本发明属保健产品,涉及一种含葡萄籽原花青素提取物的牙膏,在龋病和牙周病的预防控制和日常口腔护理保健技术领域中发挥作用。
背景技术
牙膏作为一种口腔护理保健产品已在人们日常生活和口腔辅助治疗中被广泛应用,具有预防控制龋病和牙周病,对口腔常见的牙痛、牙龈炎、牙龈出血等疾病有较好疗效,具有明显清热消炎、止血、抑菌、镇痛作用,清除口腔异味。
龋病和牙周病是最常见的口腔疾病,对这两种疾病的研究一直是我国口腔医学研究的重点。龋病生态学理论认为,与龋病发生有关的细菌都是口腔正常微生物群,在生理状态下,细菌之间、细菌与宿主之间处于动态平衡,不发生疾病;由于局部的、全身的、食物因素的改变,造成口腔生态环境的变化,正常细菌与宿主之间的生理性组合改变为病理性组合,口腔生态平衡转化为生态失调,正常细菌成为条件致病菌,引起龋病,而变形链球菌是与龋病发病关系最为密切的口腔链球菌。
天然药物来源广,毒副作用小,开发和研制天然植物药,是目前医药和食品研究中的一个重点。100多年来,葡萄一直是众多植物中经久不衰的研究课题。50年代至今,人们已从葡萄果实、叶和其它部位分离、鉴定的多酚化合物、脂肪酸、维生素、酶、碳水化合物、氨基酸、多肽和蛋白质、萜烯与挥发油成分以及脂类、果胶和蜡等物质达100多种,在众多成分之中,葡萄的医疗功效大都与原花青素的药理活性有关,作为原花青素的来源,葡萄籽原花青素提取物得到了最为深入和广泛的关注,国内外诸多研究发现其具有多种重要生物活性和药理作用,有着极强的抗氧化及清除自由基活性,能通过保护血管内皮功能、抗心肌氧化应激损伤及抑制低密度脂蛋白的氧化等多种机制抗心血管疾病,成为近年研究的热点。
细菌实验和动物实验研究发现葡萄籽原花青素提取物对变形链球菌有抑制作用。国外有学者通过对葡萄籽原花青素提取物对鼠体内活性氧和活性氮的产生,以及对牙周病鼠巨噬细胞脂多糖激化的诱生型一氧化氮合酶的表达的影响的研究提示葡萄籽原花青素提取物可以考虑作为预防牙周病的药物开发。
发明内容
本发明的目的是提供一种含葡萄籽原花青素提取物的牙膏,其中按重量百分比计葡萄籽原花青素提取物占牙膏总量的0.1%~30%,其中葡萄籽原花青素提取物中原花青素含量≥95%。
本发明的牙膏采用传统制备工艺将葡萄籽原花青素提取物添加入普通的牙膏中而制得,即将葡萄籽原花青素提取物与通常牙膏中含有的保湿剂、粘合剂、摩擦剂、发泡剂、甜味剂、香味剂、功能剂和去离子水等复配而成。葡萄籽原花青素提取物是由葡萄籽经干燥粉碎,仁壳分离后,用乙醇在壳中进行萃取、过滤、喷雾干燥得粗提物,粗提物经溶剂分级法或活性碳吸附法干燥得红褐色粉状物。
本发明的另一个目的是提供的含葡萄籽原花青素提取物的牙膏在制备辅助治疗预防龋齿和牙周炎药物中的应用。
本发明有效地利用葡萄籽提取物中的有效成分原花青素,采用传统牙膏制备工艺制备出一种对口腔常见的龋病和牙周病具有预防控制作用的含葡萄籽原花青素提取物的新型牙膏,能有效杀灭变形链球菌,影响牙周炎的进展,不但可以用于口腔常见病如龋病和牙周病的辅助治疗,也可用于日常口腔保健,预防龋齿和牙周炎。

实施例1
将葡萄籽原花青素提取物与普通牙膏中含有的保湿剂、粘合剂、摩擦剂、发泡剂、甜味剂、香味剂、功能剂和去离子水等复配而成,其中按重量百分比计葡萄籽原花青素提取物占总量的0.1%~30%,其中葡萄籽原花青素提取物中原花青素含量≥95%。
主要组分按重量计比例如下:

山梨醇 7~10%
羧甲基纤维素钠(CMC-Na) 1~2%
氢氧化铝 10~20%

实施例2
通过细菌实验测定葡萄籽原花青素提取物(GSPE)对变形链球菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC)。选用国际标准菌株,变形链球菌族的变形链球菌(S.mutans Ingbritt,血清型c)和远缘链球菌(S.Sobrinus 6715,血清型g),在含1%蔗糖的胰蛋白酶消化大豆液体培养基(TSB)中厌氧培养(CO2 80%,N2 10%,H2 10%),采用液体/琼脂稀释法体外抑菌试验分别测定葡萄籽原花青素提取物(GSPE)对两种变链菌的最低抑菌浓度(MIC)和最低杀菌浓度(MBC),结果分别为MIC 4.00mg/ml,MBC 8.00mg/ml。
实施例3
通过动物实验进一步证实葡萄籽原花青素提取物(GSPE)的防龋作用。选择21d鼠龄断乳Sprague-Dawby(SD)大鼠16只,随机分成2组,每组8只,雌雄各半,分别为葡萄籽原花青素提取物(GSPE)溶液、蒸馏水(H2O)组。21~23d鼠龄期间给予古有广谱抗生素(2kg饲料加氨苄青霉素、氯霉素各1g)的致龋饲料2000#,其它时间的饲料中不加抗生素。在鼠龄24~28d期间,连续5d于口腔内接种变形链球菌S.Sobrinus 6715(105~6CFU/ml),每次接种100ul。自29d起两组分别用MIC浓度的GSPE溶液4mg/ml、蒸馏水处理。大鼠取仰卧位,将0.3ml药物用钝针头注射器注于大鼠牙面及口腔,维持15s,每天处理1次,处理后30min内禁饮食。连续处理60d后处死大鼠,采用Keyes龋齿评分法观察、评估大鼠磨牙牙合面窝沟患龋情况,采用SPSS 13.0统计软件进行单因素方差分析和均数比较,总结GSPE对Sprague-Dawley大鼠磨牙牙合面窝沟龋的预防效果。动物实验发现GSPE能抑制大鼠龋病的发生发展(P<0.05);Dx级龋损仅见于蒸馏水组,葡萄籽原花青素提取物(GSPE)组未发生Dx级龋损。
