Im Schweizer Hauptpatent Nr. 574 956 wird die Herstellung von Piperidin-1-ylalkanonderivaten der Formel A, worin Rl' niederes Alkyl, R2' Wasserstoff oder niederes Alkyl und R3' Wasserstoff, Chlor, Brom, Fluor, Trifluormethyl, niederes Alkyl, niederes Alkoxy oder niederes Alkylthio und n' 1-3 bedeuten, beschrieben.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidin-1-ylalkanonderivate der Formel I, worin n für eine ganze Zahl von 1 bis 3 steht und R1 niederes Alkyl bedeutet, R2 Wasserstoff oder niederes Alkyl bedeutet, R3 und R4 je für Wasserstoff, Chlor, Brom, Fluor, Trifluormethyl, niederes Alkyl, niederes Alkoxy oder niederes Alkylthio stehen, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin R3 und R4 obige Bedeutung besitzen, mit Verbindungen der Formel III, worin R1 obige Bedeutung besitzt und Z für eine X-(CH2)11- CHR2-Gruppe, worin R2 und n obige Bedeutung besitzen und X den Säurerest eines reaktionsfähigen Esters bedeutet, oder eine CH2=CR2-Gruppe, worin R2 obige Bedeutung besitzt, steht, umsetzt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel I steht der Substituent R2 vorzugsweise für Wasserstoff. Falls R2 niederes Alkyl bedeutet, so enthält diese Alkylgruppe vorzugsweise 1 bis 4 Kohlenstoffatome und stellt insbesondere die Methylgruppe dar. Der Rest R1 enthält vorzugsweise 1 bis 4, insbesondere 1 bis 2 Kohlenstoffatome. Die Substituenten IR, oder R4 stellen vorzugsweise Wasserstoff oder Chlor dar. Falls R3 oder R4 für niederes Alkyl stehen, so enthält dieses vorzugsweise 1 bis 4 Kohlenstoffatome, falls R3 oder R4 für niederes Alkoxy oder Alkylthio stehen, so enthalten diese Gruppen vorzugsweise 1 bis 3 Kohlenstoffatome und stellen insbesondere Methoxy oder Methylthio dar.
Die erfindungsgemässe Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel III kann beispielsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel erfolgen. Für die Anlagerung von Vinylverbindungen der Formel IIIa, worin R1 und R2 obige Bedeutung besitzen, an Verbindungen der Formel II sind als Lösungsmittel beispielsweise niedere Alkanole wie Äthanol oder Äther, wie z. B. Dioxan, geeignet, und die Reaktionstemperatur liegt vorzugsweise bei Temperaturen zwischen ca. 20 und 1000. Vorteilhafterweise kann die Umsetzung durch Zusatz von starken Basen wie z. B. Benzyltrimethylammoniumhydroxid katalysiert werden. Die Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel IIIb, worin R1, R2, n und X obige Bedeutung besitzen, erfolgt vorzugsweise in Gegenwart eines säurebindenden Mittels.
Als unter den Reaktionsbedingungen inerte Lösungsmittel eignen sich z. B. niedere Alkanole wie Äthanol, chlorierte aliphatische Kohlenwasserstoffe wie Chloroform, aromatische Kohlenwasserstoffe wie Toluol oder auch Dimethylformamid. Als säurebindendes Mittel kann man z. B.
Alkalimetallcarbonate wie Natrium- oder Kaliumcarbonat oder eine tertiäre Stickstoffbase wie Triäthylamin verwenden.
Die Reaktion wird vorzugsweise bei Temperaturen zwischen ca. 500 und ca. 1500, insbesondere bei Siedetemperatur des Reaktionsgemisches ausgeführt. In den Verbindungen der Formel IIIb bedeutet X vorzugsweise Chlor, Brom, Jod oder den Säurerest einer organischen Sulfonsäure, z. B. den Methylsulfonyloxy- oder den p-Toluolsulfonyloxyrest.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen der Formel II können beispielsweise erhalten werden, indem man Ketone der Formel IV, worin R3 und R4 obige Bedeutung besitzen, mit der Grignard-Verbindung der Formel V umsetzt, den erhaltenen Komplex hydrolysiert, anschliessend Wasser abspaltet und aus den so erhaltenen Verbindungen der Formel VI, worin R3 und R4 obige Bedeutung besitzen, die Methylgruppe in an sich bekannter Weise abspaltet, z. B. durch Umsetzung mit einer Verbindung der Formel VII und Hydrolyse des dabei gebildeten Urethans.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere besitzen die Verbindungen der Formel I sedativ-neuroleptische und schlaffördernde sowie auch muskelrelaxierende Eigenschaften. Die sedativen und die muskelrelaxierenden Eigenschaften lassen sich z. B. an Mäusen bei der Messung der motorischen Aktivität in der Shuttle-box und im Klettertest beobachten. So hemmen die Substanzen z. B. in Dosen von 5-100 mg/kg p. o. die spontane motorische Aktivität und in Dosen von 0,3-20 mg/kg i. p. die Kletterfähigkeit der Mäuse. Die hypnotischen Eigenschaften der Verbindungen lassen sich z. B. an Mäusen und Ratten durch eine Potenzierung der durch Pentothal oder Barbital hervorgerufenen hypnotischen Wirkungen zeigen. So führen die Verbindungen der Formel I bei Mäusen in Dosen von 3100 mg/kg i. p. zu einer Verlängerung des Andauerns des durch Pentothal verursachten Verlustes des Aufrichtungsreflexes.
Bei mit einer noch nicht zur Verhinderung des Aufrichtungsreflexes ausreichenden Menge Barbital vorbehandelten Ratten führen die Verbindungen in Dosen von W80 mg/ kg i. p. zum Verlust dieses Aufrichtungsreflexes.
Aufgrund ihrer sedativen Eigenschaften können die Substanzen in der Psychiatrie zur Behandlung von Erregungszuständen Verwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 1,3 bis 50 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Aufgrund ihrer schlaffördernden Wirkungen können die Substanzen zur Behandlung von Schlafstörungen und Unruhezuständen verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden.
Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.

Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 200 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht wer den. Für grössere Säugetiere liegt die Tagesdosis bei etwa 55 bis 200 mg. Für orale Applikationen enthalten diese Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1
4-[4-(6,11-Dihydrodibenzo[b,e]oxepin-1 1-yliden)piperidino]-
2-butanon
Zu einer Lösung von 13,8 g 4-(6,11-Dihydrodibenzo[b,ej- oxepin-11-yliden)piperidin in 50 ml Äthanol tropft man innert 5 Minuten eine Lösung von 5,3 g Methylvinylketon in 10 ml Athanol. Das Reaktionsgemisch wird 2 Stunden am Rückfluss erhitzt, sodann bei reduziertem Druck vollständig eingeengt und der ölige Rückstand an 250 g Kieselgel mit einem Gemisch Chloroform/Athanol 95:5 chromatographiert.
Die so gereinigte, im Titel genannte Verbindung, wird darauf in das Hydrogenfumarat übergeführt. Smp. 173175O.
Das als Ausgangsmaterial verwendete 4-(6,11-Dihydro- dibenzo[b,e] oxepin-1 1-yliden)piperidin kann wie folgt hergestellt werden:
Zu einer Lösung von 64 g 4-(6,11-Dihydrodibenzo[b,e] oxepin-1 1-yliden) -1-methylpiperidin in 600ml Benzol tropft man eine Lösung von 96 g Chlorameisensäureäthylester in 100 ml Benzol und erhitzt das Gemisch 4 Stunden im Ölbad von 100. Nach Abkühlen wird das Reaktionsgemisch durch Hyflo filtriert, das Filtrat mit 2 N Salzsäure und Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und bei reduziertem Druck vollständig eingeengt. Das so erhaltene Urethan wird in 120 ml 4-Butanol gelöst und zu einer Lösung von 26 g Kaliumhydroxyd in 120 ml n-Butanol zugetropft.
Das Reaktionsgemisch wird sodann 5 Stunden bei einer Öl- temperatur von 1000 gerührt, dann abgekühlt und zwischen Wasser und Diäthyläther verteilt. Der Ätherextrakt wird sodann mit einer gesättigten Kochsalzlösung gewaschen, über Magnesiumsulfat getrocknet, das Lösungsmittel bei vermindertem Druck vollständig entfernt, wobei die Verbindung als gelbbraunes Öl zurückbleibt.
Beispiel 2
5-[4-(6,1 l-Dihydrodibenzo[b,eloxepin-l l-yliden)piperidin]-
2-pentanon
Zu einem Gemisch von 13,8 g 4-(6,11-Dihydrodibenzo [b,e]oxepin-11-yliden)piperidin und 12,7 g Natriumkarbonat in 200 ml Chloroform tropft man innert 10 Minuten eine Lösung von 10 g 5-Brom-2-pentanon in 40 ml Chloroform und erhitzt das Gemisch 18 Stunden am Rückfluss. Nach Abkühlen wird abfiltriert, das Filtrat mit Wasser gewaschen, über Magnesiumsulfat getrocknet und das Lösungsmittel vollständig abdestilliert. Der ölige Rückstand wird sodann an 250 g Kieselgel mit einem Gemisch Chloroform/Athanol 95:5 chromatographiert und die so gereinigte, im Titel genannte Verbindung in das Hydrogenfumarat übergeführt. A solution of 5.3 g of methyl vinyl ketone in 10 ml of ethanol is added dropwise within 5 minutes to a solution of 13.8 g of 4- (6,11-dihydrodibenzo [b, ej- oxepin-11-ylidene) piperidine in 50 ml of ethanol. The reaction mixture is refluxed for 2 hours, then concentrated completely under reduced pressure and the oily residue is chromatographed on 250 g of silica gel with a 95: 5 mixture of chloroform / ethanol.

The 4- (6,11-dihydrodibenzo [b, e] oxepin-1 1-ylidene) piperidine used as starting material can be prepared as follows:
