worin R für Wasserstoff oder einen gesättigten oder ungesättigten Kohlenwasserstoffrest, der einen ein wertigen Substituenten tragen kann, steht und X Halogen bedeutet, gelangt,
indem man die entspre chenden Ester der allgemeinen Formel II N CHs O I O=C-R worin R obige Bedeutung besitzt, bei Temperaturen von 0 bis - 2011 halogeniert.
Das Verfahren wird beispielsweise so ausgeführt, dass Ester von 8 - Oxychinaldmen der allgemeinen Formel 1I in einem geeigneten Lösungsmittel, z. B.
Äthylenchlorid, Trichloräthylen, Chloroform, Tetra chlorkohlenstoff oder Dimethylformamid, vorzugs- weise in Gegenwart eines Puffers wie Natriumacetat. Natriumborat oder Natriumphosphat bei Tempera turen vorzugsweise zwischen 0 und - 200 halogeniert werden.
Die Reaktionslösung wird dann durch Fil- tration von unverbrauchten Puffersubstanzen und während der Reaktion gebildeten anorganischen Sal zen befreit.
Das nicht verbrauchte Halogen wird durch Ausschütteln mit wässeriger Natriumbisulfit- oder Natriumnhiosulfatlösung entfernt und die orga nische Phase nach dem Nachwachen mit wässeriger Natriumacetatlösung und/oder mit Wasser, Trocknen über Natriumsulfat, im Wasserstrahlvakuum zur Trockne eingeengt.
Als Rückstand erhält man die mono-(5) halogenierten Ester der allgemeinen For mel I.
Die nach dem vorliegenden Verfahren herge stellten, bisher unbekannten Ester der 5-Halogen-8- oxychinaldine sind feste Basen. Sie sind schlechter wasserlöslich als die 5-Halogen-8-oxychinaldine und werden aus diesem Grunde schlechter als diese resor biert.
Sie zeichnen sich durch therapeutisch verwert- bare pharmakodynamische Eigenschaften aus, indem sie eine erhöhte Wirksamkeit gegen pathogene Keime und auch andere Krankheitserreger aufweisen.
Die neuen Verbindungen sollen in der Therapie als Oberflächen-Antiseptica verwendet werden, wobei ihre schlechte Wasserlöslichkeit resp. Resorbierbar- keit eine besonders gute System-Verträghchkeit ge- währleistet. Sie dienen ferner als Zwischenprodukte zur Herstellung von Medikamenten.
In den nachfolgenden Beispielen, welche die Aus- führung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, sind die Schmelzpunkte in Celsiusgraden und unkor- rigiert angegeben.
Beispiel 1 S-Chlor-8-benzoxychinaldin Zu einer Lösung von 239,0g 8-Oxychinaldüi (Smp. 73-75) in 350 ccm Pyridin werden unter Kühlen und Umschwenken portionenweise 328,
G <B>g</B> Benzoylchlorid zugegeben. Nachdem der Ansatz unter Feuchtigkeitsausschluss 4 Stunden bei Rau#m- temperatur stehengelassen wurde, wird der Ansatz im Vakuum zu einem dickflüssigen 41 eingeengt.
Dieses wird in 500 ccm Chloroform aufgenommen und mit insgesamt 750 ccm einer 5-proz. Natrium- bicarbonatlösung und anschliessend mit 500 ccm Wasser ausgezogen. Die wässerigen Teile werden mit insgesamt 500 ccm Chloroform nachgewaschen. Die vereinigten Chloroformteile werden über Na triumsulfat getrocknet und im Vakuum zur Trockne eingeengt.
Das 8-Benzoxychinaldin destilliert bei 161-1700/0,08 mm Hg. Das Destillat ergibt nach zweimaligem Umkristallisseren aus Isopropanol das analysenreine 8-Benzoxychinaddin mit dem konstan ten Smp. 76-79.
Zu einer Lösung von 39,5 g 8-Benzoxychinaldin (Smp. 76-79) in 250 ccm Dimethylformamid wer den unter Rühren und Kühlen in einem Eis-Koch- salz-Bad 26,6 g Chloreingeleitet und anschliessend wird der Ansatz bei Zimmertemperatur noch 1/2 Stunde weitergerührt. Nach dem Durchleiten von Stickstoff durch die Lösung wird diese mit 750 ccm Wasser allmählich versetzt,
wobei das zuerst schmie rig ausgefallene Produkt kristallin ausfällt. Dieses wird abgenutscht und mit kaltem Wasser solange nachgewaschen, bis das Filtrat nicht mehr sauer ist.
Das getrocknete Rohprodukt wird zweimal aus Leichtbenzin umkristallisiert und ergibt das 5-Chlor- 8-*benzoxychinaldin mit dem konstanten Smp. 93-95o. Beispiel 2 5-Brom-8-acetoxychinaldin Zu einer Lösung von 80,4- 8-Acatoxychinaldin (Smp. 63-64o) in 750 ccm Chloroform, in der 133,
0g wasserfreies Natriumacetat suspendiert sind, werden unter Rühren und Kühlen in einer Eis- Kochsalz-Mischung während 25 Minuten<B>1600g</B> Brom hinzugetropft. Der Ansatz wird noch während 21/2 Stunden bei einer Temperatur von ca.
0o weiter gerührt, das feste Material abgenutscht und mit insgesamt 11 Choroform in Portionen nachgewa- sohen. Das Filtrat wird zuerst mit :
insgesamt 1,41 einer 10-proz. wässerigen Natriumthios.ulfatlösung und dann mit 2 mal 250 ccm, im Total mit 500 ccm Wasser ausgeschüttelt. Diese wässerigen Teile wer den nach dem Nachwaschen mit 300 ccm Chloro- form verworfen. Die vereinigten Chloroformteile werden über Natriumsulfat getrocknet und dann im Vakuum zur Trockne eingeengt. Der
Rückstand wird zweimal aus Leichtbenzin. umkristallisiert und man erhält das analysenreine 5 Brom-8-acetoxychinaldin vom konstantem Smp. 83-85o. Beispiel 3 5-Brom-8-benzoxychinaldin Zu einer Lösung von 39,5 g 8-Benzoxychinaldin (Smp. 76-79) in 260 ccm Chloroform, in der 49,
8 g wasserfreies Natriumacetat suspendiert sind, werden unter Rühren und Kühlen in einer Eis-Kochsalz- Mischung während 11 Minuten 60,0 g Brom hinzuge- tropft. Nach der Bromzugabe wird der Ansatz bei Raumtemperatur während 41/z Stunden gerührt,
das feste Material abgenutscht und mit insgesamt 250ccm Chloroform nachgewaschen. Das Filtrat wird zuerst mit 180 ccm einer SO-proz. wässerigen Natriumthio- sulfatlösung, dann mit 150 ccm einer 50-proz. wäs serigen Natriumacetatlösung und am Schluss mit total 375 ccm Wasser ausgezogen.
Die wässerigen Anteile werden mit total 200 ccm. Chloroform nachgsswa- schen. Die vereinigten ChlorofoTmteile werden über Natriumsulfat getrocknet und dann im Vakuum zur Trockne eingeengt. Der Rückstand wird zweimal aus Leichtbenzin umkristallisiert und ergibt das 5-Brom- 8-benzoxychinaldin mit dem konstanten <RTI
ID="0002.0155"> Smp. 103 104. Beispiel 4 5-Brom-8-(p-chlorbenzoxy)-chinaldin Zu einer Lösung von 80,0g 8-Oxychinaldilt (Smp. 73-750) in 320 ccm Benzol und 40,0 g Pyridin werden unter Kühlen und Rühren 96,3 g p-Chlor- benzoy-Ichlorid (Sdp. 97-100/12 mm Hg)
hinzuge- geben. Nachher wird der Ansatz unter Feuchtigkeits- ausschluss 4 Stunden in einem Ölbad von 50 gerührt, das ausgefallene Pyridin-hydrochlorid abge- nutscht und mit 150 com Benzol nachgewaschen.
Das Filtrat wird im Vakuum zur Trockne eingeengt und der Rückstand mehrmals aus Leichtbenzin um.- kristallisiert. Das analysenreine 8-(p-Chlorbenzoxy)- chinaldin hat den konstanten Smp. 114-117o.
Zu einer Lösung von 42,7 g 8-(p-Chlorbenzoxy)- chinaldin (Smp. 114-1170) in 250 ccm Chloroform, in der 49,8 g wasserfreies Natriumacetat suspendiert sind, werden unter Rühren und Kühlen in einer Eis- Kochsalz-Mischung während 11 Minuten 60,
0 g Brom hinzugetropft. Nach der Bromzugabe wird der Ansatz bei Raumtemperatur während 41/2 Stunden gerührt, das feste Material abgenutscht und mit ins gesamt 200 ccm Chloroform nachgewaschen.
Das Filtrat wird zuerst mit 150 com einer 50 proz. wäs serigen Natriumthiosulfatlösung, dann mit 150 ccm einer 50-proz. wässerigen Natriumacetatlösung und am. Schluss mit 125 ccm Wasser ausgezogen. Die wässerigen Anteile werden mit total 250 ccm Chloro form nachgewaschen.
Die vereinigten Chloraformteile werden über Natriumsulfat getrocknet und dann im Vakuum zur TrockneRTI ID="0002.0225" WI="15" HE="4" LX="1464" LY="2060"> eingeengt. Der Rückstand wird dreimal aus Tetrachlorkohlenstoff umkristallisiert und ergibt das analysenreine 5 -Brom- 8 - (p - chlor benzoxy)-chinaldin mit dem konstanten Smp. 168- 170 . Beispiel 5 5-Brom-8-(p-nitrobenzoxy)
-chinaldin Eine Aufschlämmung von 80,0 g 8-Oxychinaldin (Smp. 73-75), 40,0 g Pyridin, 400 ccm Äthylen- ohlorid und 102,0 g p-Nitrobenzoylchlorid (Smp. 720) wird 4 Stunden bei einer Badetemperatur von 500 gerührt. Nach dem Abkühlen auf Zimmertempe- ratur wird das gebildete Pyridinhydrochlorid abge- nutscht,
mit 150 ccm Benzol nachgewaschen und das Filtrat im Vakuum zur Trockne eingeengt. Das Roh- Produkt wird wiederholt aus Benzol umkristallisiert und ergibt das analysenreine 8 - (p -Nitrobenzoxy)- chinaldin mit dem konstanten Smp. 145-147-.
Zu einer Lösung von 15,4 g 8-(p-Nitrobenz(>xy)- chinaldin (Smp. 145-147) in 150 ccm Chloroform, in der 16,6 g wasserfreies Natriumacetat suspendiert sind, worden unter Rühren und Kühlen in einer Eis- Kochsalz-Mischung während 8 Minuten 20,
0 g Brom hinzugetropft. Der Ansatz wird während 5 Stunden bei Zimmertemperatur weitergerührt, das feste Mate rial abgenutscht und mit 90 ccm Chloroform nach gewaschen.
Das Filtrat wird zuerst mit insgesamt 75 ccm einer SO-proz. wässerigen Natriumthiosulf,at- lösung, dann mit 55 com einer 50 proz. Natrium acetatlösung und am Schluss mit total 75 ccm Wasser ausgezogen.
Die wässerigen Anteile werden mit total 75 ccm Chloroform nachgewaschen. Die vereinigten Chloroformteile werden über Natriumsulfat getrock- net und dann im Vakuum zur Trockne .eingeengt. Der Rückstand wird dreimal aus Benzol umkristalli- siert und ergibt das 5-Brom-8-(p-nitrobenzoxy)
- chinaldin mit dem konstanten Smp. 194-1960.
