Im schweiz. Hauptpatent Nr. 574956 wird die Herstellung von Piperidin-l-ylalkanonderivaten der Formel A, worin R'1 niederes Alkyl, R'2 Wasserstoff oder niederes Alkyl und R'3 Wasserstoff, Chlor, Brom, Fluor, Trifluormethyl, niederes Alkyl, niederes Alkoxy oder niederes Alkylthio und n' 1-3 bedeuten, beschrieben.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidin-l-alkanonderivaten der Formel, worin n für eine ganze Zahl von 1 bis 3 steht, R1 niederes Alkyl bedeutet, R2 für Wasserstoff oder niederes Alkyl steht, R3 und R4 Wasserstoff, Chlor, Brom, Fluor oder niederes Alkyl bedeuten und R5 und R6 für niederes Alkyl stehen, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin R3, R4, R5 und R6 obige Bedeutung besitzen, mit Verbindungen der Formel ffi, worin Rl obige Bedeutung besitzt und Z für eine X-(CH2)n-CHR2- Gruppe, worin R2 und n obige Bedeutung besitzen und X den Säurerest eines reaktionsfähigen Esters bedeutet, oder eine CH2=CR2-Gruppe, worin R2 obige Bedeutung besitzt, steht, umsetzt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel I steht der Substituent R2 vorzugsweise für Wasserstoff. Falls R2 niederes Alkyl bedeutet, so enthält diese Alkylgruppe vorzugsweise 1 bis 4 Kohlenstoffatome und stellt insbesondere die Methylgruppe dar.
Der Rest R1 enthält vorzugsweise 1 bis 4, insbesondere 1 - 2 Kohlenstoffatome. Die Substituenten R3 und R4 stellen vorzugsweise Wasserstoff oder Chlor dar. Falls R3 oder R4 für niederes Alkyl stehen, so enthält dieses vorzugsweise 1 bis 4 Kohlenstoffatome und stellt insbesondere Methyl dar. Die Alkylgruppen R5 und R6 enthalten vorzugsweise 1 bis 3 Kohlenstoffatome und stellen insbesondere Methylgruppen dar.
Die erfindungsgemässe Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel III kann beispielsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel erfolgen. Für die Anlagerung von Vinylverbindungen der Formel IIIa, worin R1 und R2 obige Bedeutung besitzen, an Verbindungen der Formel II sind als Lösungsmittel beispielsweise niedere Alkanole wie Äthanol oder Äther wie z.B. Dioxan geeignet und die Reaktionstemperatur liegt vorzugsweise bei Temperaturen zwischen ca. 20 und 100 . Vorteilhafterweise kann die Umsetzung durch Zusatz von starken Basen wie z.B. Benzyltrimethylammoniumhydroxid katalysiert werden. Die Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel IIIb, worin R1,R2,n und X obige Bedeutung besitzen, erfolgt vorzugsweise in Gegenwart eines säurebindenden Mittels.
Als unter den Reaktionsbedingungen inerte Lösungsmittel eignen sich z.B. niedere Alkanole wie Äthanol, chlorierte aliphatische Kohlenwasserstoffe wie Chloroform, aromatische Kohlenwasserstoffe wie Toluol oder auch Dimethylformamid. Als säurebindendes Mittel kann man z.B. Alkalimetallcarbonate wie Natrium- oder Kaliumcarbonat oder eine tertiäre Stickstoffbase wie Triäthylamin verwenden. Die Reaktion wird vorzugsweise bei Temperaturen zwischen ca. 500 unnd ca.
1500, insbesondere bei Siedetemperatur des Reaktionsgemisches ausgeführt. In den Verbindungen der Formel Illb bedeutet X vorzugsweise Chlor, Brom, Jod oder den Säurerest einer organischen Sulfonsäure, z.B. den Methylsulfonyloxyoder den p-Toluolsulfonyloxyrest.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen der Formel II können beispielsweise erhalten werden, indem man Ketone der Formel IV, worin R3, R4, R5 und R6 obige Bedeutung besitzen, mit der Grignard-Verbindung der Formel V umsetzt, den ertaltenen Komplex hydrolysiert, anschliessend Wasser abspaltet und aus den so erhaltenen Verbindungen der Formel VI, worin R,, R4,R5und R6 obige Bedeutung besitzen, die Methylgruppe in an sich bekannter Weise abspaltet, z.B. durch Umsetzung mit einer Verbindung der Formel VII und Hydrolyse des dabei gebildeten Urethans.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere besitzen die Verbindungen der Formel I sedativneuroleptische und schlaffördernde sowie auch muskelrelaxierende Eigenschaften. Die sedativen und die muskelrelaxierenden Eigenschaften lassen sich z.B. an Mäusen bei der Messung der motorischen Aktivität in der Shuttle-box und im Klettertest beobachten. So hemmen die Substanzen z.B. in Dosen von 5 - 100 mg/kg p.o. die spontane motorische Aktivität und in Dosen von 0,3 - 20 mg/kg i.p. die Kletterfähigkeit der Mäuse. Die hypnotischen Eigenschaften der Verbindungen lassen sich z.B. an Mäusen und Ratten durch eine Potenzierung der durch Pentothal oder Barbital hervorgerufenen hypnotischen Wirkungen zeigen. So führen die Verbindungen der Formel I bei Mäusen in Dosen von 3 - 100 mg/kg i.p. zu einer Verlängerung des Andauerns des durch Pentothal verursachten Verlustes des Aufrichtungsreflexes.
Bei mit einer noch nicht zur Verhinderung des Aufrichtungsreflexes ausreichenden Menge Barbital vorbehandelten Ratten führen die Verbindungen in Dosen von 4 - 80 mg/kg i.p. zum Verlust dieses Aufrichtungsreflexes.
Aufgrund ihrer sedativen Eigenschaften können die Substanzen in der Psychiatrie zur Behandlung von Erregungszuständen Verwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z.B. für orale Applikationen die Teildosen etwa 1,3 bis 50 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Aufgrund ihrer schlaffördernden Wirkungen können die Substanzen zur Behandlung von Schlafstörungen und Unruhezuständen verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z.B. für orale Applikationen die Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.

Für grössere Säugetiere liegt die Tagesdosis bei etwa 55 bis 200 mg. Für orale Applikationen enthalten diese Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In dem nachfolgenden Beispiel, das die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel
5-[4-(9. 1 O-Dihydro-9,9-dimethyl-l0-anthracenylide dino]-a-pentanon
Zur Lösung von 8,5 g 4-(9,10-Dihydro-9,9-dimethyl-10- -anthracenyliden)piperidin in 200 ml Chloroform gibt man 7,2 g Natriumkarbonat, tropft dann 5,86 g 5-Brom-2-pentanon in 40 ml Chloroform zu und kocht anschliessend 17 Std.
unter Rückfluss. Nach Erkalten wird vom Niederschlag abfiltriert, das Filtrat zweimal mit Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird über eine Säule von 150 g Kieselgel filtriert. Die mit Chloroform, das 1% Methanol enthält, eluierte Fraktion wird in Isopropanol gelöst und mit ätherischem Chlorwasserstoff angesäuert, worauf das Hydrochlorid der im Titel genannten Verbindung kristallisiert. Smp. nach Umkristallisieren aus Methanol/Äther 280.2840 Zersetzung.
Das Ausgangsmaterial wird wie folgt hergestellt:
Zur Lösung von 8,2 ml Chlorameisensäureäthylester in 30 ml Toluol tropft man bei 90" während 1 Std. die Lösung von 9,5 g 4-(9,1 O-Dihydro-9,9-dimethyl- 1 O-anthracenyliden)- -1-methylpiperidin in 100 ml Toluol und kocht anschliessend 17 Std. unter Rückfluss. Nach Erkalten wird dreimal mit Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird dann 1 Std. mit einer Lösung von 13,3 g Kaliumhydroxyd in 90 ml n-Butanol gekocht.
Nach Erkalten wird mit y2 1 Wasser verdünnt und viermal mit Benzol extrahiert. Die vereinigten Extrakte werden mit gesättigter Kochsalzlösung gewaschen, getrocknet und eingedampft. Der Rückstand, ein zähes, gelbbraunes Öl, stellt rohes 4-(9,10-Dihydro-9,9-dimethyl-10-anthracenyliden)piperidin dar und wird als Rohprodukt weiterverarbeitet.
