mit einem entsprechenden Säureanhydrid,
Säure- chlorid oder Mischungen derselben umsetzt.
Das Verfahren wird beispielsweise so -ausgeführt, dass 5 -Halogen- 8 - oxychinoline mit einem Säure anhydrid in Gegenwart eines katalytischen Zusatzes, z. B. konz. Schwefelsäure oder Pyridin, auf höhere Temperatur, vorzugsweise l30-160 erhitzt werden.
Die Reaktionslösung wird sodann direkt auf Eis gegossen oder zuerst das überschüssige Acylierungs- mittel teilweise abdestilliert und erst dann auf Eis gegossen.
Nach dem Abnutschen und Nachwaschen mit Wasser erhält man. die entsprechenden Ester. Ist das für die Acylierung der 5-Halogen-8-oxychinohne verwendete Acylierungsmittel ein Säurechlorid oder eine Mischung von Säurechlorid und Säureanhydrid, dann gestaltet sich der Umsatz am besten so,
dass die 5-Halogen-8-oxychinoline mit Acylhalogenid zu sammen in einem inerten organischen Lösungsmittel, z. B.
Benzol, Todual, Xylol Äthylenchlorid, Tri- chloräthylen, Chloroform, Tetrachlorkohlenstoff, Di- methylformamid und Pyridin, Triäthylamin,
Dime- thylanilin oder Natriumbicarbonat aufgeschlämmt oder gelöst werden und die Lösung oder Suspension der Reaktionspartner wird bei Zimmertemperatur und/oder bei erhöhter Temperatur gerührt wird; schliesslich können die gebildeten Ester nach bekann ten Methoden isoliert und gereinigt werden.
Die nach dem vorliegenden Verfahren herge stellten, bisher unbekannten Ester der 5 Halogen- 8-oxychinoline sind feste Basen.
Sie sind schlechter wasserlöslich als die 5 Halogen-8-oxychinaline und werden aus dieseln. Grunde schlechter als diese resor- biert. Sie besitzen auch über einen längeren Zeitraum einen nur unwesentlichen Eigengeruch (Unterschied zu Acetat) und ihre Verseifungsgefahr ist nur minim. Sie zeichnen sich durch therapeutisch verwertbare pharmakodynamnsche Eigenschaften aus,
indem sie eine erhöhte Wirksamkeit gegen pathogene Keime und auch andere Krankheitserreger aufweisen. Die neuen Verbindungen sollen in der Therapie als Ober- flächen-Antiseptika verwendet werden, wobei ihre schlechte Wasserlöslichkeit resp. Resorbierbarkeit eine besonders gute System-Verträglichkeit gewähr leistet.
Sie dienen ferner als Zwischenprodukte zur Herstellung von Medikamenten.
In den nachfolgenden Beispielen, welche die Aus führung des Verfahrens erläutern, den Umfang der Erfindung aber in keiner Weise einschränken sollen, erfolgen alle Temperaturangaben in Celsiusgraden. Beispiel 1 5-Chlor-8-benzoxychinolin Zu einer Lösung von 12,
0g 5 -Chlor- 8 - oxy- chinolin (Smp. 124-1250, 5,5 g Pyridin und 150 ccm Benzol werden bei Raumtemperatur unter Rühren 9,5 g Benzoylchlorid (Sdp. 194-196) hinzugetropft. Der Ansatz wird hierauf bei einer Badtemperatur von 900 4 Std. gerührt.
Das gebildete Pyridin-hydro- chlorid wird abgenutscht, mit 100 ccm Benzol nach gewaschen und das Filtrat unter vermindertem Druck zur Trockne eingeengt.
Das Rohprodukt wird zwei mal aus Tetrachlorkohlenstoff umkristallisiert und man erhält das analysenreine 5-Chlor-8-benzoxy- chinolin vom konstanten Smp. 97-98,5o.
Beispiel 2 5-Chlor-8-(p-chlorbenzoxy)-chinolin Zu einer Lösung von 9,6 g 5-Chlor-8-oxychinohn (Smp. 124-125), 5,4 g Triäthylamin und 200 ccm Benzol worden unter Rühren bei Raumtemperatur 9,4 g p-Chlorbenzoylachlorid (Sdp. 97-100/12 mm Hg) hinzugetropft. Der Ansatz wird hierauf bei einer Badtemperatur von 500 4 Std. gerührt.
Das entstan dene Triäthylamin hydrochlorid wird abgenutscht, mit 100 ccm Benzol nachgewaschen und das Filtrat unter vermindertem Druck zur Trockne eingeengt. Das rohe 5-Chlor -8-(p-chlorbenzoxy)
-chinolin hat einen Smp. von 121-1260. Nach zweimaligem Um- kristallisieren der Substanz aus Tetrachlorkohlenstoff erhält man das analysenreine 5-Chlor -8-(p-chlor- benzoxy)-chinolin mit dem konstanten Smp. 126- 127,5a.
Beispiel 3 5-Chlor-8-(p-nitrobenzoxy)-chinolin Eine Suspension von 5 -Chlor - 8 - oxychinolin (Smp. 124-1250, 8,0 g Pyridm, 250 ccm Benzol und 18,5 g p-Nitrobanzaylohlorid (Smp. 72o) wird 4 Std.
bei einer Badtemperatur von 50 gerührt. Das aus gefallene Pyridin-hydrochlorid wird abgenutscht, mit 100 ccm Benzol nachgewaschen und das Filtrat unter vermindertem Druck zur Trockne eingeengt. Das Rohprodukt wird zuerst einmal aus Benzol,
anschlies- send zweimal aus Tetrachlorkohlenstoff umkristalli- siert und man erhält das analysenreine 5-Chlor-8- (p-nitrobenzaxy)-chinolin mit dem konstanten Smp. 144-146o.
