Im schweiz. Hauptpatent 574956 wird die Herstellung von Piperidin-1-ylalkanonderivaten der Formel A, worin R'1 niederes Alkyl, R' Wasserstoff oder niederes Alkyl und Ges'3 Wasserstoff, Chlor, Brom, Fluor, Trifluormethyl, niederes Alkyl, niederes Alkoxy oder niederes Alkylthio und n' 1-3 bedeuten, beschrieben.
Die vorliegende Erfindung betrifft ein Verfahren zur Her- stellung neuer Piperidin-l -ylalkanonderivate der Formel I, worin n für eine ganze Zahl von 1 bis 3 steht, R1 niederes Alkyl bedeutet, R2 für Wasserstoff oder niederes Alkyl steht, R3 Wasserstoff, Chlor, Brom, Fluor, niedus Alkyl, niederes Alkoxy, niederes Alkylthio oder Trifluormethyl bedeutet u.- A für eine Äthylen- oder Vinylengruppe steht, u. ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin A und R3 obige Bedeutung besitzen, mit Verbindungen der Formel III, worin R obige Bedeutung besitzt und Z für eine X-(CH2)n-CHR2- Gruppe, worin R und n obige Bedeutung besitzen und X den Säurerest eines reaktionsfähigen Esters bedeutet, oder eine CH2=CR2-Gruppe, worin R2 obige Bedeutung besitzt, steht, umsetzt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel I steht der Substituent R2 vorzugsweise für Wasserstoff. Falls R niederes Alkyl bedeutet, so enthält diese Alkylgruppe vorzugsweise 1 - 4 Kohlenstoffatome und stellt insbesondere die Methylgruppe dar.
Der Rest R1 enthält vorzugsweise 1-4, insbesondere 1 - 2 Kohlenstoffatome. Der Substituent R3 stellt vorzugsweise Wasserstoff oder Chlor dar. Falls R3 für niederes Alkyl stehet, so enthält dieses vorzugsweise 1 - 4 Kohlenstoffatome, falls Ra für niederes Alkoxy oder niederes Alkylthio steht, so enthalten diese vorzugsweise 1 - 3 Kohlenstoffatome und stellen insbesondere Methoxy oder Methylthio dar.
Die erfindungsgemässe Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel III kann beispielsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel erfolgen. Für die Anlagerung von Vinylverbindungen der Formel III a, worin Rl und R2 obige Bedeutung besitzen, an Verbindungen der Formel II sind als Lösungsmittel beispielsweise niedere Alkanole wie Äthanol oder Äther wie z.B. Dioxan geeignet und die Reaktionstemperatur liegt vorzugsweise bei Temperaturen zwischen ca. 20 und 100 . Vorteilhafterweise kann die Umsetzung durch Zu- satz von starken Basen wie z.B. Benzyltrimethylammoniumhydroxid katalysiert werden. Die Umsetzung von Verbindun- gen der Formel II mit Verbindungen der Formel III b, worin Rl.
Re, n und X obige Bedeutung besitzen, erfolgt vorzugsweise in Gegenwart eines säurebindenden Mittels. Als unter den Reaktionsbedingungen inerte Lösungsmittel eignen sich z.B. niedere Alkanole wie Äthanol, chlorierte aliphatische Kohlenwasserstoffe wie Chloroform oder aromatische Etoh- lenwasserstoffe wie Toluol oder auch Dimethylformamid. Als säurebindendes Mittel kann man z. B. Alkalimetallcarbonate wie Natrium- oder Kaliumcarbonat oder eine tertiäre Stickstoffbase wie Triäthylamin verwenden. Die Reaktion wird vorzugsweise bei Temperaturen zwischen ca. 50 und ca.
1500, insbesondere bei Siedetemperatur des Reaktionsgemisches ausgeführt. In den Verbindungen der Formel IIT b bedeutet X vorzugsweise Chlor, Brom, Jod oder den Säurerest einer organischen Sulfonsäure, z.B. den Methylsulfonyloxyoder den p-Toluolsulfonyloxyrest.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen der Formel II können beispielsweise erhalten werden, indem man Ketone der Formel IV, worin A und R3 obige Bedeutung besitzen, mit der Grignard-Verbindung der Formel V umsetzt, den erhaltenen Komplex hydrolysiert, anschliessend Wasser abspaltet und aus den so erhaltenen Verbindungen der Formel VI, worin A und 3 obige Bedeutung besitzen, die Methylgruppe in an sich be tvannter Weise abspaltet, z.B. durch Umsetzung mit einer Verbindung der Formel VII und Hydrolyse des dabei gebildeten Urethans.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere besitzen die Verbindungen der Formel I sedativ-neuroleptische und schlaffördernde sowie auch muskelrelaxierende Eigenschaften. Die sedativen und die muskelrelaxierenden Eigenschaften lassen sich z.B. an Mäusen bei der Messung der motorischen Aktivität in der Shuttle-box und im Klettertest beobachten. So hemmen die Substanzen z. B.
in Dosen von 5 - 100 mg/kg p.o. die spontane motorische Aktivität und in Dosen von 0,3 - 20 mg/kg i.p. die Kletterfähigkeit der Mäuse. Die hypnotischen Eigenschaften der Verbindungen lassen sich z. B. an Mäusen und Ratten durch eine Potenzierung der durch Pentothal oder Barbital hervorgeru feinen hypnotischen Wirkungen zeigen. So führen die Verbindungen der Formel I bei Mäusen in Dosen von 3 - 100 mg/ kg i.p. zu einer Verlängerung des Andauern des durch Pentothal verursachten Verlustes des Aufrichtungsreflexes. Bei mit einer noch nicht zur Verhinderung des Aufrichtungsreflexes ausreichenden Menge Barbital vorbehandelten Ratten führen die Verbindungen in Dosen von 4 - 80 mg/kg i.p. zum Verlust dieses Aufrichtungsreflexes.
Aufgrund ihrer sedativen Eigenschaften können die Substanzen in der Psychiatrie zur Behandlung von Erregungszuständen Verwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 1,3 bis 50 mg der Yerbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Aufgrund ihrer schlaffördernden Wirkungen können die Substanzen zur Behandlung von Schlafstörungen und Unruhezuständen verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 100 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z.B. für orale Applikationen die Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.

Die zu verwendenden Dosen variieren naturgemäss je nach ±t der Substanz, der Administration und des zubehandelnden Zustandes. Im allgemeinen werden jedoch be friedigende Resultate mit einer Dosis von ca. 0,1 bis 200 mg/ kg Körpergewkht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 55 bis 200 mg. Für orale Applikationen enthalten diese Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formeln neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel I 4-[4-(1O,1 I-Dihydro-5-dibenzo[a,dlcycloheptenylide piperidino]-2-butanon
Zur Lösung von 10 g 4-(10,11-Dihydro-5-dibenzo[a,d]- cycloheptenyliden)piperidin in 100 ml Äthanol tropft man bei 700 5,1 g Methylvinylketon und kocht 1 2 Std. unter Rückfluss. Dann dampft man im Vakuum ein, löst den Rückstand in Aceton, stellt mit ätherischem Chlorwasserstoff sauer, dampft nochmals ein und kristallisiert den Rückstand zuerst aus Aceton/Äther dann aus Isopropanol/Äther. Das Hydrochlorid der im Titel genannten Verbindung schmilzt erst über 3ovo.
Das Ausgangsmaterial wird wie folgt hergestellt:
Zur Lösung von 42,0 g Chlorameisensäureäthylester in 100 ml Toluol tropft man bei 90" während 2 Std. die Lösung von 37 g 4-(1 0,11 -Dihydro-5-dibenzo[a,d]cycloheptenyliden)- -1-methylpiperidin in 130 ml Toluol und kocht anschliessend 15 Std. unter Rückfluss. Dann kühlt man ab, schüttelt zweimal mit Wasser aus, trocknet über Magnesiumsulfat und dampft ein. Der zähe, ölige Rückstand wird mit der Lösung von 63 g Kaliumhydroxyd in 475 ml n-Butanol 1 Std. unter Rückfluss gekocht. Dann kühlt man ab, verdünnt mit 2,51 Wasser und schüttelt dreimal mit Chloroform aus. Der Chloroformextrakt wird mit gesättigter Kochsalzlösung gewaschen, getrocknet und eingedampft.
Der Rückstand wird in Äther gelöst und die Lösung nach Behandeln mit Tierkohle langsam mit Petroläther versetzt, worauf 4-(10,11-Dihydro-5-dibenzo- [a,d]cycloheptenyliden)piperidin kristallisiert. Smp. 113-114".
Beispiel 2 5- [4-(1O, 1 1-Dihydro -5-dibenzo[a,d]cycloheptenyliden)- piperidino]-2-pentanon
Zur Lösuung von 10 g 4-(10,11-Dihydro-5-dibenzo[a,d] -cycloheptenyliden)piperidin in 100 ml Chloroform gibt man 9,2 g Natriumkarbonat, tropft dann während 20 Min. die Lösung von 7,2 g 5-Brom-2-pentanon in 50 ml Chloroform zu und kocht anschliessend 17 Std. unter Rückfluss. Nach Erkalten wird vom Niederschlag abfiltriert, das Filtrat zweimal mit Wasser ausgeschüttelt, über Magnesiumsulfat getrocknet und eingedampft. Der Rückstand wird über eine Säule von 170 g Kieselgel filtriert. Die mit Chloroform, das 1% Methanol enthält, eluierte Fraktion wird in Aceton gelöst und mit ätherischem Chlorwasserstoff angesäuert. Nach Zusatz von mehr Äther kristallisiert das Hydrochlorid der im Titel genannten Verbindung aus. Smp. 235 - 238".
Beispiel 3 5- [4-(5-Dibenzo[a,d]cycloheptenyliden)piperidino]-2-pentanon 4-(5-Dibenzo[a,d]cycloheptenyliden)piperidin wird analog Beispiel 2 mit 5-Brom-2-pentanon umgesetzt. Smp. des Hydrochlorids der Titelverbindung 206 - 207 nach Kristallisation aus Isopropanol.
Beispiel 4
4-[4-(1O,1 1-Dihydro-5-dibenzo[a,d]cycloheptenyliden)- piperidino]-3-methyl-2-butanon
Analog Beispiel 1 wird aus 2 g 4-(10,l l-Dihydro-5-di- benzo[a,d]cycloheptenyliden)piperidin und 1,25 g Isopropenylmethylketon die rohe Titelverbindung erhalten. Nach Chromatographie des Rohproduktes an Kieselgel wird mit ätherischem Chlorwasserstoff das Hydrochlorid hergestellt und aus Methanol/Aceton umkristallisiert, Smp. über 275".
Analog Beispiel 1 können auch die folgenden [4-(10,1 1-Di- hydro-5-dibenzo [a,djcycloheptenyliden)piperidino]-alkanon- oder [4-(S-Dibenzo[a,d]cycloheptenyliden)pipen.dinol-alka- nonderivate durch Umsetzung eines Halogen-alkanons mit einem entsprechend substituierten 4-(1 0,11 -Dihydro-5-dibenzo- [a,d]cycloheptenyliden)piperidin- oder 4-(5-Dibenzo[a,d]cyclo- heptenyliden)piperidinderivat erhalten werden:

To dissolve 10 g of 4- (10,11-dihydro-5-dibenzo [a, d] -cycloheptenylidene) piperidine in 100 ml of chloroform, 9.2 g of sodium carbonate are added, then the solution of 7.2 is added dropwise over 20 min g of 5-bromo-2-pentanone in 50 ml of chloroform and then refluxed for 17 hours. After the crude product has been chromatographed on silica gel, the hydrochloride is prepared with ethereal hydrogen chloride and recrystallized from methanol / acetone, m.p. over 275 ".
