Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung neuer Indolderivate der Formel I, worin Rl niederes Alkyl, eine Cycloalkylgruppe von 3 oder 4 Kohlenstoffatomen oder die 3-Phenylpropylg'rup- pe bedeutet, R2 für eine Cycloalkylgruppe von 3 oder 4 Kohlenstoffatomen oder den Phenylrest steht und R3 Wasserstoff oder die Methylgruppe bedeutet.
Von den Verbindungen der Formel I, worin Rl für niederes Alkyl steht, sind diejenigen bevorzugt, worin dieser Alkylrest kompakt oder verzweigt, insbesondere in x-Stellung zum Stickstoff verzweigt ist, wie z.B. die Isopropyl-, sec. Butyl-, tert.Butyl-, tert.Pentyl-, 3-Pentyl- gruppe usw.
Erfindungsgemäss gelangt man zu den Verbindungen der Formel I, indem man Verbindungen der Formel II, worin Rl, R. und R3 obige Bedeutung besitzen, hydrogenolytisch debenzyliert.
Aus den freien Basen lassen sich in bekannter Weise Säureadditionssalze herstellen und umgekehrt.
Die Debenzylierung erfolgt z.B. durch Hydrierung in Gegenwart eines Katalysators, vorzugsweise eines Palladiumkatalysators in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z.B. Essigester, einem niederen Alkanol wie Äthanol usw., und wird vorzugsweise bei Raumtemperatur und Normaldruck durchgeführt.
Nach beendeter Hydrierung filtriert man gewöhnlich vom Katalysator ab und dampft das Filtrat zur Trockne ein.
Der so erhaltene Trockenrückstand kann nach bekannten Methoden aufgearbeitet und gereinigt werden.
Die Verbindungen der Formel II sind neu und können beispielsweise hergestellt werden, indem man Verbindungen der Formel III, worin R. und R3 obige Bedeutung besitzen, als Salz oder in Gegenwart einer Base mit Verbindungen der Formel IVa, worin R1 obige Bedeutung besitzt und X für Halogen, vorzugsweise Chlor oder Brom steht, oder Verbindungen der Formel IVb, worin R1 obige Bedeutung besitzt, umsetzt.
Verbindungen der Formel Wa können durch Um setzung von Aminen der Formel V, worin R1 obige Bedeutung besitzt, mit Epihalohydrinen erhalten werden und durch Behandlung mit Alkalien in Verbindiingen der Formel IV;b übergeführt werden.
Die Verbindungen der Formel II können z.B. auch hergestellt werden, indem man Verbindungen der Formel III als Salz oder in Gegenwart einer Base mit Epihalohydrinen umsetzt und die erhaltenen Reaktionsprodukte mit Aminen der Formel V kondensiert.
Zur Herstellung der Verbindungen der Formel III kann man die entsprechenden 4-Benzyloxyindol-2-carbonsäureester mit Aminen der Formel VI, worin R.
obige Bedeutung besitzt, umsetzen und anschliessend die Benzylgruppe hydrogenolytisch abspalten. Eine Variante dieses Verfahrens ist in den Beispielen 1 und 2 beschrieben.
4- Benzyloxy-3- methyl - 2-indolcarbonsäureester erhält man z.B. durch Aminomethylierung von 4-Benzyioxy-2- indolcarbonsäureestere Hydrierung der so erhaltenen Mannich-Basen und Umsetzung der so entstandenen 4 -Hydroxy-3 -methyl-2-indolcarbonsäureester mit Benzylchlorid unter alkalischen Bedingungen.
Soweit die Herstellung der Ausgangsstoffe nicht beschrieben wird, sind diese bekannt oder können nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren hergestellt werden.
Die neuen Verbindungen der Formel I oder ihre Säureadditionssalze sind in der Literatur bisher noch nicht beschrieben worden. Sie zeichnen sich durch interessante pharmakodynamische Eigenschaften aus und können daher als Heilmittel verwendet werden.
Die neuen Substanzen zeigen am spontanschlagenden, isolierten Meerschweinchenvorhof eine Hemmung der positivinotropen Adrenalinwirkung, wobei diese antagonistische Wirkung bei Badkonzentrationen von 0,005 bis 2,5 mag/ 1 auftritt. Am narkotisierten Ganztier (Katze, Hund) führen sie zu einer starken Hemmung der durch Isoproterenol 1 - (3 (3,4-Dihydroxyphenyl)-2-isopropylami- noäthanolj bedingten Tachycardie und Blutdrucksenkung. Die Verbindungen besitzen demnach eine Blocker wirkung auf die adrenergischen ,3-'Rezeptoren. Aufgrund ihrer antiarrhythmischen Wirkung sind sie ausserdem zur Behandlung von Herzrhythmusstörungen geeignet.
Die zu verwendenden Dosen variieren naturgemäss je nach der Art der verwendeten Substanz, der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch befriedigende Resultate bei Testtieren mit einer DoSis von 0,02 bis 0,6 mglkg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 100 mg. Für orale Applikationen enthalten die Teildosen etwa 0,7 bis 50 mg der neuen Verbindungen neben festen oder flüssigen Trägersubstanzen oder Verdünnungsmitteln.
In den nachfolgenden Beispielen, welche die Erfindung näher erläutern, ihren Umfang jedoch in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.

Beispiel I
4-(2-Hydroxy-3-isopropylaminopropoxy)indol-2- -carbonsäurecyclopropylamid
27,2 g rohes 4-(2,3-Epoxypropoxy)indol-2-carbonsäu- recyclopropylamid werden in 200 ml Dioxan und 41,7 g Benzylisopropylamin aufgenommen und 18 Stunden im Autoklaven auf 1500 erhitzt. Man verdampft zur Trockne, schüttelt anschliessend zwischen Essigester und 10%iger Salzsäure aus und stellt ein eventuell dabei ausfallendes Harz gemeinsam mit den salzsauren Extrakten unter Eiskühlung alkalisch. Man extrahiert mit Methylenchlorid, trocknet die organischen Phasen über Magnesiumsulfat und dampft unter vermindertem Druck ein.
Zur Entfernung eines grossen Teils des überschüssigen Benzylisopropylamins schüttelt man den erhaltenen öli- gen Rückstand mehrfach mit Petroläther und slekantiert.
Das so erhaltene ölige 42(3-Benzylisopropylamino-2-hy- droxypropoxy)indol-2-carbonsaurecyclopropylamlid wird nun in 300ml Methanol aufgenommen u. in Gegenwart von 13 g eines Palladium'katalysators (5 O Pd auf Kohle) mit Wasserstoff bis zum Stillstand der Wasserstoffaufnahme geschüttelt. Man filtriert vom Katalysator, dampft unter vermindertem Druck zur Trockne ein und kristallisiert die Titelverbindung aus Essigester /Äther.
Smp. 145 bis 147 .
Das als Ausgangsmaterial benötigte 4-(2,3-Epoxypropoxy)indol-2-carbonsäurecyclopropylamid kann wie folgt erhalten werden:
4-Benzyloxyindol-2-carbonsäurechlorid wird mit Cyclopropylamin in 4-Benzyloxyindol-2-carbonsäurecyclo- propyiamid (Smp. 199 bis 2020) überführt und dieses in Tetrahydrofuran in Gegenwart eines Palladiumkatalysators (5% Pd auf Kohle) mit Wasserstoff zum 4-Hy- droxyindol-2-cabonsäurecyclopropylamid (Smp. 244 bis 2460) entbenzyliert.
43 g 4-Hydroxyindol-2-carbonsäurecyclopropylamid werden mit 190 mol Epichlorhydrin 2 Stunden zum Sieden erhitzt und anschliessend unter vermindertem Druck eingedampft. Das erhaltene rohe 4-(2,3-Epoxypropoxy)indol-2-carbonsäurecyclopropylamid wird ohne vorherige Reinigung weiterverarbeitet.
Beispiel 2
4-(2-Hydroxy-3-isopropylaminopropoxy)indol-2- -carbonsäurearnlid
Man verfährt analog Beispiel 1, indem man statt 4 - (2,3 -Epoxypropoxy) indol - 2- carbonsäurecyclopropylamid 4-(2,3-Epoxypropoxy)indol-2-carbonsäureanilid verwendet. Die Titelverbindung kristallisiert aus Methanoll Äthanol in Prismen vom Smp. 23'8 bis 2410.
Das Ausgangsmaterial erhält man ebenfalls analog Beispiel 1, ausgehend von 4-Benzyloxyind'ol-2-cabon- säurechlorid: 4-Benzyloxyindol-2-carbonsäureanilid, Smp. 199 bis 2010; 4-Hydroxyindol-2-oarbonsäureanilid, Smp. 203 bis 2050.

The present invention relates to a process for the preparation of new indole derivatives of the formula I in which Rl is lower alkyl, a cycloalkyl group of 3 or 4 carbon atoms or the 3-phenylpropyl group, R2 is a cycloalkyl group of 3 or 4 carbon atoms or the phenyl radical and R3 is hydrogen or the methyl group.
Of the compounds of the formula I in which R 1 stands for lower alkyl, those are preferred in which this alkyl radical is compact or branched, in particular branched in the x-position to the nitrogen, e.g. the isopropyl, sec-butyl, tert-butyl, tert-pentyl, 3-pentyl, etc.
According to the invention, the compounds of the formula I are obtained by hydrogenolytically debenzylating compounds of the formula II in which R 1, R and R 3 have the above meanings.
Acid addition salts can be prepared from the free bases in a known manner and vice versa.
The debenzylation takes place e.g. by hydrogenation in the presence of a catalyst, preferably a palladium catalyst, in a solvent which is inert under the reaction conditions, e.g. Ethyl acetate, a lower alkanol such as ethanol, etc., and is preferably carried out at room temperature and normal pressure.
After the hydrogenation has ended, the catalyst is usually filtered off and the filtrate is evaporated to dryness.
The dry residue obtained in this way can be worked up and purified by known methods.
The compounds of the formula II are new and can be prepared, for example, by adding compounds of the formula III, in which R. and R3 have the above meaning, as a salt or in the presence of a base with compounds of the formula IVa, in which R1 has the above meaning and X is Halogen, preferably chlorine or bromine, or compounds of the formula IVb in which R1 has the above meaning, converts.
Compounds of the formula Wa can be obtained by reaction of amines of the formula V, in which R1 has the above meaning, with epihalohydrins, and can be converted into compounds of the formula IV; b by treatment with alkalis.
The compounds of formula II can e.g. can also be prepared by reacting compounds of the formula III as a salt or in the presence of a base with epihalohydrins and condensing the reaction products obtained with amines of the formula V.
To prepare the compounds of the formula III, the corresponding 4-benzyloxyindole-2-carboxylic acid esters can be used with amines of the formula VI in which R.
has the above meaning, convert and then split off the benzyl group hydrogenolytically. A variant of this process is described in Examples 1 and 2.
4-benzyloxy-3-methyl-2-indolecarboxylic acid esters are obtained e.g. by aminomethylation of 4-benzyioxy-2-indolecarboxylic acid esters, hydrogenation of the Mannich bases thus obtained and reaction of the 4-hydroxy-3-methyl-2-indolecarboxylic acid esters thus formed with benzyl chloride under alkaline conditions.
If the preparation of the starting materials is not described, these are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
The new compounds of the formula I or their acid addition salts have not yet been described in the literature. They are distinguished by interesting pharmacodynamic properties, so they can be used as remedies.
The new substances show an inhibition of the positively inotropic adrenaline effect on the spontaneously beating, isolated guinea pig atrium, this antagonistic effect occurring at bath concentrations of 0.005 to 2.5 mag / 1. In the anesthetized whole animal (cat, dog) they lead to a strong inhibition of the tachycardia and lowering of blood pressure caused by isoproterenol 1- (3 (3,4-dihydroxyphenyl) -2-isopropylaminoethanolj). The compounds accordingly have a blocking effect on the adrenergic, 3-'receptors. Due to their antiarrhythmic effect, they are also suitable for the treatment of cardiac arrhythmias.
The doses to be used naturally vary depending on the type of substance used, the administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals with a DoSis of 0.02 to 0.6 mg / kg body weight; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form. For larger mammals, the daily dose is around 2 to 100 mg. For oral applications, the partial doses contain about 0.7 to 50 mg of the new compounds in addition to solid or liquid carriers or diluents.

Example I.
4- (2-Hydroxy-3-isopropylaminopropoxy) indole-2-carboxylic acid cyclopropylamide
27.2 g of crude 4- (2,3-epoxypropoxy) indole-2-carboxylic acid recyclopropylamide are taken up in 200 ml of dioxane and 41.7 g of benzylisopropylamine and heated to 1500 in an autoclave for 18 hours. It is evaporated to dryness, then shaken out between ethyl acetate and 10% hydrochloric acid and any resin which may precipitate is rendered alkaline together with the hydrochloric acid extracts while cooling with ice. It is extracted with methylene chloride, the organic phases are dried over magnesium sulfate and evaporated under reduced pressure.
To remove a large part of the excess benzylisopropylamine, the oily residue obtained is shaken several times with petroleum ether and slanted.
The oily 42 (3-benzylisopropylamino-2-hydroxypropoxy) indole-2-carbonsaurecyclopropylamlid obtained in this way is then taken up in 300 ml of methanol u. shaken with hydrogen in the presence of 13 g of a palladium catalyst (5 O Pd on carbon) until the hydrogen uptake ceases. The catalyst is filtered off, evaporated to dryness under reduced pressure and the title compound is crystallized from ethyl acetate / ether.

The 4- (2,3-epoxypropoxy) indole-2-carboxylic acid cyclopropylamide required as starting material can be obtained as follows:
4-Benzyloxyindole-2-carboxylic acid chloride is converted with cyclopropylamine into 4-benzyloxyindole-2-carbonsäurecyclo- propyiamid (melting point 199 to 2020) and this in tetrahydrofuran in the presence of a palladium catalyst (5% Pd on carbon) with hydrogen to the 4-Hy- Droxyindole-2-cabonic acid cyclopropylamide (m.p. 244 to 2460) debenzylated.
43 g of 4-hydroxyindole-2-carboxylic acid cyclopropylamide are heated to boiling with 190 mol of epichlorohydrin for 2 hours and then evaporated under reduced pressure. The crude 4- (2,3-epoxypropoxy) indole-2-carboxylic acid cyclopropylamide obtained is processed further without prior purification.
Example 2
4- (2-Hydroxy-3-isopropylaminopropoxy) indole-2-carboxylic acid amide
The procedure is analogous to Example 1, using 4- (2,3-epoxypropoxy) indole-2-carboxylic anilide instead of 4- (2,3-epoxypropoxy) indole-2-carboxylic acid cyclopropylamide. The title compound crystallizes from methanol in prisms of m.p. 23'8 to 2410.
The starting material is also obtained analogously to Example 1, starting from 4-benzyloxyindole-2-cabonic acid chloride: 4-benzyloxyindole-2-carboxylic acid anilide, melting point 199 to 2010; 4-hydroxyindole-2-carboxylic acid anilide, m.p. 203-2050.
