本发明涉及用于增强肠内细菌产生牛尿酚的经口组合物,其含有用于活化肠内细菌的牛尿酚产生能力的双果糖酐作为有效成分。

含有双果糖酐的经口组合物
技术领域
本发明涉及用于增强肠内细菌产生牛尿酚的经口组合物。
背景技术
牛尿酚的抗氧化活性、雌激素活性均高于代表性异黄酮的黄豆苷元和染料木黄酮,其进一步的保健效果令人期待。牛尿酚是黄豆苷元在肠内经肠内细菌代谢后的代谢产物,通过活化肠内细菌的牛尿酚产生能力,可期待体内可利用的牛尿酚增加。
另一方面,已知双果糖酐(以下称为DFA)具有双歧杆菌增殖作用(专利文献1:日本特公平3-5788号公报)、增加钙吸收量(专利文献2:日本特开平11-43438号公报)、具有利尿作用(专利文献3:日本特开2003-321371号公报),但其增强牛尿酚产生的作用还不为人所知。

发明内容
如上所述,通过增强肠内细菌的牛尿酚产生能力,可期待保健效果。为增加体内可利用的牛尿酚,必须活化肠内细菌的牛尿酚产生能力。
本发明者潜心研究使肠内细菌的牛尿酚产生能力活化的方法,在研究过程中发现:通过摄取双果糖酐(以下称为DFA),血中的牛尿酚浓度上升。即,本发明涉及增强肠内细菌牛尿酚产生能力的经口组合物,其特征在于:含有DFA作为有效成分。
即,本申请的发明的主要构成如下所述。
(1)用于增强肠内细菌产生牛尿酚的经口组合物,其特征在于:含有双果糖酐作为有效成分。
(2)用于增强肠内细菌产生牛尿酚的经口组合物,其特征在于:含有双果糖酐和黄豆苷元及/或其衍生物。
(3)医药品或饮食品,其含有(1)或(2)所述的用于增强肠内细菌产生牛尿酚的经口组合物。
(4)宠物动物用医药品或宠物动物用饲料,其含有(1)或(2)所述的用于增强肠内细菌产生牛尿酚的经口组合物。
本发明的含有DFA的经口组合物,可增强肠内细菌的牛尿酚产生能力,使血中的牛尿酚浓度上升。
具体实施方式
本发明所述的DFA为公知物质,是两个果糖分别在两个位置相互结合的脱水环状二糖。以往,已知其存在于焦糖等中,工业上可利用菊糖酶例如Arthrobacter sp.H65-7株产生的菊糖果糖转移酶(EC2.4.1.93)使菊糖(inulin)发酵或利用Arthrobacternicotinovorans GS-9产生的左聚糖果糖转移酶(EC2.4.1.10)使左聚糖(levan)发酵来制得。根据两分子果糖的结合方式不同,存在5种衍生物,分别为DFAI、DFAII、DFAIII、DFAIV、DFAV。本发明所述的DFA指上述全部DFA,但在本发明中,主要优选使用工业生产效率和精制后稳定性等均优良的DFAIII、DFAIV。
本发明所涉及的经口组合物是含有DFA作为有效成分的组合物,可作为医药品形式及/或饮食品(包括宠物动物用饲料)形式的组合物利用。只要是最终可经口给与的形态即可,例如:人或宠物动物用的医药品、饮食品、配方奶粉、经肠营养剂、保健饮食品、饲料添加物等。本发明最优选的形态为:使DFA和黄豆苷元及/或其衍生物以未被吸收的状态抵达大肠的形态,例如:在肠溶性胶囊中含有的形态。此外,有效成分的含量不受特殊限制。
当作为饮食品形式的组合物使用时,可以直接使用DFA,或与其他的食品或食品成分合用,采用合适的常规方法即可使用。加工时,因热稳定性、酸稳定性均较高,所以一般的食品加工方法均可适用,不存在任何问题。含有DFA的食品形式的经口组合物可以是粉末、颗粒状、糊状、液体状、悬浮状、片剂、胶囊剂等,不受特殊限制。例如,可使用甜味剂、酸味剂、维生素剂及其他在营养保健液制造中常用的各种成分,制成保健饮料。
当作为医药品形式的组合物使用时,本有效成分以各种形态给药。
其给药形态可例举片剂、胶囊剂、颗粒剂、散剂、糖浆剂等经口给药。按照常规方法,在主药中添加赋形剂、粘合剂、崩解剂、润滑剂、矫味矫臭剂、助溶剂、悬浮剂、包衣剂等医药制剂技术领域中常用的已知辅料,可制成上述各种制剂。
其使用量根据症状、年龄、体重、剂型的不同而各异,通常,就成人而言,经口给药时,每日每1kg体重给与0.5~2000mg,优选1~1000mg的范围。
另外,本发明的黄豆苷元及/或其衍生物,指4’,7-二羟基异黄酮和其糖苷、及/或其经乙酰化、丙二酰化等修饰后的衍生物。特别是,本发明使用的黄豆苷元及/或其衍生物优选黄豆苷元的糖苷即黄豆苷,因其吸收缓慢、到达大肠内的量多。此外,关于本发明使用的黄豆苷元及/或其衍生物的来源,可以是化学合成品、植物提取物及其加工品、植物本身。
下面,结合实施例,更详细地说明本发明,但这只是简单的示例,并不限制本发明。
实施例1
分析DFA对肠道内产生牛尿酚的增强作用。
取27只Wistar-ST系雄性大鼠(6周龄),用固体饲料预备饲养3天,然后根据体重分层法,分成3组,各组n=9,分别给与含有0.25%的含异黄酮40%食品的基础饲料(Basal diet组)、将基础饲料的蔗糖用整体3%的DFAIII置换后的饲料(DFAIII组)、将基础饲料的蔗糖用整体3%的果寡糖置换后的饲料(FOS组),饲养20天。在第20天,从尾静脉采血,用HPLC-UV法测定血浆中的牛尿酚(Equol)浓度。
结果见表1。表中a、b表示具有不同符号的值之间存在显著性差异。
该结果表明:通过摄取DFA,DFA使血浆中的牛尿酚(Equol)浓度上升。

1.双果糖酐和黄豆苷元在制备用于增强肠内细菌产生牛尿酚的经口组合物中的应用。
2.根据权利要求1所述的应用,其特征在于所述经口组合物为医药品或饮食品。
3.根据权利要求2所述的应用,其特征在于所述医药品为宠物动物用医药品。
4.根据权利要求2所述的应用,其特征在于所述饮食品为宠物动物用饲料。
