发明背景
各种研究领域,如功能基因学、基础生命科学研究、药物研发和临床诊断,需要对样品的分子机制进行研究,如对低聚核苷酸、cDNA或蛋白质相互作用进行不同方面的监视。为了研究分子机制,可以使用一种基于微阵列的检定手段,如一种光学检定手段,实例有荧光或磷光结合检定.
可以通过由多个探查分子点组成的微阵列来进行检定,这些分子被固定在构成支持体的载物片的不同位置上。探查分子点为待分析样品的目标分子提供了结合部位(binding sites)。微阵列支持体上的测点直径通常在50微米到300微米之间,一般为100-150微米,且这些测点的厚度一般仅有几微米,通常小于10微米。当将一份含有经过荧光标示的目标的样品与微阵列支持体上的测点接触时,样品中的目标分子允许与测点的探查分子混杂。在荧光检定中,使用一种激励光源对微阵列支持体进行照明并对发出的荧光所处位置和强度进行检测。荧光团的颜色被用作一种标示,以指明样品的目标分子与探查分子点之间已经发生了相互作用。用于照明支持体和检定支持体发出的光的光学装置可能包括一种微阵列扫描仪或成像仪。

WO01/94032中公开了微阵列支持体领域的现有技术,其中描述了通过设置棱锥或圆锥形的凹陷来实现支持体表面的扩大作用,增加探针的有效表面。结果,可提供更多的结合部位,从而增大信噪比。通过扩大图案可将有效表面面积比平面支持体轻易地增大2或3倍。
美国专利号为2002/0028451的文献中介绍了一种包含聚合物微阵列支持体基片的检定设备,在该基片上开有微结构凹槽,且在凹槽中充有液晶材料。凹槽的尺寸经过精心选择,以使上述液晶材料具有一致的取向,以便在破坏这种一致取向时能检定到微粒的粘附情形。
在编号为EP 0714742的文献中则披露了另一种现有技术,该技术与具有高精度微特征的聚合物支持体有关。
然而,由于需要对基于微阵列的光学检定进行改进,本发明的目标是提供一种经过改进的聚合物微阵列支持体,以便在考虑到如信噪比等指标时,本发明与现有技术相比,能实现进一步的光学检定性能改进。
发明内容
根据整体作为本申请组成部分的所附的权利要求,可以通过用于某种光学检定装置的聚合物微阵列支持体,和包括该聚合物支持体的光学检定装置,以及通过在用于某种光学检定装置的聚合物微阵列支持体上形成微特征来实现以上目标。
用于光学检定装置的聚合物微阵列支持体包含一些具有一定的焦深的、检测从支持体发出的光的光学装置。上述支持体的厚度沿支持体表面区域按一个厚度变化值变化,且该支持体被设置了包含一种表面扩大图案的选定的微特征。该表面扩大图案包含配置成具有与上述光学装置的所述焦深和支持体的厚度变化值相适配的选定深度的凹槽。通过调整凹槽的深度,从而使凹槽深度与上述厚度变化值之和与上述光学装置的焦深基本相当,可以实现对光学检定性能的改进,如光学检定的信噪比可得到改善。信噪比的提高是通过信号的增强和减少的干扰/背景信号来实现的。信号的增强是由于凹槽导致了支持体表面积增加,从而探查分子点的结合部位也有所增加;而干扰/背景信号减弱则是因为焦深内的材料量有所减少。
凹槽可相对于与支持体材料的折射率相适配的支持体表面,具有一选定的倾斜角(α),该选定的倾斜角(α)使支持体表面具有强度和角度特性方面的理想反射性能。

可以在柱状物之间设置微粒,如实心或多孔微粒,这可以进一步提高信噪比。
在形成微特征的方法中,支持体厚度沿支持体表面区域按一个厚度变化值发生变化。上述微特征包含在一种光学检定装置的聚合物微阵列支持体中的凹槽,该光学检定装置包含具有一定焦深的、用来检测从上述支持体发出的光的光学装置。上述凹槽的深度通过选择与上述焦深和上述厚度变化值适配,以使得上述深度和上述厚度变化值之和与上述焦深基本相当,从而改善装置的性能。
凹槽相对于支持体表面的倾斜角(α)可以根据支持体材料的折射率进行调整,以达到支持体表面理想的反射性能。
这些表面扩大的凹槽的另一优点是它们能维持液体的毛细流动。

优选实施例的说明
在本说明书和权利要求书中使用的术语和表达方式具有通常业内行家所使用的含义。
根据本发明,对光学检定中聚合物微阵列支持体的性能改进是通过在聚合物微阵列支持体中加入选定的微特征来实现的,这些微特征包括一些具有选定深度的凹槽。本发明的思想是通过控制吸收的、反射的或传输的光的幅值(即强度)和或频率(波长)方面的变化,来改进包括一些光学装置的某种光学检定装置中的微阵列支持体的性能,如信噪比。而这是通过提供具有包含一些凹槽的某种微特征的微阵列的支持体实现的。上述凹槽的深度与检定装置的光学装置的焦深和支持体的厚度变化相适配,以使凹槽的选定深度和支持体的厚度变化之和与光学装置的焦深基本相当。最好是,凹槽的选定深度和支持体的厚度变化之和与光学装置的焦深相等。然而,光学装置的焦深可能稍大于或小于上述深度和厚度变化之和,这取决于支持体载物片的质量。
本发明可被应用在如荧光或磷光结合的检定中,且聚合物微阵列支持体的选定微特征能在几个方面影响检定的性能。微阵列支持体包含一个基片,该基片由一种聚合物载物片制成,并配有一层化学性质经改性的表面涂层。支持体的厚度沿支持体表面区域变化,支持体的厚度变化一般少于15-20微米,这取决于由制造方法确定的载物片质量。与平面支持体相比,由于在荧光或磷光标记所在位置的表面涂层上光子的数目有所增加,因而根据本发明设计的微特征增强了支持体的激励能力。当标记为荧光性时,由于从荧光染料发出并到达光学检定系统的光子数有所增加,因而支持体的发射能力得到了增强,同时,通过减少进入和/或离开支持体基片的光子数来避免多余的背景荧光,从而减弱了相关噪声。
因此,通过利用包含在聚合物微阵列支持体中配置的一种表面扩大图案的选定微特征,本发明被用来提高光学检定的信噪比。我们选择上述微特征并对其进行调整,以使检定装置实现理想的光学特性。其中,检定装置的光学特性可以用几何光学和/或物理光学进行描述。几何光学将光的传播视作一种直线现象,另一方面,物理光学却利用了电磁波的波动性质。在几何光学中,光的路径是直线的,而在物理光学中,引入了光的衍射和干涉现象。在几何光学中,光的波长远小于表面微特征的尺寸,而在物理光学中,光的波长对应于表面微特征的尺寸。
在本发明中,光学装置最好包含一种微阵列扫描仪,该扫描仪最好包含一个仅收集目标透反射镜的焦深内的发射光线的共焦光学器件,如通过针孔来阻止多余的光,从而减少了检测到的干扰信号。图2试图对一个扫描荧光检定装置2的实施例进行说明,该检定装置包括一个微阵列支持体1和光学装置3,4,6。这些光学装置包括一个用以将激励光引导至微阵列支持体的激光器3,一个用于检测从微阵列支持体的结合部位发出的光的PMT(光电倍增管)4,以及一个用于减少噪声的针孔6。
图3图示说明了在一台扫描仪(参见图2)中测得的来自表面平整的载物片并构成背景噪声的干扰信号和来自表面开有锥形凹槽的载物片的同类信号之间的比较。该比较是在两个不同的激励波长,即543纳米(Cy3)和633纳米(Cy5)上进行的,并采用聚苯乙烯(PS)作为支持体材料,而图示的光发射值是载物片上的一个平均值。Cy3是在543纳米处进行激励,其光发射在570纳米处进行测量;Cy5是在633纳米处进行激励,其光发射在670纳米处进行测量。表面平整的载物片在图中表示为Flat,而布满锥形凹槽的表面则被表示为Pyramid。与表面平整的载物片相比,表面布满锥形凹槽的载物片大大降低了其背景荧光。
本发明的微阵列支持体的微特征包含一些凹槽,这些凹槽具有与光学装置的焦深和支持体的厚度变化相关的选定深度。这些凹槽可以具有直边或圆边,可以是V形的、正弦形的、三角形的、梯形的或是二元形状(binary-shaped),或为柱形。这些凹槽也可以是在支持体上处于多个方向上的一些结构。例如,锥形凹槽可以被形成于两条相邻的微型凹槽之间,并与这两条凹槽的方向成90度。
各凹槽之间的距离可以是恒定的,也可以沿支持体表面区域变化。凹槽的尺寸远大于激励光的波长,且凹槽深度通常在5至10微米,有时甚至达到20微米,并可能具有一定度数的如55度的倾斜角,这取决于所采用的光学装置和所要的支持体的几何光学特性。
图1示出了本发明的微阵列支持体1的一个实施例的侧视图,该支持体的厚度7沿支持体表面区域按一个厚度变化值(图中没有示出)进行变化,该厚度变化值等于支持体的最大厚度与最小厚度之差,这取决于支持体载物片的质量。微阵列支持体具有包含一种表面扩大图案5的微特征。该图案包括具有选定深度8和与支持体表面成倾斜角α的V形凹槽。支持体可具有如25mm×75mm大小的面积,且支持体载物片的有限平整性造成支持体厚度沿支持体表面区域发生一定的变化,上述变化通常少于15-20微米,这取决于制造方法。可以用各种不同的方法制造微阵列支持体,但最好是采用从主结构形成聚合物复制物的方法,如注塑、铸造、压纹,来制造微阵列支持体。凹槽的主结构通常由硅或玻璃制成,并通过湿式或干式化学蚀刻方法、光刻法或机械加工法(磨削或车削)来形成凹槽。
根据本发明的第一实施例,支持体的微特征包括一种表面扩大图案,如V形凹槽,且凹槽的深度被用来与检定装置的光学装置的焦深相适配,以使得凹槽的选定深度与支持体的厚度变化之和与光学装置的焦深基本相当。
在一示范性实施例中,凹槽呈锥形,且凹槽主结构是在硅材上进行各向异性地蚀刻(100)而形成的,并导致凹槽与支持体表面成55度的倾斜角。如果光学装置的焦深为20-30微米,且支持体载物片的品质将支持体的厚度变化局限在10-15微米之内,则凹槽的深度可能被选定为5-10微米。从而,凹槽深度和沿支持体载物片表面区域的厚度变化之和便为15-25微米。因此,该和值便小于上述焦深值,并与上述焦深值基本相当。凹槽的深度也可选定为10-15微米,使得凹槽深度与厚度变化值之和为20-30微米,也同样与上述焦深值基本相当。
于是,由于凹槽增大了支持体的表面积,信号得到了增强,结果产生了更多的结合部位,因而荧光信号的强度变得更高。同时,干扰/背景信号强度有所减弱,因为焦深内的自身荧光材料的量有所减少。该开有凹槽的结构也导致了疏水行为的增加,因为与平面聚合体相比,具有结构的聚合体具有更高的润湿角,从而有利于在支持体上印制高密度的微阵列点。
这些凹槽也可以维持液体的毛细流动,这取决于凹槽的尺寸。
由于布鲁斯特行为,支持体中凹槽的倾斜角将影响入射到支持体上的光的入射角,并改变支持体的表面反射。从而,可以考虑支持体材料的折射率,为凹槽选择一个合适的倾斜角来控制支持体的表面反射。例如,如果聚合物的折射率是1.5,则入射到由该聚合物制成的平整表面的圆偏振光将约有4%被反射,而如果入射光的入射角为70度,则入射光将约有17%被反射,即反射率被增大到了原来的4倍。增强的表面反射率加上又有了凹槽,使得入射光在支持体表面至少被反射两次,从而,在两个不同位置进行了荧光激励。
通过考虑基片和表面涂层材料的光学常数为凹槽选择一个合适的倾斜角,可以改变在支持体表面经过反射镜面反射的光的方向,以确保发射光的路径与激励光的路径不相重合。而这一点是有利的,因为激励光能量远大于发射光能量,所以如果两者光的路径重合,微阵列扫描仪的探测器便可能饱和。
根据本发明的第二个实施例,通过配置一个由金属、半导体材料或电介质材料制成的反射层,可以进一步改善支持体的性能。对于可见光,由银、铂、钯或金制成的反射层是有益的。反射层的厚度最好与支持体的理想透明度相适配,反射层可配置在支持体基片的顶面或支持体的底部。例如,一层20纳米厚的金膜可以传送大约50%的红光。采用附加金属层的一个优点是,在金属层上比在聚合物表面更容易引入表面化学方法。另一优点是,可以利用该层的半传输特性,即通过使用在金属、半导体材料和电介质材料中发现的特定光波长吸收区域,使得反射层传输特定波长的光而反射其他波长的光。
根据本发明的第三实施例,支持体包含形成为微柱状物的凹槽,即圆柱形柱状物。根据一个示范性实施例,这些柱状物设有附加层,且该层的折射率大于支持体材料的折射率,从而实现了一种光波导。根据另一示范性实施例,合适尺寸的微粒,即0.1至50微米范围内的微粒可被放置在微柱状物之间,而这会提高支持体的信噪比。
根据本发明的第四实施例,支持体设有一介质反射镜,该反射镜包含一个由几层组成的干涉层结构。这些层的厚度可能为波长的四分之一,并包含交替设置的氧化物,如折射率较低的二氧化硅或折射率较高的氧化钛。该介质反射镜可设于支持体基片的顶面或支持体的底部。通过这种干涉层结构,可以控制在某些波长段的光的反射率,从而实现光的波长滤波。通过增大表面的反射率,减少了由于背景荧光造成的噪声,因为进入支持体基片并激励此处的自身荧光的光有所减少。
根据本发明的第五个实施例,支持体上设有叠加在凹槽上的衍射光栅。该衍射光栅型凹槽的高度和栅结构的单条凹槽之间的距离与探测光的波长相等,即处于几百纳米的范围内。通过采用衍射光栅,可以进一步扩大支持体的表面积,并提高将选定波长的光反射/传送到确定方向上的可能性。衍射光栅还可以包含一种被调整用来减少入射激励光的表面反射率的抗反射结构,或是一种被调整用来增强表面反射的增强反射结构。可以用电子束光刻法来生产衍射光栅,且栅型凹槽结构可以是正弦形、三角形、梯形或二元形。栅的凹槽可以具有直边或圆边,且栅的每条凹槽之间的距离可以恒定,也可以沿栅发生变化。
根据本发明的第六实施例,支持体基片的光吸收性能被调整用来与激励光源的激励光和/或发射光波长相适配,以使得该材料在光学上“无活性”。一个实现该目的的方法是用细小的吸光颜料为聚合物染色,以使支持体基片完全变为黑色,即对于所研究波长的光是高度吸收的。另一种实现该目的的方法是采用一种溶剂染料。同样,通过在聚合物支持体材料中混入石英粒子,可以减少支持体的自身荧光。
根据本发明的第七实施例,支持体被用来传输或吸收激励光波长和吸收经使用的荧光团的波长。从而,防止从支持体发射出自身荧光,并加入到噪声中。
本发明不局限于以上附图中描述的实施例,而可在权利要求规定的范围内自由进行修改和调整。
Claims (20)
1.一种用于光学检定装置(2)的聚合物微阵列支持体(1),所述检定装置包含具有适用于检测从所述支持体(1)发出的光的焦深的光学装置,所述支持体的厚度(7)沿支持体表面区域按等于支持体的最大厚度与最小厚度之差的厚度变化值变化,且所述支持体上设有包含一种表面扩大图案(5)的选定的微特征,其特征在于:所述表面扩大图案(5)包含一些设为具有与所述光学装置的所述焦深和所述支持体的厚度变化值相适配的深度(8)的凹槽,所述选定的深度(8)与所述支持体的厚度变化值之和与所述光学装置的焦深基本相当。
2.如权利要求1所述的聚合物微阵列支持体,其特征在于:所述凹槽具有荧光选定的与支持体表面的倾斜角(α),且所述选定的倾斜角(α)与支持体材料的折射率相适配,以达到支持体表面的理想反射率。
3.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:至少一些凹槽具有直边。
4.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:至少一些凹槽具有圆边。
5.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:凹槽沿支持体表面的多个方向设置。
6.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:各凹槽之间的距离沿支持体表面区域恒定。
7.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:各凹槽之间的距离沿支持体表面区域变化。
8.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:所述支持体的微特征上还包含一个附加层。
9.如权利要求8所述的聚合物微阵列支持体,其特征在于:所述附加层的厚度经选定以达到支持体的理想透明度。
10.如权利要求8所述的聚合物微阵列支持体,其特征在于:所述附加层的厚度经选定以达到支持体的理想反射率。
11.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:所述支持体的微特征上还包含一介质反射镜。
12.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:所述支持体的微特征还包含一个叠加在所述支持体的至少部分表面扩大图案上的衍射光栅。
13.如权利要求1或2所述的聚合物微阵列支持体,其特征在于:所述支持体设有吸光颜料。
14.如权利要求3所述的聚合物微阵列支持体,其特征在于:所述凹槽形成为柱状物。
15.如权利要求14所述的聚合物微阵列支持体,其特征在于:所述柱状物上有一附加层,所述附加层的折射率比支持体材料的折射率大,以形成一光波导。
16.如权利要求14或15所述的聚合物微阵列支持体,其特征在于:在所述柱状物之间设置微粒。
17.一种包含聚合物微阵列支持体(1)和光学装置的光学检定装置(2),所述光学装置具有适用于检测从所述支持体(1)发出的光的焦深,所述支持体的厚度(7)沿支持体表面区域按等于支持体的最大厚度与最小厚度之差的厚度变化值变化,所述支持体设有包含一种表面扩大图案(5)的选定的微特征,其特征在于:所述表面扩大图案(5)包含一些设为具有与所述光学装置的所述焦深和所述支持体的厚度变化值相适配的深度(8)的凹槽,所述选定的深度(8)与所述支持体的厚度变化值之和与所述光学装置的焦深基本相当。
18.如权利要求17所述的光学检定装置(2),其特征在于:所述凹槽具有选定的与支持体表面的倾斜角(α),且所述选定的倾斜角(α)与支持体材料的折射率相适配,以达到支持体表面的理想反射率。
19.一种形成微特征的方法,所述微特征包含在一种光学检定装置(2)的聚合物微阵列支持体(1)中的凹槽,该检定装置(2)包括具有适用于检测从所述支持体发出的光的焦深的光学装置,所述支持体的厚度(7)沿支持体表面区域按等于支持体的最大厚度与最小厚度之差的厚度变化值变化,其特征在于:通过选定一个凹槽深度(8)来使所述凹槽的深度(8)与所述焦深和所述厚度变化值相适配,从而使得所述深度与所述厚度变化值之和与光学装置的焦深基本相当。
20.如权利要求19所述的方法,其特征在于:通过使凹槽相对于支持体表面所成的倾斜角(α)与支持体材料的折射率相适配,达到支持体表面的理想反射率。
