Verfahren zur Herstellung von tertiären Diäthanolaminen
Die Erfindung betrifft ein Verfahren zur Herstellung von tertiären Diäthanolaminen, die sich durch bakterizide und fungizide Eigenschaften auszeichnen.

Als Ausgangsstoffe kommen beispielsweise folgende primäre Amine in Betracht : y, y, y-Tri- (4'-chlorphenyl)-propylamin, y-Phenyl-y, y-bis- (4'-chlbrphenyl)-propylamin, y- (3'-Chlorphenyl)-γ,γ-bis-(4'-chlorphenyl)- propylamin, y- (3'-Fluorphenyl)-y, y-bis- (4'-chlorphenyl)- propylamin, γ,γ-Bis-(4'-Fluorphenyl)-γ-(4'-chlorphenyl)- propylamin, r- (3', 4'-Dichlorphenyl)-r, r-bis-(4'-chlorphenyl)- propylamin.
Diese primären Amine können z. B. auf einfache Weise hergestellt werden, indem man Triphenylcarbinole mit Cyanessigsäure in Gegenwart einer niedrigmolekularen aliphatischen Carbonsäure und eines Salzes eines Metalls aus der II. Nebengruppe des Periodensystems unter R ckflu¯ erhitzt und das gebildete ¯,¯,¯-Triphenylpropionitril katalytisch hy driert.
Das Verfahren gemäss der Erfindung wird z. B. in der Weise durchgeführt, dass man die primären Amine mit Athylenchlorhydrin bei erhöhter Temperatur umsetzt. Vorteilhaft erhitzt man das Reaktionsgemisch auf 100-150 C. Die Verwendung eines Lösungsmittels ist zweckmässig, jedoch nicht unbedingt erforderlich. Man verwendet gegebenenfalls Alkohole, wie Athanol oder Butanol, aroma tiische Kohlenwasserstoffe, wie Toluol, oder auch höhersiedenden Ather. Es ist von Vorteil, zur Bindung der bei der Reaktion frei werdenden Halogen wasserstoffsäure eine anorganische oder organische basische Verbindung, beispielsweise ein Alkali- carbonat, gebrannten Kalk oder Dimethylanilin, zuzusetzen.
Zur Isolierung der Produkte behandelt man das Reaktionsgemisch mit verdünnten wässrigen Alkalien und einem geeigneten organischen Lösungsmittel, wie Ather oder Methylenchlorid. Nach dem Abdampfen des Lösungsmittels erhält man die Verfahrenserzeugnisse in Form der freien Basen, die gewünschtenfalls in üblicher Weise durch Behandlung mit anorganischen oder organischen Säuren, vorteilhaft in Gegenwart eines organischen Lösungsmittels, in die entsprechenden Säureadditionssalze überführt werden können. Als Lösungsmittel werden hier Alkohole, wie Athanol oder Methanol, Äther, wie Diäthyläther, oder auch Aceton verwendet.
Anstelle mit Athylenhalogenhydrinen kann man die primären Amine zur Uberführung in Diäthanol- amine in besonders einfacher Weise auch mit Athylenoxyd umsetzen. Dabei arbeitet man vorteilhaft in Gegenwart von organischen Lösungsmitteln in ge schlossenen Gefässen bei erhöhten Temperaturen, vorzugsweise zwischen 80-160 C. Als Lösungs- mittel kommen z. B. Alkohole und Äther, vorzugsweise aber aromatische Kohlenwasserstoffe, wie Benzol oder Toluol, in Betracht.
Die so erhaltenen tertiären Diäthanolamine sind sehr zÏhe, gelbe, nur unter starker Zersetzung destillierbare Öle oder kristalline Substanzen. Mit anorganischen oder organischen Säuren bilden sie Salze, die zum Teil in kristallisierter Form anfallen. Als anorganische Säuren seien beispielsweise
Chlorwasserstoffsäure, BromwasserstoffsÏure,
JodwasserstoffsÏure, Schwefelsäure,
PhosphorsÏure oder Amidosulfonsäure, genannt. Als organische Säuren kommen z. B. Essigsäure, WeinsÏure, ¯pfelsÏure oder Äthylendiamintetraessigsäure in Betracht.
Die freien tertiären Diäthanolamine können er findungsgemäss in entsprechende quaternäre Ammo niumsalze übergeführt werden. Zu diesem Zweck lässt man auf sie Alkylierungsmittel, wie reaktions fähige Ester, vorteilhaft bei erhöhten Temperaturen, einwirken. Geeignet sind hierfür beispielsweise die Ester von Alkoholen der Formel RUT-OH mit Chlor-, Brom oder JodwasserstoffsÏure, SchwefelsÏure oder einer aromatischen bzw. aliphatischen Sulfonsäure.
Die Gegenwart eines Lösungsmittels ist nicht erforderlich, aber in den meisten Fällen für die Erzielung guter Ausbeuten vorteilhaft. In Frage kommen aromatische oder aliphatische Kohlenwasserstoffe, ali phatische Ester, Äther, Ketone oder Alkohole. Beispielsweise seien genannt : Benzol, Toluol, Petrol äther, Äthylazetat, DiÏthylÏther, Dioxan, Tetrahydrofuran, Aceton, Methanol oder Äthanol. Je nach der Reaktionsfähigkeit des verwendeten Alky lierungsmittels arbeitet man bei wenig oder stark erhöhter Temperatur. Im allgemeinen verläuft die Umsetzung bei der Siedetemperatur des Lösungs- mittels genügend rasch. In manchen Fällen kann es jedoch zweckmässig sein, die Reaktion bei Temperaturen zwischen 100 und 200 C, im Autoklaven, vorzunehmen.
Die Verfahrenserzeugnisse fallen nach Beendigung der Reaktion entweder direkt in kristalliner Form an oder können durch Behandlung mit einem geeigneten Lösungsmittel zur Kristallisation gebracht werden.
Die quaternären Ammoniumsalze bilden in den meisten Fällen farblose Kristalle, die sich in Wasser je nach Art der Substitution der Benzolkerne und der Natur des eingesetzten Alkylierungsmittels mehr oder weniger gut lösen. Leicht löslich sind sie zum grössten Teil in niedrigmolekularen aliphatischen ein-oder mehrwertigen Alkoholen.
Die nach dem Verfahren gemäss der Erfindung erhältlichen tertiären und quaternären Amine bzw. deren Salze sind ungiftig und besitzen wertvolle therapeutische Eigenschaften, von denen insbesondere bakteriostatische, bakterizide, fungistatische und fungizide Eigenschaften im Vordergrund stehen.
Beispielsweise wirkt das
N- [y,-,,, ,-Tri- (4-chlorphenyl)-propyl]-N, N-bis (ss-hydroxyäthyl)-N-methyl-ammonium- methylsulfat oder das N- [y, y, y-Tri- (4'-chlorphenyl)-propyl]- diäthanolamin-hydrochlorid gegen grampositive Keime wie Staphylococcus aureus,
Streptococcus haemolyticus oder
Corynebacterium diphtheriae noch in der Verdünnung von 1-4,/ml bakteriostatisch. Gegenüber gramnegativen Keimen, wie E.
Coli oder Pseudomonas aeruginosa, liegt die untere Grenze der bakteriostatischen Wirksamkeit des N- [7,, y, 7-Tri- (4'-chlorphenyl)-propyl]-N, N-bis (/ ?-hydroxyäthyI)-N-methyl-ammonium- methyl'sulfats bei 15 r/ml.
Was die bakterizide Wirksamkeit der Verfahrenserzeugnisse anbelangt, so beträgt z. B. der Phenolkoeffizient des N- [y, -Tri- (4-chlorphenyl)-propyl]-N, N-bis (ss-hydroxyäthyl)-N-methyl-ammonium- methylsufats, der ein Mass für die bakterizide Wirkung einer Substanz darstellt, gegenüber Staphylokokken etwa 400 und gegenüber E. Col und Bacterium typhi 200.
Der Phenolkoeffizient des N- [, y, y-Tri- (4-chtorphenyl)-propyl]-diäthanol- amin-hydrochlorids gegenüber Staphylokokken liegt zwischen 400 und 800. Auf Grund dieser Eigenschaften eignen sich die neuen Verbindungen beispielsweise für die Verwendung als Desinfektionsmittel auf den verschiedensten GebSeten.
Die fungistatische Wirkung der Verfahrenserzeugnisse ist ebenfalls stark. Beispielsweise werden pathogene Hefen, wie Candida albicans, oder apathogene Schimmelpilze, wie Penicillium glaucum, noch in Konzentrationen von 50, /mol des N- [y, y,)-Tri- (4'-chlorphenyl)-propyll-N, N-bis (ss-hydroxyäthyl)-N-methyl-ammonium- methylsulfats im Wachstum gehemmt.
Gegenüber menschenpathogenen Hautpilzen, wie Microsporum gypseum, Epidermophyton-und Trichophyton-Arten, liegt die fungistatische Wirksamkeit des N- [y, y,-Tri-(4'-chlorphenyl)-propyl] 4iäthanol- amin-hydrochlorids in der Konzentration zwischen 8 und 16 γ/ml, wÏhrend die untere fungistatische Wirkungsgrenze des N-[γ,γ,γ-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-N-methyl-ammonium methylsulfats etwa 30 γ/ml betrÏgt.
Die bakterizide Wirksamkeit der neuen Verbindungen wurde im sogenannten Rideal-Walker-Test ermittelt. Die Bestimmung der bakteriostatischen Wirkung erfolgte nach der bekannten Methode von Wright (The Lancet, Jahrgang 1912) im Reihen verdünnungstest je nach Art der verwendeten Keime in Bouillon oder in Serumbouillon als Nährmedium mit kleiner Einsaat. Die Ablesung wurde nach 18bis 20stündiger Bebrütung bei 37 C an der Grenze zwischen klarer und trüber Versuchslösung vorgenommen.
Die Prüfung auf fungistatische Wirksamkeit der Verbindungen wurde in Anlehnung an die von SchraufstÏtter, Richter und Dittscheid im Archiv für Dermatologie und Syphilis , Band 188 (1949), Seite 259, beschriebene Methode im Reihenverdünnungstest vorgenommen.
Abgesehen von der guten Wirksamkeit gegen zahlreiche bakterielle Keime, Hautpilze, pathogene Hefen und apathogene Schimmelpilze, sind die Verfahrenserzeugnisse darüber hinaus wertvolle Zwi schenprodukte für die Herstellung von Arzneimitteln.
Die Verfahrenserzeugnisse können als solche oder in Form von galenischen Zubereitungen, beispielsweise als Gelees, Puder, Salben, Pasten,, Schüttelmixturen, Tinkturen, L¯sungen, Suspensio- nen oder Tabletten unter Beimischung von nichttoxischen, pharmazeutisch üblichen organischen oder anorganischen Trägersubstanzen, angewendet werden.
Die Wirkstoffe können den galenischen Zubereitungen beispielsweise in einer Dosierung von 0, 1 bis 1 % beigegeben werden. Bei Anwendung der Ver fahrenserzeugnisse in Form eines Gelees hat sich eine mittlere Dosis von 0,5 % der Wirkstoffe als vorteilhaft erwiesen. Tabletten können 1 bis 10 Milligramm der Wirkstoffe enthalten, vorzugsweise 2 bis 5 Milligramm.
Beispiel 1 N-[γ,γ,γ-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-amin
203 g y, y, y-Tri- (4'-chlorphenyl)-propylamin und 65 cm3 Athylenoxyd löst man in 600 cm3 Benzol und erhitzt die Mischung in einem Autoklaven 5 Stunden auf 120 C. Nach dem Abdestillieren des Lösungsmittels erhält man 250 g rohes N-[γ,γ,γ-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (ss-hydroxyäthyl)-amin in Form eines gelben, sehr zähen Oles, das f r die Herstellung von Salzen oder für die Quaternisierung direkt verwendet werden kann.
Löst man das rohe Amin in etwa der doppelten Menge Diisopropyl- äther und lässt den Ansatz längere Zeit stehen oder impft an, so findet Kristallisation statt, und man erhält das N- [y, y, y-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (-hydroxyäthyl)-amm als farbloses Kristallpulver vom Schmelzpunkt 109 bis 110 C.
Durch Versetzen einer ätherischen Lösung des Amins mit einem kleinen Überschuss alkoholischer Salzsäure wird das entsprechende Hydrochlorid der Verbindung erhalten, das nach dem Umkristallisieren aus Alkohol/Diisopropyläther bei 208-211 C schmilzt.
Beispiel 2 N- [y, y, y-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (ss-hydroxyäthyl)-amin
Man erhitzt eine Mischung von 78 g y, y, y-Tri (4'-chlorphenyl)-propylamin, 40 g Athylenchlorhydrin und 12 g gebranntem Kalk unter Rühren 20 Stunden auf 110 C. Nach dem Erkalten behandelt man die erhaltene zähe Masse mit Chloroform und ver dünnter Natronlauge, trennt die Chloroformschicht ab, wäscht sie mit Wasser, trocknet sie über Magne siumsulfat und destilliert schliesslich das Lösungs- mittel ab. Den Rückstand-löst man in 100 cm3 Di isopropyläther und impft an.
Das
N-[γ, γ, γ-Tri-(4'-chlorphenyl)-propyl]-N,N-bis (/?-hydroxyäthyl)-amin kri, stallisiert langsam aus und wird abgesaugt. Das Verfahrensprodukt schmilzt bei 108-110 C, die Ausbeute beträgt 70 g.
Beispiel 3 N- [y- (3'-Fluorphenyl)-y, y-bis- (4"-chlorphenyl)- propyl]-N, N-bis-(¯-hydroxyÏthyl)-amin
Man löst 109 g y- (3'-Fluorphenyl)-y, y-bis- (4"- chlorphenyl)-y, y-bis- (4"-chlorphenyl)-propylamin und 41 cm?Ätbylenoxydin 300 cm3 Benzol und erhitzt die Mischung im Autoklaven 5 Stunden auf 120 C.
Nach dem Abdestillieren des Lösungsmittels hinterbleibt das
N-[γ-(3'-Fluorphenyl)-γ,γ-bis-(4"-chlorphenyl) propyl]-N, N-bis-(ss-hydroxyäthyl)-amin in Form eines hellbraunen, sehr zähen t5les. Die Ausbeute beträgt 130 g.
Beispiel 4 N- [y, y, y-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-N-methyl-ammonium methylsulfat
Man löst 19 g N-[γ,γ,γ-Tri-(4'-chlorphenyl)-pro- pyl]-N,N-bis-(¯-hydroxyÏthyl)-amin und 5,5 g Di methylsulfat in 60 cmS Benzol und erhitzt die Mischung 15 Minuten auf dem Dampfbad unter R ckfluss. Danach setzt man 60 cm3 Diisopropyläther zu, wobei ein 01 ausfällt, das beim Stehen langsam, beim Animpfen schnell erstarrt. Nach dem Absaugen erhÏlt man 20 g N-[γ,γ,γ-Tri-(4'-chlorphenyl]-propyl]-N, N-bis (fl-hydroxyäthyl)-N-methyl-ammonium- methylsulfat vom Schmelzpunkt 110-113 C (unter schwacher Zersetzung).
Zur Reinigung kann man das Rohpro- dukt umkristallisieren, indem man es in 80 ce warmem Athylacetat löst, 100 cm3 Ather zufügt und animpft. Die erhaltenen farblosen Kristalle schmelzen bei 111-113 C unter schwacher Zersetzung.
Beispiel S N-(γ,γ,γ-Tri-(4'-chlorphenyl)-N, N-bis (ss-hydroxyäthyl)-N-allyl-ammonium-bromid
Man erhitzt eine Lösung von 24 g N-[y, y, y-Tri- (4'-chlorphenyl)-propyl]-N,N-bis-(¯-hydroxyÏthyl)amin und 7,2 g Allylbromid in 50 cm3 Alkohol 8 Stunden unter Rückfluss. Nach Zugabe von 200 cm3 Diisopropyläther findet Kristallisation statt, und man erhält 17 g N- [y, y, y-Tri- (4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-N-allyl-ammonium-bromid, das aus Alkohol/Diisopropyläther umkristallisiert werden kann. Das Verfahrenserzeugnis bildet farblose Kristalle vom Schmelzpunkt 210-211 C (unter Zersetzung).
Beispiel 6
N-[γ-(3'-Fluorphenyl)-γ,γ-bis-(4"-chlorphenyl) propyl]-N, N-bis-(ss-hydroxyäthyl)-N-allyl- ammonium-bromid
Man erhitzt eine Lösung von 23,1 g N- [y- (3'- Fluorphenyl)-γ,γ-bis-(4"-chlorphenyl)-propyl]-N,N- bis-(¯-hydroxyÏthyl)-amin und 7,2 g Allylbromid in 50 cm Alkohol 7 Stunden unter Rückfluss. Dann destilliert man das Lösungsmittel vollkommen ab und Iöst den Rückstand in 50 cm3 Athylacetat. Beim Stehen findet langsam Kristallisation statt, die durch Animpfen beschleunigt werden kann.
Nach Zugabe von 50 cm3 Diisopropyläther saugt man ab und erhÏlt 10 g N- [y- (3'-Fluorphenyl)-y, y-bis- (4"-chlorphenyl)- propyl]-N, N-bis-(¯-hydroxyÏthyl)-N-allyl ammonium-bromid vom Schmelzpunkt 190-193¯ C. Durch Umkristallisieren aus Isopropanol/Diisopropyläther kann man das Salz weiter reinigen. Es schmilzt dann bei 193 bis 194¯ C.
Beispiel 7
N-[γ,γ,γ-Tri-(4'-chlorphenyl)-propyl]N,N-bis (3-hydroxyathyl)-N-athyl-ammonium-p- toluolsulfonat
Man l¯st 24 g N-[y, y, y-Tri- (4'-chlorphenyl)- propyl]-N, N-bis-(¯-hydroxyÏthyl)-amin und 12 g p-Toluolsulfonsäurëäthylester in 50 cm3 Alkohol und erhitzt die Lösung 6 Stunden unter RückfluB. Nach Zugabe von 100 cm* Dlisopropyläther und Absaugen erhält man 26 g farbloses N- [y, y, y-Tri- (4'-chlorphenyl)-propyl]-N, N-bis (ss-hydroxyäthyl)-N-äthyl-ammonium- p-toluolsulfonat vom Schmelzpunkt 157-158 C. Das Verfahrenserzeugnis kann aus Isopropanol umkristallisiert werden,
wobei sich der Schmelzpunkt nicht mehr ver ändert.
Beispiel 8 N- [y, y, y-Tri- (4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-N-(p-chlorbenzyl)-ammonium chlorid
Man vermischt 19 g N-[γ,γ,γ-Tri-(4'-chlorphenyl)- propyl]-N,N-bis-(¯-hydroxyÏthyl)-amin und 7 g p Chlorbenzylchlorid und erhitzt das Gemisch im offenen Kolben 8 Stunden auf dem Dampfbad. Dabei wird die Mischung langsam dick und erstarrt schliesslich zu einer harten Kristallmasse. Man l¯st das Produkt in 450 cm3 siedendem Alkohol, filtriert, setzt 500 cm3 Petroläther zu und impft an.
Am nächsten Tag saugt man ab und erhält 10 g N-[γ,γ,γ-Tri-(4'-chlorphenyl)-propyl]-N, N-bis (¯-hydroxyÏthyl)-N-(p-chlorbenzyl) ammonium-chlorid vom Schmelzpunkt 208-210 C.
Beispiel 9 N-[γ,γ,γ-Tri-(4-chlorphenyl)-propyl]-N, N, N-tri (ss-hydroxyäthyl)-ammoniumchlorid
24 g N- [y, y, γ- Tri-(4-chlorphenyl)-propyl]-N, Nbis-(¯-hydroxyÏthyl)-amin und 4,8 g Äthylenchlorhydrin werden 15 Stunden lang auf 120-130 C erhitzt. Die Schmelze behandelt man dann mit etwa 50 cm3 Athylacetat, wobei Kristallisation stattfindet.
Man erhÏlt nach dem Absaugen 10 g N- [y, y, y-Tri- (4-chlorphenyl)-propyl]-N, N, N-tri (¯-hydroxyÏthyl)-ammoniumchlorid vom Schmelzpunkt 233-235 C (unter Zersetzung).
Das Salz lässt sich aus Alkohol/Diisopropyläther umkristallisieren, ohne dass sich der Zersetzungspunkt ändert.

These primary amines can e.g. B. be prepared in a simple manner by refluxing triphenylcarbinols with cyanoacetic acid in the presence of a low molecular weight aliphatic carboxylic acid and a salt of a metal from the II. Subgroup of the periodic table and catalytically hydrogenating the ¯, ¯, ¯-triphenylpropionitrile formed.
The method according to the invention is e.g. B. carried out in such a way that the primary amines are reacted with ethylene chlorohydrin at elevated temperature. The reaction mixture is advantageously heated to 100-150 ° C. The use of a solvent is advantageous, but not absolutely necessary. If appropriate, alcohols such as ethanol or butanol, aromatic hydrocarbons such as toluene or higher-boiling ethers are used. It is advantageous to add an inorganic or organic basic compound, for example an alkali carbonate, quick lime or dimethylaniline, in order to bind the hydrohalic acid liberated in the reaction.

Instead of using ethylene halohydrins, the primary amines can also be reacted in a particularly simple manner with ethylene oxide for conversion into diethanolamines. It is advantageous to work in the presence of organic solvents in closed vessels at elevated temperatures, preferably between 80-160 C. The solvents used are e.g. B. alcohols and ethers, but preferably aromatic hydrocarbons such as benzene or toluene, into consideration.
The tertiary diethanolamines obtained in this way are very viscous, yellow oils or crystalline substances that can only be distilled with severe decomposition. They form salts with inorganic or organic acids, some of which are obtained in crystallized form. Examples of inorganic acids are
