Abstract
Esta invención se refiere a construcciones nucleicas recombinantes que comprenden proteínas efectoras CRISPR–Cas Tipo V, transcriptasas inversas y ácidos nucleicos guía extendidos, y a métodos de uso de los mismos para modificar ácidos nucleicos en plantas. En un primer aspecto, se proporciona un método para modificar un ácido nucleico diana, donde el método comprende: poner en contacto el ácido nucleico diana con (a) una proteína efectora CRISPR–Cas Tipo V o una proteína efectora CRISPR–Cas Tipo II; (b) una transcriptasa inversa, y (c) un ácido nucleico guía extendido (p. ej., ARN CRISPR Tipo II o Tipo V extendido, ADN CRISPR Tipo II o Tipo V extendido, ARNcr Tipo II o Tipo V extendido, ADNcr Tipo II o Tipo V extendido), de modo de modificar así el ácido nucleico diana.
