Verfahren zur Herstellung von Ferrirubin und Desferrirubin Aus, Materialien biologischen Ursprungs, insbeson dere Mikroorganismen, ist bereits eine, grössere Anzahl eisenhaltiger Wuchsstoffe isoliert wordk,-n, beispielsweise die Ferrichrome, das Coprogen, die Ferrioxamine.
Es wurde nun gefunden, dass man aus Pilzstämmen der Familie Aspergillaceae einen neuen eisenhaltigen Wuchsstoff, das Ferrirubin, und die entsprechende eisen freie Verbindung, das Desferrirubin, gewinnen kann. Ferrirubin und Desferrirubin. bildende Stämme sind ins besondere Penicillium vanabile M 9-733, Spicaria sp. M 4622 und Paecilomyces varioti M 4646.
Gegenstand der vorliegenden Erfindung ist daher ein Verfahren zur gleichzeitigen Herstellung von Ferrirubin und Des- ferrirubin, das, dadurch gekennzeichnet ist, dass man einen Ferrirubin und Desferrirubin bildenden Stamm züchtet.
Die genannten Stämme werden in der Eidg. Techn. Hochschule, Institut für spezielle Botanik, Zürich, sowie, in unseren Laboratorien unter der angegebenen Bezeichnung aufbewahrt. Die, Bestimmung der Stämme erfolgte nach K. B. Raper and Ch. Thom, <B>A</B> Manual of the Penicillia, Baltimore, The Williams <B> & </B> Wilkiris Co-, 1949.
Penicillium variabile M<B>2733</B> wurde auseiner Erd- probe aus dem Botanischen Garten in Zürich isoliert. Der Stamm ist in die Sektion Biverticillata-Symmetrica der Penicillien einzuordnen.
Da Pelitheden oder Skle# rotien fehlen, die Kolonien eine typisch sammetige Oberfläche aufweisen und die Rück-seite der Kolonien oft rotorange bis braun gefärbt ist, gehört der Stamm in die Series Penicillium purpurogenum. Charakteristisch für<U>Stamm</U> ETH M<B>2733</B> ist weiter die Bildung steriler, gelber Luftmycelpartien,
weshalb der isolierte<U>Stamm</U> der Art P.enicillium variabfie Sopp <B>1912</B> zuzuordnen ist. Die Umschreibung,dieser Art bei Raper u. Thom (l. c. <B>S.</B> 642-644) ist ausführlich, und Stamrn M<B>2733</B> lässt sich gut in den Variationsbereich des Taxons einreihen.<B>.</B>
Spicaria sp. M 4622 wurde aus einer Probe eines infizierten Süssmostes der Zentralstelle für Obst- und Kartoffr,lverwzrtung, Wädenswil, Kanton Zürich, iso- liert. Der Organismus zeigt eine gelblich-sandfarbene Sporulation; die langen zugespitzten Steriginen entstehen in Wirteln und zeigen stark divergieren & Tendenzen;
die Konidien entstehen in Ketten an den Enden der Sterigmen. Derartige, Formen sind der Gattung Spicaria Harz<B>1871</B> zuzuordnen (Raper u. Thom, <B>S.</B> 24-25). Eine Artbestimmung innerhalb der Gattung Spicaria ist nicht mit Sicherheit möglich, da die Prinzipien der Artdifferenzierung nicht fest stehen. Die Isolierung ist daher zu bezeichnen als Spicaria species Stamm ETH M4622.
Paecilomyces varioti M 4646 wur & in der Eidg. Versuchsanstalt in Wädenswil, Kanton Zürich, isoliert. Der Organismus bildet reichlich, bräunlich-gelbe, läng lich ovale Konidien an den Enden langer, zugespitzter Konidienträger, die, in verticiller Anordnung stehen und stark divergieren.
Der Organismus ist der Gattung Paecilomyces Bainier <B>1907</B> zuzuordnen und darin der Art Paecilouiyces varioti Bainer <B>1907,</B> der vielleicht einzigen, sehr variablen Art -der Gattung (vgl. Raper und Thom, S. <B>688-692).</B>
Für die Züchtung der<U>Stämme</U> kommen als Kohlen stoff- und Stickstoffquellen in Betracht: Kohlenhydratt', z. B. Glucose" Saccharose, Lactose, Stärke, Alkohole, wie Mannit und Glycerin, Aminosäuren, z.
B. Ornithin, Peptide, Proteine und deren Abbauprodukte, wie Pepton oder Trypton, Fleischextrakte, wasserlÖsliche Anteile von Getreidekörnern, wie Mais, oder Weizen, Destilla,- tionsräckstände, der Alkohollierstellüng, Hefe, Samen, insbe,sondere der Rapsi- und Sojapflanze, der Baum- wollpflanzc, Ammoniumsalze, Nitrate.
An anorgani schen Salzen kann die Nährlösung beispielsweise Chlo ride, Carbonate, Sulfate, Nitrate. von Alkalien, Erd alkalien, Magnesium, Zink und Mangan sowie Spuren von Eisen enthalten.
Die Züchtung erfolgt aerob, also beispielsweise in ruhender Oberflächenkultux, od-,r vorzugsweise subiners unter Schütteln oder Rühren mit Luft oder Sauerstoff in Schüttelflaschen oder den bekannten Ferinentern. Als Temperatur eignet sich eine solche zwischen<B>18</B> und 40'<B>C,</B> vorzugsweise<B>27' C.</B>
Da die Bildung des Ferrirabins und Desferrirubins durch ein eisenarmes Nährmedium begünstigt wird, ist es zweckmässig, die Züchtung unter Eisenmangel, vorzugsweise bei einem Eisengehalt von höchstens<B>10-7</B> Mol/Liter, vorzunehmen. Eine wesentliche Ferrirubin- Wirkung zeigt die Nährlösung dabei im allgemeinen nach<B>12-16</B> Tagen.
Die. Ferrirubin-Aktivität kann mikroblo-logisch mittels des abgewandelten Bonifas-Testes (vgl. Zähner et al., Arch, Narobiol. <B>36, 325</B> ff. <B>[1960])</B> bestimmt werden. Als Testlösung verwendet man einem FerrimyciDlösung von<B>0,1</B> mg/ml Gehalt, als Teststamm, Staphylococcus aureus Rosenbach.
Man kann auch optisch die Fürrirubin-Konzentra- tion der Kulturlösung feststellen. Zu diesem Zweck werden<B>5</B> ml Kulturflüssigkeit mit<B><I>1,5 g</I></B> Natriumehlorid, <B>1</B> ml<B>0,1</B> %iger Ferrisulfatlösung und<B>5</B> nil Benzyl- alkohol <B>5</B> Minuten geschüttelt, abzentrifugiert, die orga nische Phase filtriert und die Extinktion der organischen Phase bei 440 mIL bestimmt.
Als Vergleichsprobe dient ein auf gleiche Weise hergestellter Extrakt aus unbe- impfter Nährlösung.
Nach<B>12-16</B> Tagen erreichen die Kulturen eine Aktivität, die<B>50-200</B> mg reinem Ferrirubin entspricht. Bei Beendigung der Züchtung kann man pro Liter <B>1 g</B> Ferrichlorid zur Nährlösung geben, wobei das vor handene, Desferrirubin in Ferrirubin übergeführt wird. Man trennt das. Mycel, vom Kulturfiltrat ab-, vorzugs weise in Gegenwart eines Fütrierhilfsmittells, z. B. Hyflo- Supereel, wonach die Hauptmenge des Ferrirabins im Kulturfiltrat gefunden wird.
Es bleiben aber trotzdem namhafte Mengendavon am Mycel haften. Es ist daher vorteilhaft, dieses gut auszuwaschen. Dazu eignen sich beispielsweisc Wasser oder wässrige Alkohole, wie wässriges. Methanol.
Für die Isolierung von Ferrirubin aus dem Kultur filtrat kann, man nach an s-ich bekannten Methoden vorgehen und z. B.. eine der nachfolgend angegebenen Arbeitsweisen oder Kombinationen davon anwenden.
<B>1.</B> Man kann Adsorptionsmittel verwenden, z. B. Aktivkohlen, wie Norit , aktivierte Erden, wie- Fran- konit, Fullererde oder Floridin, oder Harzadsorber, wie Asmit. Die Elution der Adsorbate erfolgt zweck mässig mit Gemischen von mit Wasser mischbaren orga nischen Lösungsmitteln mit Wasser, z.
B. mit Wasser- Methanol-, Wasser-Pyridin-, verdünnt-- Essigsäure#Me- thanol- oder Wasser-Methanel-Eisessig#-Butanol-Gemi- schen. Als besonders vorteilhaft für die Elution eines Frankonit- oder Noritadsorbates hat sich das Gemisch von Wasser (4 Volumteile) und Pyridin <B>(1</B> Volumte-il.) erwiesen.
2. Weiter kann Ferrirubin einer wüssrigen, Lösung mittels organischen Lösungsmitteln entzogen werden. Für diese Extraktionsverfahren haben sich höhere organische Alkohole, z. B. B.ünzylalkohol oder Iso- propylalkohol, besonders bewährt. Zweckmässigerweise wird dabei der wüssrigen Phase ein anorganisches Salz, z. B. Ammonaumsulfat oder Natriumchlorid, zugesetzt.
Aus den erhaltenen organischen Extrakten kann Ferri- rubin entweder durdh Abdampfen düs Lösungsmittels oder dürch Ausfällung mittels eines geeigneten organi schen Lösungsmittels, z. B. Äther, Petroläther oder Äthylacetat, in angereicherter Form erhalten wer-d'en.
<B>3.</B> Eine Weitere Methode der Anreicherung des Ferrirabins besteht darin, dass man es zwischen einer wässrigen Lösung und -einer Lösung von Phenol in Chloroform verteilt, wobei der Phenolg-,halt der Chloro- formlösung variiert werden kann.
4. Eine andere Methode zur Anreicherung des Ferrirubins stellt die Chromatographie dar, wie Adsorp- tionschromatographie an verschiedenen Materialien, zum Beispiel an Norit , Aluminiumoxyd, Magnesiumsilika- ten, Silicagel, Calciumsulfat, sowie Verteilungschromato- graphie mit Cellulosei, Stärke, Silicagel, Celite. und der gleichen als Trägersubstanzen.
<B>5.</B> Weiter<U>kann</U> das Ferrirubin durch Gegenstrom verteilung nach Craig zwischen zwei nicht mischbaren Lösungsmittelphasen angereichert werden. Folgendes Lösungsmittelsystem hat sich dabei besonders bewährt-. n-Butanol-Benzylalkohal-0,001-n. Salzsäure-wässrige, bei <B>19' C</B> gesättigte Natriumchloridlösung <B>(9: 9:<I>15: 5).</I></B>
Desferrirubin wird neben Ferrirubin von den ge nannten Stämmen gebildet und kann daher auch direkt aus dem Kulturfiltrat isoliert werden. Vorteilhaft ent fernt man bei der Isolierung im Kulturfiltrat vorhandenes Eisen, beaspielsweise. durch Zusatz Eisenkomplex bilden der Stoffe, z. B. 8-Oxychinohn.
Man kann die Ei#senkomplex bildenden Stoffe direkt nach Beendigung der Fermentation der Kulturbrühe zusetzen. Vorteilhaft erfolgt der Zusatz in einem späteren Stadium der Aufarbeitung, um allfällige durch die. benützten- Agenzien eingeschleppte Eisen-(111)-ionen ebenfalls zu entfernen.
Die Isolierung des Desferrirubins aus der Kultur lösung erfolgt nach den für die Isolierung von Fernrubin erwähnten Methoden.
Ferrirubin ist eine rotbraune Substanz, die bisher nur in amerphem Zustand erhalten wurde. Sie ist sehr hygroskopisch. Analysenwerte: <B>C</B> 49,01<B>%;</B> H<B>= 6,57 %;<I>N</I></B> = <B>12,70</B> Fe<B>5,25 %; 0</B> = 26,47<B>%</B> (ber.); Acetyl <B>= 0</B> Das UV-Spektrum in Athanol zeigt Maxima bei<B>216</B> my (log<B>E</B> "/- <B>= 2,58);
252</B> mg (log<B>E 1 %</B> = 2,34) und <B>1 cm</B> 450 my (log<B>E 1,52).</B> Das<B>1</B> Ick-Spektrum in Kaliumbromid. weist m u. a. Im Papierchromatogramm werden folgende Rf- Werte erhalten: 0,42 im System<B>1</B> (n-Butanol-Eisessig- Wasser, 4:<B>1 - 1)</B> und<B>0,73</B> im System II (tert.-Butanol- 0,004-n. Salzsäure7gesättigte wässrige. Natriumchlond- lösung, 2:<B>1:
1;</B> Papier imprägniert mit Aceto-n-Wass#er- gesättigte wässrige Natriumehlorid-lösung, <B>6: 3: 1).</B>
Bei der sauren Hydrolyse das Ferrirubins wird<B>-</B> im Gegensatz zu Ferrichrom, Forrichrysin und Ferricrocin <B>-</B> keine Essigsäure, sondern<B>3</B> Mol trans-5-Hydroxy-3- me,th#yl-pentDn-(2)-säure erhalten.
Bei der Behandlung des Ferrirubins mit Basen oder Säuren oder m#it Eisenkomplex bildenden Stoffen wird das Eisen aus dem Molekül entfernt, wobeJ ein farb loses Pulver, das Desferrirubin, gebUdet wird.
Wenn man dieses mit 6n Salzsäure hydrolysiert und die Hydro- lyseprodukte (zwecks Reduktion gegebenenfalls vor handener Hydroxylaminogruppen) katalytisch hydriert, erhält man drei ninhydrinpositive Produkte, die auf Grund der papierchromatographischen Untersuchung (Fliessmittel Phenol-Wasser <B>8:</B> 2 und Butanol-Eisessig- Wasser 4:<B>1 :
1</B> und Anfärben mit Ninhydrin) sowie bei der Analyse nach Moore und Stein als Omithin, Glycin und Serin identifiziert werden. Glycin und Serin sind im molekularen Verhältnis<B>1 :</B> 2 vorhanden.
Eisenfreies Ferrichrom, nüft 6n Salzsäure hydrolysiert und katalytisch hydriert, liefert nicht drei, sondem nur zwei ninhyd#,rm- positive Produkte,
nämlich Glycin und Ornithin. Co- progen gibt als einziges ninhydrin-positives Hydro- lyseprodukt Ornithän.

Die Bruttoformel des Ferrirubins ist demnach C41H64017N9Fe,.
Bei der katalytischen Hydrierung des Ferrirubins wird die trans-5-Hydroxy-3-methyl-penten-(2)-säure hy- dTiert, ohne, dass der übrige Teil der Molekel angegriffen wird. Manerhält das Hrxahydroferrirubin, dbssen Säure- kompenente die 5-Hydioxy-3-methyl-valeriansäure ist.
Hexahydroferrirubin ist identisch mit dem durch Hydrie rung von Ferrirhodin erhaltenen Hexahydroferrirhodin (vgl. französisches Patent Nr. <B>1371</B>446). Das IR-Ab- #sorptionsspektrum,der Verbindung in Nujol ist in Fig. <B>3</B> wiedergegeben.
Im UV-Abso.rptions-spektrumdes Hexa- hydrofcrrirubins ist<B>die</B> starke Absorptionsbande des Fzrrirub-ins bei<B>252</B> mg verschwunden und das breite Maximum bei 450 mg nach 425-430 mg verschoben.
Die Absorptionskurve ist praktisch deckungsgleich mit der des Ferrichrysins (dieses besitzt an Stelle der drei Im Ferrirubin sind die Aminosäuren Serin (2 Mol.), Glycin <B>(1</B> Mol) und ö-Hydroxyorm.*thin <B>(3</B> Mol) zu einem cyclischen Hexapeptid verbunden.
Die Hydroxyornithin ist N(ö)-acyhert; an die so gebildeten Hydroxamsäure# reste ist das Eisen komplex gebunden.
Ferriruhin weist folgende, Strukturformel auf: 5-Hydroxy-3-methyl-valeriansäurereste drei Acetylreste, (vgl. Patent Nr. 423<B>099).</B> Die, Rf-Werte des Hexahydro- ferrirubins in dh-n Systemen I und II sind<B>0,57</B> und<B>0,80</B> im Vergleich zu<B>0,52</B> und<B>0,
81</B> des Ferrirubins (gelb- oranger Fleck im Gegensatz zum rötlich-braunen Fleck .des Ferrirubins). Entfernt man aus dem Hexahydro- ferrirubin, das Eisen, beispielsweise mittels 8-Hydroxy- chinohn, spaltet in dem erhaltenen De#sfeni-hexahy & o- ferriirubin durch milde saure Hydrolyse- die 5-Hydroxy- 3-methyl-valeriansäurereste ab, acetyliert (am Stick stoff und Sauerstoff)
und spaltet die 0-Acetylgruppen selektiv ab, z. B. mittels Ammoniak, so erhält man Des- fenichrysin, das mit Eisen(Ill)-salz in Ferrichrysin übergeführt werden kann. Das so aus Ferrirabin er haltene Ferrichrysin zeigt im Papierchromatogramm in den Systemen<B>1</B> und II dieselben Rf-Werte wie authcn- tisches Ferrichrysin.
Bei der Elementaranalyse, des eisenfreien Ferrlrubins werden folgende Werte gefunden:<B>C</B> = <B>51,12;</B> H<B>= 7,29;</B> <B><I>N =</I> 13,00;</B> Acetyl <B>= 0 %.</B>
Ein wesentlicher Unterschied gegenüber den eisen freien Sideraminen Ferrichrysin, Ferricrocin und Ferri- chrom besteht in der Hydrolysegeschwindigkeit dee Hy- diroxamsäurebindungen. Die letzteren drei Verbindungen verlieren ihr Komplexbindungsvermögen zu Eisen(III)- Ion vollständig nach etwa<B>18</B> Minuten beim Erwärmen mit n Salzsäure im #siedenden Wasserbad.
Beim eisen- freien Ferrirubin nimmt die Intensität der Eisenehlorid- reaktion erst nach etwa 2 Stunden merküch ab. Eine ähnlich hohe Resistenz gegen Säure wurde bisher in der Sideraminreihe, nur beirn biologisch unwirksamen Ferri- chrom <B>A</B> beobachtet.
Die übrigen Eigenschaften von Forrirubin stimmen aber nüt denen von Ferrichrom. <B>A</B> nicht überein. Ferrichrom <B>A</B> ist eine Säure mit & ei freien Carboxylgruppen (ss-Methylglutaconsäurerest,--), während Ferri#rubin keine titrierbaren Gruppen besitzt.
Im NMR-Spektruin, aufgenommen mit einem Va- rian-Spektrographen in D20, weist Desferrirubih fol gende, Signale auf (vgL Fig. 2): <B>1,86</B> ppm. (14 11);<B>2,38</B> ppm. (4<B>M; 2,92</B> ppm, (111); <B>3,79</B> ppm, <B>(10</B> 1-1); 4,39 ppin (4 11); 6,-03 ppm. (2 H).
Angaben der chemischen Verschiebungen fil ö- Weiten gegenüber Tetramethylsü, an = <B>0.</B> Die durch Integration erhaltenen Protonenzahlen bedeuten reine Verhältniszahlen, wobei das kleinste Signal, als<B>1</B> an genommen wurde. Alle Signale, bilden breite Signal anhäufungen ohne deutliche Auflösung. Die Signal haufen bei<B>3,79</B> und<B>6,03</B> ppm besitzen deutliche Dop pelmaxima, sind aber offenbar keine reihen Dubletten.
Aus Desferrirubin wird bei Zusatz von Eisenchlorid bei neutralem pH das Ferrirubin zurückerhalten. Derivate des, Ferrirubins und Desferrirabins und ihrer Hydrierungsprodukte sind beispielsweise Verbin dungen, in denen die Hydroxylgruppe der Serinreste verestert ist, oder in Öenen die, Acylgruppe der Hy- droxamsäurereste durch andere Acylgruppen substituiert ist,
beispielsweise durch Niederalkanoyl- oder Nieder- alkenoyl- oder andere o)-Hydroxyalkenoylgruppen als die 5-Hydrc>xy-3-methyl-pent-enoylgruppe, z.
B. Pro- pionyl, Butyryl, Pentanoyl, Pent--noyl, 5-Hydroxy- pentenoyl. Die erstgenannten Derivate, werdlenerhalten, wenn man Ferrirubin oder Hexahydroferrirubin mit acylieTenden Mitteln behandelt, z.
B. mit Säureanhydri- den oder Säurehalogenidrn, wie, Essigsäureanhydrid oder Acetylchlorid, und, die Ester isoliert und, wenn erwÜnscht, das Eisen eliminiert. Die Desacylverbindung erhält man durch Desacylierung des Desferrirubins oder Hexahydrode,sfe,rri,rubins in mildem, saurem Medium, z.
B. in Gegenwart verdünnter Mineralsäuwen. Das so -erhaltene cyclische Hexapeptid kann in bekannter Weise a--yliert werden; dabei gebildete, O-Acylgruppen lassen sich mit Ammoniak, beispielsweise in Methanol, ab spalten. Das Acyl-analogon des Desferrirubins kann mit Eisensalzen in den entsprechenden EisenkompIcx übergeführt werden.
Ferrirubin, Desferrirubin und Analoge sind Wuchs- stoffe für verschiedene Mikroorganismen; sie können daher bei der Kultur solcher Organismen verwendet werden.
Die eisenhaltigen Verbindungen besitzen anti- anaeinische Eigens--haften und können dementsprechend als Heilmittel verwendet werden. Auch Desferrirubin, und seine Derivatz haben wertvoll-- pharmakolo#gische Eigen- .schaften. So verhindernsie die, Ablagerung eisenhaltiger Piginente. im Gewebe bzw. bewirken in Fällen von patho logischer Eisenablagerung im Organismus eine Aus- scheidunor des Eisens, z.
B. bei Hämochromatose, und Hämosiderose. Sie können dementsprechend in der Heilkunde, verwendet werden.
Die pharmazeutischen Präparate enthalten die, ge nannten Verbindungen in Mischung mit einem organi- sehen oder anorganischen pharmazeutischen Träger, der für die enterale oder parenterale Anwendung<B>ge-</B> eignet ist. Die Präparate werden aus dem Ausgangs material nach den üblichen Methoden hergestellt.
Wie andere Sideramine ist auch Ferrirubin im stande, die antibakterielle Wirkung von Antibiotika die der Gruppe dar Sideroraycine angehören, gegen über grampositiven Erregern kompetitiv aufzuheben (A,ntisideromyci,n-Wirkung).
Inden folgenden Beispielen sind die Temperaturen in Celsiusgradien angegeben.
Beigpiel <B><I>1</I></B> Der Stamm Penicillium. variabile M<B>2733</B> wird in gut belüfteter Subinerskultur bei<B>27-</B> gezüchtet. Man verwendet eine Nährlösung, die pro Liter destilliertem Wasser 20<B>g</B> Glucose (eisenfrei),<B>5 g</B> L-Asparagin, <B>1 g</B> ,sle;l#,1.-Kaliu,mpho#phat, <B>1 g</B> krist. Magnesiumsulfat (MgS04, <B>7</B> 1190) und<B>0,5 g</B> Calciumehlorid enthält. Die Nährlösung wird 20 Minuten bei 1201 sterilisiert.
Ge impft wird mit einer Sporensuspension von Reiskulturen in dest. Wasser und<B>5 %</B> Tween <B>80.</B>
Der Gehalt an Ferrirubin im Kulturmedium, wird wie folgt bestimmt: <B>10</B> mi Kulturfiltrat und <B>3 g</B> Natriumehlorid und <B>10</B> ml Benzylalkohcyl werden gut geschüttelt und zentri- fug#ert. Dieorganische Phase wird, filtnert, mit 3fachein Volumen Äther und Puffer pH <B>8</B> geschüttelt und erneut zentrifugiert.
Im wässragen Auszug wird die, Extinktion bei 440 my bestimmt und anhand -einer Eichkurve die mg rohes Ferrirubin abgelesen.
Nach<B>12-16</B> Tagen wird den Kulturen<B>1 g</B> Ferri- chlorid pro Liter zugesetzt und unter Zusatz von 2<B>%</B> Celit filtriert. Das Kulturfiltrat wird nun mit Natrium- chlorid gesätfigt und die klare Lösung zweimal mit <B>1/10</B> Volumen Benzylalkohol extrahiert. Die organische Phase wird mit Natriumsulfat getrocknet, filtriert und mit <B>3</B> Volumen Äther versetzt.
Das Benzylalkohol- Ätherg-emisch, extrahiert man bis zur Entfärbung mit destilliertem Wasser. Die wässrigen Auszüge werden vereinigt, die, Reste von Benzylalkohol mit Äther ent fernt und lyophilisiert, wobei man ein rotes Pulver erhält.
10,4<B>g</B> Rohprodukt werden einer 154stufigen Craig- Verteilung unterzogen. Als Verteilungssystern. dient ein Gemisch aus Benzylalkohol, n-Butylalkohol, gesättigte, wässrige NatriumchIK)r-i'dlösung und 0,001n wässrige Salzsäure im Voluraverhältnis <B>18: 18: 10: 30.</B>
Der rotbraune Farbstoff bildet ein einziges Ver teilungsmaximum. in Stufe,<B>128.</B> Dies entspricht ungefäh ren Verteil, ungskoeffizienten von K = 5,0- Die Fraktionen 116-140 werden zusammengefasst. Durch Zusatz von 2 Volumen Äther wird Ferrirubin in die wässrige Phase gedrängt.
Diese wird abgetrennt und die organische Schicht noch dreimal, mit wenig Wasser ausgesehüttelt. Die vereinigten wässrigen Lö- sunge,n werdien mit Natriumchlorid etwa halb gesättigt und mehrmals mit Phenol-Chloroform. <B>(1 kg</B> Phenol auf <B>1</B> Liter Chloroform) ausgeschüttelt, wobei der Farbstoff in die, organische Phase übergeht.
Die Extrakte werden an -einer Celite-Säule geklärt, mit dem doppelten Vo lumen Äther verdünnt und viermal ruit wenig Wasser ausgeschÜttelt, wobei der braunrote Farbstoff in die wässrige Phase geht. Man wäscht die wässrigen Lö sungen mehrmals mit Äther, um das Phenol vollständig zu entfernen und dampft sie im Vakuum ein. Der rot braune amorphe Rückstand (1,245<B>g)</B> wird,durch Um fallen aus Methanof-Ather weiter gereinigt.

Im Papierchromatogramm wurde Ferriruhih mit anderen Sideraminen verglichen, siehe Tabelle,. Die Methode ist folgende: Ein Blatt Whatman-Filterpapier Nr. <B>1</B> wird mit einem Gemisch aus Acoton, Wasser und gesättigter wässriger Natriumchloridlösung (Volumen-Ver- hältnis <B>6: 3: 1)</B> imprägniert und 1-0--15 Minuten an der Luft getrocknet.
Auf die Startlinie wird abwechs, lungsweise ein Tropfen einer methanolischen Lösung der Vergleichssubstanz (Startpunkte<B>1, 3, 5</B> und<B>7)</B> und des Ferrirubins (Startpunkte 2, 4,<B>6</B> und<B>8)</B> aufgetragen. Das Chromatogramm wird während etwa 20 Stunden entwickelt mit dem Fliessmittel tert.-Butanol/0,004-n, Salzsäure./gesätt. wäss. Natriumchloridlösung (2:<B>1: 1).</B> Die Flecke der Sideramine werden durch ihre Eigen farbe erkannt.
Trotz des relativ geringen Unterschiedes in den Rf-Werten einiger Sideramine ergibt sich nach der beschriebenen Methode eine eindeutige Unter scheidung.

Man extrahiert das erhaltene Kuliturfiltrat dreimal mit 1/20 Volumen Chloroform-Phenol <B>(1</B> Volumteü <B>: 1</B> Ge wichtsteil), trennt die organische Phase. ab und filtriert sie durch Celit. Dann, versetzt man die organische Lösung mit<B>3</B> Volumen Äther und extrahiert sie bis zur Ent- färbung mehrmals mit destilliertem Wasser.
Die wäss- rigen Auszüge werden vereinigt, zur Entfernung von Phenolresten mit Äther gewaschen, im Vakuum vom Äther befreit und anschliessend #lyophyhsiert. Das er haltene Rohprodukt wird wie in Beispiel<B>1</B> gereinigt.
In gleicher Weise, wie in Beispiel<B>1</B> beschrieben wird Ferrirubin erhalten, wenn man den Stamm Spiearia sp. M 4622 oder den Stamm Paecilomyces vario-ti M 4646 (bei 33') züchtet und das Kulturmedium wie beschrieben aufarbeitet.
<I>Beispiel<B>3</B></I> 435 mg Ferrirubin werden in<B>30</B> ral Wasser gelöst und<B>1,0 g</B> 8-Hydroxychino-Iin, gelöst in etwa<B>15</B> ml Methanol, zugegeben. Nach 24,ständigem -Rühren bei Zimmertemperatur wird vom schwarzen Niedtrschlag (Hydroxychinol-in-Eisenkoimplex) abfikriert und das blass geibliche Filtrat dreimal mit Chloroform ausgezogen, um das überschüssige Reagens zu -entfernen.
Darauf dampft man die. wässrige Lösung ein -und erhält das eisenfreie Ferrirubin als blassgelben amorphen Rück stand. Ausbeute<B>390</B> mg.
Eine in<B>D20</B> gelöste Probe der Verbindung zeigt im NMR-Spektrum (aufgenommen mit einem Varian- Spektrographen, Modell<B>A 60)</B> folgen65 Signale: <B>1,86</B> ppm (14 H); <B>2,38</B> ppm (4 H); <B>2,92</B> ppm <B>(1</B> H); <B>3,79</B> ppm <B>(10</B> H); 4,39 ppm (4 11); <B>6,03</B> ppm (2 H).
Angaben der chemischen Verschiebungen in ö- Werten gegenüber Tetramethylsilan = <B>0.</B> Die, durch Integration erhaltenen Protonenzahlen bedeuten reine VerhältmAszahlen, wobei das kleinste Signal als<B>1</B> an genommen, wurde.<B>Alle.</B> Signale bilden breite Signal anhäufungen ohne deutliche Auflösung. Die Signalhaufen bei<B>3,79</B> und<B>6,03</B> ppm besitzen deutliche<B>Doppel-</B> maxima, sind aber offenbar keine, reinen Dubletten.
Bei der Elementaranalyse werden folgende Werte gefunden:<B>C 51,12;</B> H<B>7,29; N 13,00;</B> Acetyl <B>0</B> <I>Beispiel 4</I> <B>150</B> mg eisenfreies. Ferrirubin werden mit 4 ml 6n Salzsäure in ein Glasrohr eingeschmolzen und 24 Stunden auf<B>110'</B> erhitzt. Den Eindampfrückstand des Hydrolysats löst man in wenig Wasser, verdünnt mit Feinsprit auf etwa<B>30</B> ml und hydriert in Gegenwart von<B>50</B> mg vorhydriertem Platinoxyd über Nacht.
Eine Probe des Eindampfrückstandes prüft man durch zwei dimensionale Paplerchromatographie (Phenol-Wasser <B>8:2</B> und, Butanol-Eisessig-Wass,er 4:<B>1: 1;</B> Ninhydrin) sowie durch eine Analyse nach Moore und Stein auf Aminosäuren. Beide- Methoden zeigen die Anwesenheit von Ornfthin, Serin und Glycin. Serin und Glycin sind im molekularen Verhältnis 2:<B>1</B> vorhanden.
<I>Beispiel<B>5</B></I> <B>28</B> mg eisenfreies Ferrirubin, gelöst in<B>1</B> ml Methanol, werden mit<B>25</B> mg Eisenchlorid in 2 ral Methanol ver mischt. Es tritt sofort eine, tiefe violettschwarze, Farbe auf, die nach Abpuffern der Lösung mit etwas krist. Natriumacetatnach rotbraun umschlägt.
Nach lstÜndilgem Stehen wird im Vakuum zur Trockne verdampft, der Rückstand in<B>5</B> ml Wasser gelöst, mit Natriumchlorid etwa halb; gesättigt und das Ferrirubin mit Benzy1- a,lkohol extrahiert. Die Extrakte, werden mit gesättigter Natriumchloridlösung gewaschen und mit Natriumsulfat getrocknet. Die Lösung wird mit Äther verdünnt und das, Sideramin wieder mit Wasser ausgezogen. Durch Eindampfen der mit Äther gewaschenen Auszüge erhält man Ferrirabin, etwa<B>25</B> mg orangprotes Pulver.
Durch Papierchromatographie in den Lösungsmittelsystemen <B>1</B> und II (siehe, Tabelle<B>1)</B> wird gezeigt, dass der auf diesem Wege gewonnene Eisenkomplex mit dem direkt isolierten identisch ist.
<I>Beispiel<B>6</B></I> Zu einer Aufschlämmung von<B>100</B> mg 10%iger Palladiumkohle in<B>5</B> ml Mothanol gibt man eine Lö sung von 149 mg Ferrirubin in<B>5</B> ml Methanoi und rührt das Gemisch bei Zimmertemperatur in einer Wasserstoffatmosphäre. Die Wasserstoffaufnahme ist anfangs sehr rasch. Nach etwa<B>1</B> Stunde sind etwa<B>3</B> Mol Wasserstoff aufgenommen, und die Hydrierung kommt zum Stillstand.
Nun wird vom Katalysator abfiltriert. Zum orangebraunen Filtrat fügt man eine Lösung von <B>100</B> mg Eisen(IR)#-chlo#r-id in Methanel, hitzu, um allen- falls reduziertes Eisen zu ersetzen und puffert die Lö sung ab durch Zugabe von etwas kristallisieritem Na- triumacetat. Nach 4stündigem Stehen wird die Lösung im Vakuum eingedampft, der Rückstand in Wasser aufgenommen und die Lösung mehrmals mit Phenol- Chloroform <B>(1<I>g .</I> 1</B> m1) ausgeschüttelt.
Der durch Celit filtrierte Phenol-Chloroform-Auszug wird mit 2 Volumen Äther verdünnt, dreimal mit dest. Wasser ausgeschüttelt und die mit Äther gewaschenen wässrigen Lösungen im Vakuum eingedarapft. Man erhält Hexahydroferriru-bin als orangegelbes amorphes Pulver; Ausbeute.<B>132</B> mg.

Die Fraktionen 12 bis<B>25</B> werden in üblicher Weise mit Phenol-Chloroform aufgearbeitet. Man erhält 202 mg Ferrichrysin, das durch Kristallisatio-n aus Äthylalkohol weiter gzreinigt wird. Die rotorangen Kristalle zersetzen sich" wie authentisches Ferrichrysin, langsam bei etwa 250#260', ohne, zu schmelzen.

Paecilomyces varioti M 4646 was isolated at the Federal Research Institute in Wädenswil, Canton Zurich. The organism forms abundant, brownish-yellow, elongated oval conidia at the ends of long, pointed conidia, which are arranged vertically and diverge strongly.
The organism belongs to the genus Paecilomyces Bainier <B> 1907 </B> and belongs to the species Paecilouiyces varioti Bainer <B> 1907 </B> perhaps the only, very variable species of the genus (cf. Raper and Thom, p . <B> 688-692). </B>

An extract prepared in the same way from non-inoculated nutrient solution is used as a comparison sample.
After <B> 12-16 </B> days, the cultures reach an activity that corresponds to <B> 50-200 </B> mg of pure ferrirubin. When the cultivation is complete, <B> 1 g </B> per liter of ferric chloride can be added to the nutrient solution, the existing desferrirubin being converted into ferricubin. The mycelium is separated from the culture filtrate, preferably in the presence of a feeding aid, e.g. B. Hyflo-Supereel, after which the majority of the ferrirabin is found in the culture filtrate.
However, significant amounts of it still stick to the mycelium. It is therefore advantageous to wash it off well. For this purpose, for example, water or aqueous alcohols such as aqueous. Methanol.
For the isolation of ferrirubin from the culture filtrate, you can proceed according to methods known to s-I and z. B .. use one of the following working methods or combinations thereof.
1. One can use adsorbents, e.g. B. activated carbons such as Norit, activated earths such as Franconite, Fuller earth or Floridin, or resin adsorbers such as Asmit. The elution of the adsorbates is expediently carried out with mixtures of water-miscible organic solvents with water, eg.
B. with water-methanol-, water-pyridine-, diluted acetic acid # methanol or water-methanel-glacial acetic acid # -butanol mixtures. The mixture of water (4 parts by volume) and pyridine (1 part by volume) has proven to be particularly advantageous for the elution of a frankonite or norite adsorbate.
2. Ferrirubin can also be removed from an aqueous solution using organic solvents. For these extraction processes, higher organic alcohols, e.g. B. B.ünzylalkohol or isopropyl alcohol, particularly proven. Conveniently, the aqueous phase is an inorganic salt, for. B. ammonium sulfate or sodium chloride added.

With iron-free ferrirubin, the intensity of the iron chloride reaction only decreases noticeably after about 2 hours. A similarly high level of resistance to acids has so far been observed in the sideramine series, only with the biologically inactive ferric chromium <B> A </B>.

Ferrirubin, desferrirubin and analogues are growth substances for various microorganisms; therefore, they can be used in the culture of such organisms.
The iron-containing compounds have anti-analgesic properties and can therefore be used as remedies. Desferrirubin and its derivatives also have valuable pharmacological properties. The content of ferrirubin in the culture medium is determined as follows: <B> 10 </B> ml of culture filtrate and <B> 3 g </B> sodium chloride and <B> 10 </B> ml benzyl alcohol are shaken well and centrifuged adds. The organic phase is filtered, shaken with 3 times one volume of ether and buffer pH 8 and centrifuged again.
The extinction at 440 my is determined in the aqueous extract and the mg of crude ferricubin is read off a calibration curve.

The culture filtrate is now saturated with sodium chloride and the clear solution is extracted twice with 1/10 volume of benzyl alcohol. The organic phase is dried with sodium sulfate, filtered and mixed with 3 volumes of ether.
The mixture of benzyl alcohol and ether is extracted with distilled water until it is discolored. The aqueous extracts are combined, the residues of benzyl alcohol are removed with ether and lyophilized, a red powder being obtained.
10.4 g of crude product are subjected to a 154-stage Craig distribution. As a distribution system. A mixture of benzyl alcohol, n-butyl alcohol, saturated, aqueous sodium chIK) r-i'd solution and 0.001N aqueous hydrochloric acid in a volura ratio of <B> 18: 18: 10: 30. </B> is used
The red-brown dye forms a single distribution maximum. in stage, <B> 128. </B> This corresponds approximately to distribution coefficients of K = 5.0- The fractions 116-140 are combined. By adding 2 volumes of ether, ferrirubin is forced into the aqueous phase.
This is separated off and the organic layer is shaken out three more times with a little water. The combined aqueous solutions are about half saturated with sodium chloride and several times with phenol-chloroform. <B> (1 kg </B> phenol to <B> 1 </B> liter of chloroform) shaken out, whereby the dye passes into the organic phase.
The extracts are clarified on a Celite column, diluted with twice the volume of ether and shaken out four times with a little water, the brownish-red dye going into the aqueous phase. The aqueous solutions are washed several times with ether in order to completely remove the phenol and evaporated in vacuo. The red-brown amorphous residue (1.245 g) is further purified by falling from methanof ether.
The pure compound is very hygroscopic. Both methods show the presence of ornfthin, serine and glycine. Serine and glycine are present in a molecular ratio of 2: <B> 1 </B>.
<I>Example<B>5</B> </I> <B> 28 </B> mg of iron-free ferrirubin, dissolved in <B> 1 </B> ml of methanol, are mixed with <B> 25 </ B > mg iron chloride mixed in 2 ral methanol. Immediately a deep purple-black color appears which, after the solution has been buffered with a little bit of crystalline. Sodium acetate turns reddish brown.

The hydrogen uptake is very rapid at first. After about <B> 1 </B> hour, about <B> 3 </B> mol of hydrogen have been absorbed and the hydrogenation comes to a standstill.
The catalyst is then filtered off. A solution of <B> 100 </B> mg iron (IR) # - chlo # r-id in methanel is added to the orange-brown filtrate, heated to replace any reduced iron and the solution is buffered by adding some crystallized sodium acetate. After standing for 4 hours, the solution is evaporated in vacuo, the residue is taken up in water and the solution is extracted several times with phenol-chloroform (1 g. 1 m1).
The phenol-chloroform extract filtered through Celite is diluted with 2 volumes of ether, three times with distilled water. Shaken out water and the aqueous solutions washed with ether are evaporated in vacuo. Hexahydroferriru-bin is obtained as an orange-yellow amorphous powder; Yield. 132 mg.
