Verfahren zur Herstellung neuer Indolderivate
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Indolderivate der Formel I, worin R1 niederes Alkyl, eine Cycloalkylgruppe von 3 oder 4 Kohlenstoffatomen oder eine Phenylalkylgruppe, deren Phenylrest durch mindestens 2 Kohlenstoffatome vom Stickstoffatom getrennt ist, bedeutet, R2 für Wasserstoff oder die Methylgruppe steht, R3 und R4 je Wasserstoff oder niederes Alkyl bedeuten, wobei R4, falls Rs Wasserstoff darstellt, auch die Amino- oder eine (nieder)Alkylaminogruppe bedeuten kann, oder R3 und R4 zusammen mit dem Stickstoffatom, woran sie gebunden sind, für den l-Pyrrolidinyl-, 1-Piperidinyl- oder 1-Morpholinylrest stehen, und ihrer Säureadditionssalze.
Von den Verbindungen der Formel I, worin R1 für niederes Alkyl steht, sind diejenigen bevorzugt, worin die Alkylgruppe verzweigt bzw. bzw. kompakt ist, wie z.B. die Isopropyl-, sekundär Butyl-, tert.Butyl-, tert.Pentyl-, 3-Pentylgrup pe usw. Stellt R1 eine Phenylalkylgruppe dar, so enthält diese insbesondere 8 bis 13 Kohlenstoffatome. Stellt R3 und/oder R4 einen niederen Alkylrest dar, so enthält dieser 1 bis 4 Kohlenstoffatome. Stellt R4 eine (nieder) Alkylaminogruppe dar, so enthält deren (nieder)Alkylrest insbesondere 1 bis 4 Kohlenstoffatome.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin R1, R2, R3 und R4 obige Bedeutung besitzen, debenzyliert und gewünschtenfalls die so erhaltenen Verbindungen in ihre Säureadditionssalze überführt.
Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Die Debenzylierung erfolgt z.B. durch Hydrierung in Gegenwart eines Katalysators, vorzugsweise eines Palladiumkatalysators in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. Essigester, einem niederen Alkanol wie Äthanol usw., und wird vorzugsweise bei Raumtemperatur und Normaldruck durchgeführt.
Nach beendeter Hydrierung filtriert man vom Katalysator ab und dampft das Filtrat zur Trockne ein.
Der so erhaltene Trocken rückstand kann nach bekannten Methoden aufgearbeitet und gereinigt werden.
Auch die Verbindungen der Formel II sind neu.
Zu den Verbindungen der Formel IIa, worin R1 und R obige Bedeutung besitzen und RI5 und R'4 für Wasserstoff oder niederes Alkyl steht, oder RIs und R'4 zusammen mit dem Stickstoffatom, woran sie gebunden sind, den l-Pyrrolidinyl-, l-Piperidinyl-, oder l-Morpholinylrest stehen, kann man z.B. erhalten, indem man Verbindungen der Formel III, worin R2, R', und R'4 obige Bedeutung besitzen, mit Epihalohydrinen, beispielsweise mit Epichlor- bzw. Epibromhydrin behandelt, und die so erhaltenen Reaktionsprodukte mit Verbindungen der Formel IV, worin R1 obige Bedeutung besitzt, umsetzt.
Zur Herstellung der Verbindungen der Formel IIb, worin R1 und R obige Bedeutung besitzen und R Wasserstoff oder niederes Alkyl bedeutet, kann man Verbindungen der Formel V, worin R1 und R2 obige Bedeutung besitzen und Alk niederes Alkyl bedeutet, mit Hydrazin bzw. (nieder)Alkylhydrazin umsetzen.
Die Verbindungen der Formel III sind ebenfalls neu.
Zu den Verbindungen der Formel IIIa, worin Re obige Bedeutung besitzt und entweder Rs und R6 je für niederes Alkyl stehen oder zusammen mit dem Stickstoffatom, woran sie gebunden sind, den 1-Pyrrolidinyl-, 1 Piperidinyl- oder l-Morpholinylrest bedeutet, können z.B.
erhalten werden, indem man 4-Benzyloxy-2-indolcarbonsäurechlorid mit Aminen der Formel VI, worin R5 und R6 obige Bedeutung besitzen, umsetzt, gewünschtenfalls die so erhaltenen Verbindungen der Formel VIIa, worin R5 und R6 obige Bedeutung besitzen, unter den Bedingungen einer Mannich-Reaktion zu den Verbindungen der Formel VIIb, worin R5 und R6 obige Bedeutung besitzen und R7 für eine di(nieder)Alkylaminomethylgruppe steht, aminomethyliert, und die so erhaltenen Verbindungen der Formel VIIa oder VIIb durch katalytische Hydrierung, z.B. in Gegenwart eines Palladiumkatalysators in die Verbindungen der Formel IIIa überführt.
Zu den Verbindungen der Formel Ilib, worin R, obige Bedeutung besitzt und Rs Wasserstoff oder niederes Alkyl bedeutet, gelangt man z.B., indem man Verbindungen der Formel VIII, worin R2 und R5 obige Bedeutung besitzen, debenzyliert, beispielsweise durch Hydrierung in Gegenwart eines Palladiumkatalysators.
Die Verbindungen der Formel VIII sind ebenfalls neu und können durch Umsetzung der Verbindungen der Formel IX, worin R2 und Alk obige Bedeutung besitzen, mit den entsprechenden Aminen hergestellt werden.
Die Ester der Formel IXa, worin Alk obige Bedeutung besitzt, können durch Veresterung von 4-Benzyloxy-2-indolcarbonsäure hergestellt werden. Durch Aminomethylierung dieser Verbindungen der Formel IXa und katalytische Hydrierung der so erhaltenen Mannich Basen erhält man 4-Hydroxyl-3-methyl-2-indolcarbonsäureester, die durch Umsetzung mit Benzylchlorid, unter alkalischen Bedingungen, in die Verbindungen der Formel IXb, worin Alk obige Bedeutung besitzt, übergeführt werden können.
Die Ester der Formel V kann man analog zu dem Verfahren zur Herstellung der Verbindungen der Formel IIa erhalten.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
Die Verbindungen der Formel I und ihre Salze sind in der Literatur bisher nicht beschrieben. Sie zeichnen sich in der pharmakologischen Prüfung durch vielfältige und stark ausgeprägte Effekte aus und können daher als Heilmittel verwendet werden.
Die neuen Substanzen zeigen am spontanschlagenden, isolierten Meerschweinchenvorhof eine Hemmung der positiv-inotropen Adrenalinwirkung, wobei diese antagonistische Wirkung bei Badkonzentrationen von 0,001 bis 20mg/l auftritt. Am narkotisierten Ganztier (Katze, Hund) führen sie zu einer starken Hemmung der durch Isoproterenol [1-(3,4-Dihydroxyphenyl)-2-isopro- pylaminoäthanol] bedingten Tachycardie und Blutdrucksenkung. Die Verbindungen besitzen demnach eine Blockerwirkung auf die adrenergischen p-Rezeptoren.
Aufgrund ihrer antiarrhythmischen Wirkung sind sie ausserdem zur Behandlung von Herzrhythmusstörungen geeignet. Die zu verwendenden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate bei Testtieren mit einer Dosis von 0,001 bis 0,6 mg/kg Körpergewicht verhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 400mg. Für orale Applikationen enthalten die Teildosen etwa 0,6 bis 200 mg der neuen Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.

Beispiel 1 2-DimethylcarOyl-4-(2-hydroxy 3-isopropyl - aminopropoxy)indol (1) Hergestellt durch Umsetzung von 4-Hydroxyindol-2-carbonsäureäthylester (Smp. 159 bis 1600) mit 1 -(N-Benzylisopropylamino)-3-chlor-2-propanol (Sdp. 110 bis 1150/0,2mm Hg) und Behandlung des so erhalte

-4-hydroxy-indol und 0,4 g Natriumhydroxid in 150ml Methanol gibt man 4,8 g 1-(N-Benzylisopropylamino) -3-chlor-2-propanol und erhitzt während 20 Stunden zum Sieden. Man verdampft unter vermindertem Druck das Lösungsmittel, nimmt in Äther auf und extrahiert mit Wasser. Die ätherische Lösung wird anschliessend bis zur beginnenden Kristallisation eingeengt: Smp. 160 bis 1620.
1 g des so erhaltenen 2-Dimethylaminocarbamoyl-4 -(2-hydroxy-3 -benzylisopropylaminopropoxy)indols werden in 50 ml Methanol aufgenommen und nach Zusatz von 0,5 g Palladiumkatalysator (5% Palladium auf Aluminiumoxid) mit Wasserstoff bis zur Beendigung der Wasserstoffaufnahme geschüttelt. Man filtriert vom Katalysator, verdampft unter vermindertem Druck das Lösungsmittel fast vollständig und kristallisiert die im Titel genannte Verbindung mit Essigester; Smp. 156- 1580.
Das als Ausgangsmaterial verwendete l-(N-Benzyl- isopropylamino)-3-chlor-2-propanol kann z.B. wie folgt hergestellt werden:
Man erhitzt ein Gemisch von 18flug Epichlorhydrin und 29,8 g N-Benzylisopropylamin in 100 ml Benzol während 24 Stunden am Rückfluss zum Sieden, verdampft hierauf das Lösungsmittel und destilliert den Rückstand im Hochvakuum; man erhält l-(N-Benzyl- isopropylamino)-3-chlor-2-propanol vom Sdp. 110-1150/ 0,2 mm Hg.
2-Dimethylcarbamoyl-4-hydroxyindol (Smp. 231 bis 2340, aus Aceton) erhält man durch Entbenzylierung von 2-Dimethylcarbamoyl-4-benzyloxyindol (= 4-Benzyloxyindol-2-carbonsäuredimethylamid) mit Wasserstoff in Gegewart eines Palladium-Katalysators (5% auf Aluminiumoxid).
Man verfährt analog zu den für Beispiel 1 gegebenen Vorschriften und erhält folgende Verbindungen: indol-2-carbonsäureäthylesters (Smp. 103 bis 1050) mit Hydrazin.
4-Hydroxyindol-2-carbonsäureäthylester wird durch Debenzylierung von 4-Hydroxyindol-2-carbonsäureäthylester (Smp. 159 bis 1600) und letztere Verbindung durch Veresterung von 4-Hydroxyindol-2-carbonsäure erhalten.
1 - (N- Benzylisopropylamino) - 3-chlor-2 propanol erhält man durch Erhitzen eines Gemisches von Epichlorhydrin und N-Benzylisopropylamin in Benzol.
(2) Hergestellt durch Umsetzung von 4-Hydroxy-2 -methylcarbamoylindol (Smp. 201 bis 2030) mit 1-(N -Benzyl-isopropylamino)-3 -chlor-2-propanol. 4-Hydroxy -2-methylcarbamoylindol erhält man durch Umsetzung von 4-Benzyloxyindol-2-carbonsäure mit Methylamin und Debenzylierung des so erhaltenen 4-Benzyloxy-2 -methylcarbamoylindols vom Smp. 209 bis 2120.
(3) Analog (2), jedoch ausgehend von 2-Carbamoyl -4-hydroxyindol. Letztere Verbindung erhält man durch Umsetzung von 4-Benzyloxyindol-2-carbonsäurechlorid mit Ammoniak und Debenzylierung des so erhaltenen 4-Benzyloxy-2-carbamoylindols vom Smp. 187 bis 1890 (4) Hergestellt durch Behandlung von 4-[3-(N-Ben- zyl - tert.butylamino) -2- hydroxypropoxyjindol.2-carbon.
säureäthylesters mit Methylhydrazin. Den eingesetzten Ester erhält man durch Umsetzung von 4-Hydroxyindol -2-carbonsäureäthylester mit Epichlorhydrin und Behandlung des so erhaltenen Reaktionsproduktes mit N -Benzyl-tert.butylamin.
(5) Analog (4), durch Umsetzung von 4-[3-(N-Ben- zyl-tert. butylamino) - 2 - hydroxypropoxy]indol-2-carbonsäureäthylesters mit Hydrazin.

Process for the production of new indole derivatives
The invention relates to a process for the preparation of new indole derivatives of the formula I, in which R1 is lower alkyl, a cycloalkyl group of 3 or 4 carbon atoms or a phenylalkyl group whose phenyl radical is separated from the nitrogen atom by at least 2 carbon atoms, R2 is hydrogen or the methyl group, R3 and R4 each denote hydrogen or lower alkyl, where R4, if Rs denotes hydrogen, can also denote the amino or a (lower) alkylamino group, or R3 and R4 together with the nitrogen atom to which they are bonded for l-pyrrolidinyl -, 1-piperidinyl or 1-morpholinyl radical, and their acid addition salts.
Of the compounds of the formula I in which R1 is lower alkyl, those are preferred in which the alkyl group is branched or compact, e.g. the isopropyl, secondary butyl, tert-butyl, tert-pentyl, 3-pentyl group, etc. If R1 represents a phenylalkyl group, it contains in particular 8 to 13 carbon atoms. If R3 and / or R4 represents a lower alkyl radical, this contains 1 to 4 carbon atoms. If R4 represents a (lower) alkylamino group, its (lower) alkyl radical contains in particular 1 to 4 carbon atoms.
According to the invention, the new compounds of the formula I and their acid addition salts are obtained by debenzylating compounds of the formula II in which R1, R2, R3 and R4 have the above meaning and, if desired, converting the compounds thus obtained into their acid addition salts.
Acid addition salts can be prepared from the free bases in a known manner and vice versa.
The debenzylation takes place e.g. by hydrogenation in the presence of a catalyst, preferably a palladium catalyst, in a solvent which is inert under the reaction conditions, e.g. Ethyl acetate, a lower alkanol such as ethanol, etc., and is preferably carried out at room temperature and normal pressure.
After the hydrogenation has ended, the catalyst is filtered off and the filtrate is evaporated to dryness.
The dry residue obtained in this way can be worked up and purified by known methods.
The compounds of the formula II are also new.
The compounds of the formula IIa, in which R1 and R have the above meaning and RI5 and R'4 are hydrogen or lower alkyl, or RIs and R'4 together with the nitrogen atom to which they are bonded, the l-pyrrolidinyl, l -Piperidinyl, or l-morpholinyl, can be, for example obtained by treating compounds of the formula III in which R2, R 'and R'4 have the above meaning with epihalohydrins, for example with epichlorohydrin or epibromohydrin, and the reaction products thus obtained with compounds of the formula IV in which R1 has the above meaning owns, implements.

The compounds of the formula VIII are likewise new and can be prepared by reacting the compounds of the formula IX, in which R2 and Alk have the above meanings, with the corresponding amines.
The esters of the formula IXa, in which Alk has the above meaning, can be prepared by esterification of 4-benzyloxy-2-indolecarboxylic acid. Aminomethylation of these compounds of the formula IXa and catalytic hydrogenation of the Mannich bases obtained in this way gives 4-hydroxyl-3-methyl-2-indolecarboxylic acid esters, which by reaction with benzyl chloride under alkaline conditions in the compounds of the formula IXb, in which Alk has the above meaning owns, can be transferred.
The esters of the formula V can be obtained analogously to the process for the preparation of the compounds of the formula IIa.
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
The compounds of the formula I and their salts have not yet been described in the literature. In pharmacological testing, they are characterized by diverse and highly pronounced effects and can therefore be used as remedies.
The new substances show an inhibition of the positive-inotropic adrenaline effect on the spontaneously beating, isolated guinea pig atrium, this antagonistic effect occurring at bath concentrations of 0.001 to 20 mg / l. In the anesthetized whole animal (cat, dog) they lead to a strong inhibition of the tachycardia and lowering of blood pressure caused by isoproterenol [1- (3,4-dihydroxyphenyl) -2-isopropylaminoethanol]. The compounds accordingly have a blocking effect on the adrenergic p-receptors.
Due to their antiarrhythmic effect, they are also suitable for the treatment of cardiac arrhythmias. The doses to be used naturally vary depending on the type of substance used, the administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals with a dose of 0.001 to 0.6 mg / kg body weight; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form. For larger mammals, the daily dose is around 2 to 400 mg. For oral applications, the partial doses contain about 0.6 to 200 mg of the new compounds in addition to solid or liquid carriers or diluents.

2-Dimethylcarbamoyl-4-hydroxyindole (m.p. 231 to 2340, from acetone) is obtained by debenzylating 2-dimethylcarbamoyl-4-benzyloxyindole (= 4-benzyloxyindole-2-carboxylic acid dimethylamide) with hydrogen in the presence of a palladium catalyst (5% on aluminum oxide).
The procedure given for Example 1 is repeated and the following compounds are obtained: ethyl indole-2-carboxylate (melting point 103 to 1050) with hydrazine.
Ethyl 4-hydroxyindole-2-carboxylate is obtained by debenzylating ethyl 4-hydroxyindole-2-carboxylate (melting point 159 to 1600) and the latter compound is obtained by esterifying 4-hydroxyindole-2-carboxylic acid.
1 - (N-Benzylisopropylamino) -3-chloro-2 propanol is obtained by heating a mixture of epichlorohydrin and N-benzylisopropylamine in benzene.
(2) Prepared by reacting 4-hydroxy-2-methylcarbamoylindole (m.p. 201 to 2030) with 1- (N -benzyl-isopropylamino) -3-chloro-2-propanol. 4-Hydroxy-2-methylcarbamoylindole is obtained by reacting 4-benzyloxyindole-2-carboxylic acid with methylamine and debenzylating the 4-benzyloxy-2-methylcarbamoylindole with a melting point of 209 to 2120.

The 4-hydroxy-3-methylindole-2-carboxylic acid dimethylamide is obtained by aminomethylation of 4-benzyloxyindole-2-carboxylic acid dimethylamide and hydrogenation of 4-benzyloxy-3-dimethylaminomethylindole-2-carboxylic acid dimethylamide with hydrogen in the presence of a palladium catalyst (5% on aluminum oxide).
