Verfahren zur Herstellung einer therapeutisch wirksamen
Steroidverbindung
Ester des 19-Nortestosterons haben in letzter Zeit durch ihre ausgeprägte anabole Wirksamkeit bei gleichzeitig nur schwachen androgenen Eigenschaften steigende Bedeutung erlangt. Besonders bevorzugt werden öllösliche Verbindungen dieser Art, weil es mit solchen Verbindungen möglich ist, in einfacher
Weise eine protrahierte Wirkung zu erzielen. Der Cyclohexylpropionsäureester des 19-Nortestosterons, dessen Herstellung bisher noch nicht beschrieben worden ist, zeigt nun in hohem Masse ausgeprägte anabolische Eigenschaften und gleichzeitig, wie im Tierversuch nachgewiesen werden konnte, eine ausgezeichnete Dauerwirkung, wobei der rasche Wirkungseintritt auffallend ist.
Im Zuge der sich ständig ausbreitenden Indikationsbreite anaboler Wirkstoffe und der relativ hohen
Gestehungskosten der Ausgangsmaterialien ist es von besonderem Interesse, ein in technischem Massstabe durchführbares und wirtschaftliches Herstellungsverfahren für diese. interessante Verbindung zu schaffen.
Besondere Schwierigkeiten bei der Herstellung des Cyclohexylpropionsäureesters des 19 - Nortestosterons ergeben sich dadurch, dass diese ausserordentlich gut öllösliche Verbindung eine nur sehr geringe Kristallsationstendenz aufweist, die schon durch kleine Mengen von Verunreinigungen völlig unterbunden wird.
Um diese Verbindung in pharmazeutischen Anforderungen entsprechender Reinheit zu erhalten, muss bei Anwendung der üblichen Veresterungsverfahren der eigentlichen Synthese eine im technischen Massstabe teure und zeitraubende chromatographische Reinigungsmethode angeschlossen werden. Zum Beispiel führte, wie gefunden wurde, eine Veresterung in üblichen wasserunlöslichen Verdünnungsmitteln unter Zusatz von Basen trotz der Möglichkeit, nicht umgesetztes Säurederivat u. a. Nebenprodukte durch Extraktion mit wässerigen Agenzien zu entfernen, unter den verschiedenen Reaktionsbedingungen nur zu einem langsam und unvollständig kristallisierenden Öl. Auch die Verwendung organischer Basen als Reaktionsmedium brachte nicht den gewünschten Erfolg.
Bezüglich der Wahl des Molverhältnisses der Reaktionspartner sind einem wirtschaftlich durchführbaren Verfahren durch die hohen Kosten von reaktionsfähigen Derivaten der Cyclohexylpropionsäure Grenzen gesetzt.
Es wurde nun gefunden, dass man den Cyclohexylpropionsäureester des 19 - Nortestosterons aus 19-Nortestosteron und reaktiven Derivaten der Cyclohexylpropionsäure ohne Anwendung eines besonders hohen Überschusses an Veresterungsmittel in hoher Ausbeute als sofort kristallisierendes Produkt erhalten kann, wenn man die Veresterung in einem mit Wasser mischbaren inerten Lösungsmittel, vorzugsweise unter Zusatz einer tertiären'organischen Base, durchführt. Zweckmässig verwendet man das reaktive Derivat der Cyclohexylpropionsäure in einer Menge von etwa 1 bis 2 Molen je Mol 19-Nortestosteron an, wobei sich Anteile an Cyclohexylpropionsäure von 1, 1 bis 1, 3 Mol je Mol 19-Nortestosteron besonders bewährt haben. Als mit Wasser mischbare inerte Lösungsmittel kommen vorzugsweise einfache Ketone, wie Aceton, in Betracht.
Als reaktionsfähige Derivate der Cyclohexylpropionsäure werden beim erfindungsgemässen Verfahren vorzugsweise die Halogenide und Anhydride dieser Säure verwendet. Zweckmässig arbeitet man bei einer Temperatur über OOC, vorzugsweise zwischen 10 und 15 C. Die Ausfällung des kristallinen 19-Nortestosteron-17ss-cyclohexylpropionats gelingt durch einfaches Verdünnen mit Wasser.

Beispiel : 450 g 19-Nortestosteron löst man in einem Gemisch von 1100 ml reinem Aceton und 350 ml abs. Pyridin, leitet Stickstoff durch die Lösung und lässt 333 ml ss-Cyclohexylpropionsäurechlorid bei einer Innentemperatur von +10 bis +15 C innerhalb 20 min zutropfen. Anschliessend wird bei Zimmertemperatur 2, 5 h gerührt und auf 0 C gekühlt ; dann lässt man 2 200 ml Wasser langsam zutropfen.
Das kristalline Produkt wird abgesaugt, mit stark verdünnter Salzsäure und Wasser gewaschen und getrocknet. Nach dem Umlösen aus Petroläther erhält man 600 g reinen Ester vom F. 80 C.
