Abstract
本发明提供一种用于日常口腔健康维护的口腔喷剂及其制备方法。本发明的口腔喷剂包括原料:表没食子儿茶素没食子酸酯、原花青素、赤藓糖醇、蜂胶提取物、三七皂苷R1、薄荷醇、乳糖醇、乙醇、无菌水。本发明制备的口腔喷剂对于多种主要口腔致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有清除和明显的抑制效果,对于口腔各种炎症具有明显的消炎功效,并且有利于口腔伤口愈合,能够减少牙龈出血等。

技术领域
本发明涉及一种用于日常口腔健康维护的口腔喷剂及其制备方法,属于医药技术领域。
背景技术
口腔疾病已经发展为日常生活中最为常见的疾病之一,各个年龄段都会产生各种各样的口腔疾病如龋齿、牙周炎、口臭、黏膜病等等,从而给患者带来精神和生理上的痛苦以及生活中的不便。大多数口腔疾病已经被证实与口腔中的致病微生物有着密不可分的关系,而且在最新的研究中,口腔疾病已经被发现与多种系统的并发症有关,如心血管疾病、糖尿病等。因此,对口腔致病微生物的抑制在口腔疾病的预防和治疗中显得愈加重要。
目前,人们对于口腔疾病的认识还处于发现疾病,治疗疾病的阶段,甚至连早发现早治疗都没有很好的做到,导致患者无故花费额外的财力和精力处理口腔疾病并遭受额外的痛苦,因此,对口腔疾病的预防非常重要。而患者对口腔疾病预防的手段目前也仅仅局限于日常刷牙,定时洁治等,而利用口腔喷剂进行日常口腔健康维护的较少。中国专利文献CN105125448A公开了一种口腔卫生喷剂,其配方由天然植物精油、表面活性剂、薄荷香精、薄荷醇、食用色素、苏打、甜叶菊、柠檬酸、木糖醇、乙醇及去离子水组成。利用该发明提供的口腔卫生喷剂可预防口腔疾病,但该发明所用原料多,并且对多种口腔疾病病源的针对作用较差,从多种口腔疾病的致病因素方面预防作用欠佳,并且抗菌、消炎、促进伤口愈合功效欠佳。
因此,急需一种用于日常口腔健康维护的口腔喷剂来有效维护口腔健康。
发明内容
针对现有技术的不足,本发明提供一种用于日常口腔健康维护的口腔喷剂。本发明的口腔喷剂对于多种主要口腔致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有清除和明显的抑制效果,对于口腔各种炎症具有明显的消炎功效,并且有利于口腔伤口愈合,能够减少牙龈出血等。
本发明还提供一种用于日常口腔健康维护的口腔喷剂的制备方法。

根据本发明,所述蜂胶提取物为蜜蜂从植物芽孢或树干上采集的树脂(树胶),混入其上腭腺、蜡腺的分泌物加工而成的一种具有芳香气味的胶状固体物,其成分复杂,主要成分为黄酮类化合物、萜类化合物、有机酸类、芳香性醛类及多种氨基酸酶、维生素、矿物质等,常用做营养补充剂,食品、饮料添加剂等。
根据本发明优选的,所述表没食子儿茶素没食子酸酯、赤藓糖醇、薄荷醇、乳糖醇均为纯度≥98%的食品或者药品级,所述原花青素为纯度≥95%的食品或者药品级,所述蜂胶提取物中蜂胶黄酮的质量含量≥40%,蜂胶提取物为食品或者药品级;所述三七皂苷R1为纯度≥80%的食品或者药品级。
上述用于日常口腔健康维护的口腔喷剂的制备方法,包括步骤:
(1)将表没食子儿茶素没食子酸酯、原花青素、赤藓糖醇和乳糖醇溶于无菌水中,混合均匀得溶液A;
(2)将三七皂苷R1、蜂胶提取物和薄荷醇溶于乙醇中,混合均匀得溶液B;
(3)将溶液A、B混匀,过滤除菌,灌装于喷瓶中即得。
根据本发明,步骤(4)中所述喷瓶可市购获得。
本发明中EGCG为天然植物提取物,为绿茶多酚中主要活性物质,具有抗菌、抗病毒、抗氧化、抗动脉硬化、抗血栓形成、抗炎以及抗肿瘤作用,对于多种主要口腔致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有明显的抑制效果,对于口腔各种炎症具有明显的消炎功效,减少牙槽骨吸收,天然健康,对正常组织无副作用;EGCG应为纯度高于99%的食品或者药品级,可市购得到。
本发明中原花青素优选为原花青素A1,为天然植物提取物,具有极好的抗氧化和消除自由基的功能,改善微循环,对口腔各种致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有较好的清除和抗黏附作用,对各类炎症也有较好的治愈效果,可减少牙槽骨吸收,天然健康,对正常组织无副作用;本发明原花青素A1优选为高纯度食品或者药品级,可市购得到。
本发明中赤藓糖醇可抑制口腔致病菌生长,并对已形成的细菌生物膜有较好的去除能力,同时可作为正常人高耐受糖醇,且不参与糖代谢不引发龋齿,对人体健康无副作用;本发明赤藓糖醇优选为高纯度食品或药品级,可市购得到。
本发明中蜂胶提取物为天然提取物,天然健康对人体无副作用,具有良好的灭菌、抑菌、防腐功效,并有较好的促细胞再生作用,可加快口腔伤口愈合;同时可辅助抗病毒、抗氧化,预防肿瘤;本发明蜂胶提取物优选为高纯度食品或药品级化合物,可市购得到。
本发明中三七皂苷R1为天然植物提取物,对人体健康无副作用,可促进细胞再生,有利于口腔伤口愈合,减少牙龈出血等,在口腔保健中起到重要作用;本发明三七皂苷R1优选为高纯度食品或药品级,可市购得到。
本发明中薄荷醇优选为左旋薄荷醇,为天然植物提取物,对人体健康无副作用,具有清新口气,清凉止痛,提神醒脑兼备杀菌功能;本发明左旋薄荷醇优选为高纯度食品或药品级,可市购得到。
本发明中乳糖醇作为甜味剂,同时不引起龋齿,兼备一定的杀菌作用,在维护口腔健康中起到重要作用;本发明乳糖醇优选为高纯度食品或药品级,可市购得到。
有益效果:
(1)本发明制备得到的口腔喷剂使人们在日常口腔清洁的同时,更加简单有效维持口腔健康,帮助人们减少可能因各种口腔疾病产生的各种痛苦和不便;减少各种因口腔不良习惯或口腔不洁造成口腔致病菌滋生而引起的各种疾病,并通过维护口腔健康来维护全身健康以减少其它系统疾病产生的风险。
(2)本发明制备得到的口腔喷剂对主要口腔致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有有效的清除、抗粘附和明显的抑制效果,对于口腔各种炎症具有明显的消炎功效,同时能够促进口腔伤口愈合;本发明各种原料发挥相互协同作用,以达到本发明的优异的抗菌、消炎、促进伤口愈合的效果。本发明EGCG、原花青素和蜂胶提取物作为主要成分,协同三七皂苷R1与赤藓糖醇,使本发明制备的口腔喷剂能够同时具备较好的杀菌、抗菌、消炎、促进伤口愈合等作用,从而能够较好的维护口腔组织健康。

下面结合具体实施例对本发明做进一步的说明,但不限于此。
同时下述实施例中所述实验方法,如无特殊说明,均为常规方法;所述试剂和材料,如无特殊说明,均可从商业途径获得。
下述中,表没食子儿茶素没食子酸酯,食品级,纯度98%,陕西森弗天然制品有限公司有售;原花青素A1,食品级,纯度95%,陕西森弗天然制品有限公司有售;赤藓糖醇,食品级,纯度99%,陕西森弗天然制品有限公司有售;蜂胶提取物,食品级,蜂胶黄酮的质量含量为40%,陕西森弗天然制品有限公司有售;三七皂苷R1,医药级,纯度85%,西安艾诺医药科技有限责任公司有售;左旋薄荷醇,食品级,纯度99%,深圳鼎诚植物香料公司有售;乳糖醇,食品级,纯度99%,河南旗诺食品配料有限公司有售;乙醇均为食用酒精,食品级,体积浓度95%,中和盛泰公司有售。
实施例1
一种用于日常口腔健康维护的口腔喷剂,包括如下百分比的原料组成:
各组分之和为百分之百。
上述用于日常口腔健康维护的口腔喷剂的制备方法,包括步骤:
a)分别称取2g EGCG、1g原花青素A1,1g赤藓糖醇,1g乳糖醇,溶于500g无菌水中,搅拌混匀,配置成溶液A;
b)分别称取1g蜂胶提取物、0.5g三七皂苷R1、3g左旋薄荷醇溶于20g食用酒精(体积浓度为95%)中,搅拌混匀,得溶液B;
c)将溶液A、B混合,加入无菌水至1000g,搅拌混匀,过滤除菌,灌装于喷瓶中即得。
实施例2
一种用于日常口腔健康维护的口腔喷剂,包括如下百分比的原料组成:
各组分之和为百分之百。
上述用于日常口腔健康维护的口腔喷剂的制备方法,包括步骤:
a)分别称取1g EGCG、0.5g原花青素A1,0.5g赤藓糖醇,0.5g乳糖醇,溶于500g无菌水中,搅拌混匀,配置成溶液A;
b)分别称取0.5g蜂胶提取物、0.2g三七皂苷R1、1g左旋薄荷醇溶于10g食用酒精(体积浓度为95%)中,搅拌混匀,得溶液B;
c)将溶液A、B混合,加入无菌水至1000g,搅拌混匀,过滤除菌,灌装于喷瓶中即得。
实施例3
一种用于日常口腔健康维护的口腔喷剂,包括如下百分比的原料组成:
各组分之和为百分之百。
上述用于日常口腔健康维护的口腔喷剂的制备方法,包括步骤:
a)分别称取5g EGCG、3g原花青素A1,3g赤藓糖醇,3g乳糖醇溶于500g无菌水中,搅拌混匀,配置成溶液A;
b)分别称取3g蜂胶提取物、1g三七皂苷R1、5g左旋薄荷醇溶于30g食用酒精(体积浓度为95%)中,搅拌混匀,得溶液B;
c)将溶液A、B混合,加入无菌水至1000g,搅拌混匀,过滤除菌,灌装于喷瓶中即得。
对比例1
一种口腔喷剂,原料组成如实施例1所述,所不同的是:组分中无表没食子儿茶素没食子酸酯(EGCG),其它与实施例1一致。
上述用于日常口腔健康维护的口腔喷剂的制备方法与实施例1一致。

一种口腔喷剂,原料组成如实施例1所述,所不同的是:组分中无原花青素A1,其它与实施例1一致。

试验例1
最小抑菌浓度MIC和最小杀菌浓度MBC的测定;
1、待测样品的制备
将实施例1和对比例1-6制备的口腔喷剂分别与无菌水混合配制不同浓度的喷雾剂溶液(8000、4000、2000、1000、500、250、125、62.5、32.15、15.625、7.8、3.9ug/ml),作为待测样品;待测样品浓度的计算方法为:口腔喷剂的密度*口腔喷剂的体积/(口腔喷剂和无菌水的总体积),不同口腔喷剂密度均约为1mg/ml。
同时分别配制不同浓度(8000、4000、2000、1000、500、250、125、62.5、32.15、15.625、7.8、3.9ug/ml)的EGCG的水溶液、原花青素A1的水溶液、赤藓糖醇的水溶液、蜂胶提取物的乙醇水溶液(乙醇水溶液中乙醇的质量浓度为10%)、三七皂苷R1的乙醇水溶液(乙醇水溶液中乙醇的质量浓度为10%),作为待测样品。
2、试验方法
将上述待测样品分别加入灭菌的96孔细胞培养板中;将具核梭杆菌、伴放线聚集杆菌和牙龈卟啉单胞菌混合,悬浊液浓度调至0.5麦氏比浊,用BHI肉汤稀释1000倍后,向上述每孔中加入100ul,密封后于37℃培养18-24h后进行观察;以孔内完全抑制细菌生长的最低药物浓度为MIC;继续培养48h后观察细菌生长情况,涂平板观察,无菌生长孔所含药物最低浓度为MBC。测试结果如下表所示:

结论:本发明制备的口腔喷剂较对比例以及单一组分拥有更好的抑制口腔致病菌的能力,说明各原料之间发挥协同作用以达到优异的抗菌性能。
试验例2
对口腔上皮细胞的粘附作用的实验:
1、待测样品的制备
将实施例1和对比例1-6制备的口腔喷剂和DMEM混合分别配制浓度为试验例1所得最小杀菌浓度的喷雾剂溶液,作为待测样品;
同时分别配制浓度为2mg/ml(足以达到最小杀菌浓度)的EGCG、原花青素A1、赤藓糖醇、蜂胶提取物、三七皂苷R1的DMEM溶液,作为待测样品。
2、试验方法
将培养24h后的具核梭杆菌、伴放线聚集杆菌和培养5-7天后的牙龈卟啉单胞菌混合,并用FITC标记,得混合致病菌;
准备口腔上皮细胞悬液,向96孔细胞培养板的孔中分别加入100ul上述细胞悬液进行过夜培养(12-16h)后,吸除培养液后加入100ul待测样品继续培养30min;向上述孔中均加入100ul混合致病菌,在37℃,5wt%二氧化碳含量的培养箱中培养4h,得培养后的混合菌;然后将未黏附于上皮细胞的混合菌吸除,PBS缓冲液冲洗2次,使未黏附于上皮细胞的混合菌彻底清除,测孔中剩余混合物的相对荧光单位代表黏附程度;同时设置对照孔,对照孔只加入100ulDMEM,测试荧光度,做为基础荧光度。

结论:本发明制备的口腔喷剂较对比例和单一组分明显减少了口腔致病菌对口腔上皮细胞的黏附,说明本发明各原料之间发挥协同作用,从而对口腔各种致病菌如具核梭杆菌,牙龈卟啉单胞菌等具有较好的清除和抗黏附作用。

7.根据权利要求1所述的用于日常口腔健康维护的口腔喷剂,其特征在于,所述表没食子儿茶素没食子酸酯、赤藓糖醇、薄荷醇、乳糖醇均为纯度≥99%的食品或者药品级,所述原花青素为纯度≥95%的食品或者药品级,所述蜂胶提取物中蜂胶黄酮的质量含量≥40%,蜂胶提取物为食品或者药品级;所述三七皂苷R1为纯度≥80%的食品或者药品级。
8.如权利要求1-7任一项所述的用于日常口腔健康维护的口腔喷剂的制备方法,包括步骤:
(1)将表没食子儿茶素没食子酸酯、原花青素、赤藓糖醇和乳糖醇溶于无菌水中,混合均匀得溶液A;
(2)将三七皂苷R1、蜂胶提取物和薄荷醇溶于乙醇中,混合均匀得溶液B;
(3)将溶液A、B混匀,过滤除菌,灌装于喷瓶中即得。
