检测方法适合筛选未知的抗老化物质及皮肤活性物质。
以下,根据实施例及实验例详细说明本发明的内容。但这些实施例仅用于更好地理解本发明,本发明的权利范围并不由这些实施例和实验例限定,本领域技术人员应该理解的是本领域常用的各种变更、替换及插入都落入本发明的范围。
实验例1 在皮肤细胞中测定klotho蛋白表达与否
为了测定klotho蛋白是否在皮肤细胞中表达,制备了能够扩增klotho基因(NCBI记录号:NM_004795)的具有SEQ ID No:1和SEQ ID No:2所示的序列的引物,利用所述引物对和具有SEQ ID No:3所示的序列的探针(TaqManTM荧光探针),通过RT-PCR观察在角质形成细胞(NHK)、成纤维细胞(NHF),NHM-增强黑(NHM-promoblack)细胞(正常人表皮黑色素细胞(Normal Human Epidermal Melanocytes,NHEM)初生包皮,源自黑人的黑色素细胞,增强细胞(Promocell)),NHM-DP细胞(NHM-暗色素(NHM-darkpigment);源自黑人的黑色素细胞,Invitrogen)和NHM-MP细胞(NHM-中等色素(NHM-moderate pigment);源自亚洲人的黑色素细胞,Invitrogen)的cDNA中是否能够检出klotho的mRNA。此时,PCR反应条件为在94℃变性30秒,在55℃退火30秒,以及在72℃延伸60秒,反复上述循环35次。阳性对照组使用已知能够表达klotho基因的人胚胎肾脏细胞HEK293(humanembryonic kidney)细胞的cDNA,结果如图1a及图1b所示。从图1a及图1b的结果可看出,本发明中所使用的角质形成细胞、成纤维细胞及黑色素细胞中均有klotho基因表达。
此外,通过RT-PCR测定klotho mRNA在各细胞中的表达量的结果如图2所示。
实验例2在经视黄酸及视黄醇处理的角质形成细胞中klotho的mRNA表达量的测量
为了测定已知作为抗老化物质的视黄酸和视黄醇在角质形成细胞中对klotho表达的影响,在与所述实验例1相同的条件进行了RT-PCR,用所述视黄酸0.1及1μM和视黄醇1及10μM处理后,在第2天及第8天测量了klotho蛋白表达量,并将结果在图3中表示。
从图3的结果可以看出,用视黄酸和视黄醇处理后的第8天klotho蛋白的表达量显著增加。

Claims (4)

1.一种检测皮肤活性物质的检测试剂盒,该试剂盒含有klotho蛋白基因、用于扩增klotho蛋白基因的引物、探针和皮肤细胞,所述用于扩增klotho蛋白基因的引物具有SEQ ID No:1和SEQ ID No:2所示的序列,所述探针具有SEQ ID No:3所示的序列,在所述皮肤细胞中能够筛选能够增加klotho蛋白表达量的物质,从而检测皮肤活性物质。
2.根据权利要求1所述的检测试剂盒,其中,所述皮肤活性物质是抗老化物质或者美白物质。
3.根据权利要求1所述的检测试剂盒,其中,所述皮肤细胞是人角质形成细胞、成纤维细胞或黑色素细胞。
4.一种检测皮肤活性物质的方法,其特征在于,该方法包括:利用权利要求1-3中任意一项所述的检测试剂盒,将皮肤细胞用候选物质处理后,测定klotho蛋白表达与否的步骤,和,将增加klotho蛋白表达的物质分离精制的步骤。
