Verfahren zur Herstellung von Ferrichrysin und Desferrychrysin Aus Materialien biologischen Ursprungs, insbe- sondere Mikroorganismen, ist bereits eine grössere Anzahl eisenhaltiger Wuchsstoffe isoliert worden, beispielsweise die Ferrichrome, das Coprogen,
die Ferrioxamine.
Es wurde nun gefunden, dass man aus einem neuen Stamm von Aspergillus melleus, nämlich Aspergillus melleus M 2853, einen neuen eisen- haltigen Wuchsstoff, das Ferrichryshn, und die ent sprechende eisenfreie Verbindung, das Desferrichry- sin, gewinnen kann.
Gegenstand des vorliegenden Patents ist daher ein Verfahren zur gleichzeitigen Herstellung von Fenichrysin und Desferrichrysin.
Der Stamm Aspergillus mellvus M 2853 wurde in Zürich aus der Luft isoliert. Er wird in unseren Laboratorien und in der Eidgenössischen Technischen Hochschule, Institut für spezielle Botanik, Zürich,
unter der angegebenen Bezeichnung aufbewahrt. Bei der Züchtung auf Malzextrakt-Agar zeichnet sich der Stamm aus durch ein vegetatives Mycel aus sep- tierten und verzweigten Hyphen, aus deren Fusszellen Konidienträger in Form von rundlich angeschwol lenen Köpfchen herauswachsen.
Die Köpfchen tragen in dichter Anordnung fertile Zellen, Sterigmen, aus denen in unverzweigten Ketten die Konidiien durch Querwandbildung abgeschnürt werden.
Dadurch ist der Stamm eindeutig als zur Gattung Aspergillus Micheli 1729 (vgl. Thom et Church, The Aspergilli , Baltimore 1926) gehörend' gekennzeichnet. Die wei tere Klassifizierung erfolgte nach Thom and Raper, A Manual of the Aspergilli , Baltimore, 1945.
Die Feststellung der Gruppenzugehörigkeit kann ent weder anhand' von Farbmerkmalen oder anhand morphologischer Kriterien vorgenommen werden. Beide Wege ergeben die Zugehörigkeit von Stamm M 2853 zur Aspergillus ochraceus-Gruppe. Innerhalb der A.
ochraceus-Gruppe erfolgt die Zuordnung vor allem auf Grund der Farbe des sporulierenden Mycels, der Ausbildung der Sklerotien und der Konidien- form. Durch die gelbliche Farbe der Konidien- köpfchen, das spärliche Auftreten von Sklerotien und die glatten,
rundlichen Konidhen ist der Stamm M 2853 all Aspergillus melleus charakterisiert.
Ferrichrysin ist eine rotbraune, kristalline Sub stanz, die in Wasser und Methanol, auch in der Kälte, gut löslich und schwer löslich ist in absolutem Äthanol. Die Verbindung hat die: Elementarzusam mensetzung C = 43,37 %; H = 6,08 %; N = 16,72 %; Fe = 7,09 %; O = 26,74 % (her.). Das UV-Spektrum in Wasser zeigt ein Maximum bei. 415 mu (10 g E i @m gleich 1,48).
Das IR-Spcktrum in Kaliumbromid weist u. a. Banden auf bei 725, 783, 975, 1060, 1140, 1200 (Schulter), 1218 (Schulter), 1237, 1275 (Schulter), 1340 (Schulter), 1368, 1430 (Schulter), 1445 (Schulter), 1465, 1495 (Schulter), 1517, 1585, 1652, 2930, 3070 (Schulter). 3420 cm-1, vgl. Fig. 1.
Die Kristalle zersetzen sich bei etwa 270 C unter Schwarzfärbung, ohne zu schmelzen. Im Papier- chromatogramm werden folgende Rf-Werte erhalten: 0,26 im System I (n-Butanol-EisessigWasser, 4 : 1 : 1) und 0,34 hm System II (tart.-Butanol-0,004-n. Salz- säure-gesättigte wässrige Natriumchloridlösung 2:1:1;
Papier imprägniert mit Aceton-Wasser-ge- sättigte wässrige Natriumchlbridlösung 6:3: 1).
Bei der sauren Hydrolyse des Ferrnchrysins ent stehen 3 Mol Essigsäure; Bernsteinsäure: und 1 Amino-5-hydroxyl-amino-pentan, die wesentliche Be= standheile der Ferrioxamine darstellen, sind im Hy- drolysegemisch des Ferrichrysins nicht nachweisbar.
Bei der Behandlung des Ferrichrysins mit Basen oder Säuren oder mit Eisenkomplex bildenden Stof fen, wie 8-Hydroxy-chinolin, wird das Eisen aus dem Mölekül entfernt, wobei ein farbloses Pulver, das Desfernehrysin, gebildet wird.
Es ist gut löslich in Wasser, Methanol, Äthanol, schwerer in Butanol, fast unlöslich in Aceton, Chloroform und llipophiIen Lösungsmitteln. Die Elementaranalyse ergibt folgende Werte: C 46,36 %; H 6,98 %; N 15,65 %; O 31,01 (bei.), was einer Summenformel C29H49N9014 ent spricht.
Das UV-Spektrum ist nicht besonders cha- rakteristisch und zeigt lediglich Endabsorption bei 214 mu. Im IR-Spektrum (aufgenommen in Kaläum- bromid.)sind Banden u. a.
bei 2,94 l#"3,43 ,u, 6,10,u, 6,537,04,u, <I>7,32</I> ,u, 8,10 u, 8,30,u, 8,62 ,u, 9,60,u und 10,14 ,
u erkennbar. Wenn man Desfer- richrysin mit 6n Salzsäure hydrolysiert und die Hydrolyseprodukte (zwecks Reduktion gegebenen falls vorhandener Hydroxylaminogruppen) katalytisch hydriert, erhält man drei ninhyd\nin-positive Pro- dukte, die mit Phenol-Wasser (4:
1) als Fliessmittel im Papierchromatogramm folgende Rf Werte auf weisen: 0,21; 0,30 und 0,38. Die Untersuchung nach der Methode von Moore und Stein zeigt, dass es sich um Ornithin, Serin und Glycin im molekularen Ver- hältnis <B>3:</B> 2: 1 handelt. Wird das saure Hydrolyse- prod'ukt ohne, nachfolgende katalytische Hydrierung direkt untersucht, so ist kein Ornithin nachweisbar.
Im Ferrichrysin sind allem nach die Amino- säuren Glycin (1 Mol), Serin (2 Mol) und d-Hydroxy- ornithin (3 Mol) zu einem cyclischen Peptid ver bunden.
Das Hydroxyornithin äst N(b)-aoetyüert; an die so gebildeten 3 Hydroxamsäurereste ist das Eisen komplex gebunden. Die Gesarntformel des eisenfreien Ferri'chrysüns ist demnach Tri-(N acetyl)-cyclo-[gly- cyl-(seryll)2-(d-hydroxyornit@hyl)3].
Aus Desferrichryssn wird bei Zusatz von Eisen- chloiid bei neutralem pH das Ferrichrysin zurück- erhalten.
Ferrichrysin ist ein Wuchsstoff für verschiedene Mikroorganismen und kann daher gegebenenfalls bei der Kultur solcher Organismen verwendet werden.
Die folgende Tabelle zeigt das relative Wachstum von Mikrobaeterium lacticum Stamm ATCC 8181, gemessen an der Extinktion der Kultur, bei Zusatz verschiedener Mengen von Ferrichrysin zum Kultur medium:

Auch Desferrichrysin hat wertvolle pharmakologische Eigenschaften. So verhindert es die Ablagerung eisen- haltigar Pigmente im Gewebe bzw. bewirkt in Fällen von pathologischer Eisenablagerung im Organismus eine Ausscheidung des Eisens, z.
B. bei Hämochro- matose und Hämosiderose. Es kann. dementsprechend in der Human- und Veterinär-Heilkunde verwendet werden.
Wie andere Sideramine ist auch Ferrichrysin imstande, die antibakterielle Wirkung von Anti biotika, die der Gruppe der Sideromycine angehören, gegenüber grampositiven Erregern kompetitiv auf zuheben (Antisideromycin-Wirkung).
Hinsichtlich der genannten biologischen Wirkun gen zeigt das Ferrichrysin gewisse Ähnlichkeiten mit anderen Sideraminen, wie z.
B. den Ferrioxaminen, den Ferrichromen und: dem Copro.gen. Hingegen unterscheidet es sich in seinen physikalisch-chemi- schen Eigenschaften von dieser Substanz in, charak teristischer Weise, was im folgenden illustriert wird.
1. Von den Ferrioxaminen, die von Actinomy- e ztenstämmen produziert werden, unterscheidet sich das Ferrichrysin nicht nur an den Hydrolyseproduk- ten, wie oben erwähnt,
sondern auch in seinen spek- troskopischen Eigenschaften. So ist sein IR-Spektrum von dem der Ferrioxamine in wesentlichen Punkten verschieden und das Maximum des UV-Spektrums liegt beim Ferrichrysin kurzwelliger (etwa 415 bis 420 mu) als beiden Ferrioxaminen (etwa 430 mrc).
2. Feerichrom, das durch Züchtung von Usti1!ago sphaerogena hergestellt werden kann, unterscheidet sich spektroskopisch nur wenig vom Ferrichrysin. Im. IR-Spektrum zeigt Ferrichrom ein mittelstarkes Maximum bei 1275 cm-1, das beim Ferrichrysin nur als schwache Schulter erscheint.
Anstelle einer breiten Bande bei 1060 cm-1 beim Ferrichrysin sind bei Fernchrom zwei gut getrennte Maxima bei 1025 und 1068 cm-1 vorhanden.
Im NMR-Spektrum weist das. Desferrichrysin ein breites Maximum bei 4,00 ppm und eine breite Anhäufung von Banden im Bereich von 4,2-5,1 ppm mit Maxima bei 4,33; 4,83 und 5,00 ppm auf, während das. eisenfreie Ferrichrom in diesem Gebiet Maxima bei 4,33; 4,52 (Schulter) und 4,74 ppm zeigt.
In der Lös lichkeit unterscheiden sich Ferrichrysin und Ferri- chrom hingegen deutlich: Ferrichrom ist im Gegen satz zum Ferrichrysin in kaltem Methanol fast un- RTI ID="0002.0228" WI="11" HE="4"LX="1130" LY="2514"> 4bsläch, es löst sich erst beim längeren Kochen in der etwa 1000fachen Menge Methanol.
Auch die Hydrolyseprodukte sind verschieden. Eisenfreies Ferrichrom, mit 6 n Salzsäure hydrolysiert und katalytisch hydriert, liefert nicht drei, sondern nur zwei ninhyd:rin-positive Produkte, nämlich Glycin und Ornithin. Auch im Papierchromatogramm sind Unterschiede vorhanden: Fernichrom hat in den Systemen I und II die Rf-Werte 0,19 bzw. 0,25.
3. Fernchrom A, ein Begleiter des Ferrichroms, weist im Gegensatz zu Ferrichrysin keine nennens werte antagonistisch-, Wirkung gegenüber Ferrimycin auf. Bei der Hydrolyse liefert es nicht 3 Mol Essig- säure, sondern 3 Mal f-Methyl-gl'utaconsäure. Im Papierchromatogramm in den Systemen I und II zeigt Ferrichrom A R f-Werte von 0,12 bzw.
0,18, und wandert demnach etwa halb so weit wie Ferri- chrysin. Schliesslich verhält sich Ferrichrom A bei der Craig-Verteilung anders als Ferrichrysin: Im System Benzylalkohol-n-Butanol-gesättigte wüssrige Natriumchlorid'lö,sung-0,001-n wässrige Saltzsäure 9 :9 :<B><I>5:
</I></B> 15 über 95 Stufen reichert es sich in den Stufen 3-15 an, während Ferrichrysin in den Frak tionen 36-60 erscheint.
4. Von Coprogen sind weder das IR-Spektrum noch Hydrolyseprodukte bekannt. Die Elementar- zusammensetzung ist jedoch deutlich von derjenigen des Ferrichrysäns verschieden; für Coprogen ist an gegeben: C = 50,9 %; H = 6,9 %; N = 10,2 %; Fe = 6,7 %. Coprogen zersetzt sich bereits bei 205 C. Im Papierchromatogramm zeigt Coprogen die Rf-Werte 0,43 (System 1) und 0,55 (System 1I).
Das Verfahren zur gleichzeitigen Herstellung von Ferrichrysin und Desferrächrysän ist dadurch gekenn zeichnet, d'ass man einen die Eigenschaften von Asper- gillus melleus M 2853 aufweisenden Aspergillus- Stamm züchtet. Die Züchtung erfolgt z.
B. in wäss riger, eine Kohlenstoffquelle, stickstoffhaltige Verbin dungen sowie anorganische Salze enthaltender Nähr lösung, aerob, also beispielsweise in ruhender Ober flächenkultur, oder vorzugsweise submers unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den bekannten Fermentern. Als Temperatur eignet sich eine solche zwischen 18 und 40 C, vorzugsweise 27 C.
Eine wesentliche Sider- amin-Wärkung zeigt die Nährlösung dabei im all gemeinen nach 4-8 Tagen.
Die Nährlösung enthält als Kohlenstoffquelle z. B. Kohlenhydrate, wie Glukose, Saccharose, Lak- tose, Stärke, Alkohole, wie Mannit oder Glycerin. A% stickstoffhaltige Nährstoffe seien genannt Amino- säuren, z.
B. Ornithin, Peptide, Proteine und deren Abbauprodukte, wie Pepton oder Trypton, Fleisch extrakte, wasserlösliche Anteile von Getreidekörnern, wie Mais oder Weizen, Destillationsrückstände der Alkoholherstellung, Hefe, Samen, insbesondere der Raps- und Sojapflanze, der Baumwollpflanze, Am- moniumsalze und Nitrate.
Von anderen anorgani schen Salzen kann die Nährlösung beispielsweise Chloride, Carbonate, Sulfate, Nitrate von Alkalien, Erdalkalien, Magnesium, Zink oder Mangan sowie Spuren von Eisensalzen ( < 10-7 MolVLiter) enthalten.
Die Ferrichrysin-Aktivität kann mikrobiologisch rm ttdls des abgewandelten Bonifas-Testes (vgl. Zähner et a1., Arch. Mikrobiol. <I>36,</I> 325 ff [1960]) bestimmt werden.
Als Testlösung verwendet man .eine Ferri- mycinlösung von 0,01 mg/ml Gehalt, als Teststamm Staphylococcus aureus.
Man kann auch optisch die Ferrichrysin-Konzen- tration der Kulturlösung feststellen. Zu diesem Zweck werden 5 ml Kulturflüssigkeit mit 1,5g Natrium- chlorid, 1 ml 0,1 % iger Ferrvsulfatlösung und 5 ml Benzylalkohol 5 Minuten geschüttelt, abzentrifugiert,
die organische Phase filtriert und die Extinktion der organischen Phase bei 410 m,u bestimmt.
Als Vergleichsprobe dient ein auf gleiche Weise hergestekter Extrakt aus unbeimpfter Nährlösung. Nach 4-8 Tagen erreichen die Kulturen eine Aktivität, die 300-400 mg reinem Ferrichrysin ent spricht.
Zur Kultur gibt man nach Beendigung der Züch tung pro Liter 1 g Ferrichlorid, wenn Ferrichrysin isoliert werden soll, nicht aber wenn das eisenfreie Desferrichrysin gewonnen werden, soll. Man trennt das Mycel vom Kulturfiltrat ab; wonach die Haupt menge des Sideramins im Kulturfiltrat gefunden wird. Es bleiben aber trotzdem namhafte Mengen davon am Mycell haften. Es ist daher vorteilhaft, dieses gut auszuwaschen.
Dazu eignen sich beispielsweise Was ser oder wässrige Alkohole, wie wässriges Methanol.
Für die Isolierung von Ferrichrysin aus dem Kulturfiltrat kann man nach an sich bekannten Me thoden vorgehen und z. B. eine der nachfolgend' an gegebenen Arbeitsweisen oder Kombinationen davon anwenden.
1. Man kann Adsorption@smättel verwenden, z. B. Aktivkohlen, wie Norit , aktivierte Erden, wie Frankonit , Fullererde oder Floridin , oder Harz adsorber, wie Asmit . Die Elution der Adsorbate erfolgt zweckmässig mit Gemischen von mit Wasser mischbaren organischen Lösungsmitteln m'it Wasser, z.
B. mit Wasser-Methanol-, Wasser-Pyridin-, ver dünnte Essigsäure-Methanol- oder Wasser-Methanol- Eisessig-Butanol-Gemischen. Als besonders vorteil haft für die El'ution eines Frankonit - oder Norit - Adsorbates hat sich das Gemisch von Wasser (4 Vo- llumteile) und Pyridin (1 Volumteil erwiesen.
2. Weiter kann das Ferrichrysin einer wässrigen Lösung mittels organischen Lösungsmitteln entzogen werden. Für diese Extraktionsverfahren haben sich höhere organische Alkohole, z. B. Benzylalkohol oder Isopropylalkohol, besonders bewährt. Zweckmässiger weise wird dabei der wässrigen Phase ein anorgani sches Salz, z. B. Ammoniumsulfat oder Natrium- chlofüd, zugesetzt.
Aus den erhaltenen organischen Extrakten kann das Ferrichrysin entweder durch RTI ID="0003.0219" WI="18" HE="4" LX="1058" LY="2364"> Abdampfen des- Lösungsmittels oder durch Ausfällung mittels eines geeigneten organischen Lösungsmittels, z. B. Äther, Petraläther oder Äthylacetat, in ange- reicherter Form erhalten werden.
3. Eine weitere Methode der Anreicherung des Ferrichrysins besteht darin, dass man es zwischen einer wässrigen Lösung und einer Lösung von Phenol in Chloroform verteilt, wobei der Phenolgehalt der Chloroformlösung variiert werden kann.
4. Eine andere Methode zur Anreicherung und/ oder Auftrennung des Ferrichrysins stellt die Chro- matographie dar, wie Adsorptionschromatographie an verschiedenen Materialien, z.
B. an Norit , Alu miniumoxyd, Magnesiumsilikaten, Silncageln>, Cal ciumsulfat sowie Verteilungschromatographie mit Cellulose, Stärke, Silicagel , Celite und der gleichen als Trägersubstanzen, oder aber Chromato- graphie an Ionenaustauscherharzen, z. B. an Dowex- 50 , Amberhte IRC-50 und dergleichen.
5. Weiter kann Ferrichrysin durch Gegenstrom- verteilung nach Craig zwischen zwei nicht mischbaren Lösungsmittelphasen angereichert werden. Folgendes Lösungsmittelsystem hat sich dabei, besonders be währt: n-Butanol-Benzylalkohol-0,001-n. Salzsäure- wässrige, bei 19 gesättigte Natriumchlorndlösung (9:9:15:5).
Desferrichrysin wird neben Ferrichrysin von Aspergillus melTeüs M 2853 gebin!det und\ kann daher auch direkt aus dem Kulturfiltrat isoliert werden. Vorteilhaft entfernt man bei der Isolierung im Kultur- filtrat vorhandenes Eisen, beispielsweise durch Zu satz Eisenkomplex bildender Stoffe, z.
B. 8-Oxy- chinoa oder Äthylendiamin tetraessigsäure.
Man kann die Eisenkomplex bildenden Stoffe direkt nach Beendigung der Fermentation der Kultur- brühe zusetzen. Vorteilhaft erfolgt der Zusatz in einem späteren Stadium der Aufarbeitung, um all- fällige durch die benützten Agenzien eingeschleppte Eisen (III)-ionen ebenfalls zu entfernen.
Die Isolierung des Desferrichrysins aus der Kul turlösung erfolgt z. B. nach den für die Isolierung von Ferrichrysin erwähnten Methoden.
Die Temperaturen sind in den folgenden Bei spielen in. Celsiusgraden angegeben. <I>Beispiel 1</I> Der Stamm Aspergillus melleus M 2853 wird in gut belüfteter Submerskultur bei 27 gezüchtet. Man verwendet eine Nährlösung, die pro Liter Leitungs wasser 20 g Glucose, 5 g L-Asparagiu, 1 g sek.
Kalumphosphat, 1 g krist. Magnesiumphosphat (M9S04; 7H20) und 0,5 g Calciumchlbrid enthält. Der Gehalt des Leitungswassers an Eisen beträgt 20-30 y/Liter. Die Nährlösung wird 20 Minuten bei 120 sterilisiert. Geimpft wird mit einer Sporen suspension, die 150-180 Millionen Sporen pro Liter enthält.
Nach 4-8 Tagen wird den Kulturen 1 g Ferri- chlorid pro Liter zugesetzt und unter Zusatz von 2 Celit fifriert. Dem Kulturfiltrat werden nun pro Liter 300g Natriumchlomd zugefügt und die bare Lösung dreimal mit 1/10 Volumen Benzylalkohol extrahiert. Die organische Phase wird mit Natrium sulfat getrocknet und mit 3 Volumen Äther versetzt.
Das Benzylalkohol-Äthergemisch extrahiert man mit einer klicinen Menge destilliertem Wasser, bis die wässrige Phase nur noch bloss rot gefäbt ist. Der wässrige Auszug wird lyophilisiert, wobei ein rotes Pulver anfällt, in einer Ausbeute von 300-4.00 mg/ Liter, enthaltend<B>80%</B> der Aktivität des Ausgangs- materials.
18,9 g rohes Ferrichrysin-Gemisch werden einer Craig Verteilung über 95 Stufen unterzogen, wobei die Substanz in die ersten 7 Gläser einer automati schen Verteilungsapparatur (Glasinhalt 25 ml pro Phase) eingefüllt wird.
Als Verteilungs-System dient ein Gemisch aus Benzylalkohol, n-Butylalkohal, ge sättigte wässriger Natriumchloridlösung und 0,001 n wässrige Salzsäure im Volumenverhältnis 9 : 9 : 5 : 15.
Der rotbraune Farbstoff wird im wesentlichen in eine Haupt- und -eine Nebenkomponente aufgetrennt, die in den Fraktionen 46 bzw. 75 ihre Verteilungs maxima haben. Dies entspricht ungefähren Ver teilungskoeffizienten von K1 = 0,9 bzw. K2 = 3,8.
Die Fraktionen 36-60 werden zusammengefasst, mit dem doppelten Volumen Äther versetzt und die wässrvge Phase abgetrennt. Die organische Schicht wird noch dreimal mit etwas Wasser ausgeschüttelt, die vereinigten wässrigen Auszüge mit Natrium.- chlorid etwa halb gesättigt und mehrmals mit Phenol- Chloroform (1 kg Phenol auf 1 Liter Chloroform) ausgeschüttelt.
Die Extrakte werden mit halbgesättig- ter Natriumchloridlösung gewaschen. Die zunächst trübe Lösung wird an einer kurzen Celit -Säule geklärt, dann mit dem doppelfiten Volumen Äther verdünnt und mehrmals mit wenig Wasser ausgeschüt telt, wobei der braunrote Farbstoff wieder in die wässrige Phase geht.
Die mehrmals mit Äther gewaschenen wässrigen Auszüge werden im Vakuum eingedampft und der Rückstand getrocknet. Man erhält 8,2 g gereinigtes Ferrichrysin. Für die Kristallisation werden 16 g eines solchen Eindampfrückstandes in etwa 200 ml heissem absolutem Äthylalkohol (über Natrium de stilliert) gelöst.
Die Kristallisation setzt rasch ein, und es fallen 11,6 g kristallines Ferrichrysin in Form rot brauner Stäbchen aus. Die Mutterlaugen werden zur Trockene eingedampft und wieder in absolutem Alkohol gelöst. Man erhält so nochmals 0,9g Kri stalle.
Zur Analyse wird eine Probe nochmals aus absolutem Alkohl umkristallisiert. Die rotbraunen Stäbchen werden bei 270 schwarz ohne zu schmelzen.
Analyse: Gef. C 43,37, H 6,08, Fe 7,09, N<B>16,72.</B> Absorptionsspektrum in Wasser: Am"" = 415 mu; 10 g Ei öm = 1,48.
Wenn der Alkohol für die Kristallisation nicht völlig trocken ist, fallen teils amorphe, teils in unregel mässigen Brocken kristallisierende Hydrate aus.
Ferrichrysin ist sowohl in Wasser wie inRTI ID="0004.0221" WI="10" HE="4" LX="1874" LY="2390"> kaltem Methanol gut löslich. Auch in absolutem Äthyl- alkohol lässt sich amorphes Ferrichrysn noch recht gut aufnehmen (bis etwa 5 %). Aus einer solchen Lö sung scheidet sich das Ferrichrysin aber rasch in rotbraunen kurzen Stärbchen ab, die auf der Nutsche kaum verfilzen. Diese Kristalle lösen sich nur noch schwer und nach längerem Erhitzen in absolutem Alkohol auf.
Das IR-Spektrum zeigt u. a. Banden beÄ 725, 783, 975, 1060, 1140, 1200 (Schulter),<B>1218</B> (Schul ter), 1237, 1275 (Schulter), 1340 (Schulter), 1368, 1430 (Schulter), 1445 (Schulter), 1465, 1495 (Schul ter), 1517, 1585, 1652, 2930, 3070 (Schulter), 3420 cm-r, vgl. Fig. 1.
Das Papierchromatogramm ergibt im System I (n-Butanol-Eisessig-Wasser, 4:1:1) den Rf-Wert 0,26, im System II (tert.-Butanol-0,04-n Salzsäure gesättigte wässrige Natriumchloridlösung, 2:1:1, Papier imprägniert mit wässrige Natriumchloridlösung 6: 3: 1) den Rf-Wert 0,34.
25 mg eisenfreies Ferrichrysin werden mit je 1 ml konzentrierter Salzsäure und Wasser 20 Stunden auf 110 erhitzt. Der Eindampfrückstand der Hydro- lyselösung wird in wenig Wasser gelöst und mit 20 ml Feinsprit verdünnt.
Diese Lösung wird in Gegenwart von 20 mg vorhydriertem Pt02 hydriert, um Hy droxylamin- zu Aminogruppen zu reduzieren. Das Hydrierungsprodukt wird nach Moore und Stein untersucht. Es ergibt sich die Anwesenheit der Anfino- säuren Ornithin, Serin und Glycin im molekularen Verhältnis 3:2:1.
<I>Beispiel 2</I> 4 g kristallisiertes Ferrichrysin werden in 100 ml Wasser gelöst, 3 g 8-Hydroxy-chdnolin in 36 nfl Methanol zugefügt und 24 Stunden bei Zimmer temperatur gerührt. Dann wird vom ,schwarzen Nie derschlag abfiltriert und das bloss gelbe Filtrat drei mal mit Chloroform ausgeschüttelt,
um überschüssiges Reagens zu entfernen. Die wässrige Phase wird im Vakuum eingedampft. Man erhält 3,3 g Desferri- chrysin in Form eines fast farblosen Rückstandes.
Das NMR-Spektrum dieser Verbindung in Tri- fluoressigsäure zeigt folgende Signale: 2,07 ppm; breites Maximum entsprechen 4 n Pro tonen; CH2-Gruppen, 2,54 ppm; Sing'ktt entsprechen 3 n Protonen;
Methyl in Acethydroxamsäure-Gruppierungen, 4,00 ppm; breites Maximum entsprechen 2 n Pro tonen, 4,2-5,1 ppm; breite Anhäufung von Banden mit erkennbaren Maxima bei 4,33; 4,83 und 5,00 ppm; total etwa 3 n Protonen, 8,02 ppm; entsprechen etwa 3 n Protonen; pepti- di,sches NH.
Die Zahl n in den obigen Angaben bedeutet wahr- scheinlich 3, entsprechend den wahrscheinlich iden tischen drei Grundeinheiten, aus denen die Molekel aufgebaut ist.
<I>Beispiel 3</I> Aspergillus melleus M 2853 wird nach dem Sub- mersverfahren in einer Nährlösung gezüchtet, die pro Liter Leitungswasser 20 g Glucose, 5 g L-Aspara- gin, 1 g sek. Kaliumphosphat, 1 g krist. Magnesium- phosphat (M9SO4, 7H20) und 0,5g Calciumchlorid enthält.
Die Nährlösung wird in den Impfkolben oder Fermentern 20-30 Minuten bei 1 atü sterilisiert. Dann beimpft man mit einer Sporensuspension wie in Beispiel 1. Man irrkubiert unter gutem Schütteln oder Rühren bei 27 , wobei die Kulturen in den Fer- mentern mit etwa 2 Volumen Luft je Volumen Lösung pro Minute belüftet werden.
Nach etwa 96 Stunden Bebrütung hat die Kulturlösung den höchsten Gehalt an Desferri-Sid eramin M 2853 erreicht. Dieser wird mittels des Antisideromycin-Testes bestimmt. Da die Nährlösung kleine Mengen Eisen enthält, liegt das gebildete Desferrichrysin teilweise in Form seines Eisen-Komplexes vor.
340 Liter aktive Kulturbrühe von Aspergillus melleus M 2853 werden mit 85 g 8-Oxy-chinolin, gelöst in 1,6 Liter Methanol, versetzt.
Nach einer Stunde wird unter Zusatz von 2 % Hyflo-Supercel als Filterhilfsmittel das Mycel abf11triert. Das Filtrat wird zur Entfernug von überschüssigem 8-Oxy-chi- nolin durch eine Säure mit 5 Liter Amberlite IR-45, OH-Form,
perkoliert. Das pH des Perkolates wird durch Zusatz von Salzsäure auf 7,5 gestellt. Die so erhaltene aktive Kulturlösung wird anschlie ssend mit Natriumchlorid gesättigt und mit Benzyl- alkohol extrahiert. Nach dem Eindampfen der Ex trakte erhält man das Desferrichrysin in Form eines gelblichen Pulvers.
70 mg Desferrichrysin werden in 2 ml Methyl- alkohol, gelöst und mit 40 ml kristallisiertem Eisen- chlorid in 2 ml Methylalkohol vermischt. Es entsteht sofort eine tief schwarzviolette Färbung, die auf Zu satz von etwas kristallisiertem Natriumacetat nach orangerot umschlägt.
Diese Lösung wird im Vakuum zur Trockene eingedampft, der Rückstand mit heissem absolutem Alkohol ausgezogen und vom Natrium chlorid abfätriert. Das Filtrat wiM eingedampft, in etwa 0,5 nfl absolutem Alkohol gelöst, filtriert und bei 0 kristallisieren gelassen.
Man erhält 53 mg kristalfsierbes Ferrichrysin, das im Papierchromato- gramm in zwei Lösungsmittelsystemen einheitlich und mit dem ursprünglichen Ferrichrysin identisch ist.
20 mg Ferrichrysin werden in 3 ml 1,2 n Salz säure gelöst und 5 Minuten im kochenden Wasser bad erwärmt. Die anfänglich rotbraune Lösung wird dabei fast farblos. Sie wird im Vakuum zur Trockene eingedampft. Im Rückstand lä,sst sich weder Bern- steinsäure noch 1-Amino-5-hyd'roxylaminopentan nachweisen.
20 mg Ferrichrysin werdenRTI ID="0005.0230" WI="5" HE="4" LX="1570" LY="2157"> mit 20%iger Schwe- felsäure am Rückfluss erhitzt. Die Hydrolyselösung wird unter reduziertem Druck destilliert, wobei man einige Male mit Wasser verdünnt. Das unter guter Kühlung aufgefangene Filtrat,
das die flüchtigen Säuren enthält, wird mittels Natronlauge auf Phenol- phthalain neutralisiert und im Vakuum zur Trockene eingedampft. Durch Umsetzung des Rückstandes mit p-Phenylphenacylbromid erhält man die p Phenyl- phenacylacetat-Mengen,
die drei Mol Essigsäure ent sprechen.
20 mg Desferrichrysin werden 3 Stunden mit 1 ml 6 n Salzsäure erhitzt und der Eindampfrück- stand, gelöst in 96 % igem Äthanol, in Gegenwart von Platinoxyd-Katalysator (Adams) hydriert. Im Reaktionsprodukt kann man 3 ninhyd'rin-positive Produkte nachweisen,
die im Papiercbromatogramm mit Phenol-Wasser (4: 1) als Fliessmittel folgende Rf- Werte zeigen: 0,21; 0,30 und 0,38.
Process for the production of ferrichrysin and desferrychrysin A large number of iron-containing growth substances have already been isolated from materials of biological origin, in particular microorganisms, for example ferrichromes, coprogens,
the ferrioxamines.
It has now been found that a new iron-containing growth substance, Ferrichryshn, and the corresponding iron-free compound, Desferrichrysin, can be obtained from a new strain of Aspergillus melleus, namely Aspergillus melleus M 2853.
The present patent therefore relates to a process for the simultaneous production of fenichrysin and desferrichrysin.
The Aspergillus mellvus M 2853 strain was isolated from the air in Zurich. It is used in our laboratories and in the Swiss Federal Institute of Technology, Institute for Special Botany, Zurich,
kept under the specified name. When cultivated on malt extract agar, the strain is characterized by a vegetative mycelium made up of septate and branched hyphae, from whose foot cells conidia carriers in the form of rounded, swollen heads grow.
The heads carry fertile cells, sterigms, in a dense arrangement, from which the conidia are cut off in unbranched chains by the formation of transverse walls.
As a result, the trunk is clearly identified as belonging to the genus Aspergillus Micheli 1729 (cf. Thom et Church, The Aspergilli, Baltimore 1926). Further classification was based on Thom and Raper, A Manual of the Aspergilli, Baltimore, 1945.

Ferrichrysin is a red-brown, crystalline substance that is readily soluble in water and methanol, even in the cold, and is sparingly soluble in absolute ethanol. The UV spectrum in water shows a maximum at. 415 mu (10 g E i @m equal to 1.48).

If the acidic hydrolysis product is examined directly without subsequent catalytic hydrogenation, no ornithine can be detected.
In ferrichrysine, the amino acids glycine (1 mol), serine (2 mol) and d-hydroxyornithine (3 mol) are combined to form a cyclic peptide.
The hydroxyornithine is N (b) -aoetyüert; The iron is bound in a complex to the 3 hydroxamic acid residues formed in this way. The overall formula of the iron-free Ferri'chrysüns is accordingly tri- (N acetyl) -cyclo- [glycyl- (seryl) 2- (d-hydroxyornit @ hyl) 3].
When ferric chloride is added, the ferric chrysin is recovered from desferrichrysin at neutral pH.
Ferrichrysin is a growth substance for various microorganisms and can therefore optionally be used in the culture of such organisms.

Like other sideramines, ferrichrysin is also able to competitively cancel the antibacterial effect of antibiotics, which belong to the group of sideromycins, against gram-positive pathogens (antisideromycin effect).
With regard to the biological effects mentioned, the ferrichrysin shows certain similarities with other sideramines, such as.
B. the ferrioxamines, the ferrichromes and: the Copro.gen. On the other hand, it differs from this substance in its physico-chemical properties in a characteristic way, which is illustrated below.
1. Ferrichrysin differs from the ferrioxamines produced by actinomycete strains not only in terms of the hydrolysis products, as mentioned above,
but also in its spectroscopic properties. Its IR spectrum differs from that of the ferrioxamines in essential points and the maximum of the UV spectrum for ferrichrysin is shorter-wave (about 415 to 420 mu) than both ferrioxamines (about 430 mrc).
2. Feerichrome, which can be produced by cultivating Usti1! Ago sphaerogena, differs only slightly from ferrichrysin in terms of spectroscopy. In the IR spectrum, ferrichrome shows a medium-strength maximum at 1275 cm-1, which in the case of ferrichrysin only appears as a weak shoulder.
Instead of a broad band at 1060 cm-1 for ferrichrysin, Fernchrom has two well-separated maxima at 1025 and 1068 cm-1.
In the NMR spectrum, the desferrichrysin has a broad maximum at 4.00 ppm and a broad accumulation of bands in the range from 4.2-5.1 ppm with maxima at 4.33; 4.83 and 5.00 ppm, while the iron-free ferrichrome in this area maxima at 4.33; Shows 4.52 (shoulder) and 4.74 ppm.

The hydrolysis products are also different. Iron-free ferric chromium, hydrolyzed and catalytically hydrogenated with 6N hydrochloric acid, provides not three, but only two ninhyd: rin-positive products, namely glycine and ornithine. There are also differences in the paper chromatogram: Fernichrom has Rf values 0.19 and 0.25 in systems I and II.
3. Fernchrom A, a companion of ferrichrome, has, in contrast to ferrichrysin, no noteworthy antagonistic effect on ferrimycin. During hydrolysis it does not produce 3 moles of acetic acid, but 3 times f-methyl-glutaconic acid. In the paper chromatogram in systems I and II, Ferrichrom A R f values of 0.12 and
0.18, and therefore migrates about half as far as ferrichrysin. Finally, ferrichrome A behaves differently than ferrichrysine in the Craig distribution: In the benzyl alcohol-n-butanol-saturated aqueous sodium chloride solution, 0.001-n aqueous hydrochloric acid 9: 9: <B> <I> 5:
</I> </B> 15 over 95 levels, it accumulates in levels 3-15, while ferrichrysine appears in fractions 36-60.
4. Neither the IR spectrum nor hydrolysis products are known of Coprogen. The elemental composition, however, is clearly different from that of Ferrichrysän; for coprogen the following is given: C = 50.9%; H = 6.9%; N = 10.2%; Fe = 6.7%. Coprogen already decomposes at 205 C. In the paper chromatogram, Coprogen shows the Rf values 0.43 (system 1) and 0.55 (system 1I).
The process for the simultaneous production of ferrichrysin and desferrächrysan is characterized in that an Aspergillus strain having the properties of Aspergillus melleus M 2853 is bred. The breeding takes place z.
B. in aqueous, a carbon source, nitrogenous connec tions and nutrient solution containing inorganic salts, aerobic, so for example in dormant surface culture, or preferably submerged with shaking or stirring with air or oxygen in shaker bottles or the known fermenters. A suitable temperature is between 18 and 40 C, preferably 27 C.
The nutrient solution generally shows a significant increase in sideramine after 4-8 days.
The nutrient solution contains z. B. carbohydrates such as glucose, sucrose, liquorice, starch, alcohols such as mannitol or glycerol. A% nitrogen-containing nutrients are called amino acids, e.g.
B. ornithine, peptides, proteins and their breakdown products such as peptone or tryptone, meat extracts, water-soluble fractions of cereal grains such as maize or wheat, distillation residues from alcohol production, yeast, seeds, especially the rapeseed and soy plants, the cotton plant, ammonium salts and nitrates.
The nutrient solution can contain other inorganic salts, for example, chlorides, carbonates, sulfates, nitrates of alkalis, alkaline earths, magnesium, zinc or manganese as well as traces of iron salts (<10-7 mol / liter).
The ferrichrysin activity can be determined microbiologically using the modified Bonifas test (cf. Zahner et al., Arch. Mikrobiol. <I> 36, </I> 325 ff [1960]).
A ferricin solution with a content of 0.01 mg / ml is used as the test solution, and Staphylococcus aureus as the test strain.
The ferrichrysin concentration of the culture solution can also be determined optically. For this purpose, 5 ml of culture liquid with 1.5 g of sodium chloride, 1 ml of 0.1% ferric sulfate solution and 5 ml of benzyl alcohol are shaken for 5 minutes, then centrifuged off,
the organic phase is filtered and the absorbance of the organic phase is determined at 410 m u.
An extract made in the same way from uninoculated nutrient solution serves as a comparison sample. After 4-8 days, the cultures reach an activity equivalent to 300-400 mg pure ferrichrysin.
After cultivation is complete, 1 g of ferric chloride per liter is added to the culture if ferric chrysin is to be isolated, but not if the iron-free desferrichrysin is to be obtained. The mycelium is separated from the culture filtrate; after which the main amount of sideramine is found in the culture filtrate. Nevertheless, significant amounts of it stick to the Mycell. It is therefore advantageous to wash it off well.
For example, what water or aqueous alcohols such as aqueous methanol are suitable.
For the isolation of ferrichrysin from the culture filtrate you can proceed according to methods known per se and z. B. apply one of the working methods given below or combinations thereof.
1. You can use Adsorption @ smättel, z. B. activated carbons such as Norit, activated earths such as Frankonite, Fuller's earth or floridin, or resin adsorbers such as Asmit. The adsorbates are expediently eluted with mixtures of water-miscible organic solvents with water, e.g.
B. with water-methanol, water-pyridine, ver thinned acetic acid-methanol or water-methanol-glacial acetic acid-butanol mixtures. The mixture of water (4 parts by volume) and pyridine (1 part by volume) has proven to be particularly advantageous for the elution of a Frankonite or Norit adsorbate.
2. Furthermore, the ferrichrysin can be removed from an aqueous solution by means of organic solvents. For these extraction processes, higher organic alcohols, e.g. B. benzyl alcohol or isopropyl alcohol, particularly proven. Conveniently, the aqueous phase is an inorganic cal salt, for. B. ammonium sulfate or sodium chloride added.
The ferrichrysin can be extracted from the organic extracts obtained either by RTI ID = "0003.0219" WI = "18" HE = "4" LX = "1058" LY = "2364"> evaporation of the solvent or by precipitation using a suitable organic solvent, z. B. ether, petroleum ether or ethyl acetate, can be obtained in enriched form.
3. Another method of enriching ferrichrysin is to distribute it between an aqueous solution and a solution of phenol in chloroform, it being possible to vary the phenol content of the chloroform solution.
4. Another method for the enrichment and / or separation of ferrichrysin is chromatography, such as adsorption chromatography on various materials, e.g.
B. on Norit, aluminum miniumoxyd, magnesium silicates, Silncageln>, Cal ciumsulfat and partition chromatography with cellulose, starch, silica gel, Celite and the like as carrier substances, or chromatography on ion exchange resins, eg. At Dowex-50, Amberhte IRC-50 and the like.
5. Ferrichrysin can also be enriched by countercurrent distribution according to Craig between two immiscible solvent phases. The following solvent system has proven to be particularly effective: n-butanol-benzyl alcohol-0.001-n. Hydrochloric acid aqueous solution of sodium chloride saturated at 19 (9: 9: 15: 5).
In addition to ferrichrysin, desferrichrysin is bound by Aspergillus melTeüs M 2853 and can therefore also be isolated directly from the culture filtrate. It is advantageous to remove iron present in the culture filtrate during the isolation, for example by adding iron complex-forming substances, e.g.
B. 8-Oxy- quinoa or ethylenediamine tetraacetic acid.
The substances forming the iron complex can be added to the culture broth immediately after the fermentation has ended. The addition is advantageously carried out at a later stage of the work-up in order to also remove any iron (III) ions entrained by the agents used.
The isolation of Desferrichrysins from the culture solution takes place, for. B. by the methods mentioned for the isolation of ferrichrysin.
In the following examples, the temperatures are given in degrees Celsius. <I> Example 1 </I> The strain Aspergillus melleus M 2853 is grown in a well-aerated submerged culture at 27. A nutrient solution is used which contains 20 g of glucose, 5 g of L-Asparagiu, 1 g of sec per liter of tap water.
Potassium phosphate, 1 g crystall. Contains magnesium phosphate (M9S04; 7H20) and 0.5 g calcium chloride. The iron content of tap water is 20-30 y / liter. The nutrient solution is sterilized at 120 for 20 minutes. It is vaccinated with a spore suspension that contains 150-180 million spores per liter.
After 4-8 days, 1 g of ferric chloride per liter is added to the cultures and frozen with the addition of 2 Celite. 300 g of sodium chloride per liter are then added to the culture filtrate and the exposed solution is extracted three times with 1/10 volume of benzyl alcohol. The organic phase is dried with sodium sulfate and mixed with 3 volumes of ether.
The benzyl alcohol-ether mixture is extracted with a small amount of distilled water until the aqueous phase is only red in color. The aqueous extract is lyophilized, whereby a red powder is obtained, in a yield of 300-4.00 mg / liter, containing <B> 80% </B> of the activity of the starting material.
18.9 g of crude ferrichrysine mixture are subjected to a Craig distribution over 95 stages, the substance being poured into the first 7 glasses of an automatic distribution apparatus (glass content 25 ml per phase).
A mixture of benzyl alcohol, n-butyl alcohol, saturated aqueous sodium chloride solution and 0.001N aqueous hydrochloric acid in a volume ratio of 9: 9: 5: 15 is used as the distribution system.
The red-brown dye is essentially separated into a main component and a secondary component, which have their distribution maxima in fractions 46 and 75, respectively. This corresponds to approximate distribution coefficients of K1 = 0.9 and K2 = 3.8.
Fractions 36-60 are pooled, double the volume of ether is added and the aqueous phase is separated off. The organic layer is extracted three times with a little water, the combined aqueous extracts are approximately half saturated with sodium chloride and extracted several times with phenol-chloroform (1 kg of phenol to 1 liter of chloroform).
The extracts are washed with half-saturated sodium chloride solution. The initially cloudy solution is clarified on a short Celite column, then diluted with twice the volume of ether and shaken out several times with a little water, the brownish-red dye going back into the aqueous phase.
The aqueous extracts, washed several times with ether, are evaporated in vacuo and the residue is dried. 8.2 g of purified ferrichrysin are obtained. For crystallization, 16 g of such an evaporation residue are dissolved in about 200 ml of hot absolute ethyl alcohol (distilled over sodium).
Crystallization sets in rapidly and 11.6 g of crystalline ferric chrysin precipitate in the form of red-brown rods. The mother liquors are evaporated to dryness and redissolved in absolute alcohol. This gives another 0.9 g of crystals.
For analysis, a sample is recrystallized again from absolute alcohol. The red-brown rods turn black at 270 without melting.
Analysis: Found C 43.37, H 6.08, Fe 7.09, N 16.72. Absorption spectrum in water: Am "" = 415 mu; 10 g egg öm = 1.48.
If the alcohol is not completely dry for crystallization, hydrates which are partly amorphous and partly in irregular chunks precipitate.
Ferrichrysin is readily soluble both in water and in RTI ID = "0004.0221" WI = "10" HE = "4" LX = "1874" LY = "2390"> cold methanol. Amorphous Ferrichrysn can also be absorbed quite well in absolute ethyl alcohol (up to about 5%). From such a solution, however, the ferrichrysin quickly separates in reddish-brown short thicknesses that hardly become matted on the suction filter. These crystals dissolve only with difficulty and after prolonged heating in absolute alcohol.
The IR spectrum shows u. a. Gangs beÄ 725, 783, 975, 1060, 1140, 1200 (shoulder), <B> 1218 </B> (shoulder), 1237, 1275 (shoulder), 1340 (shoulder), 1368, 1430 (shoulder), 1445 (Shoulder), 1465, 1495 (shoulder), 1517, 1585, 1652, 2930, 3070 (shoulder), 3420 cm-r, cf. Fig. 1.
The paper chromatogram gives in system I (n-butanol-glacial acetic acid-water, 4: 1: 1) the Rf value 0.26, in system II (tert-butanol-0.04-n hydrochloric acid, saturated aqueous sodium chloride solution, 2: 1: 1, paper impregnated with aqueous sodium chloride solution 6: 3: 1) the Rf value 0.34.
25 mg of iron-free ferrichrysin are heated to 110 for 20 hours with 1 ml of concentrated hydrochloric acid and water. The evaporation residue of the hydrolysis solution is dissolved in a little water and diluted with 20 ml of fine spirits.
This solution is hydrogenated in the presence of 20 mg of pre-hydrogenated Pt02 in order to reduce hydroxylamine to amino groups. The hydrogenation product is examined according to Moore and Stein. The result is the presence of the amino acids ornithine, serine and glycine in a molecular ratio of 3: 2: 1.
<I> Example 2 </I> 4 g of crystallized ferric chrysin are dissolved in 100 ml of water, 3 g of 8-hydroxychdnoline in 36 nfl methanol are added and the mixture is stirred for 24 hours at room temperature. Then the black precipitate is filtered off and the yellow filtrate is extracted three times with chloroform,
to remove excess reagent. The aqueous phase is evaporated in vacuo. 3.3 g of desferric chrysin are obtained in the form of an almost colorless residue.
The NMR spectrum of this compound in trifluoroacetic acid shows the following signals: 2.07 ppm; broad maximum correspond to 4 n protons; CH2 groups, 2.54 ppm; Sing'ktt correspond to 3 n protons;
Methyl in acethydroxamic acid moieties, 4.00 ppm; broad maximum correspond to 2 n protons, 4.2-5.1 ppm; broad accumulation of bands with noticeable maxima at 4.33; 4.83 and 5.00 ppm; total about 3n protons, 8.02 ppm; correspond to about 3 n protons; peptic NH.
The number n in the above information probably means 3, corresponding to the three probably identical basic units that make up the molecule.
<I> Example 3 </I> Aspergillus melleus M 2853 is grown according to the submerged method in a nutrient solution which contains 20 g glucose, 5 g L-asparagine, 1 g sec per liter of tap water. Potassium phosphate, 1 g crystall. Contains magnesium phosphate (M9SO4, 7H20) and 0.5g calcium chloride.
The nutrient solution is sterilized in the inoculation flask or fermenter for 20-30 minutes at 1 atm. A spore suspension is then inoculated as in Example 1. It is incubated at 27, with good shaking or stirring, the cultures in the fermenters being aerated with about 2 volumes of air per volume of solution per minute.
After about 96 hours of incubation, the culture solution has reached the highest content of Desferri-Sid eramin M 2853. This is determined by means of the antisideromycin test. Since the nutrient solution contains small amounts of iron, the desferrichrysin that is formed is partly in the form of its iron complex.
340 liters of active culture broth from Aspergillus melleus M 2853 are mixed with 85 g of 8-oxyquinoline, dissolved in 1.6 liters of methanol.
After one hour, the mycelium is filtered off with the addition of 2% Hyflo-Supercel as a filter aid. To remove excess 8-oxy-quinoline, the filtrate is acidified with 5 liters of Amberlite IR-45, OH-Form,
percolated. The pH of the percolate is adjusted to 7.5 by adding hydrochloric acid. The active culture solution thus obtained is then saturated with sodium chloride and extracted with benzyl alcohol. After evaporation of the extracts, the desferrichrysin is obtained in the form of a yellowish powder.
70 mg desferrichrysin are dissolved in 2 ml methyl alcohol and mixed with 40 ml crystallized iron chloride in 2 ml methyl alcohol. A deep black-violet color develops immediately, which turns orange-red when some crystallized sodium acetate is added.
This solution is evaporated to dryness in vacuo, the residue is extracted with hot absolute alcohol and the sodium chloride is filtered off. The filtrate was evaporated, dissolved in about 0.5 nfl absolute alcohol, filtered and left to crystallize at 0.
53 mg of crystalline ferrichrysin are obtained, which in the paper chromatogram in two solvent systems is uniform and identical to the original ferrichrysin.
20 mg of ferrichrysine are dissolved in 3 ml of 1.2N hydrochloric acid and heated in a boiling water bath for 5 minutes. The initially red-brown solution becomes almost colorless. It is evaporated to dryness in vacuo. Neither succinic acid nor 1-amino-5-hydroxylaminopentane can be detected in the residue.
20 mg of ferrichrysine are refluxed with 20% sulfuric acid, RTI ID = "0005.0230" WI = "5" HE = "4" LX = "1570" LY = "2157". The hydrolysis solution is distilled under reduced pressure, diluting a few times with water. The filtrate collected with good cooling,
which contains the volatile acids, is neutralized on phenol phthalain using sodium hydroxide solution and evaporated to dryness in a vacuum. By reacting the residue with p-phenylphenacyl bromide, the p-phenylphenacyl acetate amounts are obtained,
the three moles of acetic acid correspond.
20 mg of desferrichrysin are heated for 3 hours with 1 ml of 6N hydrochloric acid and the evaporation residue, dissolved in 96% ethanol, is hydrogenated in the presence of platinum oxide catalyst (Adams). In the reaction product one can detect 3 ninhyd'rin-positive products,
which show the following Rf values in the paper bromatogram with phenol-water (4: 1) as flow agent: 0.21; 0.30 and 0.38.
