Die Erfindung betrifft ein Verfahren zur Herstellung neuer 9-(4-Piperidyliden)-thioxanthenderivate der Formel I, worin Rl Wasserstoff oder Chlor bedeutet und R2 für die Cyano-, eine COR3- oder COOR-Gruppe, wobei R3 eine niedere Alkylgruppe bedeutet, steht, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I und ihren Säureadditionssalzen, indem man die Verbindungen der Formel II, worin R, obige Bedeutung besitzt, mit Verbindungen der Formel III, worin R2 obige Bedeutung besitzt, umsetzt und die erhaltenen Verbindungen der Formel I in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
Vorzugsweise steht der Rest R2 für eine Alkylcarbonylgruppe. In dem Rest R2 enthaltene Alkylgruppen R3 besitzen vorzugsweise 1-4 Kohlenstoffatome und stellen insbesondere Methyl dar.
Die Anlagerung einer Verbindung der Formel III - Acrylnitril, eines Vinylketons oder Acrylesters - an die Verbindungen der Formel II kann in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. in einem niederen Alkanol, wie Äthanol, vorzugsweise bei erhöhter Temperatur, z.B. bei 70 bis 100 insbesondere bei Siedetemperatur des Reaktionsgemisches, ausgeführt werden.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen der Formel II können beispielsweise erhalten werden, indem man Ketone der Formel IV, worin Rl obige Bedeutung besitzt, mit der Grignard-Verbindung der Formel V umsetzt, den erhaltenen Komplex hydrolysiert, anschliessend Wasser abspaltet und aus den so erhaltenen Verbindungen der Formel VI, worin Rl obige Bedeutung bssitzt, die Methylgruppe in an sich bekannter Weise abspaltet, z.B. durch Umsetzung mit einer Verbindung der Formel VII und Hydrolyse des dabei gebildeten Urethans.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher'als Heilmittel verwendet werden. Insbesondere besitzen sie schlaffördernde Eigenschaften. So führen sie im Schlafversuch nach der Methode von G. Stille, H. Lauener und E. Eichenberger (I1 Farmaco Vol. 26, 10, 603-625 [1971]) bei Ratten in Dosen von ca. 1 bis 10 mg/kg Körpergewicht p. o.
zur Vermehrung des Schlafes. In Dosen von 0,1 bis 3,0 mg/kg Körpergewicht i. v. bewirken die Substanzen beim Kaninchen im Weckversuch nach der Methode von G. Stille, H. Ackermann, E. Eichenberger und H. Lauener (int. J. Neuropharmacol. 4, 375-391 [1965J) eine Steigerung der elektrographischen Weckschwelle.
Aufgrund ihrer schlaffördernden Wirkung können die Substanzen zur Behandlung von Schlafstörungen und Unruhezuständen verwendet werden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca.
0,7 bis 10 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 50 bis 500 mg. So enthalten z.B. für orale Applikationen die Teildosen etwa 12 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Ausserdem besitzen die Substanzen muskelrelaxierende Eigenschaften. So bewirken sie am Kaninchen in Dosen von ca. 0,01 bis 1,0 mg/kg Körpergewicht i. v. eine Herabsetzung des Muskeltonus, wie sich bei Messungen des reflektorischen Muskeltonus nach der Methode von H. J. Teschendorf, R. Kretzschmar und A. Ladous (Arch. exp. Pharmacol. 266, 467-468 [1970]) zeigt.
Aufgrund ihrer muskelrelaxierenden Wirkungen können die Substanzen zur Behandlung von Muskelspasmen Verwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,01 bis 7 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 50 bis 500 mg. Für orale Applikationen enthalten diese Teildosen etwa 12 bis 250 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 9-[1 -(3 -Oxobutyl)-4-piperidyliden]-thioxanthen
Zur Lösung von 10 g 9-(4-Piperidyliden)-thioxanthen in 100 ml abs. Äthanol werden bei 70O unter gutem Rühren während 10 Minuten 5.9 ml Methylvinylketon getropft. Man rührt noch eine Stunde bei 70al kühlt dann auf 0O ab und filtriert das auskristallisierte 9-[ 1 -(3-Oxobutyl)-4-piperidyli- den]-thioxanthen ab. Es schmilzt nach Kristallisation aus Äthanolbei 151-153
Beispiel 2 9-[1 -(3-Oxopentyl)-4-piperidylidenj-thioxanthen
Die Lösung von 10 g 9-(4-Piperidyliden)-thioxanthen in 100 ml abs. Äthanol wird mit 6 g Äthylvinylketon versetzt und anschliessend 15 Stunden unter Rückfluss gekocht.
Dann kühlt man auf 0O ab, filtriert das auskristallisierte 9-[1-(3-Oxopen tyl)-4-piperidyliden]-thioxanthen ab und kristallisiert es aus Äthanol um. Smp. 132-1340.
Beispiel 3 9-[1 -(2-Methoxycarbonyläthyl)-4- piperidyliden]-thioxanthen
Man verfährt wie in Beispiel 1, verwendet jedoch 10 g 9-(4 Piperidyliden)-thioxanthen und 6,4 ml Acrylsäuremethylester.

Beispiel 4 9-[1- (2-Cyanoäthyl)-4-piperidyliden ]-thioxanthen
Man verfährt wie in Beispiel 1, verwendet jedoch 10 g 9-(4 Piperidyliden)-thioxanthen und 7,5 g Acrylnitril. Smp. der Titelverbindung 153-155 nach Kristallisation aus Methanol.
Zur Lösung von 28,8 g (0,036 Mol) 2-Chlor-9-(4-piperidyliden)-thioxanthen in 300 ml Alkohol gropft man während 10 Minuten eine Lösung von 13,0 g (0. 185 Mol) Methyl-vinylketon in 20 ml Alkohol, rührt 1'/2 Stunden bei 100ound dampft im Vakuum ein. Man löst den Eindampfrückstand in Aceton und versetzt mit ätherischem Chlorwasserstoff bis zur schwach kongosauren Reaktion. Das sofort ausfallende 2-Chlor-9-[1 (3-oxobutyl)-4-piperidyliden]-thioxanthen-hydrochlorid wird aus Aceton umkristallisiert. Smp. 307-3100.
Das Ausgangsmaterial wird wie folgt erhalten:
Zu einer Lösung von 17,6 g Chlorameisensäureäthylester in
60 ml absolutem Toluol tropft man bei 90O während 2 Stunden die Lösung von 17,7 g 4-(2-Chlor-thioxanthenyliden)- 1methylpiperidin in 100 ml absolutem ToluoL Dann wird 15 Stunden bei 120 gerührt, die Lösung im Vakuum eingedampft und der Rückstand mit einer Lösung von 28,0 g Kaliumhydro xyd in 200 ml n-Butanol eine Stunde unter Rückfluss gekocht.
Man kühlt ab, verdünnt mit viel Wasser und extrahiert mit Äther. Das nach Abdampfen des Äthers zurückbleibende rohe 4- (2-Chlor-9-thioxanthenyliden)-piperidin wird ohne weitere Reinigung weiterverarbeitet.

Example 1 9- [1 - (3 -oxobutyl) -4-piperidylidene] thioxanthene
To dissolve 10 g of 9- (4-piperidylidene) thioxanthene in 100 ml of abs. Ethanol are added dropwise 5.9 ml of methyl vinyl ketone at 70 ° with thorough stirring for 10 minutes. The procedure is as in Example 1, except that 10 g of 9- (4 piperidylidene) thioxanthene and 7.5 g of acrylonitrile are used. Mp. Of the title compound 153-155 after crystallization from methanol.
