Im Schweiz. Hauptpatent Nr. 574 956 wird die Herstellung von Piperidin-1-ylalkanonderivaten der Formel A, worin R1, niederes Alkyl, R2' Wasserstoff oder niederes Alkyl und R3' Wasserstoff, Chlor, Brom, Fluor, Trifluormethyl, niederes Alkyl, niederes Alkoxy oder niederes Alkylthio und n' 1-3 bedeuten, beschrieben.
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer Piperidin- 1-ylalkanonderivate der Formel I, worin n für eine ganze Zahl von 1 bis 3 steht, R1 niederes Alkyl bedeutet, R2 Wasserstoff oder niederes Alkyl bedeutet, R3 für Wasserstoff, Chlor, Brom, Fluor, niederes Alkyl oder niederes Alkoxy steht, R4 Wasserstoff, Chlor, Brom oder niederes Alkyl bedeutet und A für eine Aethylen- oder Vinylengruppe steht, und ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin A, Rs und R4 obige Bedeutung besitzen, mit Verbindungen der Formel III, worin Rl obige Bedeutung besitzt und Z für eine X-(CH2)n-CHR2- Gruppe, worin R2 und n obige Bedeutung besitzen und X den Säurerest eines reaktionsfähigen Esters bedeutet, oder eine CH2 = CR2-Gruppe, worin R2 obige Bedeutung besitzt, steht, umsetzt und die erhaltenen Verbindungen der Formel 1 in Form der freien Basen oder ihrer Säureadditionssalze gewinnt.
In den Verbindungen der Formel 1 steht der Substituent R2 vorzugsweise für Wasserstoff. Falls R2 niederes Alkyl bedeutet, so enthält diese Alkylgruppe vorzugsweise 1-4 Kohlenstoffatome und stellt insbesondere die Methylgruppe dar. Der Rest R1 enthält vorzugsweise 1-4, insbesondere 1-2 Kohlenstoffatome. Die Substituenten R3 und R4 stellen vorzugsweise Wasserstoff oder Chlor dar. Falls R3 und R4 für niederes Alkyl stehen, so enthält dieses vorzugsweise 14 Kohlenstoffatome, falls R4 für niederes Alkoxy steht, so enthält dieses vorzugsweise 1-3 Kohlenstoffatome und stellt insbesondere Methyl dar.
Die erfindungsgemässe Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel III kann beispielsweise in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel erfolgen. Für die Anlagerung von Vinylverbindungen der Formel IIIa, worin Rt und R2 obige Bedeutung besitzen, an Verbindungen der Formel II sind als Lösungsmittel beispielsweise niedere Alkanole wie Aethanol oder Aether wie z. B. Dioxan geeignet und die Reaktionstemperatur liegt vorzugsweise bei Temperaturen zwischen ca. 20 und 1000.
Vorteilhafterweise kann die Umsetzung durch Zusatz von starken Basen wie z. B. Benzyltrimethylammoniumhydroxid katalysiert werden. Die Umsetzung von Verbindungen der Formel II mit Verbindungen der Formel IIIb, worin Rt, R2, n und X obige Bedeutung besitzen, erfolgt vorzugsweise in Gegenwart eines säurebindenden Mittels. Als unter den Reaktionsbedingungen inerte Lösungsmittel eignen sich z.B.
niedere Alkanole wie Aethanol, chlorierte aliphatische Kohlenwasserstoffe wie Chloroform oder aromatische Kohlenwasserstoff wie Toluol oder auch Dimethylformamid. Als säurebindendes Mittel kann man z. B. Alkalimetallcarbonate wie Natrium- oder Kaliumcarbonat oder eine tertiäre Stickstoffbase wie Triäthylamin verwenden. Die Reaktion wird vorzugsweise bei Temperaturen zwischen ca. 50 und ca. 1500, insbesondere bei Siedetemperatur des Reaktionsgemisches ausgeführt. In den Verbindungen der Formel IIIb bedeutet X vorzugsweise Chlor, Brom, Jod oder den Säurerest einer organischen Sulfonsäure, z.B. den Methylsulfonyloxy- oder den p-Toluolsulfonyloxyrest.
Die Verbindungen der Formel I können auf an sich bekannte Weise aus dem Reaktionsgemisch isoliert und gereinigt werden, die freien Basen lassen sich in üblicher Weise in ihre Säureadditionssalze überführen und umgekehrt.
Die Ausgangsverbindungen der Formel II können beispielsweise erhalten werden, indem man Ketone der Formel IV, worin A, R3 und R4 obige Bedeutung besitzen, mit der Grignard-Verbindung der Formel V umsetzt, den erhaltenen Komplex hydrolysiert, anschliessend Wasser abspaltet und aus den so erhaltenen Verbindungen der Formel VI, worin A, R3 und R4 obige Bedeutung besitzen, die Methylgruppe in an sich bekannter Weise abspaltet, z. B. durch Umsetzung mit einer Verbindung der Formel VII und Hydrolyse des dabei gebildeten Urethans.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Insbesondere besitzen die Verbindungen der Formel I sedativneuroleptische und schlaffördernde sowie auch muskelrelaxierende Eigenschaften. Die sedativen und die muskelrela xierenden Eigenschaften lassen sich z. B. an Mäusen bei der Messung der motorischen Aktivität in der Shuttle-box und im Klettertest beobachten. So hemmen die Substanzen z. B. in Dosen von 5-100 mg/kg p.o. die spontane motorische Aktivität und in Dosen von 0,3-20 mg/kg i.p. die Kletterfähigkeit der Mäuse. Die hypnotischen Eigenschaften der Verbindungen lassen sich z. B. an Mäusen und Ratten durch eine Potenzierung der durch Pentothal oder Barbital hervorgerufenen hypnotischen Wirkungen zeigen. So führen die Verbindungen der Formel I bei Mäusen in Dosen von 3-100 mg/kg i.p. zu einer Verlängerung des Andauerns des durch Pentothal verursachten Verlustes des Aufrichtungsreflexes.
Bei mit einer noch nicht zur Verhinderung des Aufrichtungsreflexes ausreichenden Menge Barbital vorbehandelten Ratten führen die Verbindungen in Dosen von 4-80 mg/kg i. p. zum Verlust dieses Aufrichtungsreflexes.
Aufgrund ihrer sedativen Eigenschaften können die Substanzen in der Psychiatrie zur Behandlung von Erregungszuständen Verwendung finden. Die zu verwendenden Dosen variieren naturgemäss je nach Art der Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate mit einer Doses von ca.
0,1 bis 100 mg/kg Körpergewicht erhalten. Diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 5 bis 200 mg. So enthalten z. B. für orale Applikationen die Teildosen etwa 1,3 bis 50 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.

Im allgemeinen werden jedoch befriedigende Resultate mit einer Dosis von ca. 0,1 bis 200 mg/kg Körpergewicht erhalten, diese Dosis kann nötigenfalls in 2 bis 4 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 55 bis 200 mg.
Für orale Applikationen enthalten diese Teildosen etwa 1,3 bis 100 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw.
ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden.
Beispiel 1 4-[4- (9,1 O-Dihydro-4H-benzo[4,5] cyclohepta[l ,2-b]- thio phen-4-yliden) piperidino]-2-butanon
Zur Lösung von 15,0 g 9,10-Dihydro-4-(4-piperidyliden) 4H-benzo[4,5]cyclohepta [1,2-b]thiophen in 60 ml Dioxan und 0,3 ml Benzyltrimethylammoniumhydroxyd tropft man innert 10 Minuten eine Lösung von 5,8 g Methylvinylketon in 15 ml Dioxan. Das Reaktionsgemisch wird 1 Stunde bei Raumtemperatur gerührt, über Nacht stehen gelassen, mit Aktivkohle entfärbt und zur Trockne eingedampft. Der ölige Rückstand, die im Titel genannte Verbindung, wird in das Hydrobromid übergeführt. Smp.: 234-235" (aus Aethanol).
Beispiel2 5-[4-(9,1 O-Dihydro-4H-benzo[4,5] cyclohepta[1,2-b] thio phen-4-ylid en) piperidillo]-2-pentanon
Zu einem Gemisch von 18,0 g 9,10-Dihydro-4-(4-piperidy liden)- 4H-benzo[4,5] cyclohepta[1,2-b] thiophen-hydrobromid und 12,0 g Natriumkarbonat in 200 ml Chloroform tropft man innert 10 Minuten eine Lösung von 10,0 g 5-Brom2-pentanon in 100 ml Chloroform und erhitzt das Gemisch 7 Stunden am Rückfluss. Nach Zugabe von weiteren 5 g Natriumkarbonat wird das Gemisch bei Siedetemperatur noch 16 Stunden gerührt, mit 100 ml Dimethylformamid verdünnt, heiss filtriert und das Filtrat mit Wasser gewaschen.
Die organische Lösung wird über Natriumsulfat getrocknet, das Lösungsmittel abgedampft und der ölige Rückstand, die im Titel genannte Verbindung, in das Hydrobromid übergeführt. Smp.: 218-220 (aus Aethanol).
Beispiel 5 5-[4- (9,1 O-Dihyd ro-6-methyl-4H-benzo-[4,5] cyclohepta[l,2- bfthiophen-4-yllden) - piperidinol-2,2-dimethyl-3-pentanon
Die Titelverbindung wird analog Beispiel 1 erhalten und chromatographisch gereinigt. Rf-Wert*: 5,0
Beispiel 3 4-[4- (4H-Benzo[4,5]cyclohepta [1,2-b]thiophen-4.yliden) pipendino]-2 -butanon
Gemäss Beispiel 1 wird aus 7,5 g 4-(4-Piperidyliden)- 4Hbenzo[4,5]cyclohepta [1,2-b]thiophen und 0,15 g Benzyltrimethylammoniumhydroxid in 30 ml Dioxan und 2,9 g Methylvinylketon in 10 ml Dioxan die Titelverbindung hergestellt.
Smp. des Hydrobromids: 133-136 (aus Aethanol/Aether).
Beispiel 4 5-[4-(4H-Benzo[4,51cyclohepta [1,2 -b]tlziophen-4-yliden) piperidinol-2-pentanon
Gemäss Beispiel 2 wird aus 7,5 g 4-(4-Piperidyliden) 4Hbenzo[4,5]cyclohepta [1,2-b]thiophen, 7,lg Natriumcarbonat und 5,3 g 5-Brom-2-pentanon in 300 ml Chloroform die Titelverbindung hergestellt. Smp. des Hydrobromids: 248-251" (aus Methanol/Aethanol).
