Abstract
测定体内和体外细胞生长率的方法。用花青染料标记细胞,用质膜上花青染料水平的变化测定生长率。利用细胞生长率测定检测移植的骨髓细胞的植入和手术后角膜上皮细胞的生长。所发明的方法还用于测定肿瘤细胞对肿瘤治疗剂的敏感性、酵母对抗真菌剂的敏感性和细菌对抗菌剂的敏感性。

本项发明涉及测定体内和体外细胞生长速率的新方法。
目前有两种测定细胞生长的普通方法。一种方法是在分析阶段开始时计细胞的数量,而后在该阶段结束时再计细胞的数量,从而测定出细胞数量的增多。可应用借助血球计数器的显微镜方法或应用仪器辅助的方法,例如应库尔特(Coulter)计数器或其它的流式血细胞计数器来进行细胞计数。测定细胞生长的另一种方法是用β计数法测定细胞对氚标记的胸腺嘧啶核苷的吸收。按照此方法,在实验开始时测定细胞的数量,而后在细胞中加入氚标记的胸腺嘧啶核苷,以定期的间隔取出一份培养液,计数,洗去未结合的氚标记胸腺嘧啶核苷,并将洗过的这份细胞用三氯乙酸(TCA)沉淀,然后对放射标记了的固体沉淀物进行闪烁计数,从而测定氚标记胸腺嘧啶核苷对DNA的掺入。此方法测定的只是DNA的合成速度,而并未测定细胞生长本身。但由于这个方法比较简单,因此此法通常是测量细胞生长的优选方法。用于测定细胞增殖活动的另一种方法是测定在所考察的任何组织中,每一百个细胞中进行有丝分裂的数量。这种方法并不十分精确,因为在制造过程中会损失细胞。一般说来,这些方法能较好地测定体外细胞生长,但对于体内测定细胞生长则是十分困难的。
取出组织并在体外监测有规律的氚标记胸腺嘧啶核苷的掺入,就可以判断组织的生长速率。将组织切成30μm的切片,与氚标记的胸腺嘧啶核苷接触30分钟,洗去未掺入的氚标记胸腺嘧啶核苷,在切片上涂一
层核乳胶,并对胶片曝光放射性同位素的衰变。对乳胶显影和定影。组织用苏木精和伊红染色,然后镜检以测出被标记的部分。这种方法既费时又费力。
花青染料已广泛应用于生物学,二噁羰花青染料(dioxacar-bocyanine dyes)已用于进行白血细胞分类计数,Gunter Valet,Max Planck Ges Wissensch;登记专利号84-102307/17,“用选择性染色并测定体积和荧光法同时定量测定血细胞”。但是,在这些研究中,所用的染料是一些短链的羰花青染料(少于10个碳),对细胞膜电位的变化有反应。同时,这种短链的羰花青染料进入细胞的线粒体,有细胞毒性,当洗涤细胞时,无论细胞膜电位是否发生变化,这种染料都易于从细胞中漏出来。其它的短脂肪链花青染料用于许多其它生物学分析中。但是这些短链的分子对细胞膜电位的变化有反应并穿过细胞膜进入线粒体。H.M.Shapiro,美国专利号4,343,782,1982年8月10日。这些短链染料对细胞也是有毒的,不能用于测定细胞的生长速率。
三羰花青染料(Fox,I.J.,et al.,Proc.Mayo Clinic,32∶478-484,1957)和伊文思蓝染料(Schad,H.,et al.,Pfluegers Arch.Eur.J.Physiol.,370(2)∶139-144,1977)已经以稀释法用于体内测心输出量。Dow(Dow,P.,Physiol.Rev.,36∶77-102,1956)描述了这个方法,其做法是在肺的静脉侧注射已知量的某种血管内指示剂,并测定动脉血管内指示剂浓度随时间的变化,从而确定注射点和取样点之间的体积。这些染料不能用来将细胞染色。
本发明是测定细胞生长速率的新方法。按照本发明,首先用花青染料标记活细胞。测定被花青染料标记的母细胞所产生的子细胞在细胞质膜中花青染料量的变化,以确定细胞生长速率。应用本发明的测定生长的方法,例如在体内监测角膜上皮的愈合、移植骨髓细胞的植入。本发明的方法在体外的用途包括测定肿瘤细胞对各种化疗剂的敏感程度。

在本发明的测定细胞生长速率方法中,用花青染料来标记细胞。本文涉及的具有下述结构的化合物为花青染料:

本发明所用的烷基取代的亚甲基是指具有与甲基、乙基或丙基取代基的任何结合形式的单或双取代的亚甲基。

Sims,P.J.,et al.,Biochem,13∶3315(1974)。因此,例如其中Y是硫,并具有将环和2个14碳的脂肪链联接起来的3个碳原子的化合物,可写成DiSC14(3)。同样地,DiIC14(5)是指其中Y是异丙基,并有将环和2个14碳的脂肪链联接起来的5个碳原子的化合物。
在本文中所述的花青染料的化合物包括具有一个或一个以上的取代基的上述结构的化合物,其条件是此种取代的化合物至少在进行标记
所需要的时间内始终溶于细胞标记介质中,并且还具有足够高的膜分配系数,使其保持与标记的细胞膜相联结。在进行标记所要求的浓度范围内,此种化合物还必须不显著影响细胞的活力。花青染料在细胞标记介质中的溶解度的测定方法如下:将花青染料分散到标记介质中,用标准的荧光分光光度技术,测定荧光强度随时间的变化。荧光强度减小说明染料沉淀并粘附在容器壁上。测定该染料是否仍然与细胞膜结合的方法是,例如应用公知的流式血细胞计数方法监测标记后重新注射入供血动物的红血细胞的荧光强度。重新注入后,标记细胞荧光强度基本不变说明染料在细胞膜上的稳定性。
本发明所用的花青染料可从多处购得,例如Molecular Probes,Inc.,Eugene,Oregon,或用已知的合成方法从现有的原始材料制备,Hamer,F.M.,“花青染料及有关化合物”,Interscience Publishers(1964)。
应用所述的步骤,任何活细胞均可用花青染料进行标记。本文中中所用的术语“细胞”包括有核细胞如白血细胞、各种肿瘤细胞、其它哺乳动物细胞(例如组织培养细胞),酵母和细菌。如果一个细胞能够生长并基本上能发挥该类细胞所应有的功能,那么它就是活细胞。
细胞标记是在对细胞无毒并使细胞标记可重复的介质中进行的。为了使该介质具有这些必要的特性,应用渗透压调节剂使花青染料至少在进行标记所需要的时间内,在介质中形成稳定的溶液。适用的渗透压调节剂包括例如糖,象单糖,如葡萄糖、果糖、山梨糖、木糖、核糖、和二糖、如蔗糖,糖醇,如甘露醇、甘油、肌醇、木糖醇和阿东糖醇,氨基酸,如甘氨酸和精氨酸,以及一些Good′s缓冲剂如N-三(羟甲基)-甲基-3-氨基丙磺酸和如下面表2中所列的化合物。Good,N.E.,et al.,Biochem.15,467-477(1966),Good,N.E.and S.Izawa,Methods Enzymol.,24,Part B,53(1968),Feguson,W.J.,et al.,Anal.
Biochem.104∶301-310(1980)。但某些细胞系可能对一种或多种渗透压调节剂敏感,特别是对糖醇。因此,在标记前,要进行一些标准的实验以确定细胞在这些所要用的渗透压调节剂中是有活力的。此外,在标记介质中可加少量缓冲剂以调节氢离子的浓度。
接触各种渗透压调节剂对细胞生存力的影响可由测量Yac细胞在接触各种渗透压调节剂30分钟后的倍增时间来确定。Yac细胞是小鼠淋巴瘤组织培养细胞系,公众可由American Type Culture Collection得到,并由Kiessling,R.,European J.Immunology 5∶112-117(1975)描述。表1的数据说明,和磷酸盐缓冲液相比,与蔗糖、葡萄糖、Good氏缓冲液(TAPS、CAPS、EPPS、HEPPSO和DIPSO)接触对细胞倍增时间产生可忽略的影响,这表明,未产生因接触这些试剂而出现的细胞毒性。

表2列举了用来检定花青染料溶解度的各种渗透压调节剂。所有的浓度的测定都是在离心除去沉淀和为了进行荧光分光光度分析而将少量含有花青染料的渗透压调节剂溶于乙醇后进行的。所用的染料是DiSC14(5)和DiOC14(3),而渗透压调节剂的浓度为对哺乳动物细胞来说是等渗的浓度,根据乙醇溶液标准,荧光强度的降低,直接与花青染料的溶解度降低相关联。

从表2可以看出,花青染料在一般盐中的溶解度要比在糖(除了来苏糖)、糖醇、氨基酸、和Good氏缓冲剂(TAPS、HEPPO、DIPSO、CAPS和EPPS中)的溶解度小得多。另外,已经测定了DiSC14(5)在糖溶液中,如葡萄糖、果糖、核糖、山梨糖、蔗糖和木糖,糖醇如甘油、肌醇、木糖醇和阿东糖醇以及氨基酸如甘氨酸、精氨酸溶液中的稳定性。花青染料在测试溶液中至少能稳定20分钟,这个时间足够进行可重复的标记,在许多试剂中花青染料在溶液中的量在60分钟内无明显减少。
此外,还测定了花青染料在含有一般盐和染料能在其中溶解的渗透压调节剂的介质中的溶解度。DiSC(5)在等渗葡萄糖溶液中的溶解度并不显著地受到用蒸馏水进行稀释的影响。但是,DiSC14(5)在等渗葡萄糖溶液中的溶解度,才用大约20%的等渗氯化钠溶液来稀释,则其溶解度就显著地降低。因此,在仅含有少量的一般盐类,如氯化钠、氯化钾、氯化钙、乙酸钠、乙酸钾、硫酸钠、碘化钠、氯化胆碱或磺化胆碱中,用花青染料即可进行可重复的细胞标记。而最好是在不用一般盐来调节渗透压的介质中进行。
对按本发明程度用花青染料标记的细胞进行分析以确定标记对细胞活力的影响。将从American Type Culture Collection,Rockville,Maryland得到的V79细胞(这种细胞由Prescott,D.M.,Ann.New York
Acad.Sci.,397∶101-109(1982)描述)用含DiOC14(3)10-5或4×10-5M的溶液进行标记,并将染色细胞的生长动力学与未染色的细胞及染色与未染色细胞各占一半的混合物的生长动力学进行比较。用花青染料进行标记并不影响细胞倍增时间。因此,标记对细胞生长并无影响。另外,其它一些测定细胞活力的标准实验方法,如台盼蓝排除实验和Propidium iodide排除实验也证明,按所述程度进行的花青染料标记对细胞活力并无影响。
为了测试按本发明方法用花青染料标记的细胞在体内的稳定性,抽取兔子的红血细胞,用DiSC14(5)进行标记,再回输入兔体内。此后定期采血样,分析标记细胞的百分比以及标记细胞的荧光强度。循环的红血细胞的数量随时间而线性降低,所测出的标记细胞的寿命为52天,这与已报导的兔红细胞的平均寿命为40-60天十分接近。因此,花青染料标记并不影响红血细胞的清除率。
在受试的5只兔子中,除一只外,其余动物的染色细胞的荧光强度在标记并重新注入后的60天基本上保持不变。在第5只动物中,在兔子的循环系统中60天后,有不超过20%的花青染料从标记细胞中流出。这些数据与在组织培养中显示未见染料从标记细胞转移到未标记细胞的数据一起,证明了细胞被这种染料稳定地标记。
花青染料标记的活细胞在所发明的方法中用于测定细胞的生长率。测定花青染料在细胞质膜中的量的变化从而确定细胞生长速率,每次当一个细胞分裂时,与质膜结合的花青染料均等地分布于两个子细胞上。因此,测定一系列标记的、生长着的细胞的质膜中花青染料的量,可用于计算细胞的生长速率。
应用标准技术的流式血细胞计数法,适用于测定不粘附壁的细胞或可从其生长基质中取出并可以以单个细胞状态悬浮的细胞质膜上花青染料的量。一种粘附壁的细胞的血细胞计数器(Meridian ACAS 470)适用
于细胞不易或不能从生长基质中取出的情况。
可将细胞生长速率的测定用于许多用途。例如:测定组织培养细胞的生长速率可用来选择最适生长条件。测定肿瘤细胞在含化疗剂的介质中的生长速率就可确定肿瘤细胞对化疗剂的敏感度。同样地,测定酵母细胞在含各种抗真菌剂的介质中的生长速率就可确定酵母细胞对各种抗真菌剂的敏感度。
本发明方法也用于监测体内组织的生长速率。例如测定骨髓细胞在被移植后的生长速率就能确定骨髓移植物的植入情况,测定其它体内细胞如角膜上皮细胞的生长率可以确定创伤后或手术后的愈合情况。
以下实施例说明本发明,但是不能将本发明的范围限制在以上所述的范围和以下所述的权利要求书的范围内。
实施例1
组织培养细胞的染色方法
Ⅰ.细胞的准备
利用对数期的培养细胞以得到最佳结果。从培养器皿中取出悬浮培养物,放到聚丙烯离心管中。
如果用的是单层培养,必须除去上清液,用不含钙和镁的磷酸盐缓冲液洗涤粘附壁的细胞,以除去培养瓶中的血清蛋白。加入胰蛋白酶-EDTA溶液(Gibco Laboratories,Grand Island,New York,*610-5300),覆盖培养瓶的底部并令其在室温下培养,直到细胞单层松动并解聚。将所得的细胞悬浮液转移到聚丙烯离心管中,加入等体积的含10%小牛血清(FBS)(Hazelton)的培养液,以终止胰蛋白酶的酶作用。
细胞在室温下以400xg离心10分钟。吸出上清液,并换用等体积的等渗甘露醇以重新悬浮细胞片状沉淀物。用甘露醇洗涤的目的是除去细胞悬浮液中的血浆蛋白,并准备对细胞进行染色。细胞在室温下以400xg再离心10分钟。吸出上清液,所得的细胞片状物再悬浮于甘露醇
溶液中,使每毫升悬浮液中含2×106个细胞,以便染色。但有些细胞系对糖醇(甘露醇)的使用敏感,在这种情况下可用等渗的葡萄糖溶液(分子量180.16,54.05克/升)。
Ⅱ.染料原液的制备
用无水乙醇配制2×10M的原液如下:
DiO-C14(3) 分子量800(1.600毫克/毫升)

DiO-C14(3) 分子量936(1.872毫克/毫升)
DiI-C14(5) 分子量850(1.700毫克/毫升)
从美国俄勒冈尤金Molecular Probes公司得到所有的染料。
染料原液经超声处理以保证染料完全溶解并使其对试管的粘附减少到最低程度。用聚苯乙烯管来制备原液以便能观察到染料的溶解情况。但是,用聚丙烯管来进行细胞染色,因为与聚苯乙烯相比,花青染料在水性环境中对聚丙烯的粘附要少得多。
Ⅲ.细胞染色
将细胞在等渗甘露醇溶液中调到浓度为2×106个细胞/毫升。按每毫升细胞悬浮液中加2×10-3M的染料原液5μl将染料原液加到染色溶液中,使终浓度达到10μM,将染色的样品用吸管吹吸或搅拌使其充分混合。将细胞与染料一起温育十分钟后,取一小份样品在荧光显微镜下观察,以确保已产生强烈而均匀的染色。对于DiO系列的染料,使用对488nm的激光具有选择性的显微镜滤光片,而DiS和DiI染料系列则需在靠近575nm激发,以观察荧光。
保温后,于染色细胞悬浮液中加等体积的磷酸盐缓冲的盐水(PBS)。细胞在20℃下以400xg离心10分钟。吸出上清液,用磷酸盐缓冲的盐水(PBS)将片状沉淀物重新悬浮。重复离心过程,观察所得上清液中是否有染料存在。如果上清液中仍有染料存在,重复洗涤直到用荧光分光光
度法测不出上清液中有染料为止。最后一次洗涤后,除去上清液,并将该片状沉淀物重新以所需浓度悬浮于合适的培养液中。全部操作均在无菌条件下进行。
实施例2
组织培养细胞生长速率的测定
按实施例1中所述的方法用DiOC14(3)将V79细胞进行染色。用EPICS753型流式血细胞计数器测定荧光强度(Coulter Electronics,Inc.)。染色后立即取一份被染色的细胞测定荧光强度,其余的细胞在标准完全培养介质中,37℃于湿润的空气-CO(7.5%CO)培养箱中培养,在染色后的第一天、第二天、第三天分别取细胞样品进行荧光强度测定。
图1显示生长中的V79细胞荧光强度对数的曲线。在每一个峰上面标出的数值是在那一天的平均荧光强度值的对数值。如图1所示,随着培养细胞的生长,荧光强度的平均对数值逐天减小。
通过对荧光的测量,从与荧光强度的对数值对时间所做的图的线性部分配合的回归线的斜率,可测定出生长速率。
为了确认细胞染色并不影响其生长速率,比较了被染色的V79细胞和未染色的V79细胞的生长率。图2的结果表示,未杂色的细胞与用10-5或4×10-5M染料染色的细胞,或与未染色细胞和染色细胞的等量混合物,以同样的速率生长。
因为染料由染色细胞向未染色细胞转移将引起生长率测定不正确,所以将染色与未染色的细胞放在一起共同培养。将一种用DiOC14(3)染色的人类结肠癌细胞系HT29(可由American Type Culture Collection,Rockville,Maryland得到,由J.Fogh and G.Tremp,《人类体外癌细胞》,pp.115-159,Plenum Press,New York(1975)描述)与未染色的人前骨髓白血病细胞HL60(可由American Type Culture Collection,
Rockville,Maryland得到,由Collins,S.J.,et al.,Nature,270∶347-349(1977)描述)以1∶1的染色细胞/未染色细胞的比例共同培养。所得的结果示于图3。
图3中,单独培养的染色的HT29细胞的荧光强度(菱形点)表明荧光强度随培养时间而降低的特征。未染色的HL60细胞在4天内几乎无荧光,4天后荧光强度显示大约中等程度的增加。此外,与HL60细胞共同培养的染色的HT29细胞(方形点),直到大约第5天以前,其荧光强度的损失与没有和HL60细胞一起培养的TH29细胞荧光强度的损失完全一样,此后只出现一些额外的、但只是极小量的荧光强度的损失。从这些数据可清楚地看到,HL60细胞在与HT29细胞培养4天后也许也吸收一些荧光染料。但是应该注意到HL60细胞是前髓细胞,具有某种吞噬细胞和细胞碎片的能力。培养4天后,在混和培养物中的未染色亚种群所经历的荧光强度的增强或许实际上是由于它们吞噬了有荧光的碎片的结果。更重要的是在4天的培养中,在未杂色的HL60细胞中荧光强度没有增加。再者,在同一混合物中染色的HT29细胞的荧光动态与未经混合的HT29细胞的荧光动态表现一致。因此,并未因细胞与细胞间染料的转移而导致错误的生长率测定。
实施例3
肿瘤细胞生长率的测定
如果研究的肿瘤细胞是适应于体外培养条件的组织培养系,那么就按实施例1及实施例2中所述的方法进行细胞染色和测定。如果研究的细胞来自肿瘤组织外植体,那么就用一般技术将其打散成单个细胞,并把它们涂到Lab-Tech组织培养箱的载片上。按实施例1中所述的方法用花青染料将细胞染色。然后将其放在37℃湿润的空气-CO2培养箱中保温进行平衡。每隔一段时间,将载片放在粘附壁的细胞计数器(即Meridian ACAS470)的显微镜镜台上测定荧光强度。细胞相对于参比
标记的位置可用计算机进行测定,所以能进行一系列的荧光强度测定。将载片放回培养箱中,使细胞继续生长。显微镜载片上应用荧光标准,例如库尔特全光聚苯乙烯微球(coulter fullbright polystrene microspheres)(美国弗罗里达海厄利里库尔特电子仪器公司),该微求在培养过程中荧光强度不随时间而变化。此标准是用来进行荧光强度测量比较。
可用显微镜上的相镜片测量形态学参数,或用荧光标记的对肿瘤细胞特异的单克隆抗体,将肿瘤细胞从正常基质细胞中鉴定出来。细胞生长的测定则是监测肿瘤细胞在每次分裂后花青染料荧光的稀释,并应用实施例9中的公式。
也可用流式血细胞计数器来测定荧光强度。但是在进行流式细胞分析前需将细胞从显微镜载片上取下来。这种方法不能用同一个细胞每天进行测定而进行染料稀释的一系列定量分析。但在某些例子中,如白血病细胞,这种方法较好。
测定体外细胞生长是用在最适培养基质中培养的细胞或用在各种不同量的治疗肿瘤的制剂的存在下培养的细胞。由抑制荧光强度的降低所测出的治疗剂抑制肿瘤细胞生长的能力,是对该制剂杀死肿瘤细胞的效力的一种度量。
实施例4
白血细胞生长速率的测定
用常规技术由脾脏切除术或静脉抽血法取淋巴细胞。用实施例1的方法用花青染料来标记细胞,但是用葡萄糖或蔗糖代替甘露醇作为渗透支持介质。然后将被染色的细胞分置到微量滴定板的每一个孔中,每孔5×105个细胞(Mazumder,A.,Grimm,E.A.,Zhang,H.Z.,and Rosenberg,S.A.,Cancer Res.42,918(1982)),与适当的细胞分裂素如白细胞介素-2、高碘酸钠(IO- 4)、植物凝集素、伴刀豆球蛋白A、
美洲商陆(Pokeweed)-细胞分裂素和B细胞生长因子(BCGF)一起培养。将细胞放在37℃湿润的空气-CO2培养箱中使细胞生长。隔一定的时间将细胞从培养容器中取出,用流式血细胞计数技术测定。得到的数据与实施例2得到的数据类似,并用实施例9中的方程进行分析。
实施例5
细菌生长率的测定
用实施例1中的方法以花青染料标记细胞,但是用葡萄糖或蔗糖代替甘露醇作为渗透支持介质。然后将被染色的细胞按每一小孔中5×105个细胞的量分置于微量滴定板的各个含有肉汤培养液的小孔中。将细胞放入37℃湿润的培养箱中使细胞生长。隔一定时间,将细胞从培养容器中取出,用流式血细胞计数方法测定。测得的数据与实施例2中所得的数据类似,并用实施例9中的方程分析。
测定体外细胞生长的方法是用在最适培养基质中培养的细胞或在有不同量的抗生素存在下培养的细胞进行的,加入抗生素是为了测定它们的抗菌能力。由抑制细胞荧光强度的降低所测出的杀菌剂抑制细胞细胞生长的能力,是对该制剂杀死细胞效力的一种度量。
实施例6
酵母生长率的测定
用实施例1中的方法用花青染料标记细胞,但用葡萄糖或蔗糖代替甘露醇作为渗透支持介质。然后将被染色的细胞以每个小孔中5×105个细胞的量分置于微量滴定板上的每个含有肉汤培养液的小孔中。将细胞置于培养箱中使其生长。隔一定时间,从培养容器中取出细胞,并用流式血细胞计数法或粘附壁的细胞计数法(Meridian ACAS470)进行测定。所得的数据与实施例2得到的数据类似,并用实施例9中的方程进行分析。
测定体外细胞生长的方法是用在最适生长基质中培养的细胞,或
是在有待测其抗真菌能力的化合物以各种所选量存在下培养的细胞进行的。由抑制细胞荧光强度的降低所测出的杀真菌剂抑制真菌细胞生长的能力是对该制剂杀真菌效力的一种度量。
实施例7
骨髓细胞生长率的测定
在胸骨或髂骨嵴处用吸出法(Illinois针头)或中心活组织检查法(Jamshiti针头)取出骨髓细胞。按实施例1中的方法用花青染料标记细胞,但是用葡萄糖或蔗糖代替甘露醇作为渗透支持介质。在将骨髓细胞输给受体以前,先有流式血细胞分析技术测定其荧光强度的大小。将标记的细胞由静脉注入,在从外周血和骨髓中取样前要等待适当的时间。由受体采血和骨髓,与抗凝剂混合,按常规技术制备细胞以用于流式血细胞分析。这些样品中含有正在生长的细胞,也含有未进入细胞周期的细胞。直方图将是很复杂的,但是,是通过应用实施例9的方程式,用来分析与在实施例2的方法中所测得的数据相类似的细胞生长的。
由于示踪染料是荧光绿,所以应用另一种与单克隆抗体偶联的染料来鉴定在骨髓中的每一个细胞谱系。应用染色红细胞及其前体的单克隆抗体,用红色的荧光来鉴定此谱系的细胞,然后再监测在该红血细胞谱系中的细胞的绿色荧光(及细胞生长)的降低。
与此相似的双色法被用于测定淋巴样细胞、髓样细胞和单核细胞的生长。
实施例8
角膜上皮生长率的测定
此方法用来检测角膜上皮移植后的生长情况,在检测细胞生长时,此法应用不伤害细胞生长、眼睛不痛的技术。移植后立即用眼科配方的花青染料洗眼睛,此花青染料吸收大于680nm波长的光。此操作也可用吸收低于680nm波长的光的染料进行,但激发光束在检查组织时会引起
剧烈头痛。
在零时刻,在将结合到角膜上的花青染料激发时进行红外照相(波长大于染料的最大吸收波长)。荧光强度的大小是在零时刻细胞染色情况的一种度量。之后再给眼睛照相,并将图象与零时刻拍的照片进行比较。在细胞生长的区域内,细胞的荧光强度减小。荧光强度的定量测定用于测定细胞倍增的数目。
实施例9
细胞生长率的计算
下列数学公式用于计算细胞倍增时间:
其中:
TD是细胞倍增时间,
t2和t1是细胞对数生长期的两个任意时刻,
F(t2)和F(t1)分别是在t2和t1时刻的细胞平均荧光强度,
ln表示自然对数(底数是e)。
某生长期中的细胞倍增数由下列公式决定:
其中:
N是细胞倍增数目,
F0是初始时刻的荧光强度,
F(t)是细胞生长期后任一时刻的荧光强度。为了测定细胞倍增数,不必一定在细胞对数生长期测荧光。
下面的推论说明上述公式能精确算出细胞倍增时间及细胞倍增数,并说明用数学模拟预测的在生长中的细胞质膜上花青染料的量与实测结
果相平行。
推论基于以下假设:
1.细胞以低密度培养,具有起始数目(N0)。平均起始荧光强度(F0)及细胞周期时间(T2)。
2.在细胞分裂后,染料均匀地分布于子细胞中。
3.细胞培养过程中无滞后时间。
4.细胞的死亡是可忽略的。
5.染色是永久的,不存在细胞与细胞之间的染料转移。
基于以上的第2个假设,细胞群体的平均荧光强度应与任何时刻t的群体中细胞数目成反比。这个关系可用方程1表示:

其中F(t)和N(t)分别是t时刻细胞群体的平均荧光强度和细胞数。K是比例常数。如果我们在零时刻计算这个方程,就成为K=FN。把它代入方程1解F(t),得到:

F0和N0在假设1中定义。
从方程2中可明显看出,如果所用的染料以理想的方式作用,荧光动力学定与生长动力学直接有关。现在,可确定定义荧光动力学的关系式。方程2中的细胞数目遵从如下定义的一般生长动力学。

其中A是群体生长率与种群中细胞数之间关系的一个比例常数,方程4-7给出方程3的简单解法:

对这个方程进行解释时要慎重考虑。首先,因我们已假定忽略细胞死亡,所以T代表细胞周期时间而不是细胞倍增时间。这一点将在下面更仔细地考虑。如果有细胞死亡,只要结合在死亡细胞上的染料不被活细胞重新吸收,这个方程仍然正确。发生重吸收的情况将在下文中进行处理。如果细胞群中有一部分细胞不生长时,方程9只对生长的部分正确。因此,F0和N0只分别适用于生长部分。
细胞死亡对荧光动力学的影响
让我们考虑有明显的细胞死亡并且染料能很快移入活细胞的情况。细胞死亡的动力学可由方程10表示:

上式中,B是细胞从群体中消失的速率与群体大小的比例常数。将导数加注下标:下标1表示细胞死亡的过程。让我们以相似的方式重写方程3。

在方程17a和17b中,参考TD有时间单位,代表实际的细胞倍增时间而不是细胞周期时间。方程17b将限定培养物中的荧光的分布,该培养物中有明显的细胞死亡并有快速的染料重吸收。如果不满足这两个条件中的任何一项,就用方程9。比较方程17a和方程17b表明:细胞动力学和荧光动力学成反比关系,这样细胞动力学曲线与荧光动力学倒数的曲线可以重叠。这一事实在图4中清楚地说明。
细胞生长滞后时间的加入
我们现在假定当细胞进入培养时有一段滞后时间。进一步还假定种群将从生长率为零一步就达到最大生长率,从而简化处理过程。在这些条件下,限定细胞生长的关系式就可以容易地表示为:

式中,TL定义为滞后时间。将方程3与上述关系式合并,得出:

利用这些方程,已知F0并在指数生长期间测三或四次F(t),倍增时间和滞后时间就都可测出。
生长份额的测定
让我们考虑种群中部分细胞不生长的情况。首先让我们假定没有滞后时间。随后我们再将滞后时间加入。为进行推导,需一个定义。在方程21中:

下标g和n分别代表生长和不生长的部分。在任意时刻t,种群中细胞的数目应是生长的和不生长的种群之和。

种群中生长细胞的部分可用方程25测定。因为涉及到亚种群,这个测定比前面的情况复杂。为测定生长部分,必须等到可以区别两个亚种群。在这一点上,不生长部分的荧光强度可直接测定。将生长部分的荧光强度随时间的变化外推到零时刻,就可计算出生长部分在零时刻的荧光强度。生长部分的份额即简单地是生长部分零时刻的荧光强度与种群总荧光强度的比值。
以上说明了本发明的优选实施例,但是,本发明并不限于本文所公开的内容,同时本发明人还保留在以下的权利要求书的范围内,所进行的全部修正的所有权利。
Claims (20)
1、一种测定细胞生长速率的方法,其特征在于,此法包括:
获得将要被花青染料标记的母细胞;
用一种花青染料将所述母细胞标记;
测定由所述的花青染料标记的母细胞产生的子细胞的质膜中的花青染料量的变化;和
应用所述的测得的花青染料量的变化,以计算细胞生长速率。
2、一种权利要求1的方法,其中用荧光来测定花青染料水平的变化。
3、一种权利要求2的方法,其中的细胞是组织培养细胞。
4、一种权利要求3的方法,其中的细胞是人类肿瘤细胞。
5、一种权利要求4的方法,其中,在含有肿瘤治疗剂的介质中培养人类肿瘤细胞,以测定肿瘤细胞对这些制剂的敏感性。
6、一种权利要求3的方法,其中的细胞是白血细胞。
7、一种权利要求3的方法,其中的花青染料是DiSC14(5)或DiOC14(3)。
8、一种权利要求2的方法,其中的细胞是细菌。
9、一种权利要求8的方法,其中,在含有抗菌剂的介质中培养细菌以测定细菌对这些制剂的敏感性。
10、一种权利要求9的方法,其中的花青染料是DiSC14(5)或DiOC14(3)。
11、一种权利要求2的方法,其中的细胞是酵母。
12、一种权利要求2的方法,其中,在含有抗真菌剂的介质中培养酵母,以便可以测定对这些制剂的敏感性。
13、一种权利要求12的方法,其中的花青染料是DiSC14(5)或DiOC14(3)。
14、一种权利要求2的方法,其中的细胞是体内组织细胞。
15、一种权利要求14的方法,其中的体内组织细胞是移植的细胞。
16、一种权利要求15的方法,其中移植的细胞是骨髓细胞。
17、一种权利要求14的方法,其中的体内组织细胞是角膜上皮细胞。
18、一种权利要求2的方法,其中的花青染料是DiSC14(5)或DiOC14(3)。
19、一种权利要求2的方法,其中花青染料吸收波长大于680nm的光。
20、一种权利要求17的方法,其中花青染料吸收波长大于680nm的光。
