Abstract

Description
技术领域
本发明通常涉及对水分敏感的产品的包装,以延长这种产品的保质期。更具体地,本发明涉及延长与口服再水化溶液(ORS)粉剂混合的冻干乳酸菌的保质期。所述产品是包括两种干燥剂的包装:一种干燥剂整合到箔材料中,第二种干燥剂是无水的ORS。本发明还涉及生产这种产品的方法。
技术背景
益生菌是通过改善宿主的肠道的微生物平衡来有益地影响宿主的活的微生物食品添加物。现今,许多种不同的细菌被用作益生菌,例如,产乳酸细菌,例如乳杆菌(Lactobacillus)和双歧杆菌(Bifidobacteria)的菌株。产乳酸细菌不仅因为它们对人类或动物健康的有益效果而被使用,它们还在食品工业中广泛地用于发酵过程。一般地,为此目的销售的微生物被制成处于低水环境中的冻干粉剂。
在这种冻干微生物制品的应用中遇到的一般性难题是细胞的有限的保存稳定性。例如,随着时间的过去,微生物变得少数可存活,导致必需使用高剂量来补偿这种活性损失。
一般已知的是利用铝箔作为包装材料用于微生物的保藏,最优选的箔具有在隔层外侧的一个表面上的塑料层,例如,层压聚乙烯的铝箔,作为包装材料以减少冻干的乳酸菌对水分和氧气的暴露。
虽然常规包装的防潮层在限制水分进入包装的运动方面可能是有用的,但是某些水分分子仍然可以找到途径进入包装,有害地影响其中包含的产品。此外,即使阻挡材料在限制水分子通过包装的输送方面是有效的,包装的某些特征仍然允许水分子的输送,例如,沿着热密封的包装的边缘。并且,就水分吸收而言,包装冻干微生物的时刻是特别弱的时点。
维持包装内特别低的或者几乎不存在的水分水平的一种方案是将小袋的干燥剂材料包裹入包装的内部间隙以从包装的顶部空间移除水分。小袋中含有的干燥剂材料一般为粉剂或颗粒形式,可能从小袋漏出,或者溢出,从而污染产品或包装内含有的产品。市场上已知的可摄取的干燥剂不适合于例如患有腹泻的人。相反,它可能产生有害影响。然而,在此本发明解决了这个问题,其中在小袋内部的干燥剂的目的是消耗所述干燥剂。
典型的干燥剂材料是“物理”干燥剂材料,例如,分子筛,其在材料的孔隙空间内结合水分子。一般地,物理干燥剂材料在所有湿度水平下吸收水,但是当物理干燥剂的空隙被填满时将停止吸水。因而,物理干燥剂材料在高湿度水平下可能是无效的。
另一类干燥剂材料是水化物形成试剂,例如盐。可以用作干燥剂材料的典型的盐是硫酸镁、磷酸二氢钠、氯化铵、碳酸钾、硫酸铝钾、氯化镁、硫酸二铵、硝酸钠、氯化钙和硫酸钙,许多其他的也是已知的。这些材料的干燥能力大大地受到包装内的相对湿度的影响。一般地,直到相对湿度达到第一水化物形成的值时,水化物形成试剂才会吸水。例如对于氯化钙来说,在小于约百分之二的相对湿度(RH)下产生第一水化物。然后水化物形成盐吸水直到第一水化物由盐完全地形成。直到相对湿度达到第二水化物形成的第二水平,盐不会进一步吸收水。这个过程一直继续到试剂形成许多水化物,在此时物质开始溶解并形成饱和溶液。饱和溶液然后将继续吸水。
虽然这些盐在从包装顶部空间内含有的一定数量气体中移除水分子方面是有效的,但是由于盐仅在盐内部结合水分子,水分子可能容易逃逸回到包装中。这被称为呼吸,可能导致包装内的潮解(水滴和液化)。一般地,在包装的装运或保存期间可能发生的,如果盐变得饱和,以及包装的温度升高或者包装的压力降低,可能发生包装内的潮解。
此外,盐不能容许包装内的水分水平降至保护包装内包含的水分敏感性产品所必需的水平。一般地,由于盐具有不同的水化水平,在水化改变之前,湿度水平可能保持在一定水平而不会降低。可以利用这些盐来维持包装顶部空间内一定的湿度水平。例如,某些产品可能需要在包装顶部空间内维持一定水平的水分或湿度。产品的顶部空间湿度控制可以通过掺入合适的水化物形成试剂来控制。
还可以使用不形成水化物的干燥剂材料,例如,食盐(NaCl)或溴化钾(KBr)。例如,食盐将在低于约百分之75的相对湿度下不吸水。当达到百分之75的相对湿度时,形成饱和溶液,其继续吸水。
另一种干燥剂利用化学干燥剂技术。化学干燥剂材料一般在所有湿度水平下吸收水,在高相对湿度水平下将继续吸水。美国专利申请No.20070160789A1描述了多层的塑料聚合柔性包装箔,具有包括在箔层内的化学干燥剂物质。箔优选地是选自由超低密度聚乙烯、低密度聚乙烯、线性低密度聚乙烯、中密度聚乙烯和高密度聚乙烯构成的组的聚乙烯。本发明利用这样的干燥剂,如氧化钙(优选的)、氧化镁、氧化钡、氧化锶、氧化铝、部分水化的氧化铝、硫酸镁、磷酸二氢钠、氯化铵、碳酸钾、硫酸钾铝、氯化镁、硫酸二铵、硝酸钠、氯化钙、硫酸钙、氯化钠、溴化钾、分子筛、粘土,或对本发明有用的任何其他干燥剂材料。
口服再水化治疗(ORT)是用于与腹泻相关的脱水的简单、便宜和有效的治疗。ORT由口服施用的盐和糖溶液组成,也称为口服再水化溶液(ORS)或口服再水化制剂(ORF)。ORT在全世界使用,但最重要的是在发展中世界,在此它将数百万儿童从由于腹泻的死亡中拯救出来,腹泻是5岁以下儿童死亡的第二大主因。
已经进行了几项实验来评估在口服再水化溶液中施用的乳酸菌的功效。例如,进行了多中心的试验来评估向所有病因的急性发作腹泻的患者在口服再水化溶液中施用的乳杆菌GG(Lactobacillus GG)的功效,显示了向患有急性腹泻的儿童施用含有乳杆菌GG的口服再水化溶液是安全的,产生更短的腹泻延续时间、延长病程的几率更低,以及更快的出院(J Pediatr Gastroenterol Nutr,2000Jan;30(1):54-60)。然而,如前面提及的,难题是冻干的乳酸菌需要在粉剂基质中的低水活性(通常αw<0.2),在没有活性的显著损失的情况下,以在环境温度下具有合理的保质期。
由于口服再水化溶液粉剂的极度吸湿的(从周围环境吸引水分子的能力)性质,迄今为止不可能将冻干的乳酸菌与ORS粉剂混合并在环境保存期间维持细菌存活较长的时间。活的益生培养物可变的保存质量引起向急性水泻患者施用的制品中剂量和活性的不可预见的损失,特别是在发展中国家这是难以解决的技术难题。
从保存的角度,在本发明中测试了一种方法来评估与冻干的路氏乳杆菌(Lactobacillus reuteri)混合,是否有任何盐-混合物可以作为包装袋内的干燥剂成功地起作用。包装袋由US20070160789A1中描述的箔材料制成。令人惊讶地,与无水的ORS混合的路氏乳杆菌(L.reuteri)的冻干的培养物成功地达到了30℃下至少12个月的保质期,已知的是意味着在更低的温度下更长的保存时间。这种类型的有益的保存稳定性迄今为止没有被描述。就我们所知,在世界上没有这样的产品出售。
总之,迄今为止不可能将冻干的乳酸菌与ORS粉剂混合并仍然在环境保存期间保持细菌在任何形式的包装中存活持续更久的时间。在此本发明通过用两种不同的干燥剂两个步骤除去水分,解决了这个难题。第一种干燥剂是作为干燥剂起作用的无水的ORS,在包装时围绕着微生物,第二种干燥剂处在箔中吸收在保存期间可能出现的水分。当使用无水的ORS时,它作为不需要移除的干燥剂起作用,同时它满足了其作为口服再水化溶液的目标。
发明内容
本发明提供了对水分敏感的产品的包装方法,从而延长这种产品的保质期,更具体地,延长与口服再水化溶液(ORS)粉剂混合的冻干乳酸菌的保质期。所述包装包含两种干燥剂:一种干燥剂整合在箔材料中,第二种干燥剂是无水的ORS。
本发明的主要目的是提供延长与口服再水化溶液(ORS)粉剂混合的冻干乳酸菌的保质期的产品。
本发明的另一个目的是提供用于冻干的乳酸菌的保存的、在两个步骤中移除水分的两种干燥剂的包装。
另一个目的是提供具有两种干燥剂的小袋:一种干燥剂整合到如US20070160789A1中描述的箔材料中,另一种干燥剂是在围绕产品的、在小袋内自由存在的无水ORS。
本发明的其他目的和优点对于读者将变得显而易见,意图是这些目的和优点处在本发明的范围之内。

发明及其优选实施方式的详细说明
本发明的方法和设备提供了对水分敏感的产品的包装,从而延长这种产品的保质期,更具体地,延长与口服再水化溶液(ORS)粉剂混合的冻干乳酸细菌的保质期。所述包装包括两种干燥剂:一种干燥剂整合到铝箔中,第二种干燥剂是无水的ORS。所述箔优选地由铝制成,具有适合的保护包被例如聚乙烯或漆。理由是,铝箔是已知最好的针对水分的阻挡物,在此讨论的产品对水分非常敏感。包装的特征性的阻挡物的测试如本领域已知的来进行(Allinson et al,InternationalJournal of Pharmaceutics Volume 221,Issues 1-2,19June 2001,Pages49-5)。
如在此使用的术语“无水的ORS”是指,ORS粉剂的成分是无水性质的,即,没有水。成分可以通过本领域已知的方法变为无水的形式或无水地制备。
在此本发明的目标是提供方法和设备,其使得冻干的乳酸菌存活到以约+30℃的保存12个月的末尾,在这个温度下和起始点,有至少约10E+07CFU/克(当以5×10E+09CFU/克开始时)但优选的至少10E+08CFU/克,最好的10E+09CFU/克。当产品保存在+25℃时,在大约18个月时,这种稳定性已知转变相同损失数量的最大值,当温度更低时则更少。这将意味着,在+25℃下在18个月时的目标,在这个温度下和起始点,有至少10E+07CFU/克(当以5×10E+09CFU/克开始时),但是优选的至少10E+08CFU/克,以及最好的10E+09CFU/克。
通过参考以下实施例,将更清楚地理解本发明的特征,实施例不被看作是限制本发明。
实施例1
测试的参数
在此处的实施例中,测试本发明的方法来评估ORS溶液盐是否可以与冻干的路氏乳杆菌在小袋中混合,目的是实现在30℃下至少12个月的保质期,作为一种加速的研究,已知的是这意味着在更低温度下更久的保存时间。新的生产方法包括使用具有在US 20070160789A1中描述的干燥剂的新的箔材料。
由五种不同的测试参数组成的试验如表1中所示:

实施例2

无水葡萄糖:3781.000mg/小袋
无水柠檬酸钠:866.000mg/小袋
氯化钾:379.000mg/小袋
氯化钠:365.000mg/小袋
硫酸锌:4,000mg/小袋
总共:5463.000mg/小袋
在BioGaia Lund的微生物学实验室中,在LAF工作台(来自Heto-Holten A/S,Denmark的Holten Lamiriair Model S-2010 1.2)上,共混物在环境温度下填充到具有干燥剂的铝箔袋中(10cm×12cm,使用来自Alcan的包装材料PET12/PE/ALU 12/PE/PE+干燥剂/PE)。向每个袋中,使用来自Denver Instrument GmbH,Germany的平衡XP-600添加5.46g的具有路氏乳酸杆菌的ORS粉剂。填充的铝箔袋然后用来自Kettenbaum Folienschweisstechnik GmbH&Co.KG.Germany的箔密封设备型号F460/2热密封。由于实际上完全没有通过铝箔的水分穿透,特别是如果铝厚于0.025mm,因而最优选的是,封闭是完全封闭的敏感部分,这意味着使用的密封设备必需有高质量,得到完整的密封而没有任何渗漏,并且是如本领域已知的充分的。这些袋子也用于实施例2中描述的研究中。
实施例3
在30℃下在12个月期间,在口服再水化溶液(ORS)粉剂中保存期间冻干的路氏乳杆菌DSM17938的稳定性研究
在来自Norfoods AB,Sweden的口服再水化溶液粉剂中评估路氏乳杆菌DSM 17938粉剂art no 1012 BioGaia AB,Stockholm,Sweden(产品规格PS137)的生存力。无水ORS粉剂根据实施例1制得。测验参数根据表1进行。样品保存在Lund Sweden的BioGaia AB处30℃/RH的环境柜中。
实施例4
结果在图1中示出。保存在具有整合的干燥剂和无水ORS的箔中的微生物令人惊讶地(附图1中的曲线2)显示了与没有ORS而保存的微生物几乎相同的生存力(附图1中的曲线1)。
与保存在没有干燥剂的箔中的微生物相比,保存在具有整合的干燥剂的箔中的微生物显示了保存期间更好的生存力,因为箔中的干燥剂在保存期间吸收了袋子中几乎所有的水分。但是仅具有干燥剂的箔和非无水的ORS(附图1中的曲线3)显示不是足够的,因为与保存在具有整合的干燥剂和无水的ORS中的微生物(附图1中的曲线2)相比生存力是更差的。
结论性地,单独的无水的ORS或单独的整合干燥剂的箔都不足以为产品提供足够的保质期。包括它们两者的保存一起满足了在保质期末尾的活性目标。
虽然参考具体实施方式描述了本发明,要理解的是,许多变体、修改和实施方式是可能的,因而,所有这样的变体、修改和实施方式被认为处于本发明的精神和范围之内。
Claims (19)
1.一种益生菌产品,所述益生菌产品为:与无水的口服再水化溶液粉剂混合的冻干的乳酸菌,所述冻干的乳酸菌和所述无水的口服再水化溶液粉剂包装在具有整合到箔中的干燥剂的箔中。
2.如权利要求1所述的益生菌产品,其中,所述箔包括铝。
3.如权利要求2所述的益生菌产品,其中,所述箔进一步包括聚乙烯层。
4.如权利要求1所述的益生菌产品,其中,所述整合到箔材料中的干燥剂包括氧化钙。
5.如权利要求1所述的益生菌产品,其中,所述冻干的乳酸菌是路氏乳杆菌。
6.如权利要求1所述的益生菌产品,其中,所述无水的口服再水化溶液粉剂含有:3781.000mg无水葡萄糖、866.000mg无水柠檬酸钠、379.000mg氯化钾、365.000mg氯化钠以及4000mg硫酸锌。
7.如权利要求1所述的益生菌产品,其中,在30℃下所述益生菌产品的12个月保存之后,当以5×10E+09CFU/克的路氏乳杆菌开始时,存在至少10E+07CFU/克的路氏乳杆菌。
8.如权利要求7所述的益生菌产品,其中,在12个月的保存后存在至少10E+08CFU/克的路氏乳杆菌。
9.如权利要求8所述的益生菌产品,其中,在12个月的保存后存在10E+09CFU/克的路氏乳杆菌。
10.一种用于对水分敏感的产品的活性保存的产品,所述产品为无水的口服再水化溶液粉剂,所述无水的口服再水化溶液粉剂用于与所述对水分敏感的产品混合和用于所述对水分敏感的产品的包装,以及掺合在箔层内的化学干燥剂材料;其中所述对水分敏感的产品包括活的冻干乳酸菌。
11.如权利要求10所述的产品,其中,所述箔包括铝。
12.如权利要求11所述的产品,其中,所述箔进一步包括聚乙烯层。
13.如权利要求10所述的产品,其中,所述无水的口服再水化溶液粉剂含有:3781.000mg无水葡萄糖、866.000mg无水柠檬酸钠、379.000mg氯化钾、365.000mg氯化钠以及4000mg硫酸锌。
14.一种活的冻干益生菌乳酸菌的长期保存的方法,所述方法包括:
a)提供无水的口服再水化溶液粉剂;
b)混合冻干的乳酸菌和所述无水的口服再水化溶液粉剂,
c)在掺入到箔层之内的化学干燥剂材料的包装中,包装所述混合的冻干乳酸菌和口服再水化粉剂;以及
d)密封所述包装使得所述密封是完整的。
15.如权利要求14所述的方法,其中,所述箔包括铝箔。
16.如权利要求15所述的方法,其中,所述箔进一步包括聚乙烯层。
17.如权利要求14所述的方法,其中,所述整合到箔材料中的干燥剂包括氧化钙。
18.如权利要求14所述的方法,其中,所述冻干的乳酸菌是路氏乳杆菌。
19.如权利要求14所述的方法,其中,所述无水的口服再水化溶液粉剂含有:3781.000mg无水葡萄糖、866.000mg无水柠檬酸钠、379.000mg氯化钾、365.000mg氯化钠以及4000mg硫酸锌。
