Description
技术领域
本发明涉及能够有效地除去与消化性溃疡的发生有关的幽门螺杆菌(Helicobacter pylori)的幽门螺杆菌的除菌剂。
背景技术
幽门螺杆菌是革兰氏阴性杆菌,直径为约0.5μm,长度为3-5μm。其保持弯曲2-3次的螺旋菌的形状,在显微镜下观察到为S字状、或被称为海欧状的弯弯曲曲的形态。在长轴的两端(极)各具有4-8根鞭毛(称为极鞭毛),通过该鞭毛的旋转运动,可以在溶液内、粘液中游动而进行移动。由于为微好氧性且营养要求性也严格,因此是难以分离、培养类群的细菌,可以通过在氧浓度5%、二氧化碳浓度5-10%的条件下使用专用的培养基来培养。
幽门螺杆菌在人体中存活于胃粘膜上。幽门螺杆菌是胃炎、胃溃疡、十二指肠溃疡的病因菌,而且还与胃MALT淋巴瘤、萎缩性胃炎、胃增生性息肉等疾病相关。幽门螺杆菌一旦固定附着在胃粘膜上,尽管对感染的免疫应答强烈,但却不会被除去而在胃内持续存活。而且,胃内由于盐酸而保持非常低的pH,因此多数抗生素被钝化。
基于这样的理由,在进行幽门螺杆菌的除菌时,组合使用抗生素和强烈抑制胃酸分泌的质子泵抑制剂。根据平成15(2003)年2月的日本幽门螺杆菌学会的幽门螺杆菌感染的诊断与治疗的指导方针修订版,将质子泵抑制剂+阿莫西林+克拉霉素给药1周的3剂合用疗法是首选的。
但是,在长期给与抗生素时,可怕的是耐药菌增加这一非常重大的问题。在日本,克拉霉素耐药菌的出现率从2000年开始骤增,在2004年达到了25~30%(日本幽门螺杆菌学会的监测),近年来有进一步增加的趋势。在耐药菌感染例中,报告有除菌率显著降低,且除菌不成功后发生获得克拉霉素耐性的例子,轻易进行不充分的除菌治疗被认为会增加耐药菌的出现。进而,对于过去长期使用大环内酯系药剂的病例,有可能使菌对克拉霉素获得耐药性(上述幽门螺杆菌感染的诊断与治疗的指导方针)。
因此,期待开发出没有这样的缺点的幽门螺杆菌的除菌剂。
发明内容
本发明提供能有效排除幽门螺杆菌的除菌剂。
本发明的发明人等,为了解决上述问题而进行了深入努力地研究,结果发现,野拔子蜂蜜能够有效地除去幽门螺杆菌,从而完成了本发明。
即,本发明涉及下述内容:
1.一种幽门螺杆菌除菌剂,含有野拔子蜂蜜作为有效成分;
2.上述1所述的除菌剂,用于预防或改善与幽门螺杆菌相关的疾病;
3.一种幽门螺杆菌的除菌方法,包括如下步骤:食用上述1所述的除菌剂;以及
4.一种用于除去幽门螺杆菌的食品添加用组合物,含有野拔子蜂蜜。
本发明的抗幽门螺杆菌除菌剂对幽门螺杆菌具有优异的除菌作用,且安全性高。进而,在短期内得到除菌效果。因此,对于幽门螺杆菌的除菌、与幽门螺杆菌相关的疾病的预防或改善是非常有用的。
具体实施方式
在本发明中,所谓幽门螺杆菌除菌剂,是指能够抑制幽门螺杆菌的增殖,从而从胃内除去幽门螺杆菌的组合物。本发明的除菌剂可以用于预防或改善与幽门螺杆菌相关的疾病,更具体来说,可以用于预防或改善胃溃疡和十二指肠溃疡等消化性溃疡、胃炎以及胃癌等。
所谓野拔子蜂蜜,是从野拔子中采蜜而得的蜂蜜。所谓野拔子(野巴子,Elsholtzia rugulosa Hemsl.),是唇形科的灌木,野生生长于中国云南省。高度为0.5~1.5m,茎为灰褐色且多分枝,花冠为白色,有时为淡黄色或紫色,5裂,为二唇形。从野拔子中采蜜而得的蜂蜜的特征在于自然地凝固而成为固体。
本发明的野拔子蜂蜜可以使用通过常法精制而成的蜂蜜,也可以直接使用市售的蜂蜜。野拔子蜂蜜还可以添加到任意的食品原料,例如牛奶、橙、柠檬等的果汁、酸奶等乳制品、面包、其它食品中后使用。
此外,关于本发明的除菌剂,成人每天的野拔子蜂蜜的摄取量通常为0.01~10.0g。通常,除菌剂中的野拔子蜂蜜的浓度根据其形态而异,当为片剂(tablet)、咀嚼片、颗粒、胶囊(例如硬胶囊)等固体形态时,相对于总质量为10~100重量%的范围。优选以在胃内的最终浓度达10~100μg/ml来配制。
作为本发明的除菌剂的形态,可以举出片剂(tablet)、咀嚼片、颗粒、胶囊(例如硬胶囊)等固体食品和流食等各种形态,进而还可以举出汤类、果汁类、茶饮料、乳饮料、发酵乳饮料、豆浆、可可饮料和啫喱状饮料等液状饮料食品;布丁和酸奶等半固体食品;面包类;乌冬等面类;曲奇、巧克力、糖果和脆饼等点心、干燥调味粉(ふりかけ),黄油和果酱等涂抹类等的形态。此外,本发明的除菌剂可以制成保健用食品或医疗用食品,对其形态没有特别限制,除了片剂(tablet)、咀嚼片、颗粒、胶囊等之外,还优选点心类、汤、饮料和流食等能持续摄取的形态。
进而,本发明的除菌剂中可以配合各种食品添加剂,例如各种营养素、各种维生素、矿物质、食物纤维、多元不饱和脂肪酸、分散剂和乳化剂等稳定剂、甜味剂、呈味成分、风味剂等。此外,为液状时,可以最开始即制成液状,也可以制成粉末或糊状,并溶解在规定量的水性液体中。
本发明的除菌剂能够除去胃肠内的幽门螺杆菌,因此可以用于预防或改善与幽门螺杆菌相关的疾病。作为与幽门螺杆菌相关的疾病,包括由于幽门螺杆菌存在于胃肠内而引起的疾病。具体来说,包括胃溃疡和十二指肠溃疡等消化性溃疡、胃炎以及胃癌等(幽门螺杆菌感染的诊断与治疗的指导方针(修订版),日本幽门螺杆菌学会,2003年)。日本幽门螺杆菌学会在2000年6月发表了《诊断与治疗的指导方针》,提出幽门螺杆菌阳性的胃溃疡、十二指肠溃疡均适于除菌治疗的建议。
通过下述实施例更详细地说明本发明,但本发明并不受它们的限定。
实施例
野拔子蜂蜜对幽门螺杆菌的抗菌效果如下进行评价。
实验材料和实验方法
1.使用从人(34人)采集的幽门螺杆菌的菌株(克拉霉素敏感性菌株23株,克拉霉素耐药菌株11株)。此外,还研究了对于属于普通细菌的从人(5人)采集的大肠菌(5株)的抗菌效果。野拔子蜂蜜的抗菌活性使用纸片法以最低抑菌浓度(Minimum InhibitoryConcentration,MIC)为指标进行研究。作为研究对象的蜂蜜是野拔子蜂蜜(皇蜜(注册商标)中国产2组),作为对照组是一直以来被认为对幽门螺杆菌具有抗菌效果的麦芦卡(Manuka)蜂蜜(JRJ株式会社,新西兰产,UMF10+),将两者进行比较研究。抗菌力试验以单盲法进行。将各蜂蜜用40度~50度的加热灭菌水溶解,依次将稀释原液(10%浓度)稀释10倍,以纸片法实施。进而,为了观察因加热所致的抗菌力变化,还使用在85度已加热30分钟、60分钟的稀释原液进行研究。
2.对于已获得充分的研究内容的说明和承诺的幽门螺杆菌携带者且未治疗的志愿者进行麦芦卡蜂蜜的临床试验。基于1中得到的MIC浓度在每餐前和就寝前摄取4次下面的溶液,即,在水150ml溶解有相当量(3g或5g)的野拔子蜂蜜而成的溶液,每周进行作为幽门螺杆菌除菌判断试验的尿素呼气试验。
实验结果
1.在采用纸片法进行的抗菌力试验结果中,可以确认,野拔子蜂蜜2组、麦芦卡蜂蜜在培养基中的浓度100μl(10%稀释)/mL时均阻止了克拉霉素敏感性HP菌株(23株)、克拉霉素耐性HP菌株(11株)中的所有菌株的生长。野拔子蜂蜜具有与麦芦卡蜂蜜同等的抗菌力,可以确认不仅对药剂敏感性HP菌而且对耐药性HP菌也具有阻止菌生长的效果。关于对大肠菌的抗菌力,野拔子蜂蜜在任意浓度下均未发现有抗菌作用,麦芦卡蜂蜜在100μl(10%稀释)/mL下发现对大肠菌也有抗菌作用。即使在85度加热30分钟、60分钟,野拔子蜂蜜、麦芦卡蜂蜜对幽门螺杆菌的抗菌效果均没有变化,在100μl(10%稀释)/mL下所有菌株的生长均被阻止。麦芦卡蜂蜜对大肠菌的抗菌力消失。
2.在每天12g(1次3g×4次)连续摄取野拔子蜂蜜1周后,采用尿素呼气试验测得的菌数与摄取前相比减少率为50%。另一方面,在每天20g(1次5g×4次)连续摄取1周后,在尿素呼气试验中可以确认菌完全被除去。
片剂和胶囊
将野拔子蜂蜜250g、结晶纤维素580g、还原麦芽糖118g、作为粘多糖类的果胶12.5g混合,向其中添加蔗糖脂肪酸酯40g并进一步混合,制成粉末。将该粉末用打片机打片,制备每片500mg的片剂。此外,将该粉末填充在2号硬胶囊中,制备胶囊剂。
发酵乳饮料
在温水(40℃)7kg中溶解脱脂奶粉800g、黄油100g,其次是作为粘多糖类的瓜尔胶25g、野拔子蜂蜜150g后,加入温水(40℃),使总体量达到10kg。向该溶液中添加酸奶发酵剂培养菌1质量%,一边搅拌一边在40℃保持,直到乳酸酸度达到1%。乳酸酸度达到1%时,冷却至10℃,包装为每个200ml,制备乳酸菌饮料。
本发明的幽门螺杆菌除菌剂用于与消化性溃疡的发生有关的幽门螺杆菌(Helicobacter pylori)的除菌是有用的。
Claims (2)
1.野拔子蜂蜜在制备用于预防或改善与幽门螺杆菌相关的疾病的除菌剂中的应用。
2.野拔子蜂蜜在制备用于除去幽门螺杆菌的食品添加用组合物中的应用。
