Bei Anwendung von Nicotinsäure erfolgt die Veresterung zweckmässig in Gegenwart von wasserabspaltenden Mitteln, insbesondere von Phosphoroxychlorid oder Phosphorpentoxyd und von säurebindenden Mitteln, wie Pyridin, Chinolin od. dgl. Bei Anwendung funktioneller Derivate der Nicotinsäure für die Veresterung kommen als solche Nicotinsäurehalogenide, vorzugsweise in der Form ihrer halogenwasserstoffsauren Salze, wie z. B. Nicotinsäurechlorid-hydrochlorid oder Nicotinsäureanhydrid in Betracht.
Auch im Falle der Anwendung von Nicotinsäurehalogeniden wird die Veresterung in Gegenwart säurebindender Mittel, beispielsweise der vorher genannten Art, durchgeführt. Bei Durchführung der Veresterung mit Nicotinsäureanhydrid kann diese Umsetzung sowohl durch Schmelzen einer Mischung der Komponenten oder aber ebenso wie auch die vorher angeführten Reaktionen in Gegenwart von Lösungsmitteln, wie aromatischen und aliphatischen Kohlenwasserstoffen, z. B. Benzol, ferner Halogenkohlenwasserstoffen, z. B. Chloroform, Trichloräthylen, Tetrachlorkohlenstoff od. dgl., ferner tertiären Basen, z. B.
Pyridin, Dimethylanilin od. dgl. ausgeführt werden.
Die nach dem erfindungsgemässen Verfahren erhältlichen und im nachstehenden beschriebenen neuen Nicotinsäureester synthetischer Oestrogene zeigen eine ganz vorzügliche orale Wirksamkeit, da diese Körper dem enzymatischen Abbau in der Leber praktisch nicht unterliegen. Die neuen Ester bedeuten somit einen wichtigen Fortschritt für die Therapie, da sie oral mindestens gleich wirksam sind, wie parenteral.
Das Verfahren wird im nachfolgenden an Hand mehrerer Ausführungsbeispiele näher erläutert.
Beispiel 1 : Hexoestrol-4', 4"-bis-nicotinsäureester.
28 Gew.-Teile Nicotinsäureanhydrid werden geschmolzen und bei einer Badtemperatur von etwa 140 C 14 Gew.-Teile Hexoestrol in Anteilen zugefügt. Nach beendetem Eintragen wird unter Rühren 30 min bei dieser Temperatur gehalten, die Schmelze erkalten gelassen und zerkleinert ; hierauf wird mit 500 Vol.Teilen Wasser aufgenommen und Natriumbicarbonat bis zum Aufhören der CO2-Entwicklung eingetragen.
Das sich ausscheidende Reaktionsprodukt wird abgesaugt, mit Wasser gewaschen und getrocknet. Ausbeute : 20 Gew.-Teile, d. s. 75% der Theorie.
Durch Umkristallisieren aus Aceton wird der Hexoestrol-4', 4"-bis-nicotinsäureester in reiner Form gewonnen, F = 175-177 C.

Diäthylstilboestrol vermengt und die Mischung 15 min auf dem Dampfbade erhitzt ; nach dem Erkalten wird die entstandene Lösung in 800 Vol.-Teile Wasser gegossen, mit Natriumbicarbonat versetzt, die ausgefallenen Kristalle auf dem Filter gesammelt und mit Wasser gewaschen. Ausbeute : 16 Gew.-Teile.
Nach dem Umkristallisieren aus Alkohol schmilzt die erhaltene Verbindung bei 179, 0-180, 5 C.
Beispiel 4 : Diäthylstilboestrol-bis-nicotinsäureester.

Umkristallisieren aus Alkohol schmilzt die auf diesem Wege erhaltene Verbindung, welche mit der nach Beispiel 3 erhaltenen identisch ist, ebenfalls bei 179, 0-180, 5 C.
Beispiel 5 : Diäthylstilboestrol-bis-nicotinsäureester.
5 Gew.-Teile Diäthylstilboestrol werden mit 20 Gew.-Teilen Nicotinsäureanhydrid und 60Vol.-Teilen Pyridin 2 h auf dem Dampfbade erwärmt. Die klare Lösung wird hierauf mit 600 Vol.-Teilen Wasser versetzt und mit Natriumbicarbonat neutralisiert. Die ausgeschiedenen Kristalle werden filtriert, mit Wasser gewaschen, im Vakuum getrocknet und hierauf aus Alkohol umkristallisiert. Man erhält 8, 5 Gew.-Teile Diäthylstilboestrol-bis-nicotinsäureester vom F = 179, 0-180, 5 C. Die Verbindung ist identisch mit der nach den Beispielen 3 und 4 erhaltenen Verbindung.
Beispiel 6 : Dienoestrol-bis-nicotinsäureester.
28 Gew.-Teile Nicotinsäurechlorid-hydrochlorid werden in 120 Vol.-Teile Pyridin eingetragen und 15 Gew.-Teile Dienoestrol zugefügt. Man lässt t h auf dem Dampfbade reagieren und hierauf noch 2 h bei gewöhnlicher Temperatur. Anschliessend wird mit 1200 Vol.-Teilen Wasser aufgenommen, mit Natriumbicarbonat neutralisiert, die gebildeten Kristalle abgesaugt, mit Wasser gewaschen und getrocknet. Nach dem Umkristallisieren aus Äthanol erhält man 18 Gew.-Teile Dienoestrol-bis-nicotinsäureester vom F == 161-163 C.
Beispiel 7 : Benzoestrol-bis-nicotinsäureester.
