La présente invention a pour objet un procédé de préparation de la phtalazino[2,3-b]-phtalazine-5(14H),12(7H)-dione décrite dans le brevet USA No. 3 557 108 qui est une substance anti-inflammatoire.
Conformément au brevet précité on prépare le composé par des réactions qui conduisent d'abord au composé clé : la 3,4dihydro-1(2H)-phtalazinone qu'on fait réagir à son tour avec le chlorure de 2-bromométhylbenzoyle pour obtenir le composé tétracyclique final.
Bien qu'on puisse conduire le stade final à l'échelle industrielle avec des rendements tout-à-fait satisfaisants, la prépara
tion de la 3,4-dihydro-1(2H)-phtalazinone intermédiaire en
grandes quantités présente de nombreuses difficultés et limite
les rendements globaux.
La présente invention a pour objet un procédé de prépara
tion de la phtalazino[2,3-b]phtalazine-5(14H),12(7H)-dione
en des quantités plus grandes qu'à l'échelle du laboratoire.
Le présent procédé consiste à cycliser la 2-benzyl-3,4
dihydro-1(2H)-phtalazinone en phtalazino[2,3-b]phtalazine5(14H),12(7H)-dione par la réaction avec un composé de
formule XCOY selon le schéma:

dans lequel X et Y représentent chacun un groupe qui est éliminé au cours de la réaction et choisi indépendamment parmi un radical halogéno, alcoxy inférieur, vinyloxy, p-nitrophénoxy, benzyloxy, benzhydryloxy, azido, imidazolyle, striazolyle ou ensemble représentent un radical éthylènedioxy, la cyclisation étant effectuée en présence d'un catalyseur de
Lewis.
De préférence on conduit la réaction en deux stades. On fait d'abord réagir la 2-benzyl-3,4-dihydro-1(2H)-phtalazinone avec le composé XCOY sans catalyseur, de préférence en présence d'un accepteur d'acide halogénohydrique lorsque la réaction forme un acide halogénohydrique, pour favoriser la carbonylation de l'atome d'azote en position 3; ensuite, on ajoute un catalyseur de Lewis qui peut être choisi parmi ceux utilisés habituellement pour des réactions d'acylation arénique, et on chauffe le mélange pour accélérer la cyclisation en la phtalazino[2,3-b]phtalazine-5(14H),12(7H)-dione. Des catalyseurs de cyclisation utiles sont les acides de Lewis utilisés habituellement pour l'acylation arénique de Friedel-Kraft, par exemple ZnCl2, SbCls, SbFs, AlCl3, BF3, HF, H2SO4, H3PO4 etc.
Au lieu de la 2-benzyl-3,4-dihydro-1(2H)-phtalazinone, on peut utiliser un de ses sels d'acide comme matière de départ en présence d'une base pour libérer le dérivé de phtalazine I.
Dans la pratique, on conduit la réaction en ajoutant le dérivé de phtalazine I ou un de ses sels d'acide, à un solvant organique inerte, par exemple le tétrachloroéthane qui dans le deuxième cas contient aussi une quantité suffisante de triéthylamine ou d'une base organique tertiaire équivalente, puis en ajoutant à la solution un excès du réactif XCOY.
Selon une forme d'exécution préférée, on utilise le phosgène, le carbonate de chloroéthyle et le carbonate de diéthyle comme réactif XCOY.
On maintient le mélange réactionnel à la température ordinaire pendant environ une à deux heures, puis on le filtre si, conformément au procédé réactionnel un halogénohydrate s'est formé.
On enlève alors l'excès du réactif XCOY par barbottage dans un courant d'azote ou par évaporation sous pression réduite. On ajoute environ 2 à 3 parties équimoléculaires de chlorure d'aluminium ou d'un autre acide de Lewis équivalent, par exemple ZnCk, et on laisse la température monter jusqu'à 70-130"C pendant environ 2 heures. En utilisant des catalyseurs de cyclisation qui sont volatils, tels que BF3 ou l'acide fluorhydrique anhydre, les conditions réactionnelles sont différentes pour ce qui est de la température et du temps. Par exemple, la température peut varier entre 0 et 200C et le temps jusqu'à ce que la réaction soit complète peut être de 2 à 20 heures.
Lorsque la réaction est complète, on verse le mélange dans de l'eau et on sépare la couche organique. Après évaporation de la solution on obtient la phtalazino[2,3-b]phtala:iine- 5(14H),12(7H) brute.
On peut encore purifier le produit par cristallisation dans les solvants usuels, par exemple l'éthanol.
Le composé de formule I de base pour la préparation de la phtalazino[2,3-b]phtalazine-5 (14H), 12(7H)-dione selon le procédé décrit, est un nouveau composé qu'on peut préparer par la réduction de la 2-benzyl-1(2H)-phtalazinone, qu'on peut obtenir à son tour de diverses manières, par exemple la benzylation directe de la 1(2H)-phtalazinone ou par la réaction de l'acide phtalaldéhydique avec la benzylhydrazine.
Avantageusement on conduit la réduction de la 2-benzyl1(2H)-phtalazinone avec de la poudre de zinc en milieu acide à 50-80 C, ou de l'hydrogène gazeux en présence d'un catalyseur d'hydrogénation.
Les exemples suivants illustrent l'invention sans la limiter.
Exemple 1 Phtalazino[2,3-b]phtalazine-5(14H), 12(7H)-dione
A 14 g (0,05 mole) de chlorhydrate de 2-benzyl-3,4-dihydro-1(2H)-phtalazinone dans 300 ml de tétrachloroéthane, on a ajouté 7 ml de triéthylamine puis on a saturé la solution avec du phosgène. On a maintenu le mélange réactionnel à la température ordinaire pendant environ 2 heures, puis on a enlevé l'excès de phosgène par barbottage dans un courant d'azote.
On a ajouté 20 g d'A1C13 anhydre au mélange réactionnel et on a maintenu la température à environ 900C pendant environ 2 heures. On a alors refroidi le mélange réactionnel, on l'a dilué avec du dichlorométhane puis on a versé dans un litre d'eau glacée. On a évaporé la phase organique après l'avoir lavée à l'eau.
On a obtenu 7,8 g (59%) du composé désiré qui fondait à 225oC.
Exemple 2
Phtalazino[2,3-b]phtalazine-5 (14H),12(7H)-dione
A 12 g (0,05 mole) de 2-benzyl-3,4-dihydro-1(2H)-phtala- zinone dans 200 ml de tétrachloroéthane, on a ajouté 4 ml de triéthylamine puis on a ajouté 8 g de chlorocarbonate d'éthyle à cette solution. On a gardé le mélange réactionnel à la température ordinaire pendant environ 2 heures. Après avoir filtré les sels, on a évaporé à sec la solution sous pression réduite.
On a versé le résidu dans 200 ml d'acide fluorhydrique anhydre à 10oC. Après 15 heures à 10oC on a chasé l'acide fluorhydrique par un courant d'azote et on a lavé le résidu avec du carbonate de sodium acqueux puis avec de l'eau.
On a purifié le résidu par cristallisation dans l'éthanol. On a obtenu 7,5 g (61%). Selon une variante, on peut conduire le stade de cyclisation en chauffant à reflux pendant une heure dans un mélange de 100 ml d'acide acétique anhydre et 60 ml d'acide acétique contenant 2 g de ZnCl2. Après refroidissement on a versé le mélange réactionnel dans de l'eau glacée, puis on a séparé sur un filtre le solide précipité. Le rendement était de 53 % après cristallisation dans l'éthanol.
Exemple 3 Phtalazino[2,3-b]phtalazine-5(14H),12(7H)-dione
On a chauffé 12 g de 2-benzyl-3,4-dihydro-1(2H)-phtalazinone dans 100 ml de carbonate de diéthyle au bain marie pendant 5 heures. On a évaporé à sec le mélange réactionnel et on a cyclisé le résidu en le chauffant à reflux dans un mélange d'acide acétique et d'anhydride acétique en présence de ZnCl2 comme décrit dans l'exemple 2.
Exemple 4
2-benzyl-3 ,4-dihydro- 1 (2H)-phtalazinone
On a mis en suspension 10 g de 2-benzyl-1(2H)-phtalazinone et 6,2 g de poudre de zinc dans 200 ml d'un mélange 1 : 1 d'éthanol et d'eau.
On a alors ajouté graduellement 10 ml d'HCl à 37% et on a maintenu la température à 600C en agitant. Après environ 30 minutes on a refroidi le mélange et on l'a neutralisé avec une solution diluée d'hydroxyde d'ammonium, puis on a chassé l'éthanol par distillation.
On a alors extrait la phase aqueuse avec du dichlorométhane et on a acidifié la phase organique avec une solution d'éthanol saturée avec de l'HCl sec. Après l'évaporation du solvant, on a obtenu le chlorhydrate de 2-benzyl-3,4-dihydro1(2H)-phtalazinone qui, après cristallisation dans de l'éthanol à 95 %, fondait à 169-1700C. On a obtenu 9,6 g (82 %).
Exemple 5
2-benzyl- (2H)-phtalazinone
On a préparé le composé en faisant réagir des quantités équimoléculaires d'acide phtalaldéhydique et de benzylhydrazine dans l'acide acétique à 50OC pendant une heure. Le rendement était de 93%. Le produit fondait à 104-1060C.

Procédé selon la revendication I, caractérisé en ce que X et Y représentent tous les deux un radical chloro et en ce qu'on conduit la réaction à la température ordinaire pendant environ 2 heures en présence de l'accepteur d'acide halogénohydrique, puis, après avoir ajouté l'acide de Lewis catalyseur, à 70-110oC pendant encore 2 heures.

Example 1 Phthalazino [2,3-b] phthalazin-5 (14H), 12 (7H) -dione
To 14 g (0.05 mole) of 2-benzyl-3,4-dihydro-1 (2H) -phthalazinone hydrochloride in 300 ml of tetrachloroethane was added 7 ml of triethylamine and then the solution was saturated with phosgene . The reaction mixture was kept at room temperature for about 2 hours, then the excess phosgene was removed by bubbling in a stream of nitrogen.
20 g of anhydrous A1Cl3 was added to the reaction mixture and the temperature was maintained at about 900C for about 2 hours. The reaction mixture was then cooled, diluted with dichloromethane and then poured into one liter of ice water. The organic phase was evaporated after washing it with water.
7.8 g (59%) of the desired compound were obtained which melted at 225oC.
Example 2
Phthalazino [2,3-b] phthalazin-5 (14H), 12 (7H) -dione
To 12 g (0.05 mole) of 2-benzyl-3,4-dihydro-1 (2H) -phtala-zinone in 200 ml of tetrachloroethane, 4 ml of triethylamine was added and then 8 g of chlorocarbonate d ethyl to this solution. The reaction mixture was kept at room temperature for about 2 hours. After filtering the salts, the solution was evaporated to dryness under reduced pressure.
The residue was poured into 200 ml of anhydrous hydrofluoric acid at 10oC. After 15 hours at 10 ° C., the hydrofluoric acid was removed with a stream of nitrogen and the residue was washed with aqueous sodium carbonate and then with water.
The residue was purified by crystallization from ethanol. 7.5 g (61%) were obtained. The reaction mixture was evaporated to dryness and the residue cyclized by heating to reflux in a mixture of acetic acid and acetic anhydride in the presence of ZnCl2 as described in Example 2.
Example 4
2-benzyl-3, 4-dihydro- 1 (2H) -phthalazinone
10 g of 2-benzyl-1 (2H) -phthalazinone and 6.2 g of zinc powder were suspended in 200 ml of a 1: 1 mixture of ethanol and water.
10 ml of 37% HCl were then gradually added and the temperature was maintained at 600C with stirring. After about 30 minutes the mixture was cooled and neutralized with dilute ammonium hydroxide solution, then the ethanol was distilled off.
The aqueous phase was then extracted with dichloromethane and the organic phase was acidified with an ethanol solution saturated with dry HCl. After evaporation of the solvent, 2-benzyl-3,4-dihydro1 (2H) -phthalazinone hydrochloride was obtained which, after crystallization from 95% ethanol, melted at 169-1700C. 9.6 g (82%) were obtained.
Example 5

The compound was prepared by reacting equimolecular amounts of phthalaldehyde acid and benzylhydrazine in acetic acid at 50OC for one hour. The yield was 93%. The product melted at 104-1060C.
