Verfahren zur Herstellung neuer Dibenzothiepinderivate
Die Erfindung betrifft ein Verfahren zur Herstellung neuer Dibenzothiepinderivate der Formel I, worin R für eine niedere Alkylgruppe steht, sowie ihrer Säureadditionssalze.
Erfindungsgemäss gelangt man zu den neuen Verbindungen der Formel I, und ihren Säureadditionssalzen, indem man Verbindungen der Formel II, worin R obige Bedeutung besitzt, in neutraler oder schwach saurer Lösung mit einem Alkali- oder Erdalkalisalz der Perjodsäure oder mit der stöchiometrischen Menge einer organischen Persäure oder mit der stöchiometrischen Menge Wasserstoffperoxid umsetzt und die erhaltenen Verbindungen der Formel I gewünschtenfalls in ihre Säureadditionssalze überführt.
Die Umsetzung wird vorzugsweise bei niederen Temperaturen, insbesondere zwischen 0 und 50 , und gegebenenfalls in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in Essigsäure oder Aceton durchgeführt. Nach Beendigung der Reaktion wird das Reaktionsprodukt nach bekannten Methoden isoliert und gegebenenfalls gereinigt.
Bei der vorliegenden Reaktion bleibt die sterische Konfiguration unverändert, so dass man bei Verwendung von 1 1-(8-Alkyl-3 a-nortropanyloxy)-6, 11 -dihydro- dibenzo[b,e]thiepin die entsprechenden 11-(8-Alkyl-3a nortropanyloxy)-6,11-dihydrodibenzo[b,e]thiepin-5- oxide erhält.
Die Verbindungen der Formel I können in ihre Säureadditionssalze überführt werden und umgekehrt.
Die durch R symbolisierte niedere Alkylgruppe besitzt vorzugsweise 1 bis 4 Kohlenstoffatome.
Die Verbindungen der Formel I und ihre pharmakologisch verträglichen Säureadditionssalze besitzen bei geringer Toxizität interessante pharmakodynamische Eigenschaften und können daher als Heilmittel verwendet werden. Insbesondere sind sie zur vorbeugenden und heilenden Behandlung von Magengeschwüren geeignet, wie sich durch die Ergebnisse des Acetylcholin- und Histamintoxizitätstests am Meerschweinchen, den Mydriaseeffekt an der Maus und die Hemmung der durch Phenylbutazon induzierten Ulcusbildung an der Ratte zeigt.
Die zu verwendenden Dosen variieren naturgemäss je nach der Art der Administration und des zu behandelnden Zustandes. Im allgemeinen werden jedoch bei Testtieren befriedigende Resultate mit einer Dosis von 0,05 bis 10 mg/kg Körpergewicht erhalten; diese Dosis kann nötigenfalls in 2 bis 3 Anteilen oder auch als Retardform verabreicht werden. Für grössere Säugetiere liegt die Tagesdosis bei etwa 2 bis 10 mg.
Für orale Applikationen enthalten die Teildosen etwa 0,3 bis 5 mg der Verbindungen der Formel I neben festen oder flüssigen Trägersubstanzen.
Als Heilmittel können die Verbindungen der Formel I bzw. ihre physiologisch verträglichen Säureadditionssalze allein oder in geeigneter Arzneiform mit pharmakologisch indifferenten Hilfsstoffen verabreicht werden.
Die Verbindungen der Formel II können hergestellt werden, indem man Verbindungen der Formel III, worin X für den Säurerest eines reaktionsfähigen Esters steht, mit Verbindungen der Formel IV, worin R obige Bedeutung besitzt, umsetzt. Das Verfahren kann beispielsweise folgendermassen ausgeführt werden:
Man lässt eine Lösung einer Verbindung der Formel III, worin X vorzugsweise für Chlor, Brom, Jod, Fluor oder den Rest einer organischen Sulfonsäure steht, in einem unter den Reaktionsbedingungen inerten Lösungsmittel, z. B. in einem aromatischen Kohlenwasserstoff wie abs. Toluol oder abs. Benzol bei erhöhter Temperatur, z.B. bei Siedetemperatur des Reaktionsgemisches, gegebenenfalls in Anwesenheit eines säurebindenden Mittels, z.
B. eines Alkalimetallkarbonates wie Natrium- oder Kaliumkarbonat, oder einer tertiären or ganischen Base, wie Triäthylamin, zu einer Lösung eines Nortropin- bzw. Pseudonortropinalkylderivates (Nortropan-3a-d-, bzw. Nortropan-3ss-ol-derivat) im gleichen Lösungsmittel zutropfen. Nach Beendigung der Reaktion wird das Reaktionsprodukt nach bekannten Methoden isoliert und gegebenenfalls gereinigt.
Die Verbindungen der Formel III können hergestellt werden, indem man die Verbindung der Formel V auf an sich bekannte Weise verestert, z. B. indem man a) zur Herstellung von Verbindungen der Formel IIIa, worin XI für den Rest einer organischen Sulfonsäure steht, die Verbindung der Formel V mit einem organischen Sulfonsäurehalogenid in einem unter den Reaktionsbedingungen inerten Lösungsmittel umsetzt, oder b) zur Herstellung von Verbindungen der Formel IIIb, worin XlI für -Chlor, Brom oder Jod steht, in die Lösung der Verbindung der Formel V in einem unter den Reaktionsbedingungen inerten Lösungsmittel Chlor-, Brom- oder Jodwasserstoff einleitet, oder c) zur Herstellung der Verbindung der Formel IIIc Verbindungen der Formel IIId, worin XIIt für Brom oder Chlor steht, mit Metalllluoriden behandelt,
oder d) zur Herstellung von Verbindungen der Formel IIIb die Verbindung der Formel V mit Thionyl- oder Phosphorhalogeniden umsetzt.
Bei Verfahren a) wird als unter den Reaktionsbe dingungen inertes Lösungsmittel vorzugsweise ein organisches basisches Lösungsmittel verwendet, z. B. Pyridin oder ein niederes Trialkylamin, wie Triäthylamin, oder die Reaktion wird in einem unter den Reaktionsbedingungen inerten organischen Lösungsmittel unter Zusatz von Pyridin oder eines niederen Trialkylamins ausgeführt.
Als Lösungsmittel in Verfahren b) kann z. B. ein wasserfreier aromatischer Kohlenwasserstoff wie Benzol eingesetzt werden. Das 11-Jod-Derivat kann auch aus dem 11-Brom-Derivat durch Umsetzen mit Natriumjodid in Aceton hergestellt werden.
In Verfahren d) kann als Thionylhalogenid z. B.
Thionylchlorid oder -bromid, als Phosphorhalogenid z. B. Phosphortrichlorid -tribromid oder -pentachlorid eingesetzt werden.
Soweit die Herstellung der Ausgangsverbindungen nicht beschrieben wird, sind diese bekannt oder nach an sich bekannten Verfahren bzw. analog zu den hier beschriebenen oder analog zu an sich bekannten Verfahren herstellbar.
In den nachfolgenden Beispielen, die die Erfindung näher erläutern, ihren Umfang aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden und sind unkorrigiert.
Beispiel 1
6,1 1-Dihydro-1 1-(3a-tropanyloxy) dibenzo [b,e]thiepin-5-oxid
Zu einer Lösung von 10,5 g 6,11-Dihydro-11-(3a tropanyloxy)dibenzo[b,e]thiepin in 45 ml Eisessig gibt man 2,8 ml 400/oige Wasserstoffperoxidlösung und rührt das Gemisch 17 Stunden bei Raumtemperatur. Darauf wird die Reaktionslösung mit 70 ml Wasser verdünnt, abgekühlt und mit 250/oiger Ammoniaklösung alkalisch gestellt. Die wässerige Phase wird dreimal mit je 100 ml Äther ausgeschüttelt, die vereinigten Atherextrakte- über Magnesiumsulfat getrocknet und das Lösungsmittel bei vermindertem Druck vollständig eingeengt. Der ölige Rückstand, die im Titel genannte Verbindung, wird sodann mit Fumarsäure in Äthanol in das Hydrogenfumarat übergeführt.
Nach Umkristallisieren aus Methanol schmilzt die Verbindung bei 125127O.
Das als Ausgangsprodukt verwendete 6,11 -Dihydro- 11-(3a-tropanyloxy)dibenzo[b,e]thiepin kann folgendermassen hergestellt werden:
14,0 g 6,1 1-Dihydrodibenzo [b,e] thiepin-1 1-ol werden in 40 ml abs. Xylol mit 7,3 g Thionylchlorid versetzt und 1 Stunde auf 70" erhitzt. Nach Abkühlen auf Zimmertemperatur werden 3,0 g Calciumchlorid zugegeben und die Xylolphase unter Feuchtigkeitsausschluss filtriert. Man lässt sie anschliessend innerhalb von 15 Minuten in ein siedendes Gemisch von 8,6 g Tropen (Tropan-3a-ol) und 24,6 g Natriumkarbonat in 50 ml abs. Xylol zutropfen.
Das Reaktionsgemisch wird während 6 Stunden am Rückfluss erhitzt und nach Abkühlen mit 200 ml Diäthyläther verdünnt und zweimal mit je 100 ml Wasser ausgeschüttelt. Die organische Phase wird mit 100 ml 1 N Salzsäure extrahiert, der Salzsäureauszug mit 2 N Natronlauge alkalisch gestellt und dreimal mit je 300 ml Diäthyläther ausgeschüttelt.
Nach Trocknen des Atherauszuges über Natriumsulfat wird eingeengt und der ölige Rückstand destilliert, wobei die im Titel genannte Verbindung bei 190-2000 und 0,02 Torr übergeht.
Beispiel 2
11-(8-Athyl- 3 a-nortropanyloxy)-6,1 1- dihydro-dibenzo [b,e]thiepin-5-oxid
Wird analog wie in Beispiel 1 beschrieben erhalten.
Smp. des Hydrogenfumarats 1340 (Zers.).
Das als Ausgangsprodukt verwendete 11-(8-Äthyl- 3 a-nortropanyloxy)-6, 11 -dihydrodibenzo [b, e] thiepin kann folgendermassen hergestellt werden:
Zu 22,7 g 6,1 1-Dihydrodibenzo [b,e] thlepin-1 1-ol in 50 ml abs. Benzol leitet man unter Eiskühlung während 15 Minuten Chlorwasserstoff ein. Anschliessend wird die Lösung vollständig eingeengt, der Rückstand in 50 ml abs. Benzol gelöst, über Magnesiumsulfat getrock net und nochmals vollständig eingeengt. Der Rückstand wird sodann in.80 ml abs. Xylol gelöst und diese Lösung zu einer siedenden Lösung von 15,5 g N-Athylnortropin in 50 ml abs. Xylol innerhalb 15 Minuten zugetropft.
Das Reaktionsgemisch wird sodann 1 Stunde am Rückfluss zum Sieden erhitzt, dann auf Raumtemperatur abgekühlt, mit 100 ml Diäthyläther verdünnt und zweimal mit je 50 ml 2 N Salzsäure ausgeschüttelt. Der wässerige Auszug wird mit 2 N Natronlauge alkalisch gestellt, dreimal mit je 100 ml Äther ausgeschüttelt, die vereinigten Ätherextrakte über Magnesiumsulfat getrocknet, mit Tierkohle gereinigt und das Lösungsmittel vollständig abdestilliert. Der ölige Rückstand wird in das Methansulfonat übergeführt, welches nach Umkristallisieren aus Äthanol bei 198-200" schmilzt.

Process for the preparation of new dibenzothiepine derivatives
The invention relates to a process for the preparation of new dibenzothiepine derivatives of the formula I, in which R is a lower alkyl group, and their acid addition salts.
According to the invention, the new compounds of the formula I and their acid addition salts are obtained by mixing compounds of the formula II, in which R has the above meaning, in neutral or slightly acidic solution with an alkali or alkaline earth metal salt of periodic acid or with the stoichiometric amount of an organic peracid or reacts with the stoichiometric amount of hydrogen peroxide and, if desired, converts the resulting compounds of the formula I into their acid addition salts.
The reaction is preferably carried out at low temperatures, in particular between 0 and 50, and optionally in a solvent which is inert under the reaction conditions, e.g. B. carried out in acetic acid or acetone. After the reaction has ended, the reaction product is isolated and, if necessary, purified by known methods.
In the present reaction, the steric configuration remains unchanged, so that when 1 1- (8-alkyl-3 a-nortropanyloxy) -6, 11 -dihydrodibenzo [b, e] thiepin is used, the corresponding 11- (8- Alkyl-3a (nortropanyloxy) -6,11-dihydrodibenzo [b, e] thiepin-5-oxide.
The compounds of the formula I can be converted into their acid addition salts and vice versa.
The lower alkyl group symbolized by R preferably has 1 to 4 carbon atoms.
The compounds of the formula I and their pharmacologically acceptable acid addition salts have interesting pharmacodynamic properties with low toxicity and can therefore be used as medicaments. In particular, they are suitable for the preventive and curative treatment of gastric ulcers, as shown by the results of the acetylcholine and histamine toxicity tests on guinea pigs, the mydriase effect on mice and the inhibition of the ulcer formation induced by phenylbutazone on rats.
The doses to be used naturally vary depending on the type of administration and the condition to be treated. In general, however, satisfactory results are obtained in test animals at a dose of 0.05 to 10 mg / kg body weight; if necessary, this dose can be administered in 2 to 3 portions or as a sustained-release form. For larger mammals, the daily dose is around 2 to 10 mg.
For oral administration, the partial doses contain about 0.3 to 5 mg of the compounds of the formula I in addition to solid or liquid carrier substances.
The compounds of the formula I or their physiologically tolerable acid addition salts can be administered as medicaments alone or in a suitable medicinal form with pharmacologically inert auxiliaries.
The compounds of the formula II can be prepared by reacting compounds of the formula III in which X is the acid radical of a reactive ester with compounds of the formula IV in which R has the above meaning. The method can be carried out as follows, for example:
A solution of a compound of the formula III, in which X is preferably chlorine, bromine, iodine, fluorine or the remainder of an organic sulfonic acid, in a solvent which is inert under the reaction conditions, e.g. B. in an aromatic hydrocarbon such as abs. Toluene or abs. Benzene at elevated temperature, e.g. at the boiling point of the reaction mixture, optionally in the presence of an acid-binding agent, e.g.
B. an alkali metal carbonate such as sodium or potassium carbonate, or a tertiary or ganic base such as triethylamine, to a solution of a nortropine or pseudonortropine alkyl derivative (nortropan-3a-d-, or nortropan-3ss-ol derivative) in the same solvent drop. After the reaction has ended, the reaction product is isolated and, if necessary, purified by known methods.
The compounds of formula III can be prepared by esterifying the compound of formula V in a manner known per se, e.g. B. by a) for the preparation of compounds of the formula IIIa, wherein XI is the residue of an organic sulfonic acid, reacting the compound of the formula V with an organic sulfonic acid halide in a solvent inert under the reaction conditions, or b) for the preparation of compounds of Formula IIIb, wherein XlI is -chlorine, bromine or iodine, introduces chlorine, bromine or hydrogen iodide into the solution of the compound of formula V in a solvent inert under the reaction conditions, or c) for the preparation of the compound of formula IIIc compounds of Formula IIId, in which XIIt is bromine or chlorine, treated with metal fluorides,
or d) for the preparation of compounds of the formula IIIb, the compound of the formula V is reacted with thionyl or phosphorus halides.
In process a), an organic basic solvent is preferably used as the inert solvent under the reaction conditions, eg. B. pyridine or a lower trialkylamine such as triethylamine, or the reaction is carried out in an inert organic solvent under the reaction conditions with the addition of pyridine or a lower trialkylamine.
As a solvent in process b), for. B. an anhydrous aromatic hydrocarbon such as benzene can be used. The 11-iodine derivative can also be prepared from the 11-bromo derivative by reacting it with sodium iodide in acetone.
In process d), as thionyl halide, for. B.
Thionyl chloride or bromide, as a phosphorus halide z. B. phosphorus tribromide or pentachloride can be used.
If the preparation of the starting compounds is not described, they are known or can be prepared by processes known per se or analogously to those described here or analogously to processes known per se.
In the following examples, which explain the invention in more detail but are not intended to restrict its scope in any way, all temperatures are given in degrees Celsius and are uncorrected.
example 1
6.1 1-Dihydro-1 1- (3a-tropanyloxy) dibenzo [b, e] thiepin-5-oxide
2.8 ml of 400% hydrogen peroxide solution are added to a solution of 10.5 g of 6,11-dihydro-11- (3a tropanyloxy) dibenzo [b, e] thiepin in 45 ml of glacial acetic acid, and the mixture is stirred for 17 hours at room temperature. The reaction solution is then diluted with 70 ml of water, cooled and made alkaline with 250% ammonia solution. The aqueous phase is extracted three times with 100 ml of ether each time, the combined ether extracts are dried over magnesium sulfate and the solvent is completely concentrated under reduced pressure. The oily residue, the compound named in the title, is then converted into the hydrogen fumarate using fumaric acid in ethanol.
After recrystallization from methanol, the compound melts at 125127O.
The 6,11-dihydro-11- (3a-tropanyloxy) dibenzo [b, e] thiepin used as the starting product can be prepared as follows:
14.0 g of 6,1 1-dihydrodibenzo [b, e] thiepin-1 1-ol are dissolved in 40 ml of abs. 7.3 g of thionyl chloride are added to xylene and the mixture is heated to 70 "for 1 hour. After cooling to room temperature, 3.0 g of calcium chloride are added and the xylene phase is filtered with exclusion of moisture. It is then left in a boiling mixture of 8.6 within 15 minutes Add dropwise g of tropics (tropan-3a-ol) and 24.6 g of sodium carbonate in 50 ml of absolute xylene.
The reaction mixture is refluxed for 6 hours and, after cooling, diluted with 200 ml of diethyl ether and extracted twice with 100 ml of water each time. The organic phase is extracted with 100 ml of 1N hydrochloric acid, the hydrochloric acid extract is made alkaline with 2N sodium hydroxide solution and extracted three times with 300 ml of diethyl ether each time.
After drying the ether extract over sodium sulfate, it is concentrated and the oily residue is distilled, the compound mentioned in the title passing over at 190-2000 and 0.02 torr.
Example 2
11- (8-Ethyl-3 a -nortropanyloxy) -6.1 1- dihydro-dibenzo [b, e] thiepin-5-oxide
Is obtained analogously to that described in Example 1.

The reaction mixture is then refluxed for 1 hour, then cooled to room temperature, diluted with 100 ml of diethyl ether and extracted twice with 50 ml of 2N hydrochloric acid each time. The aqueous extract is made alkaline with 2N sodium hydroxide solution, extracted three times with 100 ml of ether each time, the combined ether extracts are dried over magnesium sulfate, cleaned with animal charcoal and the solvent is completely distilled off. The oily residue is converted into the methanesulfonate, which, after recrystallization from ethanol, melts at 198-200 ".
